The oyster mushroom(Pleurotus spp.)is one of the most widely cultivated mushroom species globally.The present study investigated the effect of synbiotics on the growth and quality of Pleurotus ostreatus and Pleurotus ...The oyster mushroom(Pleurotus spp.)is one of the most widely cultivated mushroom species globally.The present study investigated the effect of synbiotics on the growth and quality of Pleurotus ostreatus and Pleurotus pulmonarius.Different synbiotics formulations were applied by spraying mushroom samples daily and measuring their growth parameters,yield,biological efficiency,proximate composition,mineral content,total phenolic content(TPC),and diphenyl-1-picryl-hydrazyl(DPPH)radical scavenging activity.Results demonstrated that the most significant yield of oyster mushrooms was harvested from synbiotics sprayed with inulin and Lactobacillus casei(56.92 g).Likewise,the highest biological efficiency obtained with a similar synbiotic was 12.65%.Combining inulin and L.casei was the most effective method of improving the mushrooms'growth performance and nutrient content in both samples.Furthermore,synbiotics that combined inulin and L.casei resulted in the highest TPC(20.550 mg gallic acid equivalent(GAE)/g dry extract(DE))in white oyster mushrooms(P.ostreatus).In comparison,in grey mushroom(P.pulmonarius)the highest TPC was yielded by L.casei(1.098 mg GAE/g DE)followed by inulin and L.casei(1.079 mg GAE/g DE).The DPPH results indicated that the oyster mushroom could be an efficient antioxidant.The results revealed that applying synbiotics improved the mushrooms'quality by increasing their antioxidant capacity with higher amounts of phenolic compounds and offering better health benefits with the increased levels of mineral elements.Together,these studies demonstrated the potential of using synbiotics as a biofertilizer,which is helpful for mushroom cultivation;therefore,it might solve the challenge of inconsistent quality mushroom growers face.展开更多

Bioremediation, particularly through the use of organisms like Pleurotus ostreatus (mushroom) and Eisenia fetida (earthworm), has emerged as a promising approach to mitigate crude oil pollution. Understanding the effectiveness of these organisms in reducing hydrocarbon levels in contaminated soil is crucial for devising sustainable remediation strategies. Objectives: This study aimed to evaluate the efficacy of Pleurotus ostreatus and Eisenia fetida in remediating crude oil-polluted soil. Specifically, it sought to assess the hydrocarbon profiles in soil treated with these organisms across varying concentrations of crude oil pollution. Method: Crude oil concentration levels ranging from 0% to 10% were applied to soil samples alongside control treatments, including soil only, soil with earthworms, and soil with mushrooms. Each treatment was replicated five times using a randomized complete block design. Standard methods were employed to determine the hydrocarbon contents of the soil. Results: The results indicated a significant increase (P Pleurotus ostreatus and Eisenia fetida exhibited noteworthy reductions in these hydrocarbon levels. At the three-month mark, mushrooms demonstrated a remarkable ability to reduce hydrocarbon content by 70% - 90% compared to the pollution treatment. In contrast, earthworms exhibited minimal potential for hydrocarbon reduction, particularly at both three and six-month intervals. For instance, TOC reduction reached a maximum of 96% with mushroom treatment and 85% with earthworm treatment at 5% crude oil pollution over six months. Conclusion: The findings highlight the effectiveness of Pleurotus ostreatus in significantly reducing hydrocarbon levels in crude oil-polluted soil compared to Eisenia fetida. Mushroom-treated soils consistently exhibited substantial reductions in TOC, TPH, TOG, PAH, and THC over the study period, suggesting their potential as a viable bioremediation agent. In contrast, while earthworms showed some capability in reducing hydrocarbon content, their effectiveness was comparatively limited. Recommendation: Based on the results, it is recommended to utilize Pleurotus ostreatus for the bioremediation of crude oil-polluted soils. Further research could explore optimizing remediation protocols involving mushroom-based treatments for enhanced efficiency. Statement of Significance: This study contributes valuable insights into the application of bioremediation techniques for mitigating crude oil contamination in soil. The demonstrated efficacy of Pleurotus ostreatus underscores its potential as a sustainable and eco-friendly solution for remediating hydrocarbon-polluted environments, offering a promising avenue for environmental restoration and conservation efforts.展开更多
