Leucine-rich alpha-2 glycoprotein as a superior biomarker to Creactive protein for detecting small bowel lesions in Crohn’s disease

BACKGROUND Achievement of endoscopic healing(EH)is significant in the clinical practice of inflammatory bowel disease as it is correlated with improved prognosis.Existing biomarkers,including C-reactive protein(CRP),have relatively low accuracy for predicting EH,especially in small intestinal lesions in Crohn’s disease(CD);thus,noninvasive and more accurate biomarkers are required.Leucine-rich alpha-2 glycoprotein(LRG),a 50-kD protein,is produced under inflammatory conditions and has been reported to be useful in assessing disease activity in inflammatory bowel disease.However,the usefulness of LRG in small intestinal lesions in CD remains inconclusive.AIM To determine the usefulness of LRG for EH in small bowel lesions in CD and compare it with CRP.METHODS This study included 133 consecutive patients with CD who underwent balloonassisted enteroscopy between June 2021 and March 2024 at Shiga University of Medical Science Hospital(Otsu,Japan).We retrospectively analyzed endoscopic scores in each of the ileum and colon and four markers including LRG,CRP,albumin,and Harvey-Bradshaw index(HBI).Spearman’s rank correlation coefficient and receiver operating characteristic analysis were performed.RESULTS Either active ileal or colonic lesions exhibited significant differences in LRG,CRP,albumin,and HBI compared with EH.CRP,albumin,and HBI showed a worse correlation with endoscopic activity in the ileum than that in the colon;however,LRG did not show a worse correlation(colon,r=0.5218;ileum,r=0.5602).Receiver operating characteristic analysis revealed that LRG for EH in the ileum and colon had the same cutoff values of 12.4μg/mL.Comparing the areas under the curve of LRG and CRP for predicting EH in the ileum revealed a significantly higher areas under the curve of LRG(95%confidence interval,0.017-0.194;P=0.024),whereas the two showed no significant difference in the colon.CONCLUSION LRG is a useful biomarker in assessing the endoscopic activity of CD and is more useful than CRP in the small intestine. 展开更多

In Situ Growth of 2D Metal–Organic Framework Ion Sieve Interphase for Reversible Zinc Anodes

Zinc metal anodes are gaining popularity in aqueous electrochemical energy storage systems for their high safety,cost-effectiveness,and high capacity.However,the service life of zinc metal anodes is severely constrained by critical challenges,including dendrites,water-induced hydrogen evolution,and passivation.In this study,a protective two-dimensional metal–organic framework interphase is in situ constructed on the zinc anode surface with a novel gel vapor deposition method.The ultrathin interphase layer(~1μm)is made of layer-stacking 2D nanosheets with angstrom-level pores of around 2.1Å,which serves as an ion sieve to reject large solvent–ion pairs while homogenizes the transport of partially desolvated zinc ions,contributing to a uniform and highly reversible zinc deposition.With the shielding of the interphase layer,an ultra-stable zinc plating/stripping is achieved in symmetric cells with cycling over 1000 h at 0.5 mA cm−2 and~700 h at 1 mA cm^(−2),far exceeding that of the bare zinc anodes(250 and 70 h).Furthermore,as a proof-of-concept demonstration,the full cell paired with MnO_(2) cathode demonstrates improved rate performances and stable cycling(1200 cycles at 1 A g−1).This work provides fresh insights into interphase design to promote the performance of zinc metal anodes. 展开更多

为了探究辣椒C_(2)H_(2)型锌指蛋白家族在响应涝胁迫中的作用,本研究通过生物信息学方法对辣椒C_(2)H_(2)型锌指蛋白家族蛋白理化性质、基因结构、染色体定位、顺式作用元件进行了全面分析。结果表明,辣椒C_(2)H_(2)型锌指蛋白家族包含9... 为了探究辣椒C_(2)H_(2)型锌指蛋白家族在响应涝胁迫中的作用,本研究通过生物信息学方法对辣椒C_(2)H_(2)型锌指蛋白家族蛋白理化性质、基因结构、染色体定位、顺式作用元件进行了全面分析。结果表明,辣椒C_(2)H_(2)型锌指蛋白家族包含91个成员,多数成员定位于细胞核。91个成员可被分为6个亚族,不同亚族的成员基因结构存在差异。C_(2)H_(2)型锌指蛋白家族基因启动子区域主要包含植物生长发育元件、植物激素响应元件及逆境胁迫响应元件。对耐涝辣椒资源GC41和涝胁迫敏感型辣椒资源GC39在涝胁迫下的转录组数据进行分析,结果表明,在GC41中,CaC_(2)H_(2)-17、CaC_(2)H_(2)-87在涝胁迫处理3 d时相对表达量高,CaC_(2)H_(2)-17、CaC_(2)H_(2)-87在GC39中未表达。筛选出CaC_(2)H_(2)-16(Capana01g003556)、CaC_(2)H_(2)-32(Capana04g001616)、CaC_(2)H_(2)-72(Capana11g000176)、 CaC_(2)H_(2)-73(Capana11g000178)、CaC_(2)H_(2)-87(Capana12g000773)5个转录因子,这5个转录因子分别调控了419个、526个、566个、305个、480个差异表达基因。本研究结果为辣椒C_(2)H_(2)家族成员的功能解析及耐涝辣椒资源的分子育种提供了重要的理论依据和数据支持。 展开更多

