Endothelin 3(EDN3) has been known as a key gene causing hyperpigmentation in black bone chicken, the Silky fowl. Methods: In this study, EDN3 was employed as a candidate gene for regulating black color pigmentation. First,EDN3 was cloned from sheep to obtain the full-length cDNA by using the rapid amplification of cDNA ends(RACE).Genomic EDN3 was screened and a total of thirty predicted single nucleotide polymorphisms(SNPs) were genotyped for allele and genotype frequency analysis in a case-control study involving two black bone sheep populations. Genomic copy number analysis of EDN3 in sheep was conducted to measure the variation in copy number. EDN3 expression levels were observed among the groups in adult liver, lymph node, and kidney tissues, as well as embryo kidney samples. Also, among the tissues of black bone and non-black bone sheep. Results: The size of the full-length cDNA was 1,578 bp, which included 426 bp of 5′-untranslated region(5′-UTR),an open reading frame(ORF) of 639 bp encoding a protein of 212 amino acids, and a 3′-UTR of 513 bp. Genotype and allele frequencies of all the discovered SNPs were found insignificantly different in black bone and non-black bone sheep(P > 0.05). Genomic copy number analysis of EDN3 in sheep revealed no significant difference between the two sheep groups. No significant variations were found in the adult liver and kidney embryo samples. However, the expression in lymph node and kidney tissue was significantly higher in black bone sheep than that in non-black bone sheep(P < 0.05). Significant variations in the EDN3 expression levels were observed among the tissues of nonblack bone sheep. Conclusions: The findings of the present study indicate that unlike in Silky chickens, EDN3 is not responsible for hyperpigmentation but may play a key functional role in immune and excretory systems of black bone sheep.展开更多

Canonical RNA editing(i.e., A-to-I and C-to-U editing) mediated by the adenosine deaminases acting on RNA(ADAR) family and apolipoprotein B mRNA editing catalytic polypeptide-like(APOBEC)family during the transcriptional process is considered common and essential for the development of an individual. To date, an increasing number of RNA editing sites have been reported in human, rodents, and some farm animals;however, genome-wide detection of RNA editing events in sheep has not been reported. The aim of this study was to identify RNA editing events in sheep by comparing the RNA-seq and DNA-seq data from three biological replicates of the kidney and spleen tissues. Results: A total of 607 and 994 common edited sites within the three biological replicates were identified in the ovine kidney and spleen, respectively. Many of the RDDs were specific to an individual. The RNA editing-related genes identified in the present study might be evolved for specific biological functions in sheep, such as structural constituent of the cytoskeleton and microtubule-based processes. Furthermore, the edited sites found in the ovine BLCAP and NEIL1 genes are in line with those in previous reports on the porcine and human homologs, suggesting the existence of evolutionarily conserved RNA editing sites and they may play an important role in the structure and function of genes. Conclusions: Our study is the first to investigate RNA editing events in sheep. We screened out 607 and 994 RNA editing sites in three biological replicates of the ovine kidney and spleen and annotated 164 and 247 genes in the kidney and spleen, respectively. The gene function and conservation analysis of these RNA editing-related genes suggest that RNA editing is associated with important gene function in sheep. The putative functionally important RNA editing sites reported in the present study will help future studies on the relationship between these edited sites and the genetic traits in sheep.展开更多

The cows were fed a basal ration of 35% concentrate feed mix-ture with 40% berseem and 25% rice straw ( on a dry matter (DM) basis) without supplementation ( G1 ) or supplemented with 5 and 10 g zinc methionine per head per day (G2 and G3, respectively). Nutrient digestibility coefficients and nutritive values were significantly increased (P 〈0.05) by zinc methionine supplementation. Cows in G2 had the highest feed intake of total digestible nutrients (TDN) and digestible crude protein (DCP), followed by Ga and G1, respectively ( P 〈 0.05 ). The ruminal total volatile fatty acid concentration increased and ammonia nitrogen decreased significantly ( P 〈 0.05 ) by zinc methio-nine supplementation. Zinc methionine supplementa- tion led to significant decreases ( P 〈 0.05 ) in the activities of aspartate aminotransferase and alanine aminotransferase. A significant increase ( P 〈 0.05 ) was observed in the milk yield for G2 (2.54 kg/d or 18. 1% ) and G3 ( 1.95 kg/d or 14.2% ) compared with the control group. The corresponding increase in 4% fat-corrected milk (FCM) was 2.85 and 2.03 kg/d (21.63% and 15.74% ) for G2 and G3, respectively. Cows in G2 had the highest fat, total protein, and total solids contents, and the yield of all milk constitu- ents and animals in G3 showed the highest lactose, solids-not-fat, and ash contents. Zinc methionine sup- plementation improved feed conversion, reducing the quantities of DM, TDN, and DCP required to pro- duce 1 kg of 4% FCM (P〈0.05). The average dai- ly feed cost per kilogram of 4% FCM decreased and the average income from milk production increased with zinc methionine supplementation ( P 〈 0.05 ). Quadratic analysis showed that the optimal level of zinc methionine supplementation is 6 g per head per day in terms of milk yield and net revenue.展开更多

