Tomato brown rugose fruit virus(ToBRFV) is a novel tobamovirus firstly reported in 2015 and poses a severe threat to the tomato industry. Tomato brown rugose fruit virus(ToBRFV) is a novel tobamovirus firstly reported in 2015 and poses a severe threat to the tomato industry. So far, it has spread to 10 countries in America, Asia, and Europe. In 2019, ToBRFV was identified in Shandong Province(ToBRFV-SD), China. In this study, it was shown that ToBRFV-SD induced mild to severe mosaic and blistering on leaves, necrosis on sepals and pedicles, and deformation, yellow spots, and brown rugose necrotic lesions on fruits. ToBRFV-SD induced distinct symptoms on plants of tomato, Capsicum annumm, and Nicotiana benthamiana, and caused latent infection on plants of Solanum tuberosum, Solanum melongena, and N. tabacum cv. Zhongyan 102. All the 50 tomato cultivars tested were highly sensitive to ToBRFV-SD. The complete genomic sequence of ToBRFV-SD shared the highest nucleotide and amino acid identities with isolate IL from Israel. In the phylogenetic tree constructed with the complete genomic sequence, all the ToBRFV isolates were clustered together and formed a sister branch with tobacco mosaic virus(TMV). Furthermore, a quadruplex RT-PCR system was developed that could differentiate ToBRFV from other economically important viruses affecting tomatoes, such as TMV, tomato mosaic virus, and tomato spotted wilt virus. The findings of this study enhance our understanding of the biological and molecular characteristics of ToBRFV and provide an efficient and effective detection method for multiple infections, which is helpful in the management of ToBRFV. 展开更多

Anthocyanin is one of water-soluble natural pigments widely existing in flowers, fruits, stems, leaves and seeds of plants, and it is the major factor conferring pink or red to the petals of Rosa rugose. MYB TFs play an important role in the anthocyanin synthesis in plants. This work aimed to clone the MYB gene related to anthocyanin synthesis in the petals of Rosa rugose, and explore the relationship between them to lay a good foundation for gene engineering improvement of R. rugose. Based on the transcriptional data, a full-length cDNA sequence of MYB Gene, RrMYB113 (GenBank accession Nos MG720012), was cloned at the first time from the petals of Rosa rugose “Zi zhi” with RT-PCR and RACE methods. The full-length cDNA is 885 bp with an open reading frame of 654 bp, encoding 216 amino acids. The derived RrMYB113 protein has a molecular weight of 25,297.64 Da, a calculated pI of 9.61, a R2R3-MYB domain and bHLH binding domain, and it also has the signature motifs ((A/S/G)NDV and KPRPR(T/S)), thus belonging to Sg6 R2R3-MYB subfamily. In the secondary structure of RrMYB113 protein, there is 37.04% α-helix, 39.81% random coil, 14.81% extended peptide chain, and 8.33% β-corner. There is no transmembrane domain and no signal peptide cleavage site, seventeen Ser phosphorylation sites, fifteen Thr phosphorylation sites, four Tyr phosphorylation sites, and no O-glycosylation sites. The expression of RrMYB113 increased with the color deepening in petals, and it expressed at a higher level in petals than in other tissues of R. rugose “Zi zhi”. These results are meaningful to reveal that RrMYB113 might be an important regulator in anthocyanin biosynthesis and coloration in the petals of R. rugose. 展开更多

In order to find a method to break dormancy of Rosa rugose seeds and interspecific hybrid seeds between Rosa rugose and Rosa hybrid quickly, and accelerate the breeding process of interspecific hybridization between Rosa rugose and Rosa hybrid, the influence of concentrated acid, seed maturity, GA3 (gibberellin) and low temperature (4°C) on seed germination of Rosa rugose from Muping was researched under aseptic condition. The results showed that aseptic germination can significantly shorten the germination time of Rosa rugose seeds and raise its germination ratio. Before inoculation, concentrated acid treatment greatly increased the germination rate and reduced the contamination rate of the seeds. The higher the degree of maturity of seeds is, the lower the germination rate would be, and the best time for seed to aseptic germination is 60 d after pollination. The addition of GA3 in 1/2MS medium could promote seeds germination better, and when the concentration of GA3 was 0.15 mg/L, the seed germination ratio was the highest;the germination time decreased and the seed germination ratio increased gradually as the treatment time at 4°C lasted longer. 展开更多