BACKGROUND Cases of myelin oligodendrocyte glycoprotein(MOG)antibody-related disease have a history of coronavirus disease 2019 infection or its vaccination before disease onset.Severe acute respiratory syndrome virus 2(SARS-CoV-2)infection has been considered to be a trigger of central nervous system autoimmune diseases.CASE SUMMARY Here we report a 20-year male with MOG-associated transverse myelitis after a SARS-CoV-2 infection.The patient received a near-complete recovery after standard immunological treatments.CONCLUSION Attention should be paid to the evaluation of typical or atypical neurological symptoms that may be triggered by SARS-CoV-2 infection. 展开更多

BACKGROUND Zinc-α2-glycoprotein 1 (AZGP1) plays important roles in metabolism-related diseases. The underlying molecular mechanisms and therapeutic effects of AZGP1 remain unknown in non-alcoholic fatty liver disease (NAFLD). AIM To explore the effects and potential mechanism of AZGP1 on NAFLD in vivo and in vitro. METHODS The expression of AZGP1 and its effects on hepatocytes were examined in NAFLD patients, CCl4-treated mice fed a high fat diet (HFD), and human LO2 cells. RESULTS AZGP1 levels were significantly decreased in liver tissues of NAFLD patients and mice. AZGP1 knockdown was found to activate inflammation;enhance steatogenesis, including promoting lipogenesis [sterol regulatory elementbinding protein (SREBP)-1c, liver X receptor (LXR), fatty acid synthase (FAS), acetyl-CoA carboxylase (ACC), and stearoyl CoA desaturase 1 (SCD)-1], increasing lipid transport and accumulation [fatty acid transport protein (FATP), carnitine palmitoyl transferase (CPT)-1A, and adiponectin], and reducing fatty acid β-oxidation [farnesoid X receptor (FXR) and peroxisome proliferator-activated receptor (PPAR)-α];accelerate proliferation;and reverse apoptosis in LO2 cells. AZGP1 overexpression (OV-AZGP1) had the opposite effects. Furthermore, AZGP1 alleviated NAFLD by blocking TNF-α-mediated inflammation and intracellular lipid deposition, promoting proliferation, and inhibiting apoptosis in LO2 cells. Finally, treatment with OV-AZGP1 plasmid dramatically improved liver injury and eliminated liver fat in NAFLD mice. CONCLUSION AZGP1 attenuates NAFLD with regard to ameliorating inflammation, accelerating lipolysis, promoting proliferation, and reducing apoptosis by negatively regulating TNF-α. AZGP1 is suggested to be a novel promising therapeutic target for NAFLD. 展开更多

V_(3)O_(7)·H_(2)O(VO)is a high capacity cathode material in the field of aqueous zinc ion batteries(AZIBs),but it is limited by slow ion migration and low electrical conductivity.In this paper,polypyridine(PPyd)intercalated VO with nanoribbon structure was prepared by a simple in-situ pre-intercalation,which is noted VO-PPyd.The total density of states(TDOS)shows that after the pre-intercalation of PPyd,an intermediate energy level appears between the valence band and conduction band,which provides a step that can effectively reduce the band gap and enhance the electron conductivity.Furthermore,the density functional theory(DFT)results found that Zn^(2+)is more easily de-intercalated from the V-O skeleton,which proves that the embeddedness of PPyd improves the diffusion kinetics of Zn^(2+).Electrochemical studies have shown that VO-PPyd cathode materials exhibit excellent rate performance(high specific capacity of 465 and 192 mA h g^(-1)at 0.2 and 10 A g^(-1),respectively)and long-term cycling performance(92.7%capacity retention rate after 5300 cycles),due to their advantages in structure and composition.More importantly,the energy density of VO-PPyd//Zn at 581 and 5806 W kg^(-1)is 375 and 247 W h kg^(-1),respectively.VO-PPyd exhibits excellent electrochemical properties compared to previously reported vanadium based cathodes,which makes it highly competitive in the field of high-performance cathode materials of AZIBs. 展开更多