Fingerlings with an average weight of 1.65 ± 0.01 g/fish stocked at a rate of 10 fish/aquarium for 84 days experimental period. All experimental diets were isocaloric (437.69 kcal/100 g Dry Matter, DM) and isonitrogenous (30.5% crude protein) and supplemented with 0.5% L-Methionine and 0.5% L-Lysine Hcl. Results showed that there were a significant differences (P 〈 0.05) in final body weights, average daily gain (g fish^-1 dayl), specific growth rate (SGR, % day^-1), feed conversion ratio (FCR), protein efficiency ratio (PER), protein productive value (PPV%) and energy retention (ER%) among the tested groups. Nile tilapia fingerlings fed on the diet containing 25% S. chinensis protein exhibited comparable growth performance to those fed FM protein based diet. Carcass composition of crude protein decreased significantly (P 〈_ 0.05) with increasing JM replacement level above 50%, while replacement 25% JM does not affect DM and Crude Protein of fish flesh. On the other hand, increasing JM replacement level up to 75% increased Ether Extract % significantly (P 〈 0.05) and increasing JM up to 100% increased ash content significantly (P ≤ 0.05) while energy content (EC) decreased. It could be concluded the replacement of 25% Jojoba meal instead of fishmeal in Nile tilapia fingerlings diets is possible without any adverse effects on its growth performance or feed utilization.展开更多

Fr...In this (modest) study, we developed artificial neural network (ANN) models for predicting body weight using various independent (input) variables in eight-week old New Zealand white purebred and crossbred rabbits. From the whole data sets of similar age groups, 75 percent were used to train the neural network model and 25 percent were used to test the effectiveness of the model. Five predictor variables were used viz, breed, sex, heart girth, body length and height at wither as input variables and body weight was considered as dependent variable from the model. The ANN used was multilayer feed forward network with back propagation of error for efficient learning. Our ANN models (with R2 = 0.68 at ten thousand iterations, and R2 = 0.71 one million iterations) performed better than traditional multivariate linear regression (MLR) models (R2 = 0.66) indicating that the ANN models were able to more accurately capture how the variations in input variables explained the variations in body weight. It is concluded that ANN models are more powerful than MLR models in predicting animals’ body weight. Nonetheless, we recognize that fitting an ANN model requires more computation resources than fitting a tradition MLR model but the benefits of its accuracy outweigh any demerit from the associated computation overhead.展开更多

A 2 x 4 factorial arrangement of treatments was employed with 6 replicates of 15 birds per pen. Factors were: diets-positive and negative control(PC, NC); phytase-0, 500,1,000.1,500 FTU/kg. The PC was formulated to meet the 2014 Ross 308 nutrient specifications, whereas the NC was formulated with lower Ca(-1.4 g/kg), available P(-1.5 g/kg), Na(-0.3 g/kg), dLys(-0.2 g/kg) and MEn(-0.28 MJ/kg) equivalent to nutrient matrix values for 500 FTU/kg phytase in the starter, grower and finisher periods(i.e.,downspec diet). On d 24, phytase decreased FCR by 1.6, 4.3 and 4.6 points at inclusion levels of 500.1,000 and 1,500 FTU/kg, respectively(P < 0.01) across all diets. Phytase by diet interactions on BW gain were observed on d 24 and 35(P < 0.01). The effect of phytase was much more pronounced in the NC diet as compared with the PC diet.On d 24, phytase increased BW gain by 37, 55 and 68 g in the PC and 127.233 and 173 g in the NC at 500,1,000 and 1,500 FTU/kg, respectively. Diet by phytase interactions were also observed for tibia ash, litter quality and water to feed intake ratio(P < 0.01) with higher phytase effect in NC as compared with PC.Neither diet nor phytase impacted excreta moisture content on d 18 or 21(P> 0.05). Solid phase microextraction gas chromatography-mass spectrometry(SPME-GC-MS) analysis of gaseous emissions on d 39 indicated no difference in the emission of alcohols, aldehydes, ketones, volatile fatty acids and phenols between treatments(P> 0.05). The results indicate that phytase has greater benefits when formulated using nutrient matrix values as compared with adding it over the top in an already nutrient sufficient diet. The later method would be expected to increase feed costs without concomitant performance benefits.展开更多

