Sweet potato leaf tips have high nutritional value,and exploring the differences in the metabolic profiles of leaf tips among different sweet potato varieties can provide information to improve their qualities.In this study,a UPLC-Q-Exactive Orbitrap/MS-based untargeted metabolomics method was used to evaluate the metabolites in leaf tips of 32 sweet potato varieties.Three varieties with distinct overall metabolic profiles(A01,A02,and A03),two varieties with distinct profiles of phenolic acids(A20 and A18),and three varieties with distinct profiles of flavonoids(A05,A12,and A16)were identified.In addition,a total of 163 and 29 differentially expressed metabolites correlated with the color and leaf shape of sweet potato leaf tips,respectively,were identified through morphological characterization.Group comparison analysis of the phenotypic traits and a metabolite-phenotypic trait correlation analysis indicated that the color differences of sweet potato leaf tips were markedly associated with flavonoids.Also,the level of polyphenols was correlated with the leaf shape of sweet potato leaf tips,with lobed leaf types having higher levels of polyphenols than the entire leaf types.The findings on the metabolic profiles and differentially expressed metabolites associated with the morphology of sweet potato leaf tips can provide useful information for breeding sweet potato varieties with higher nutritional value. 展开更多

Twelve percent (12%) of Ghanaians are food insecure, and climate-smart crops like sweet potatoes are required to help end poverty. Small-scale farmers in Ghana who produce low-technology, subsistence crops, such as sweet potatoes, are more food secure than those who do not. This study was initiated to investigate the effect of chicken manure, compost, and cow dung on the growth and yield of “apomuden”, “SARI-Nyoriberigu”, “SARI-Nan” and “kufour” sweet potato under the Guinea Savannah agroecological zone of Ghana. Organic fertilizer increased leaf chlorophyll content and leaf area index. The application of cow dung, chicken manure and compost in 2015 significantly increased total storage root yield by 38%, 55% and 98%, 62%, 45% and 37%, 52%, 61% and 44%, and 33%, 36% and 28% for SARI-Nyoriberigu, Kufour, SARI-Nan and Apomuden, respectively, when compared to the untreated check. In 2016, and in comparison with the untreated check, the application of cow dung, chicken manure and compost increased total storage root yield by 42%, 61% and 93%, 69%, 49% and 41%, 57%, 67% and 48%, and 36%, 39% and 30% for SARI-Nyoriberigu, Kufour, SARI-Nan and Apomuden, respectively. Hence, the application of organic fertilizers will increase sweet potato yield, give higher returns to resource-poor smallholder farmers and contribute to enhancing food and nutrition security. 展开更多

Increasing access to locally produced, safe, nutritious and affordable complementary foods is essential to combat micronutrient deficiencies in young children in low- and middle-income countries. Two formulations of sweets based on jujube, baobab fruit powder and sweet potato puree were produced, and their nutritional values and sensory properties such as taste, smell, color and acceptability were assessed. The formulation containing the most jujube powder, baobab fruit and sweet potato puree gave the best nutritional value. The sweets are rich in calcium (259.80 mg/100 g), magnesium (67.98 mg/100 g), potassium (782.04 mg/100 g), vitamin C (32.37 mg/100 g), iron (6.45 g mg/100 g) and zinc (0.97 mg/100 g). Sensory tests on the two formulations showed acceptability scores ranging from 81.26% to 84.58%, well above the minimum acceptable threshold. Candies with jujube, baobab fruit powder and sweet potato puree could be an alternative for preventing micronutrient deficiencies. 展开更多

However, some constraints related to climate change limit its production. This study evaluated the agronomic and technological performances of eight varieties of orange-fleshed of sweet potato (Ipomoea batatas (L.) to identify the most promising ones. The results show three groups of varieties with different agronomic performances. Group 1 composed of Irene and Vita is characterized by low production of small and large tuberous roots, low number of productive plants and low yield. Group 2 consisting mainly of Kabode is characterized by high production and weight of large tuberous roots as well as high yield. Group 3, composed of Tacha-2 Ininda, Ejumula Gloria, TIB-440060, CIP-199062-1, Bela bela is characterized by high production of small and large tuberous roots, high number of productive plants and average yield. The TIB-440060 variety has a recovery rate of 100%. The Irene, Kabode, Vita, Kabode varieties were resistant to viral and fungal diseases. In terms of technological quality, the TIB-440060 and CIP-199062-1 varieties have the best organoleptic qualities. Given the agronomic performances of the Kabode, TIB-440060 and CIP-199062-1 varieties, could be selected as the elites varieties and distributed to producers. 展开更多