The main active components of ginseng are ginsenosides,which play significant roles in treating cardiovascular diseases,cancer,and providing antioxidant effects.Ginsenosides are primarily synthesized through the mevalo-nate pathway and the methylerythritol phosphate pathway.Many key enzyme genes involved in this biosynthetic process have been cloned and validated,yet the regulatory functions of transcription factors remain unclear.The C_(2)H_(2)-type zincfinger protein family,one of the largest families of transcription factors,is crucial in plant growth and development,response to biotic and abiotic stresses,and regulation of secondary metabolism.This study,based on the ginseng transcriptome database from Jilin,conducted a correlation analysis between the expression levels of PgZFPs genes in the Jilin ginseng C_(2)H_(2)-type zincfinger protein family and ginsenoside content,a gen-ome-wide association study of PgZFPs,and co-expression analysis of PgZFPs with validated key enzyme genes.Ultimately,five candidate genes involved in ginsenoside biosynthesis were identified.The involvement of PgZFP27 and PgZFP-59-02 genes from the PgZFPs family in the biosynthesis of ginsenosides was validated through in vitro methyl jasmonate(MeJA)induction experiments.This result provides new genetic resources for the biosynthesis of ginsenosides. 展开更多

目的目的探究子宫内膜癌(EC)组织中富含亮氨酸α-2糖蛋白1(LRG1)的表达情况及其与EC肿瘤细胞生物学行为及患者预后的关系。方法方法选取96例EC患者为研究对象,检测其EC组织和癌旁组织中LRG1水平。比较不同LRG1表达情况EC患者的临床特征... 目的目的探究子宫内膜癌(EC)组织中富含亮氨酸α-2糖蛋白1(LRG1)的表达情况及其与EC肿瘤细胞生物学行为及患者预后的关系。方法方法选取96例EC患者为研究对象,检测其EC组织和癌旁组织中LRG1水平。比较不同LRG1表达情况EC患者的临床特征,分析LRG1表达对EC患者预后的影响。将EC组织分为con-trol组、NC组(转染si-NC的细胞)、LRG1组(转染siRNA-LRG1的细胞),检测其生物学行为。结果结果EC组织中LRG1表达水平与阳性率均高于癌旁组织,差异均有统计学意义(P﹤0.05)。LRG1阳性组中国际妇产科联盟(FI-GO)分期为Ⅲ~Ⅳ期、发生淋巴结转移患者的比例均高于LRG1阴性组,差异均有统计学意义(P﹤0.05)。死亡组EC患者中LRG1阳性表达、年龄≥55岁、肌层浸润≥1/2、腺癌、低分化、FIGO分期为Ⅲ~Ⅳ期、有淋巴结转移患者的比例均高于生存组,差异均有统计学意义(P﹤0.05)。LRG1阳性表达和分化程度为低分化是EC患者发生不良预后的独立危险因素(P﹤0.05)。在48、72 h,LRG1组的EC细胞活力显著低于control、NC组(P﹤0.05);相比control、NC组,LRG1组细胞侵袭数、细胞迁移率均更低,凋亡水平更高(P﹤0.05)。结论结论EC组织中LRG1表达升高,是EC患者不良预后的危险因素,抑制LRG1表达可抑制EC细胞活力及侵袭、迁移能力,促进细胞凋亡。 展开更多

Objective:To investigate the levels of zinc-α-2-glycoprotein(ZAG) among Omani AIDS patients receiving combined antiretroviral therapy(cART).Methods:A total of 80 Omani AIDS patients(45 males and 33 females),average age of 36 vears.who were receiving cART at the Saltan Qaboos University Hospital(SQUH).Muscat,Oman,were tested for the levels of ZAG.In addition,SO healthy blood donors(46 males and 34 females),average age of 26 years,attending the SOUH Blood Bank,were tested in parallel as a control group.Measurement of the ZAG levels was performed using a competitive enzyme—linked immunosorbent assay and in accordance with the manufacturer's instructions.Results:The ZAG levels were found to he significantly higher among AIDS patients compared to the healthy individuals(P=0.033).A total of 56(70%) of the AIDS patients were found to have higher levels of ZAG and 16(20%) AIDS patients were found to have high ZAG levels,which are significantly(P>0.031) associated with weight loss.Conclusions:ZAG levels are high among Omani AIDS patients on cART and this necessitales the measurement of ZAG on routine basis,as it is associated with weight loss. 展开更多