As an important signal transduction protein, G protein beta subunit gene encoded by oligonucleotides plays an important role in many physiological, biochemical and environmental stresses in plants. In order to understand the action mode of G protein beta subunit gene, this paper cloned a Wd40 gene related to G protein beta subunit gene, named RrG-beta1, based on the R. rugose—transcriptome data, using Rosa rugose “Zi zhi” as experimental materials. The full length of cDNA of the gene was obtained by RT-PCR and RACE methods. The total length of this gene is 981 bp, and it encodes 326 amino acids. After bioinformatics analysis, the molecular formula C1601H2520N450O486S11 was predicted;the relative molecular weight was 36,201.00 Da;the theoretical isoelectric point PI value was 6.71;and its instability index was 30.44. The total average hydrophobic index was -0.847. In the secondary structure of RrG-beta1 protein, there are 17 α-helix, 131 Random coil, and 141 extended peptide chain. Gene Bank Blast results showed that the amino acid sequence encoded by RrG-beta1 was more than 90% homologous with the beta-like protein of Rosa chinensis, Fragaria, Malus, Pyrus, Prunus, Arabidopsis and tobacco, so it can be inferred that the RrG-beta1 Gene is guanine nucleotide-binding protein subunit beta-like protein. Fluorescence quantitative expression analysis of RrG-beta1 protein decreased with the development of flower color, and it was speculated that it could exert negative regulation effect on flower color. The leaf expression was highest in the tissue part, so it was inferred that the signal was transmitted through the stoma on the leaf. 展开更多

The northern flank of the Tindouf Syncline in southern Morocco exhibits a continuous, well exposed Carboniferous succession with limestones of Late Asbian to Early Bashkirian age containing rich and diverse foraminiferal and rugose coral assemblages. Analysis of these assemblages provides new data on the relatively poorly known Saharan basins. The palaeobiogeographical relationship of the Tindouf Basin with other Palaeotethyan basins is complex. Although there is a predominance of cosmopolitan taxa for the Palaeotethys, it is recognized that there was an influence of basins from NW Europe, such as the UK and Ireland. Some taxa are recorded in both NW Europe and Tindouf without any characteristic contributions from intermediate basins in northern Morocco. The neighbouring Béchar Basin in Algeria presents distinct assemblages. The bulk of the data analyzed suggest that this sector of the western Palaeotethys can be subdivided into four palaeobiogeographical subprovinces: the Atlantic Subprovince (UK, Ireland, N France and Belgium), the Mediterranean Subprovince (Pyrenees, Montagne Noire, Betic Cordillera, Rif (N Morocco) and Balearic Islands), and the Saharan Subprovince (Béchar, Reggan, Ahnet-Mouydir and Tindouf). In between, mobile belts of mixed faunal assemblage characteristics are observed (e.g., SW Spain and Central Meseta) forming the West peri-Gondwanan Subprovince.Analysis of the Tindouf Basin faunas shows that, as in other Saharan basins, there is a high diversity and abundance of foraminiferal taxa, with a higher proportion of survivors and longer stratigraphic ranges; these features also are mirrored by rugose corals. This emphasizes the longevity of the carbonate platform in a tropical setting, where periodic transgressions introduced new assemblages, and oceanic currents are interpreted as one of the main controlling factors for the distribution of the taxa in these subprovinces. Moreover, not only were water temperatures on the platform higher, but also tectonic stability greater. It is considered that the effects of the first phases of the Gond-wanan glaciation were minimal on the Tindouf faunas. 展开更多

The control of external and internal canopy structural heterogeneity on diversity and productivity relationship in a subtropical forest