Blocking the development of edible mushrooms will affect the production cycle and yield of fruiting bodies.Phenylalanine ammonia lyase(PAL,EC 4.3.1.24.)is an enzyme that catalyzes the deamination of phenylalanine to f...Blocking the development of edible mushrooms will affect the production cycle and yield of fruiting bodies.Phenylalanine ammonia lyase(PAL,EC 4.3.1.24.)is an enzyme that catalyzes the deamination of phenylalanine to form trans-cinnamic acid.Previous studies have shown that a decrease in pal1 gene transcription delays fruiting body development in Pleurotus ostreatus.Herein,we used wild type(WT)and RNA interference(RNAi)strains to study the molecular regulation of pal1 by RNA sequencing and Agrobacterium-mediated genetic transformation.Our results showed that interference with the pal1 gene resulted in reductions in the total PAL enzyme activity and the total phenol content,as well as an increase in the intracellular H_(2)O_(2)content.RNA-Seq data demonstrated that the significantly enriched KEGG terms were mainly related to the peroxisome pathway,MAPK signaling pathway-yeast and three other pathways,and the catalase(CAT)gene cat1 is also involved in multiple pathways that were enriched above.Exogenous H_(2)O_(2)significantly enhanced the transcription of the cat1 gene and elevated total CAT enzymatic activity.Moreover,the levels of cat1 gene transcription and the total CAT enzymatic activity in the RNAi-pal1 strains gradually become closer to those in the WT strain through the removal of H_(2)O_(2),which indicated that pal1 regulated the expression of cat1 by affecting the intracellular H_(2)O_(2)content.Finally,the overexpression of the cat1 gene in P.ostreatus caused growth retardation,especially during the process of primordia formation.In conclusion,this study demonstrated that PAL1 affects cat1 gene expression through the signaling molecule H_(2)O_(2)and regulates the development of P.ostreatus.The findings of this study enhance our understanding of the molecular developmental mechanism of edible mushrooms.展开更多
Cronobacter spp.has strong resistance to desiccation and high permeability in Enterobacteriaceae,and powdered infant formula(PIF)is one of the main contamination routes.In recent years,the contamination of Cronobacter...Cronobacter spp.has strong resistance to desiccation and high permeability in Enterobacteriaceae,and powdered infant formula(PIF)is one of the main contamination routes.In recent years,the contamination of Cronobacter spp.in PIF incidents occurs from time to time,causing infant serious diseases or death.In this investigation,matrix-assisted laser desorption/ionization time of flight mass spectrometry was used to identify the phenotypes of 35 Cronobacter strains isolated from PIF and its processing environment.Subsequently,the isolates were evaluated for drying and osmotic pressure tolerance.The results showed that the deactivation rate of the strains ranged from 9.01%to 77.57%,and the highest osmotic pressure condition the strains could tolerate was 6 g/100 mL Na Cl.In addition,there was a positive correlation between biofilm formation ability and desiccation resistance.Combined with transcriptomics,Cronobacter spp.could activate biofilm synthesis,produce more trehalose,accumulate betaine and electrolytes to stabilize intracellular structure under the two treatment conditions.A total of 31 and 43 genes were found related to desiccation and permeability resistance,respectively.And some genes(cysM,thuF,ycjO,etc.)were found to be associated with two tolerances for the first time.展开更多
Objective:To evaluate the predictive value of secreted phosphoprotein 1(SPP1)gene expression for postoperative survival in patients with advanced liver cancer undergoing hepatic artery interventional chemoembolization...Objective:To evaluate the predictive value of secreted phosphoprotein 1(SPP1)gene expression for postoperative survival in patients with advanced liver cancer undergoing hepatic artery interventional chemoembolization treatment.Method:Bioinformatics methods,including gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis,were used to identify genes related to survival prognosis in hepatocellular carcinoma(HCC)patients.A retrospective analysis of 115 advanced liver cancer patients treated between January 2016 and October 2017 was conducted.Patients were categorized into SPP1 high-expression(n=89)and low-expression groups(n=26).Additionally,115 healthy individuals served as the control group.The relationship between SPP1 expression and clinical pathological features was analyzed.A 60-month follow-up and logistic regression analysis identified risk factors affecting survival.Results:SPP1 mRNA expression was significantly higher in liver cancer patients compared to healthy controls(P<0.05).SPP1 expression levels were significantly associated with tumor size,Child-Pugh grading,lymph node metastasis,and BCLC staging(P<0.05).High SPP1 expression,along with tumor size,Child-Pugh grading,lymph node metastasis,and BCLC staging,were independent risk factors for survival(P<0.05).The 60-month survival rate was 17.39%,with a median survival of 40 months in the low-expression group versus 18 months in the high-expression group(P<0.05).Conclusion:SPP1 expression is significantly upregulated in advanced liver cancer patients and has predictive value for postoperative survival following hepatic artery chemoembolization treatment.SPP1,combined with clinical indicators such as tumor size,Child-Pugh grading,lymph node metastasis,and BCLC staging,may serve as a prognostic biomarker for interventional treatment outcomes.展开更多