[Objective] The aim was to resolve the issue of sparsely planting (37 500-40 500 plants/hm2) of sweet potato in hilly areas. [Method] The starch-oriented Jishu No.21 and raw-eating oriented Jishu No.22 were studied to explore effects of planting density on yield and sink and source characteristics of sweet potato. [IRe- suit] Leaf area index of Jishu No.21 and Jishu No.22 were increasing upon planting density. Leaf area index of the same planting density showed a single-peak curve. Specifically, leaf area index grew fast during the 40th-80th d after planting, and reached the peak on the 80th d after planting, followed by decreasing. What's more, ventilation and sunshine transmission both declined upon planting density, as well as the number of leaf, the number of branch, the length of vine, dry and fresh weights of stem and leaf. When planting density exceeded 75 000 plants/hm2, the yield of sweet potato dropped dramatically. Besides, the optimal planting density tended to be volatile upon cultivars. For example, the range of 45 000-60 000 plants/hm2 is the optimal planting density of Jishu No.21 and the range of 60 000-75 000 plants/hm2 is the optimal planting density of Jishu No.22. [Conclusion] It can be concluded that rational planting densities would well coordinate sweet potato growth of ground parts and underground parts to get a high yield by providing a rational group structure. Considering the optimal planting density differs upon cultivars, it is necessary to take genotype, environment, soil fertility and planting density into consideration in determining planting density. 展开更多

The degenerate primers were designed based on the conserved NBS-LRR motifs among the known disease-resistance genes. A fragment of about 500 bp was amplified from genomic DNA of sweet potato using the specifically designed degenerate primers. After cloning and sequencing, 20 NBS-LRR type of disease-resistance gene analogue (RGAs) in sweet potato were observed. The deduced amino acid sequence of DNA fragment contains the conserved motifs of NBS-LRR type RGAs, such as P-loop, Kinase-2α, Kinase-3α and GLPL domain. The 20 RGAs could be sorted into two subclasses, namely TIR- NBS-LRR type and non-TIR-NBS-LRR type. Compared with the known resistance genes including N, L6 and M, the percentages of homologous amino acid sequence in 10 TIR-NBS-LRR range between 21% -44%. While other 10 non-TIR-NBS-LRR assume 15% -46% homology with the known resistance genes (Prf, RPM1, RPS2, etc. ). Consequently the RGAs may further be used as molecular marker for screening the candidate disease-resistance genes in sweet potato. 展开更多

The root quality of sweet potato cultivated during the summer season is poor in northern China;thus,this study was conducted to determine whether root quality could be improved through mulching with plastic film(MPF).The effect of MPF on root starch and its composition,the activity of starch synthesis enzymes,and other quality-related parameters were investigated in two purple flesh sweet potato cultivars,Jishu 18 and Ayamurasaki(Aya).The results indicated that root dry matter,anthocyanin content,adenosine triphosphate(ATP),and starch content were higher in both cultivars under the MPF treatment than those under the control treatment.The root adenosine diphosphate glucose pyrophosphorylase/uridine diphosphate glucose pyrophosphorylase(ADPGPPase/UDPGPPase)activity and adenosine triphosphatease(ATPase)activity were increased using MPF.However,under the MPF treatment,the amylose content,soluble sugar content,and granule-bound synthase(GBSS)activity increased in Jishu 18 but decreased in Aya,and the amylopectin content,protein content,and soluble starch synthase(SSS)activity decreased in Jishu 18 but increased in Aya.Therefore,MPF seems benifit to improve the quality of sweet potato,but the effects of this treatment condition may be dependent on the cultivar. 展开更多