目的探讨结直肠癌(CRC)患者血清胰岛素样生长因子结合蛋白-3(IGFBP-3)、高尔基体跨膜糖蛋白73(GP73)、结直肠癌特异性抗原-2(CCSA-2)的表达与临床病理特征和预后的关系。方法选取2019年1月至2020年12月于宝鸡高新医院及宝鸡市中医医院... 目的探讨结直肠癌(CRC)患者血清胰岛素样生长因子结合蛋白-3(IGFBP-3)、高尔基体跨膜糖蛋白73(GP73)、结直肠癌特异性抗原-2(CCSA-2)的表达与临床病理特征和预后的关系。方法选取2019年1月至2020年12月于宝鸡高新医院及宝鸡市中医医院就诊的CRC患者80例为CRC组,另纳入同期的体检健康者80例作为对照组,采用酶联免疫吸附试验检测两组血清IGFBP-3、GP73和CCSA-2水平,收集CRC患者的临床资料,对所有患者进行为期3年的随访,采用Cox比例风险回归模型分析预后影响因素。结果CRC组血清IGFBP-3水平低于对照组(P<0.05),GP73、CCSA-2水平高于对照组(P<0.05)。血清IGFBP-3、GP73和CCSA-2水平与患者肿瘤最大径、分化程度、TNM分期及有无淋巴结转移有关(P<0.05)。IGFBP-3高表达、GP73和CCSA-2低表达患者的总生存率和无进展生存率均高于IGFBP-3低表达、GP73和CCSA-2高表达患者(P<0.05)。CRC患者的肿瘤最大径、分化程度、TNM分期、有无淋巴结转移及血清IGFBP-3、GP73、CCSA-2水平是CRC患者预后的影响因素(P<0.05)。结论血清IGFBP-3、GP73和CCSA-2水平与CRC患者的临床病理学特征及预后密切相关,有望成为预测和评估患者预后的重要生物学指标,可为临床早期诊断病情,尽早发现复发和转移,评估和预测患者预后提供一定的参考。 展开更多

血清LRG1及FGF-21水平与新生血管性青光眼的相关性

Rapeseed(Brassica napus L.)is a global oil crop.Salinity stress impedes the growth of rapeseed,especially during seed germination.The key genes mediating salinity stress response during seed germination in B.napus remain largely unknown.Here,we found that all six paralogs of C2H2 zinc finger transcription factor WIP DOMAIN PROTEIN 2(BnaWIP2)showed increased expression during the initial 12 hours of germination,and expression was further enhanced by salinity stress.Under NaCl treatment,knocking out all six BnaWIP2 paralogs in B.napus led to significantly reduced germination,while overexpression of BnaC06.WIP2 promoted germination.Transcriptomic analysis revealed that BnaC06.WIP2 downregulated a series of genes related to abscisic acid(ABA)biosynthesis and signaling,among which BnaA05.NCED3,BnaC04.ABI5-2,BnaA03.EM6,and BnaA05.EM6 were directly repressed by BnaC06.WIP2.Further analysis showed that in germinating seeds,BnaC06.WIP2 was induced by ABA and in turn restrained ABA production,indicating that BnaC06.WIP2 forms a negative feedback loop with ABA to promote seed germination under salinity stress in B.napus.Collectively,these results enhance our understanding of the novel function of BnaWIP2 and provide valuable genetic resources for breeding salinity-tolerant rapeseed varieties. 展开更多

设计了一种纳米颗粒状MoO_(2)@碳纳米管(CNT)复合纤维电极结构。通过简单的电化学沉积方法和一系列表征技术,将直径为10~50 nm MoO_(2)纳米颗粒均匀分散于CNT纤维束的表面及内部,获得了柔韧性、稳定性优异的纤维电极材料。研究了其作为... 设计了一种纳米颗粒状MoO_(2)@碳纳米管(CNT)复合纤维电极结构。通过简单的电化学沉积方法和一系列表征技术,将直径为10~50 nm MoO_(2)纳米颗粒均匀分散于CNT纤维束的表面及内部,获得了柔韧性、稳定性优异的纤维电极材料。研究了其作为水系锌离子电池正极材料的电性能和电化学行为。研究结果表明该纤维电极材料在900 mA/cm^(3)(4.7 A/g)电流密度下,常温循环1700周后,容量保持率为109%,-30℃低温下循环1000周后,容量保持率为103%,具有优异的常温和低温循环稳定性;1800 mA/cm^(3)(9.4 A/g)电流密度下,放电容量仍有17.5 mA·h/cm^(3)(91.5 mA·h/g),表现出优异的倍率性能。此外,研究得出Zn^(2+)嵌入/脱出过程主要发生在Mo—O—C键上,且整个充放电过程中主要在Mo^(6+)、Mo^(5+)和Mo^(4+)间发生了氧化还原反应,为开发新型纤维电极材料提供了新思路。 展开更多