[Method] Six strains or species of Penicilliurn and Trichoderma were inocu- lated into PDA plates growing with P. ostreatus to observe the growth of their mycelia. And the inhibitory effects of hydrogen peroxide, bromogeramine bromide, 84 disinfectant, lysol, potassium permanganate, and 75% medical alcohol on Trichoderma pleuroticola, Trichoderma viride, Trichoderma harzianum, Penicillium citrinum, Penicillium thomii and Penicillium brevicompactum were detected. [Result] Confrontation test revealed that Penicillium spp. and Trichoderma spp. competed for nutrients with P. ostreatus mycelia, antagonized against the growth of P. ostreatus mycelia, invaded and twined around P. ostreatus mycelia, and also produced toxins poisoning P. ostreatus mycelia. The six disinfectants exhibited different inhibitory effects against different species of Penicillium and Trichoderma. Among them the in- hibitory effects of bromogeramine bromide against the six species of Penicillium and Trichoderma were significantly different. However, the inhibitory effects of 3% hydrogen peroxide, 84 disinfectant, 5% lysol, potassium permanganate and 75% medical alcohol were significantly different among Penicillium spp., but did not among Tri- choderma spp. [Conclusion] Bromogeramine bromide can be used to control the bacteria and fungi in mushroom production for it has significant inhibitory effects on the six species of both Penicillium and Trichoderma.展开更多
Pleurotus spp.was one of the most precious and common edible fungi,polysaccharides were the main active component.The structural,biological activities and structure-activity relationship(SAR)of polysaccharides from Pl...Pleurotus spp.was one of the most precious and common edible fungi,polysaccharides were the main active component.The structural,biological activities and structure-activity relationship(SAR)of polysaccharides from Pleurotus spp.were systematically reviewed.On the basis of structure and biological activity,structure-activity relationships were also analyzed and discussed to look forward to its future research direction and application prospect.In the past 5 years,about 30 kinds of polysaccharides were isolated from Pleurotus spp.,and their preliminary structures were studied,but the fine structures seldomly reported.The polysaccharides showed the activities of immunomodulatory,hypoglycemic,anti-inflammatory,antioxidant,anti-ageing,hepatoprotective,anti-tumor,hypolipidemic and regulating intestinal flora,but the mechanism was needed further study.There were few studies on the SAR of polysaccharides from Pleurotus,and the polysaccharides of P.eryngii were studied.It was found that the biological activities of polysaccharides were affected by molecular weight,monosaccharide composition,sugar chain structure and configuration.In addition,sulfonation and selenization could significantly increase the bioactivities of polysaccharides.These findings might help to better understand the research status of polysaccharides from Pleurotus spp.and provided a scientific basis for their application as functional foods.展开更多
Oyster mushrooms of genus Pleurotus are well known as valuable edible mushrooms which are broadly cultivated in the world as well as wildly propagated in forests of the temperate climate zones including Russia. Indoor...Oyster mushrooms of genus Pleurotus are well known as valuable edible mushrooms which are broadly cultivated in the world as well as wildly propagated in forests of the temperate climate zones including Russia. Indoor cultivation conditions adapted for a laboratory with non-specialized facilities were suggested for effective solid-state cultivation and fruit bodies’ production of Pleurotus species. Seven of 9 Pleurotus species of different origin produced fruit bodies successfully. The best fructification was shown for species P. sajor-caju, P. ostreatus, and P. pulmonarius with fruit body yields of 74%, 72%, and 61% by dry substrate weight in the first flushing cycle. Fruit bodies appeared in five to 10-day flushes. “Spawn run” stage was completed within 17 to 24 days, the longest colonization stage (26 days) being for the pink oyster, P. djamor. The cultivation parameters proposed in this study can be employed with ease for laboratory and “home” cultivation of oyster mushrooms. The species and strains’ identification was confirmed by restriction analysis of ITS region of rRNA gene cluster. Molecular barcodes based on restriction enzyme (AluI, and BsuRI) profiles of ITS sequences were shown to be applicable for molecular genotyping of Pleurotus species of different origin.展开更多
