目的探究环状RNA hsa_circ_0085576对口腔鳞状细胞癌(OSCC)细胞迁移和侵袭的影响及其分子机制。方法使用实时荧光定量聚合酶链反应(qRT-PCR)和蛋白印迹法检测OSCC细胞中hsa_circ_0085576、微小RNA-498(miR-498)以及B细胞特异性莫洛尼鼠... 目的探究环状RNA hsa_circ_0085576对口腔鳞状细胞癌(OSCC)细胞迁移和侵袭的影响及其分子机制。方法使用实时荧光定量聚合酶链反应(qRT-PCR)和蛋白印迹法检测OSCC细胞中hsa_circ_0085576、微小RNA-498(miR-498)以及B细胞特异性莫洛尼鼠白血病病毒整合位点1(BMI-1)的表达水平。使用CCK-8、划痕实验、Transwell实验以及qRT-PCR、蛋白印迹法分别检测SCC-15细胞增殖活力、迁移及侵袭能力以及相关基因和蛋白的相对表达量。结果OSCC细胞中hsa_circ_0085576与BMI-1表达上调,miR-498表达下调(P<0.05)。下调hsa_circ_0085576表达或过表达miR-498后SCC-15细胞的增殖活性、划痕愈合率、侵袭细胞数目以及细胞周期蛋白D1、波形蛋白表达水平下调,miR-498和E-钙黏蛋白表达水平上调(P<0.05);抑制miR-498表达可减弱下调hsa_circ_0085576表达对OSCC细胞增殖、迁移及侵袭的抑制作用;上调BMI-1表达可减弱过表达miR-498对OSCC细胞增殖、迁移及侵袭的抑制作用。结论下调hsa_circ_0085576表达可通过激活miR-498/BMI-1轴抑制OSCC细胞增殖、迁移及侵袭。 展开更多

Expression and clinical value of circular RNA Hsa_circ_0046701 in gliomas

Objective:To explore the expression of hsa_circ_0046701 in patients with glioma and its correlation with clinicopathological features and prognosis of glioma,and to study its clinical value.Methods:qRT-PCR was used to detect the expression of hsa_circ_0046701 in glioma tissues and normal brain tissues,and chi-square test was used to analyze the effects of different levels of hsa_circ_0046701 expression on the clinicopathological features of patients.The survival curve was drawn by Kaplan-Meier method,and the univariate and multivariate analysis of prognosis was performed by Cox proportional hazard regression model.Results:The expression level of hsa_circ_0046701 in gliomas was significantly higher than that in normal brain tissues,and the expression in gradeⅢ-Ⅳgliomas was higher than that gradeⅠ-Ⅱgliomas in WHO grade.The expression of hsa_circ_0046701 was significantly correlated with WHO grade,KPS score and postoperative recurrence,but not with age,sex,tumor size,number and location.The five-year survival rate of the group with high expression of hsa_circ_0046701 was lower than that of the group with low expression of hsa_circ_0046701.The patients with high expression of hsa_circ_0046701,had high WHO grade and the KPS score was less than 80.Postoperative recurrence was an independent prognostic factor for glioma patients.Conclusion:Hsa_circ_0046701 is highly expressed in gliomas,which is positively correlated with the malignant degree of gliomas,and can be used as a potential biological marker.High expression of hsa_circ_0046701,high WHO grade,KPS score<80,postoperative recurrence are the key factors affecting the prognosis of patients,which can be used as independent prognostic factors for patients. 展开更多