Forest canopy structure is closely related to species diversity,crown packing efficiency,and ecological processes,while influencing ecosystem functions.However,most existing structural diversity indices only consider internal heterogeneity within the canopy but often neglect the heterogeneity of the external canopy.While,the external canopy heterogeneity can increase the exchange surface area between canopy and external atmosphere,capture more light,which is closely linked to tree growth and ecosystem functions.Comprehensively and accurately estimating canopy structural diversity is crucial in understanding the underlying mechanisms of productivity change.Therefore,in this study,we apply a topographic index–canopy surface rugosity(CSR)–to capture canopy external heterogeneity,and use crown complementary index(CCI)and other canopy structural variables to quantify canopy internal heterogeneity.We find that CSR varies significantly among quadrats(20 m×20 m)and it decreases with species diversity and wood net primary productivity(NPP_(wood)).Species diversity increases NPP_(wood) directly and indirectly through CSR and CCI.Functional diversity increases NPP_(wood) through CSR and CCI.Community weighted mean of light demand decreases with NPP_(wood) through CCI.The mediating effect of CSR on the relationship between species(functional)diversity and NPP_(wood) is greater than the mediating effect of CCI.We find soil fertility positive effects on NPP_(wood) mainly through species richness and CSR.Conversely,water availability affects NPP_(wood) by a direct positive effect and indirect negative effects through species richness,CSR and CCI.Overall,our CSR and CCI provide strong support for a space-based niche partitioning mechanism regulating the relationship between species diversity and NPP_(wood).Additionally,CSR is sensitive to gap dynamics,suggesting mechanistic relationships between local disturbance,species diversity,and NPP_(wood). 展开更多

Plants have evolved and perfected a series of light receptors to feel the light at different bands and regulate the expression, modification and interaction of related genes in plants through signal transduction. So far, many photoreceptors have been identified in plants, UVR8 has recently been identified as a receptor for UV-B light. This paper cloned a WD40 gene related to UVR8 protein subunit, named RrRUP2, based on the Rosa rugose transcriptome data, using Rosa rugose “Zi zhi” as experimental materials. The full length of cDNA of the gene was obtained by RT-PCR and RACE methods. The total length of this gene is 1173 bp, and it encodes 390 amino acids. After bioinformatics analysis, the molecular formula C3415H5659N1173O1434S313 was predicted;the relative molecular weight was 96129.27 Da;the theoretical isoelectric point PI value was 5.00;and its instability index was 47.06. The total average hydrophobic index was 0.750. In the secondary structure of RrRUP2 protein, there are 10 α-helix, 45 β-helix, 181 Random coil, and 154 Extended strand. Gene Bank Blast results showed that the amino acid sequence encoded by RrRUP2 was more than 90% homologous with the RUP2 protein of Rosa chinensis, Fragaria, Malus, Pyrus, Prunus, Juglans, Arabidopsis and Tobacco, so it can be inferred that the RrRUP2 gene is a WD repeat-containing protein. Regarding to fluorescence quantitative expression analysis of RrRUP2, we find its experssion pattern is corresponded with the accumulation of anthocyanins. 展开更多

Rosa rugosa is widely cultivated in China,and the blossom of R.rugosa is used to extract rose essential oil or directly make into various foods,such as mei gui hua cake,mei gui hua sauce.Rose dregs are mostly used as fertilizer after extracting essential oil and the polar part has not been fully utilized.There is still much to be done in the products and deep processing of rose.In this study the content of total flavonol glycosides,the extractable polyphenols,the non-extractable polyphenols,the total phenolic and the total anthocyanins in R.rugosa from five major production areas in China was analyzed;the biological activities of the total extract,the polyphenols fraction,the flavonol glycosides fraction and the isolated compounds of R.rugosa were evaluated,including antioxidant,anti-ultraviolet,inhibitory activities ofα-glucosidase,lipase,alveolar epithelial cell senescence and tyrosinase,together with regulating activities on intestinal flora.The components content of R.rugosa in different producing areas was significantly different.The total flavonol glycosides in Yunnan,the extractable polyphenols and the total phenolics in Shandong,the non-extractable polyphenols in Xinjiang,the total anthocyanins in Gansu was the higher than other areas.The polyphenols fraction showed antioxidant and anti-ultraviolet properties,inhibition of alveolar epithelial cell senescence,lipase and tyrosinase activities,and regulation of intestinal flora.The flavonol glycosides fraction showedα-glucosidase inhibitory activity.The results showed the polyphenols fraction and the flavonol glycosides fraction of R.rugosa could be used as skin care products and healthy food ingredient. 展开更多