BACKGROUND Neuropathic pain(NP)is the primary symptom of various neurological condi-tions.Patients with NP often experience mood disorders,particularly depression and anxiety,that can severely affect their normal live...BACKGROUND Neuropathic pain(NP)is the primary symptom of various neurological condi-tions.Patients with NP often experience mood disorders,particularly depression and anxiety,that can severely affect their normal lives.Microglial cells are as-sociated with NP.Excessive inflammatory responses,especially the secretion of large amounts of pro-inflammatory cytokines,ultimately lead to neuroinflam-mation.Microglial pyroptosis is a newly discovered form of inflammatory cell death associated with immune responses and inflammation-related diseases of the central nervous system.METHODS Two models,an in vitro lipopolysaccharide(LPS)-stimulated microglial cell model and a selective nerve injury model using BTX-A and SPP1 knockdown treatments,were used.Key proteins in the pyroptosis signaling pathway,NLRP3-GSDMD,were assessed using western blotting,real-time quantitative polymerase chain reaction,and immunofluorescence.Inflammatory factors[interleukin(IL)-6,IL-1β,and tumor necrosis factor(TNF)-α]were assessed using enzyme-linked immuno-sorbent assay.We also evaluated microglial cell proliferation and apoptosis.Furthermore,we measured pain sensation by assessing the delayed hind paw withdrawal latency using thermal stimulation.RESULTS The expression levels of ACS and GSDMD-N and the mRNA expression of TNF-α,IL-6,and IL-1βwere enhanced in LPS-treated microglia.Furthermore,SPP1 expression was also induced in LPS-treated microglia.Notably,BTX-A inhibited SPP1 mRNA and protein expression in the LPS-treated microglia.Additionally,depletion of SPP1 or BTX-A inhibited cell viability and induced apoptosis in LPS-treated microglia,whereas co-treatment with BTX-A enhanced the effect of SPP1 short hairpin(sh)RNA in LPS-treated microglia.Finally,SPP1 depletion or BTX-A treatment reduced the levels of GSDMD-N,NLPRP3,and ASC and suppressed the production of inflammatory factors.CONCLUSION Notably,BTX-A therapy and SPP1 shRNA enhance microglial proliferation and apoptosis and inhibit microglial death.It improves pain perception and inhibits microglial activation in rats with selective nerve pain.展开更多

The leaves are rich in proteins, vitamins and essential amino acids. Molecular characterization of Jute mallow with focus on improvement of leaf yield is scarcely reported. In the present study, inter sequence simple repeats (ISSR) molecular markers were employed to assess genetic diversity and relationships of 83 accessions of Jute mallow from different parts of Africa and Asia conserved at the World Vegetable Center East and Southern Africa. A total of 89 bands were amplified by 8 ISSR primers. Number of polymorphic bands per primer ranged from 2 to 6 with an average of 2.75 bands per primer. Polymorphic information content (PIC) values ranged from 0.390 to 0.760 with average of 0.53. Average Nei’s gene diversity (h) and Shannon’s information index (I) were 0.335 and 0.494 respectively. The highest pairwise genetic distance was 0.431 observed in a population from East Africa accessions. PC1 and PC2 axis explained 21.69% and 11.66% of the total variation respectively. UPGMA cluster analysis grouped the accessions into six main clusters at genetic similarity coefficient of 0.53 as standard value for classification. These results have important implications for jute mallow breeding and conservation.展开更多

However, the increasing global development of antibiotic resi...Urinary tract infections (UTIs) caused by uropathogens are a significant public health problem, and their treatment primarily relies on antibiotic therapy. However, the increasing global development of antibiotic resistance necessitates updating diagnostic techniques to ensure higher sensitivity and specificity, especially with advancements in science and medicine. This study aimed to evaluate the prevalence of UTIs and antibiotic resistance profiles through urine culture, as well as to identify Klebsiella pneumoniae, Klebsiella oxytoca, and Acinetobacter spp. in urine samples using a molecular approach with multiplex real-time PCR. From May 3 to July 25, 2023, at the Pietro Annigoni Biomolecular Research Center (CERBA) and Saint Camille Hospital of Ouagadougou (HOSCO), 209 urine samples collected from patients with suspected UTIs were analyzed using both urine culture and multiplex real-time PCR. Among the 209 patients, 52.15% were male and 47.85% female, with an average age of 46.87 ± 21.33 years. Urine cultures revealed an overall UTI prevalence of 23.44%, with a prevalence of 8.13% in men versus 15.31% in women (P = 0.023). The bacterial prevalence rates were as follows: Escherichia coli (12.92%), Klebsiella spp. (7.18%), Enterobacter cloacae (1.44%), Staphylococcus aureus (0.96%), and other bacteria. Klebsiella spp. demonstrated 100% resistance to Amoxicillin and Amoxicillin/Clavulanic Acid, while Escherichia coli showed 96.2% and 65.4% resistance to Amoxicillin and Amoxicillin/Clavulanic Acid, respectively. PCR analysis of the target bacteria revealed mono-infection prevalence rates of Klebsiella pneumoniae (10.39%), Klebsiella oxytoca (7.79%), and Acinetobacter spp. (7.79%), along with a co-infection prevalence rate of Klebsiella pneumoniae/Acinetobacter spp. (1.30%). This study demonstrated that PCR, with its high sensitivity and specificity, could effectively distinguish Klebsiella pneumoniae from Klebsiella oxytoca and detect Acinetobacter spp. in less than 24 hours—something urine culture alone could not achieve. The relative ease of automating urine PCR testing, combined with its diagnostic accuracy and rapid turnaround time, makes it a valuable addition to modern medical practice for the laboratory diagnosis of UTIs.展开更多