Compared with the control group, both QT levels decreased(P < 0.05) fat-corrected milk(FCM)yield, but no significant effects were found on percentages of milk fat and protein. Contrariwise, yields of milk fat, lactose and milk protein were decreased(P < 0.05) when QT_(200) was supplemented. The solids nonfat(SNF) percentage and yield were decreased(P < 0.05) with QT100 supplementation. Moreover, QT tended to numerically reduce total number of ovarian follicles, number of small follicles, peripheral progesterone concentration and conception rate. Supplementation of QT_(200) numerically increased number of large follicles, mean diameter of large follicle, number and diameters of corpora lutea. The inclusion of QT_(200) shortened days open(DO) and decreased number of services per conception.Contrariwise, QT did not show significant effects on serum total protein, albumin, globulin, glucose, cholesterol and triglycerides concentrations. Supplementation of QT100 caused an increase(P < 0.05) of serum urea compared with that in control and QT_(200) groups. Generally, QT decreased(P < 0.05) serum creatinine concentration. Therefore, the supplementation of a commercial QT to early lactating Egyptian buffalo cows displayed negative consequences on their productive and reproductive performances.展开更多

文摘The therapeutic effect of gamma-aminobutyric acid(GABA)on diabetes was spread as one of the alarming epidemics worldwide. The study aims to investigate the function of Lactobacillus brevis KLDS_(1.0727) and KLDS_(1.0373) strains as glutamic acid decarboxylase 65(GAD65)carriers capable of generating GABA by comparing in vitro free and freeze-dried models and GABA intervention in vivo.PCR amplification of gad and in vitro i.e.,(growth rate, viability at different pH,bile tolerance, and survivability in simulated gastric juice)were performed. In vivo experiments were conducted in 7 groups of C57BL/6J mice. Each group was injected with streptozotocin(Cont_(STZ),INSSTZ,LAC1_(STZ),LAC_(1MFDSTZ),LAC_(2STZ),LAC_(2MFDSTZ))daily except for the control(Cont). One group was injected with insulin(INSSTZ).The body weight and hyperglycemia in the blood were assessed weekly,post-euthanasia blood plasma parameters, insulin, and histological examination were evaluated. Results indicated L.brevis strains demonstrated a great tolerance to bile and simulated gastric juice in vitro(P<0.05).Cont_(STZ) had the highest average glucose level(6.84±6.46)mmol/L while INS_(STZ) expressed dramatically decreed in glucose level and displayed a significant decline in the average of weekly blood glucose(−5.74±3.08)mmol/L.The lowest body weight(ContSTZ)was(19.30±0.25)g.Based on the blood plasma analysis,L.brevis strains improved good cholesterol properties, liver and kidney functions, where most of these parameters fall within the average the reference range and prevent the development of symptoms of type 1 diabetes in vivo.As recommended,L.brevis should be commonly distributed as a postbiotic GABA in pharmaceutical and nutritional applications.
文摘The present study was carried out to investigate the total and partial replacement of Jojoba meal (JM), Simmondsia chinensis, with fishmeal (FM) at different levels (0%, 25%, 50%, 75%, and 100%) on growth performance, feed utilization, and carcass composition of Nile tilapia, Oreochromis niloticus fingerlings. It could be concluded the replacement of 25% Jojoba meal instead of fishmeal in Nile tilapia fingerlings diets is possible without any adverse effects on its growth performance or feed utilization.
文摘Objective:To test the effects of extenders and packaging methods on morphological and functional characteristics of frozen thawed Ossimi ram semen. Methods:Ram semen was pooled, diluted in 3 different extenders:Tris-egg yolk(TEY),Tris-soybean lecithin(TSBL),and Tris-butylatedhydroxytoluene(TBHT),equilibrated at 5℃for 4 h,and packaged in straws or pellets for freezing.Semen was evaluated for sperm progressive motility, viability, abnormality, and membrane integrity after dilution, equilibration and thawing. The percentages of viable, early apoptotic, apoptotic, and necrotic spermatozoa as well as comet assay parameters were determined in post-thawed semen.Total antioxidants capacity, malondialdehyde and lactic dehydrogenase were assayed in thawed seminal plasma. Results:After equilibration, only sperm membrane integrity was significantly higher(P<0.05)in TEY and TSBL than in TBHT.After thawing,TEY or TSBL in straws significantly improved sperm progressive motility and vitality(P<0.05).In thawed seminal plasma,TBHT or TSBL in straws and TBHT in pellets significantly reduced malondialdehyde, and TBHT in pellets significantly increased lactic dehydrogenase(P<0.05).TEY in straws increased viable sperm, while significantly decreased early apoptotic and apoptotic sperm(P<0.05).DNA damage was significantly decreased(P<0.05)in straws with TEY and TSBL,and tail moment decreased in straws with all extenders(P<0.05).Conclusions:Despite the disadvantages of dilution of cryopreserved semen with egg yolk, ram semen cryopreserved with TEY gives the best physical, morphological and functional characteristics in straws compared with pellets, followed by semen diluted with TSBL.However, semen diluted with TBHT or TSBL,regardless of packaging method, showed the highest impact on antioxidant status of cyopreserved ram semen.
文摘This study accessed the reproductive performance of Bunaji cows in an Ovsynch protocol involving ovatide.Bunaji cows(n=16)aged 4-6 years and weighing between 250-350 kg with body condition scores(BCS)of 2.5-3.5 were used. There were two treatment groups for synchronization of ovulation.Treatment group 1 comprising Bunaji(n=8)received 50μg of gonadotropin releasing hormone(GnRH)(Cystorelin)and 25 mg of PGF2α.While,treatment group 2 comprising Bunaji(n=8)received 50μg of ovatide and 25 mg of PGF2α.All cows from both treatment groups were inseminated at 16 h after each second GnRH or ovatide injections.Cows that showed mucus discharge from the vagina on the day of artificial insemination(AI)were recorded as well as those that had patent cervix.Cervical dilation was measured by taking note of the cows that had mid cervix insemination due to non-passage of the AI gun through the cervix.Transrectal palpation was conducted twice at a month interval to select cycling cows within 5-12 d of the estrous cycle before initiating Ovsynch protocol. It was repeated on day 45 post AI to confirm pregnancies in animals. Results showed that the rate of mucus discharge from the vagina was 37.5%and 87.5%for ovatide and Cystorelin, respectively;while the rate of cervical dilation were 75%and 87.5%for ovatide and Cystorelin,respectively(p>0.05),and pregnancy rates were 0%and 12.5%for ovatide and Cystorelin, respectively. There were no significant differences between the two groups. It was concluded that treatment of Bunaji cows with 50μg ovatide in Ovsynh protocol has heat and ovulation synchronization potentials and zero pregnancy rate.It was recommended that further studies be carried out using graded doses of 50,100 and 150μg of ovatide in a fixed time AI synchronization protocol in Bunaji cows,to tap the potentials of the hormone in manipulation of bovine reproduction.
文摘In this (modest) study, we developed artificial neural network (ANN) models for predicting body weight using various independent (input) variables in eight-week old New Zealand white purebred and crossbred rabbits. Nonetheless, we recognize that fitting an ANN model requires more computation resources than fitting a tradition MLR model but the benefits of its accuracy outweigh any demerit from the associated computation overhead.

