Potato invertase inhibitor(StInvInh2)positively regulates cold-induced sweetening(CIS)resistance by inhibiting the activity of vacuolar invertase.The distinct expression patterns of StInvInh2 have been thoroughly char...Potato invertase inhibitor(StInvInh2)positively regulates cold-induced sweetening(CIS)resistance by inhibiting the activity of vacuolar invertase.The distinct expression patterns of StInvInh2 have been thoroughly characterized in different potato genotypes,but the related CIS ability has not been characterized.The understanding of the regulatory mechanisms that control StInvInh2 transcription is unclear.In this study,we identified an ERF‐Ⅶ transcription factor,StRAP2.3,that directly regulates StInvInh2 to positively modulate CIS resistance.Acting as a nuclear-localized transcriptional activator,StRAP2.3 directly binds the ACCGAC cis-element in the promoter region of StInvInh2,enabling promoter activity.Overexpression of StRAP2.3 in CIS-sensitive potato tubers induced StInvInh2 mRNA abundance and increased CIS resistance.In contrast,silencing StRAP2.3 in CIS-resistant potato tubers repressed the expression of StInvInh2 and decreased CIS resistance.We conclude that cold-responsive StInvInh2 is due to the binding of StRAP2.3 to the ACCGAC cis-element in the promoter region of StInvInh2.Overall,these findings indicate that StRAP2.3 directly regulates StInvInh2 to positively modulate CIS resistance,which may provide a strategy to improve the processing quality of potatoes.展开更多
A new space era is bringing deployments of thousands of networked assets in near-Earth and deep-space.Delay-Tolerant Networking(DTN)protocols and algorithms such as Contact Graph Routing(CGR)are state-of-the-art techn...A new space era is bringing deployments of thousands of networked assets in near-Earth and deep-space.Delay-Tolerant Networking(DTN)protocols and algorithms such as Contact Graph Routing(CGR)are state-of-the-art technologies certainly capable of achieving this ambitious objective.Nevertheless,in this paper,we highlight and analyze the roots of current scalability limits of CGR,to then propose more efficient alternatives.In particular,we leverage a different graph modeling approach that enables optimizations such as priority queues and spanning-tree searches in the core route determination process.While such abstract data types are difficult to adapt to CGR,we incorporate them into a novel Shortest-Path Tree Approach for Routing in Space Networks(SPSN),which is presented as a compute-efficient alternative.This paper thoroughly describes the SPSN approach including route object modeling,capacity-oriented search,and potential route management techniques.Prototype evaluations analyzed in this paper present significant evidence that SPSN outperforms legacy CGR in more than an order of magnitude in contact plans comprising thousands of contacts.展开更多

Structural variants(SVs)are a type of genetic variation that contribute substantially to phenotypic diversity and evolution.Further study of SVs will help us understand the influence of SVs associated with tea quality...Structural variants(SVs)are a type of genetic variation that contribute substantially to phenotypic diversity and evolution.Further study of SVs will help us understand the influence of SVs associated with tea quality and stress resistance and provide new insight into tea plant breeding improvement and genetic research.However,SVs have not been thoroughly discovered in the tea plant genomes.Herein,we constructed a large-scale SV map across a population of 107 resequenced genomes,including both ancient and cultivated tea plants.A total of 44,240 highconfident SVs were identified,including 34,124 DEL(deletions),4,448 DUP(duplications),2,503 INV(inversions),544 INS(insertions)and 2,621 TRA(translocations).In total,12,400 protein-coding genes were overlapped with SVs,of which 49.5%were expressed in all five tea tissues.SVbased analysis of phylogenetic relationships and population structure in tea plants showed a consistent evolutionary history with the SNP-based results.We also identified SVs subject to artificial selection and found that genes under domestication were enriched in metabolic pathways involving theanine and purine alkaloids,biosynthesis of monoterpenoid,phenylpropanoid,fatty acid,and isoflavonoid,contributing to traits of agronomic interest in tea plants.In addition,a total of 27 terpene synthase(TPS)family genes were selected during domestication.These results indicate that these SVs could provide extensive genomic information for tea quality improvement.展开更多

文摘Structural variants(SVs)are a type of genetic variation that contribute substantially to phenotypic diversity and evolution.Further study of SVs will help us understand the influence of SVs associated with tea quality and stress resistance and provide new insight into tea plant breeding improvement and genetic research.However,SVs have not been thoroughly discovered in the tea plant genomes.Herein,we constructed a large-scale SV map across a population of 107 resequenced genomes,including both ancient and cultivated tea plants.A total of 44,240 highconfident SVs were identified,including 34,124 DEL(deletions),4,448 DUP(duplications),2,503 INV(inversions),544 INS(insertions)and 2,621 TRA(translocations).In total,12,400 protein-coding genes were overlapped with SVs,of which 49.5%were expressed in all five tea tissues.SVbased analysis of phylogenetic relationships and population structure in tea plants showed a consistent evolutionary history with the SNP-based results.We also identified SVs subject to artificial selection and found that genes under domestication were enriched in metabolic pathways involving theanine and purine alkaloids,biosynthesis of monoterpenoid,phenylpropanoid,fatty acid,and isoflavonoid,contributing to traits of agronomic interest in tea plants.In addition,a total of 27 terpene synthase(TPS)family genes were selected during domestication.These results indicate that these SVs could provide extensive genomic information for tea quality improvement.

文摘A new linear phased array on a concave cylindrical transducer is designed for meeting the specific requirements of applications for interstitial thermal ablation.Using the array,a focal line can be generated rapidly and the focal position can be adjusted in the proper range without the use of complex mechanical structures.The focused acoustic field distributions in the axial,radial and azimuthal directions of the transducer are investigated theoretically by numerical simulation.Effects of the focal distance,steering angle,element arc-width,arc-space between adjacent elements and number of elements on the acoustic field are also thoroughly studied.Many important results are obtained.

文摘The efficiency and quality of human induced pluripotent stem cell-derived cardiomyocytes(hiPSC CMs)is crucial for regenerative medicine,disease modeling,drug screening and the study of the development events during cardiac specification.However,their applications have been hampered by the differentiation efficiency,poor maturation and high interline variability.Recent studies have reported that histamine plays important roles in hema topoietic stem cell proliferation and neutrophil maturation.However,its roles in cardiovascular tissue regeneration have not been thoroughly investigated.In the current study,we identified a novel physiological function of the histamine/histamine 1 receptor(H,R)signal in regulating the differentiation of hiPSC-CMs and heart development.