Understan ding the mechanism of harmful algal bloom formation is vital for effectively preventing algal bloom outbreaks in coastal environments.Karenia spp.blooms in the East China Sea show a significant correlation w...Understan ding the mechanism of harmful algal bloom formation is vital for effectively preventing algal bloom outbreaks in coastal environments.Karenia spp.blooms in the East China Sea show a significant correlation with nutrient regimes.However,the impact of key components of nutrients,especially dissolved organic nitrogen(DON),on the blooms of Karenia spp.is not clear.Quantitative research is still lacking.In this study,the cruise observations,field mesocosm-flask culture experiments,and a multinitrogen-tri-phytoplankton-detritus model(NTPD) are combined to reveal the quantitative influence of nutrient regimes on the shift of Prorocentrum donghaiense and Karenia spp.in the East China Sea.It has a synchronism rhythm of diatom-P.donghaienseKarenia spp.-diatom loop in the field culture experiment,which is consistent with the results of the cruise observation.The results showed that the processes of terrigenous DON(TeDON) and dissolved inorganic nitrogen(DIN:NO_(3)^(-)-N,NH_(4)^(+)-N) absorption promoted P.donghaiense to become the dominant algae in the community;whereas the processes of DON from P.donghaiense absorption promoted Karenia spp.to become the dominant algae in ambient DIN exhaustion.In addition,the three-dimensional fluorescence components of humus C,tyrosine and fulvic acid can indicate the processes of growth and extinction of P.donghaiense and Karenia spp.,respectively.This study infers that P.donghaiense and Karenia spp.regime shift mechanism associated with the nutrient regime in coastal waters,which provides a scientific basis for the environmental management of coastal eco system health.展开更多

To have an inventory of its cultivation in Cameroon, a survey study was carried out among eight farms of Fragaria spp. from January 2021 to February 2022. The plant was introduced in Cameroon in 2018. There are 13 varieties of Fragaria spp. currently cultivated. Among these 13 varieties, eleven are hybrids of Fragaria x ananassa (“Amiga”, “Amine”, “Camarosa”, “Chandler”, “Charlotte”, “Elsanta”, “Gariguette”, “Madame Moutot”, “Ostara”, “Ruby gem” and “San Andreas”), and two of the hybrids of Fragaria vesca (“Maestro” and “Mara des bois”). The cropping system, irrigation system, and type of fertilizers applied differ from one strawberry farm to another. Biofertilizers (such as mycorrhizal), inorganic and organic fertilizers are actually used to improve production. The potential annual production of strawberries from January 2021 to February 2022, estimated based on the survey data, was 21.216 tons for all growers. Among these eight production farms, the Lolodorf BIO Farm presents 6000 kg (six tons) of strawberries and 100,000 stolons (seedlings) produced, from seven varieties of Fragaria spp. cultivated, with 6 varieties which are hybrids variety Fragaria x ananassa (“Amiga”, “Amine”, “Chandler”, “Gariguette”, “Madame Moutot”, and “Ruby gem”), and one which is a hybrid of Fragaria vesca (“Mara des bois”). Certain diseases were also observed and recorded depending on the growing areas.展开更多

文摘The oyster mushroom(Pleurotus spp.)is one of the most widely cultivated mushroom species globally.The present study investigated the effect of synbiotics on the growth and quality of Pleurotus ostreatus and Pleurotus pulmonarius.Different synbiotics formulations were applied by spraying mushroom samples daily and measuring their growth parameters,yield,biological efficiency,proximate composition,mineral content,total phenolic content(TPC),and diphenyl-1-picryl-hydrazyl(DPPH)radical scavenging activity.Results demonstrated that the most significant yield of oyster mushrooms was harvested from synbiotics sprayed with inulin and Lactobacillus casei(56.92 g).Likewise,the highest biological efficiency obtained with a similar synbiotic was 12.65%.Combining inulin and L.casei was the most effective method of improving the mushrooms'growth performance and nutrient content in both samples.Furthermore,synbiotics that combined inulin and L.casei resulted in the highest TPC(20.550 mg gallic acid equivalent(GAE)/g dry extract(DE))in white oyster mushrooms(P.ostreatus).In comparison,in grey mushroom(P.pulmonarius)the highest TPC was yielded by L.casei(1.098 mg GAE/g DE)followed by inulin and L.casei(1.079 mg GAE/g DE).The DPPH results indicated that the oyster mushroom could be an efficient antioxidant.The results revealed that applying synbiotics improved the mushrooms'quality by increasing their antioxidant capacity with higher amounts of phenolic compounds and offering better health benefits with the increased levels of mineral elements.Together,these studies demonstrated the potential of using synbiotics as a biofertilizer,which is helpful for mushroom cultivation;therefore,it might solve the challenge of inconsistent quality mushroom growers face.