文摘The effect of graded levels of phytase on performance, bone characteristics, excreta/litter quality and odorant emissions was examined using 720 Ross 308 male d-old broilers. The later method would be expected to increase feed costs without concomitant performance benefits.

文摘This study was conducted to investigate the effects of 10 weeks supplementation of Quebracho tannins(QT; 0 [control], 100 [QT100] or 200 g/[cow$d] [QT_(200)]) to 30 multiparous postpartum buffalo cows(10 cows per group) on milk yield and composition, blood metabolites and reproductive performance.Supplementation of QT100 had no significant effect on milk yield, whereas QT_(200) decreased(P < 0.05) this trait. Therefore, the supplementation of a commercial QT to early lactating Egyptian buffalo cows displayed negative consequences on their productive and reproductive performances.
文摘The current study presents magnetic nanoparticles(MNPs)dispersed polyurethane(PU)nanofiber nanocoating with exceptional magnetic, thermal, hydrophobicity, and textural properties.X-ray photoelectron spectroscopy(XPS)was used to clarify the Fe3O4 MNPs synthesis, which had previously been accomplished using a single-step green method using molasses as a biowaste. The MNPs particles were then encapsulated in PU nanofibers with varying concentrations of 0.1,0.3,and 0.5 wt%.SEM revealed a homogenous, smooth, and consistent fiber morphology with a diameter of 173.61(size,nm),and the diameter rises with the addition of MNPs.With statistical significance(P<0.05),PU/MNPs nanofiber showed intense antibacterial activity against different microbes.Notably,PU/0.5 wt%MNPs exhibit advantages as antimicrobial packaging materials by maintaining the quality and freshness of the Karish cheese and extending its shelf life to 40 days while also boosting its gumminess(5.9,N),hardness(7.93,N),cohesiveness(0.6),and adhesiveness(1.14,mg).Iron content is quantified using inductively coupled plasma mass spectrometry(ICP-MS)to determine whether the developed materials are safe for food. These findings showed that PU/MNPs could be a valid alternative for controlling Karish cheese storage.Further research into the effectiveness of this method with various smear cheeses is possible.