目的:探究环状非编码RNA hsa_circ_001842作为癌基因在肾细胞癌(renal cell carcinoma,RCC)患者肾癌组织及细胞中的表达情况,以及其对肾癌细胞增殖、凋亡、侵袭与成瘤能力的影响。方法:收集2018年9月至2019年10月绵阳市中心医院泌尿外... 目的:探究环状非编码RNA hsa_circ_001842作为癌基因在肾细胞癌(renal cell carcinoma,RCC)患者肾癌组织及细胞中的表达情况,以及其对肾癌细胞增殖、凋亡、侵袭与成瘤能力的影响。方法:收集2018年9月至2019年10月绵阳市中心医院泌尿外科进行手术切除的肾癌标本及癌旁组织标本各64例。利用qRT-PCR检测所有RCC组织与癌旁正常组织中hsa_circ_001842的表达情况。将Caki-1肾癌细胞分别转染hsa_circ_001842基因过表达质粒(ox-circ_001842)、沉默质粒(shcirc_001842)和空质粒(ox-NC和sh-NC),采用细胞克隆形成实验、流式细胞术、细胞划痕实验和细胞侵袭实验,检测hsa_circ_001842对肾癌细胞增殖凋亡和迁移侵袭能力的影响。使用Western blot检测各组肾癌细胞上皮间充质转化(epithelial mesenchymal transition,EMT)相关蛋白与MMP-9蛋白表达水平。将转染sh-NC和sh-circ_001842的Caki-1肾癌细胞分别接种于裸鼠皮下,每7天测量一次肿瘤体积及体质量,4周后处死各组小鼠,并使用免疫组化技术和qRT-PCR检测肿瘤组织中EMT相关蛋白的表达情况。结果:RCC患者肾癌组织中hsa_circ_001842的表达量明显高于癌旁正常组织(P<0.05)。体外实验表明,与ox-NC组和sh-circ_001842组相比,ox-circ_001842组和sh-NC组Caki-1肾癌细胞的集落形成率、划痕愈合比率、细胞侵袭能力和E-cadherin蛋白的表达均明显增加(P<0.05),而细胞凋亡率、N-cadherin和MMP-9蛋白的表达明显降低(P<0.05)。裸鼠成瘤实验表明,与sh-NC组相比,sh-circ_001842组裸鼠形成的肿瘤体积和体质量均小于sh-NC组形成的肿瘤(P<0.05),并且E-cadherin蛋白表达水平明显上升,N-cadherin蛋白表达水平明显下降(P<0.05)。结论:hsa_circ_001842在肾癌组织及细胞中表达上调,抑制hsa_circ_001842的表达可以抑制癌细胞的增殖、迁移、侵袭和成瘤能力。 展开更多

Role of hsa_circ_0007482 in pterygium development: insights into proliferation, apoptosis, and clinical correlations

AIM:To investigate the impact of hsa_circ_0007482 on the proliferation and apoptosis of human pterygium fibroblasts(HPFs)and its correlation with the severity grades of pterygium.METHODS:Pterygium and normal conjunctival tissues were collected from the superior area of the same patient’s eye(n=33).The correlation between pterygium severity and hsa_circ_0007482 expression using quantitative reversetranscription polymerase chain reaction(RT-qPCR)were analyzed.Three distinct siRNA sequences targeting hsa_circ_0007482,along with a negative control sequence,were transfected into HPFs.Cell proliferation was assessed using the cell counting kit-8.Expression levels of Ki67,proliferating cell nuclear antigen(PCNA),Cyclin D1,Bax,B-cell lymphoma-2(Bcl-2),and Caspase-3 were measured via RT-qPCR.Immunofluorescence staining was employed to detect Ki67 and vimentin expressions.Apoptosis was evaluated using flow cytometry.RESULTS:Hsa_circ_0007482 expression was significantly higher in pterygium tissues compared to normal conjunctival tissues(P<0.001).Positive correlations were observed between hsa_circ_0007482 expression and pterygium severity,thickness,and vascular density.Knockdown of hsa_circ_0007482 inhibited cell proliferation,reducing the mRNA expression of Ki67,PCNA,and Cyclin D1 in HPFs.Hsa_circ_0007482 knockdown induced apoptosis,increasing mRNA expression levels of Bax and Caspase-3,while decreasing Bcl-2 expression in HPFs.Additionally,hsa_circ_0007482 knockdown attenuated vimentin expression in HPFs.CONCLUSION:The downregulation of hsa_circ_0007482 effectively hampers cell proliferation and triggers apoptosis in HPFs.There are discernible positive correlations detected between the expression of hsa_circ_0007482 and the severity of pterygium. 展开更多