文摘Rationale: The contamination of soil with crude oil poses significant environmental and ecological threats. Bioremediation, particularly through the use of organisms like Pleurotus ostreatus (mushroom) and Eisenia fetida (earthworm), has emerged as a promising approach to mitigate crude oil pollution. Understanding the effectiveness of these organisms in reducing hydrocarbon levels in contaminated soil is crucial for devising sustainable remediation strategies. Objectives: This study aimed to evaluate the efficacy of Pleurotus ostreatus and Eisenia fetida in remediating crude oil-polluted soil. Specifically, it sought to assess the hydrocarbon profiles in soil treated with these organisms across varying concentrations of crude oil pollution. Method: Crude oil concentration levels ranging from 0% to 10% were applied to soil samples alongside control treatments, including soil only, soil with earthworms, and soil with mushrooms. Each treatment was replicated five times using a randomized complete block design. Standard methods were employed to determine the hydrocarbon contents of the soil. Results: The results indicated a significant increase (P Pleurotus ostreatus and Eisenia fetida exhibited noteworthy reductions in these hydrocarbon levels. At the three-month mark, mushrooms demonstrated a remarkable ability to reduce hydrocarbon content by 70% - 90% compared to the pollution treatment. In contrast, earthworms exhibited minimal potential for hydrocarbon reduction, particularly at both three and six-month intervals. For instance, TOC reduction reached a maximum of 96% with mushroom treatment and 85% with earthworm treatment at 5% crude oil pollution over six months. Conclusion: The findings highlight the effectiveness of Pleurotus ostreatus in significantly reducing hydrocarbon levels in crude oil-polluted soil compared to Eisenia fetida. Mushroom-treated soils consistently exhibited substantial reductions in TOC, TPH, TOG, PAH, and THC over the study period, suggesting their potential as a viable bioremediation agent. In contrast, while earthworms showed some capability in reducing hydrocarbon content, their effectiveness was comparatively limited. Recommendation: Based on the results, it is recommended to utilize Pleurotus ostreatus for the bioremediation of crude oil-polluted soils. Further research could explore optimizing remediation protocols involving mushroom-based treatments for enhanced efficiency. Statement of Significance: This study contributes valuable insights into the application of bioremediation techniques for mitigating crude oil contamination in soil. The demonstrated efficacy of Pleurotus ostreatus underscores its potential as a sustainable and eco-friendly solution for remediating hydrocarbon-polluted environments, offering a promising avenue for environmental restoration and conservation efforts.

文摘Blocking the development of edible mushrooms will affect the production cycle and yield of fruiting bodies.Phenylalanine ammonia lyase(PAL,EC 4.3.1.24.)is an enzyme that catalyzes the deamination of phenylalanine to form trans-cinnamic acid.Previous studies have shown that a decrease in pal1 gene transcription delays fruiting body development in Pleurotus ostreatus.Herein,we used wild type(WT)and RNA interference(RNAi)strains to study the molecular regulation of pal1 by RNA sequencing and Agrobacterium-mediated genetic transformation.Our results showed that interference with the pal1 gene resulted in reductions in the total PAL enzyme activity and the total phenol content,as well as an increase in the intracellular H_(2)O_(2)content.RNA-Seq data demonstrated that the significantly enriched KEGG terms were mainly related to the peroxisome pathway,MAPK signaling pathway-yeast and three other pathways,and the catalase(CAT)gene cat1 is also involved in multiple pathways that were enriched above.Exogenous H_(2)O_(2)significantly enhanced the transcription of the cat1 gene and elevated total CAT enzymatic activity.Moreover,the levels of cat1 gene transcription and the total CAT enzymatic activity in the RNAi-pal1 strains gradually become closer to those in the WT strain through the removal of H_(2)O_(2),which indicated that pal1 regulated the expression of cat1 by affecting the intracellular H_(2)O_(2)content.Finally,the overexpression of the cat1 gene in P.ostreatus caused growth retardation,especially during the process of primordia formation.In conclusion,this study demonstrated that PAL1 affects cat1 gene expression through the signaling molecule H_(2)O_(2)and regulates the development of P.ostreatus.The findings of this study enhance our understanding of the molecular developmental mechanism of edible mushrooms.