Anthocyanidin in plants, an important pigment, is of great interest to researchers, consumers, and commercial entities due to its physiological functions. Anthocyanin content and mRNA levels of anthocynin biosynthesis genes were investigated in storage root of different purple-fleshed sweet potatoes (PFSP) genotypes to understand the regulation mechanism of anthocyanin under weak light conditions. Anthocyanin content, its amount of accumulation, and the expression of CHS, DFR, F3H, GT, and ANS genes in the PFSP storage root under weak light conditions were studied. The results demonstrated that the anthocyanin content of the treatments was decreased and was obviously lower than that of the control until 30 days after shading in Ayamurasaki, while it was lower than that of the control from the beginning of shading in Jishu 18. Their accumulation rates of both treatmeants were lower than its control before 10-20 d of shading in Jishu 18, while those of Ayamurasaki weren't in their treatments. This indicated that Jishul 8 is more sensitive to light as compared to Ayamuraska. Under the different weak light conditions, mRNA levels for ibCHS, ibF3H, ibDFR, and ibANS were obviously decreased, while the expression of ibGT was increased. These results indicated that anthocyanin content was regulated by light at the mRNA levels and the enzymatic level in sweet potato. Therefore, the development dynamic response to anthocyanin content varied in different genotypes of PFSP, and mRNA levels of anthocyanin biosynthesis were inhibited under the weak light condition. 展开更多

Sequence-related amplification polymorphism (SRAP) markers closely linked to stem nematode resistance gene were developed in sweetpotato, lpomoea batatas (L.) Lam. Using bulked segregant analysis (BSA), 200 SRAP primer combinations were screened with the resistant and susceptible bulked DNA from the 196 progenies of an F1 single-cross population of resistant parent Xu 781xsusceptible parent Xushu 18, 77 of them showed polymorphic bands between resistant and susceptible DNA. Primer combinations detecting polymorphism between the two bulks were used to screen both parents and 10 individuals from each of the bulks. The results showed that primer combination A9B4 produced 3 specific bands in the resistant plants but not in the susceptible plants, suggesting that the markers, named Nspl, Nsp2 and Nsp3, respectively, linked to a gene for stem nematode resistance. Primer combination A3B6 also produced a SRAP marker named Nsp4 linking to the resistance gene. Amplified analysis of the 196 F1 individuals indicated that the genetic distance between these markers and the resistance gene was 4.7, 4.7, 6.3, and 9.6 cM, respectively. 展开更多

Sweet potato (Ipomoea batatas Lam.) leaf stalks were cut into 20 cm length, and stored at 20, 6, and 2℃. The respiration rate, ethylene production, ascorbic acid, free amino acid, total chlorophyll content, freshness, and shelf life were determined during storage to investigate the effect of storage temperatures on the quality and physiological responses in sweet potato leaf stalks. Wound responses were observed as high respiration rate and ethylene production immediately after cutting. Sweet potato leaf stalks were found to be sensitive to chilling injury manifested as browning and water-soaking on the surface at 2℃. In contrast, sweet potato leaf stalks were susceptible to senescence, exhibited by etiolating and yellowing, at 20℃. Loss in weight and chlorophyll was minimized under low temperatures. High temperatures also caused the accumulation of amino acids with a significant loss of ascorbic acid and chlorophyll. Sweet potato leaf stalks had a storage life of 16 days at 6℃, 8 days at 2℃, and 6 days at 20℃, respectively. 展开更多

Sweet potato(Ipomoea batatas,Lam.)is an important root vegetable in developing countries.After its domestication in Neotropical America,human migration led to the distribution of the sweet potato plant throughout the world.Both leaf and storage root are high in compounds of nutritional value.Yet,the storage roots are of particular value due to their significant content of provitamin A(β-carotene).The breeding effort for elite sweet potato lines led to the reduction of genetic diversity and the potential to improve other traits.The focus of the present study was to assess the metabolic diversity of 27 sweet potato cultivars including landraces and improved varieties.A metabolite profiling approach was optimised for sweet potato leaf and storage root tissue and 130 metabolites identified with three different analysis platforms.The data highlighted a lack of correlation between storage root phenotype and leaf metabolism.Furthermore,the metabolic diversity of storage roots was based on the secondary metabolism,including phenylpropanoids and carotenoids.Three cultivars of three different flesh colouration(yellow,orange and purple)showed a significant difference of the primary metabolism.This data demonstrates the value of metabolite profiling to breeding programs as a means of identifying differences in phenotypes/chemotypes and characterising parental material for future pre-breeding resources. 展开更多