Ida2--H2O system(iminodiacetate aqueous solution) was used to leach a low grade zinc oxide ore for Zn extraction.The effects of leaching time,liquid-solid ratio(L/S),total concentration of Ida2-([Ida2-]T),leaching temperature and pH on Zn leaching recovery and the dissolution of impurities such as Ca,Mg,Cu,Ni,Fe,Pb and Cd were investigated.Results show that Ca,Mg and Fe in ores were hardly dissolved in alkalescent iminodiacetate aqueous solution,while valuable metals such as Cu,Ni,Pb and Cd were partly dissolved into leaching liquor with Zn.The recovery of Zn reaches 76.6% when the ores were leached for 4 h at 70 ℃ by 0.9 mol/L iminodiacetate aqueous solution with pH of 8 and L/S of 5:1. 展开更多

The selective recovery of lead from the zinc oxide dust using an alkaline Na2EDTA solution was investigated. The effects of temperature, leaching time, Na2EDTA concentration and initial NaOH concentration on the leaching rates of lead and zinc were studied. The following optimized leaching conditions were obtained: liquid-to-solid ratio 5:1 mL/g, stirring speed 650 r/min, Na2EDTA concentration 0.12 mol/L, initial NaOH concentration 0.5 mol/L, leaching temperature 70 ℃, leaching time 120 min. Under the optimized conditions, the average leaching rates of lead, zinc, fluoride and chloride are 89.92%, 0.94%, 62.84% and 90.02%, respectively. The filtrate was used to electrowin lead powders. The average current efficiency of electrowinning is about 93% and lead content is higher than 98% under the conditions of temperature of 60 ℃, current density of 200 A/m2, H3PO4 concentration of 1.5 g/L, and lead ion concentration of above 5 g/L. The consumption of Na2EDTA and the direct current are about respectively 0.218 kg and 0.958 kW·h for per kilogram of lead powder. 展开更多

目的构建小鼠锌α2糖蛋白(mZAG)真核表达载体,在体外培养细胞中鉴定其mRNA和蛋白水平表达。方法提取小鼠肝组织总RNA,采用RT-PCR法扩增mZAG全基因表达序列,酶切法将mZAGcDNA全序列插入表达质粒载体pcDNA3.1(-)中构建pcDNA3.1(-)-mZAG真... 目的构建小鼠锌α2糖蛋白(mZAG)真核表达载体,在体外培养细胞中鉴定其mRNA和蛋白水平表达。方法提取小鼠肝组织总RNA,采用RT-PCR法扩增mZAG全基因表达序列,酶切法将mZAGcDNA全序列插入表达质粒载体pcDNA3.1(-)中构建pcDNA3.1(-)-mZAG真核表达质粒。采用阳离子脂质体转染法将不同浓度mZAG表达质粒(0、0.4、0.8、1.6μg)pcDNA3.1(-)-mZAG和4对mZAG小干扰RNA(siRNA)序列转染到3T3-L1前脂肪细胞中,实时荧光定量RT-PCR测定mZAGmRNA表达水平,Western blot检测mZAG蛋白表达情况。结果测序鉴定证实成功构建mZAG真核表达质粒pcDNA3.1(-)-mZAG。实时荧光定量RT-PCR检测结果显示,0.4、0.8、1.6μg mZAG转染组3T3-L1细胞中的mZAGmRNA表达水平分别是0μg mZAG转染组的2.58(P=0.002)、3.67(P=0.000)、5.19倍(P=0.001);mZAG-siRNA1组和mZAG-siRNA4组小鼠3T3-L1细胞中的mZAGmRNA表达水平显著减少,分别是不转染mZAGsiRNA干扰序列对照组的49%(P=0.002)和41%(P=0.000)。Western blot检测结果显示,0.8μg mZAG质粒转染组的体外mZAG蛋白表达水平是不转染mZAG质粒对照组的2.75倍(P=0.017);mZAG-siRNA1和mZAG-siR-NA4干扰序列转染组的体外mZAG蛋白表达水平仅为对照组的55%(P=0.004)和62%(P=0.025)。结论成功构建mZAG真核表达载体,该载体能够在体外细胞中良好表达。筛选出能够显著抑制mZAG表达的siRNA1和siRNA4,为今后进一步深入研究ZAG提供了有用工具。 展开更多