文摘Cronobacter spp.has strong resistance to desiccation and high permeability in Enterobacteriaceae,and powdered infant formula(PIF)is one of the main contamination routes.In recent years,the contamination of Cronobacter spp.in PIF incidents occurs from time to time,causing infant serious diseases or death.In this investigation,matrix-assisted laser desorption/ionization time of flight mass spectrometry was used to identify the phenotypes of 35 Cronobacter strains isolated from PIF and its processing environment.Subsequently,the isolates were evaluated for drying and osmotic pressure tolerance.The results showed that the deactivation rate of the strains ranged from 9.01%to 77.57%,and the highest osmotic pressure condition the strains could tolerate was 6 g/100 mL Na Cl.In addition,there was a positive correlation between biofilm formation ability and desiccation resistance.Combined with transcriptomics,Cronobacter spp.could activate biofilm synthesis,produce more trehalose,accumulate betaine and electrolytes to stabilize intracellular structure under the two treatment conditions.A total of 31 and 43 genes were found related to desiccation and permeability resistance,respectively.And some genes(cysM,thuF,ycjO,etc.)were found to be associated with two tolerances for the first time.

文摘Objective:To evaluate the predictive value of secreted phosphoprotein 1(SPP1)gene expression for postoperative survival in patients with advanced liver cancer undergoing hepatic artery interventional chemoembolization treatment.Method:Bioinformatics methods,including gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis,were used to identify genes related to survival prognosis in hepatocellular carcinoma(HCC)patients.A retrospective analysis of 115 advanced liver cancer patients treated between January 2016 and October 2017 was conducted.Patients were categorized into SPP1 high-expression(n=89)and low-expression groups(n=26).Additionally,115 healthy individuals served as the control group.The relationship between SPP1 expression and clinical pathological features was analyzed.A 60-month follow-up and logistic regression analysis identified risk factors affecting survival.Results:SPP1 mRNA expression was significantly higher in liver cancer patients compared to healthy controls(P<0.05).SPP1 expression levels were significantly associated with tumor size,Child-Pugh grading,lymph node metastasis,and BCLC staging(P<0.05).High SPP1 expression,along with tumor size,Child-Pugh grading,lymph node metastasis,and BCLC staging,were independent risk factors for survival(P<0.05).The 60-month survival rate was 17.39%,with a median survival of 40 months in the low-expression group versus 18 months in the high-expression group(P<0.05).Conclusion:SPP1 expression is significantly upregulated in advanced liver cancer patients and has predictive value for postoperative survival following hepatic artery chemoembolization treatment.SPP1,combined with clinical indicators such as tumor size,Child-Pugh grading,lymph node metastasis,and BCLC staging,may serve as a prognostic biomarker for interventional treatment outcomes.
文摘[Objective] This study was conducted to investigate the effects of Penicillium spp. and Trichoderma spp. on the growth of Pleurotus ostreatus (oyster mushroom) and to screen effective disinfectants to control them and other pathogenic microbes. [Method] Six strains or species of Penicilliurn and Trichoderma were inocu- lated into PDA plates growing with P. ostreatus to observe the growth of their mycelia. And the inhibitory effects of hydrogen peroxide, bromogeramine bromide, 84 disinfectant, lysol, potassium permanganate, and 75% medical alcohol on Trichoderma pleuroticola, Trichoderma viride, Trichoderma harzianum, Penicillium citrinum, Penicillium thomii and Penicillium brevicompactum were detected. [Result] Confrontation test revealed that Penicillium spp. and Trichoderma spp. competed for nutrients with P. ostreatus mycelia, antagonized against the growth of P. ostreatus mycelia, invaded and twined around P. ostreatus mycelia, and also produced toxins poisoning P. ostreatus mycelia. The six disinfectants exhibited different inhibitory effects against different species of Penicillium and Trichoderma. Among them the in- hibitory effects of bromogeramine bromide against the six species of Penicillium and Trichoderma were significantly different. However, the inhibitory effects of 3% hydrogen peroxide, 84 disinfectant, 5% lysol, potassium permanganate and 75% medical alcohol were significantly different among Penicillium spp., but did not among Tri- choderma spp. [Conclusion] Bromogeramine bromide can be used to control the bacteria and fungi in mushroom production for it has significant inhibitory effects on the six species of both Penicillium and Trichoderma.