To determine the genetic diversity and population structure of sweet potato accessions cultivated in China, and to establish the genetic relationships among their germplasm types, a representative collection of 240 accessions was analyzed using inter-simple sequence repeat (ISSR) markers. The mean genetic similarity coefifcient, Nei’s gene diversity, and shared allele distance of tested sweet potato accessions were 0.7302, 0.3167 and 0.2698, respectively. The 240 accessions could be divided into six subgroups and ifve subpopulations based on neighbor-joining (NJ) clustering and STRUCTURE results, and obvious genetic relationships among the tested sweet potato accessions were identiifed. The marker-based NJ clustering and population structure showed no distinct assignment pattern corresponding to lfesh color or geographical ecotype of the tested sweet potato germplasm. Analysis of molecular variance (AMOVA) revealed small but signiifcant difference between white and orange-lfeshed sweet potato accessions. Small but signiifcant difference were also observed among sweet potato accessions from the Southern summer-autumn sweet potato region, the Yellow River Basin spring and summer sweet potato region and the Yangtze River Basin summer sweet potato region. This study demonstrates that genetic diversity in the tested sweet potato germplasm collection in China is lower than that in some reported sweet potato germplasm collections from other regions. Pedigree investigations suggest that more diverse Chinese sweet potato varieties should be formed by broadening the selection scope of breeding parents and incorporating the introduced varieties into future breeding programs. 展开更多

Sporamin is a soluble protein in sweet potato, and falls into two distinct homology groups, subfamilies A and B. In this research, a sporamin B was purified and its amino acid sequences, trypsin inhibitor activity (Ti activity) were analyzed. This sporamin B was isolated from sweet potato tubers [Ipomoea batatas (L.) Lam cv. 55-2] through extraction of the water-soluble fraction, dialysis, ultrafiltration and ion-exchange chromatography. Homology determined by polyacrylamide gel electrophoresis showed that mainly one bond appeared in gel after being reduced by SDS (sodium dodecyl sulfate), or by SDS and 2-mercaptoethanol, or in native situation. By comparing the data of the polypeptide mass Matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry with those of the mass of the theoretical amino acid sequences from NCBI protein database, it was revealed that it was Q40091|Q40091_IPOBA, sweet potato sporamin B - Ipomoea Batatas (sweet potato) (Batate). The sequence coverage was 70.6%. N-terminal sequence was SETPV (Ser-Glu-Thr-Pro-Val). There is a linear relationship between trypsin inhibitor activity (Ti activity) and amounts of this sporamin B (3-18 μg mL-1). The equation of linear regression was y = 2.5809x + 17.049 (r2 = 0.9966). There was a curvilinear relationship between Ti activity and amounts of this sporamin B (21-150 μg mL-1). The equation of curvilinear regression is y = 14.417ln(x) + 23.26 (r2 = 0.9924). The concentration of sporamin B with Ti activity after heating at 40°C may induce part denature of this sporamin B, and there was no statistic difference after heating at 40, 50, 60°C for 20 min. Heat treatment at more than 90°C leads to a dramatic decrease of trypsin inhibitor efficiency. The results suggested that Q40091|Q40091_IPOBA was the major sporamin B in sweet potato tubers [Ipomoea Batatas (L.) Lam cv. 55-2], which had strong Ti activity, and was stable to both thermal and DTT (DL-dithiothreitol) relatively. 展开更多

An experiment was carried out at Mkuranga Research Station of National Coconut Development Programme (NCDP) to study the effect of light regimes on three different crops which are sweet potato, cowpea and pineapple between 1995 and 1996. Four light intensities with nominal values of light transmission namely L0 = 100%, L 1 = 75%, L2 = 50%, L3 = 25% were provided by nylon netting materials which filtered sunlight to varying light percentages. The experiment was a split-plot on randomized complete block design, replicated four times. Growth measurements and yield were recorded at a regular interval in all crops. Leaf morphological characteristics were carried out only on sweet potato. Result on the leaf morphological characteristics showed light intensity strongly influenced growth and development of sweet potato. Specific leaf area values in full light were smaller than those in under heavy shade. The light intensity increased the cowpea seed yield significantly (P 〈 0.01) and the interaction between seasons (year) x light regimes was significant (P 〈 0.01). In low intensity, pineapple flowered earlier and yielded more than in high intensity. These results have provided useful information in planning intercropping models in coconut based-farming systems in Tanzania. 展开更多

This study aimed to evaluate the effect of water extracts from sweet potato [Ipomoea batatas (L.) Lamarck] on seed germination of Ageratum conyzoides L. under controlled conditions. The aqueous was produced from plant parts i.e., roots, stems, and leaves of sweet potato at concentrations of 0.025, 0.050, 0.075, and 0.100 g·mL<sup>-1</sup>. The results showed that the plant parts of sweet potato all contained allelopathic substances, which showed high-concentration inhibition and low-concentration promotion of seed germination of A. conyzoides. When the aqueous extract concentrations were 0.050, 0.075, and 0.100 g·mL<sup>-1</sup>, the germination of A. conyzoides seeds was inhibited, while the germination was promoted at a concentration of 0.025 g·mL<sup>-1</sup>. This shows that when the planting density of sweet potato is large, it can form an obvious prevention and control effect on A. conyzoides, and thus improve herbicide resistance management. 展开更多

Sweet potato is a multifunctional root crop with many essential nutrients and bioactive compounds.Due to its genetic complexity and lack of genomic resources,efficient genetic studies and cultivar development lag far behind other major crops.Simple sequence repeats(SSRs)offer an effective molecular marker technology for molecular-based breeding and for locating important loci in crop plants,but only a few have previously been developed in sweet potato.To further explore new SSR markers and accelerate their use in sweet potato genetic studies,genome-wide characterization and development of SSR markers were performed using the recently published genome of sweet potato cultivar,Taizhong6.In this study,a set of 2,431 primer pairs were developed from 133,727 SSRs identified in the sweet potato genome using the Perl script MISA software.The average frequency was one SSR per 6.26 kb,with dinucleotides(38.5%)being the most dominant repeat motif.The main motif types in all repeats were AT/AT,AAT/ATT,A/T,AAAT/ATTT,AAAAT/ATTTT and AAAAAT/ATTTTT accounting for 78.29%of the total SSRs.50%of the 100 randomly selected primer pairs amplified 251 alleles,and the average number of alleles was 5.02 per locus for values ranging between 1 and 13.The UPGMA cluster analysis grouped the 24 sweet potato genotypes into four clusters at a similarity coefficient of 0.68.The SSR markers currently developed will provide valuable genetic resources for germplasm identification,genetic diversity analysis,and functional genomics studies in sweet potato and related species. 展开更多

Allelopathic compounds have the potential to inhibit the growth and development of other organisms in a diverse manner ranging from shifting nutrients and enhancing their growth to inflicting diseases. In addition, these compounds influence seedling growth and seed germination of various crops. The goal of this study was to identify and quantify different allelochemicals in various sweet potato cultivars through high-performance liquid chromatography techniques. Selected sweet potato slips (weight: 2.0 - 2.5 grams/slip) were propagated in separate glass tubes filled with 10.0 mL distilled water. Water extract from each glass tube was collected after 2, 4, and 6 weeks after transplanting (WAP) to identify and quantify allelochemical compounds by comparing their peaks with the retention time of standards. Results show that the concentration of allelochemicals in water extract was increased from 2 to 4 WAP but remained constant in the sixth week. Quantitative analysis revealed that the amount of chlorogenic acid was higher in all sweet potato cultivars compared to other allelochemicals. Some sweet potato cultivars, A5 and A39, exhibited higher allelopathy (18.28 - 19.37 ppm/slip) and reduced the height and biomass of Palmer amaranth the most due to the presence of increased concentration of combined allelochemicals, while other cultivars produced lesser allelochemicals (10.90 ppm/slip) and did not reduce the growth of the weed species. Allelopathic sweet potato cultivars high in chlorogenic acid production can effectively suppress Palmer amaranth with minimal dependence on chemicals to manage weeds and harmful pests under sustainable agricultural system. 展开更多

In Martinique (F.W.I.), biodiversity of vegetable varieties remain unexploited due to public unawareness or supply problems. To further enhance the sweet potato, physicochemical, sensory and nutritional characteristics of eight varieties of sweet potato were established. The eight sweet potatoes cultivars were noticeably diverse with phenotypic, sensory, physicochemical and nutritional characteristics which were very varied. The eight cultivars grown in Martinique were advantageously positioned in relation to the globally identified sweet potato, regarding fibre, starch, potassium, copper, vitamin B1, B3 and B9 contents. Compared to the ordinary potato, Martiniquan sweet potatoes have a much better nutritional potential. Energy value and vitamin C levels were twice as much as in normal potatoes. Martiniquan sweet potatoes cultivars were also richer in minerals and vitamins, with 2.7 times more vitamin B9 and 63 times more beta-carotene than the ordinary potato. 展开更多

Objective: To determine antioxidant activity and phytochemical content from various tubers extracts of sweet potato(Ipomoea batatas) and to explore the correlation of phytochemical content with their antioxidant activities. Methods: Antioxidant activities were tested using DPPH and FRAP assays. Total phenolic was calculated by Folin-Ciocalteu reagent, flavonoid content by Chang's method and correlation with their antioxidant activities were analyzed by Pearson's method. Results: PO2 showed highest antioxidant activity, which had the lowest IC50 DPPH(10.54 μg/m L) and the lowest EC50 FRAP(11.14 μg/m L). PO2 showed the highest total phenolic(11.91 g GAE/100 g) and total flavonoid content(17.83 g QE/100 g). There were significantly negative correlation between total phenolic content and flavonoid content in sample PO with their IC50 DPPH and EC50 FRAP. IC50 DPPH of sample PP and PO showed significantly positive correlation with their EC50 FRAP. Conclusions: Result of DPPH method shows that all different ethyl acetate and ethanolic tubers extracts of four varieties of sweet potato are classified as strong and very strong antioxidant. Result of DPPH and FRAP methods indicates that phenolic and flavonoid compounds in sample PO contributes together to antioxidant activities. Antioxidant activities of sample PP and PO by DPPH method are linear to FRAP method. 展开更多

Effect of Tillage Systems and Vine Orientation on Yield of Sweet Potato (<i>Ipomoea batatas</i>L.)

In Zimbabwe sweet potato (Ipomoea batatas L.) production is practiced under different tillage systems with varying vine orientations, which contribute to variable yields. Limited research on yield loss due to these different practices is available despite that the majority of farmers are growing sweet potatoes to sustain their livelihoods. A field study was carried out at Midlands State University in the 2013/14 rainy season, to determine the effect of tillage systems and vine orientation on yield of sweet potato. A 2 × 3 factorial treatment structure in a randomized complete block design (RCBD), with three replications was used. Tillage systems had two factors (ridge and mound) and three vine orientations were used (horizontal, fold and loop). Data on storage root length, storage root diameter and storage root weight was subjected to Analysis of Variance (ANOVA) at 5% significance level. There was no interaction (p < 0.05) between tillage systems and vine orientations on storage root diameter, storage root length and storage root yield. The horizontal vine orientation gave statistically significant (p < 0.05) storage roots diameter of width 405 mm. Conversely the loop vine orientation had statistically the least (316 mm) storage root diameter. Horizontal vine orientation had significantly (p < 0.05) wider storage roots than the loop vine orientation. The ridge recorded longer (134.2 mm) mean storage root length, while those from mounds had shorter (115.9 mm) root length. The loop and the horizontal vine orientations recorded statistically the highest (35.5 t/ha and 34.8 t/ha respectively) sweet potato storage root yield. On the other hand, the fold vine orientation obtained significantly the lowest (28.7 t/ha) storage root yield. The research concluded that the horizontal and fold vine orientations had the widest storage root diameter and the ridge had longer storage root lengths. The loop and horizontal vine orientations are recommended in sweet potato production if high yields are to be achieved. 展开更多