文摘Pleurotus spp.was one of the most precious and common edible fungi,polysaccharides were the main active component.The structural,biological activities and structure-activity relationship(SAR)of polysaccharides from Pleurotus spp.were systematically reviewed.On the basis of structure and biological activity,structure-activity relationships were also analyzed and discussed to look forward to its future research direction and application prospect.In the past 5 years,about 30 kinds of polysaccharides were isolated from Pleurotus spp.,and their preliminary structures were studied,but the fine structures seldomly reported.The polysaccharides showed the activities of immunomodulatory,hypoglycemic,anti-inflammatory,antioxidant,anti-ageing,hepatoprotective,anti-tumor,hypolipidemic and regulating intestinal flora,but the mechanism was needed further study.There were few studies on the SAR of polysaccharides from Pleurotus,and the polysaccharides of P.eryngii were studied.It was found that the biological activities of polysaccharides were affected by molecular weight,monosaccharide composition,sugar chain structure and configuration.In addition,sulfonation and selenization could significantly increase the bioactivities of polysaccharides.These findings might help to better understand the research status of polysaccharides from Pleurotus spp.and provided a scientific basis for their application as functional foods.
文摘Oyster mushrooms of genus Pleurotus are well known as valuable edible mushrooms which are broadly cultivated in the world as well as wildly propagated in forests of the temperate climate zones including Russia. Indoor cultivation conditions adapted for a laboratory with non-specialized facilities were suggested for effective solid-state cultivation and fruit bodies’ production of Pleurotus species. Seven of 9 Pleurotus species of different origin produced fruit bodies successfully. The best fructification was shown for species P. sajor-caju, P. ostreatus, and P. pulmonarius with fruit body yields of 74%, 72%, and 61% by dry substrate weight in the first flushing cycle. Fruit bodies appeared in five to 10-day flushes. “Spawn run” stage was completed within 17 to 24 days, the longest colonization stage (26 days) being for the pink oyster, P. djamor. The cultivation parameters proposed in this study can be employed with ease for laboratory and “home” cultivation of oyster mushrooms. The species and strains’ identification was confirmed by restriction analysis of ITS region of rRNA gene cluster. Molecular barcodes based on restriction enzyme (AluI, and BsuRI) profiles of ITS sequences were shown to be applicable for molecular genotyping of Pleurotus species of different origin.
文摘BACKGROUND Neuropathic pain(NP)is the primary symptom of various neurological condi-tions.Patients with NP often experience mood disorders,particularly depression and anxiety,that can severely affect their normal lives.Microglial cells are as-sociated with NP.Excessive inflammatory responses,especially the secretion of large amounts of pro-inflammatory cytokines,ultimately lead to neuroinflam-mation.Microglial pyroptosis is a newly discovered form of inflammatory cell death associated with immune responses and inflammation-related diseases of the central nervous system.METHODS Two models,an in vitro lipopolysaccharide(LPS)-stimulated microglial cell model and a selective nerve injury model using BTX-A and SPP1 knockdown treatments,were used.Key proteins in the pyroptosis signaling pathway,NLRP3-GSDMD,were assessed using western blotting,real-time quantitative polymerase chain reaction,and immunofluorescence.Inflammatory factors[interleukin(IL)-6,IL-1β,and tumor necrosis factor(TNF)-α]were assessed using enzyme-linked immuno-sorbent assay.We also evaluated microglial cell proliferation and apoptosis.Furthermore,we measured pain sensation by assessing the delayed hind paw withdrawal latency using thermal stimulation.RESULTS The expression levels of ACS and GSDMD-N and the mRNA expression of TNF-α,IL-6,and IL-1βwere enhanced in LPS-treated microglia.Furthermore,SPP1 expression was also induced in LPS-treated microglia.Notably,BTX-A inhibited SPP1 mRNA and protein expression in the LPS-treated microglia.Additionally,depletion of SPP1 or BTX-A inhibited cell viability and induced apoptosis in LPS-treated microglia,whereas co-treatment with BTX-A enhanced the effect of SPP1 short hairpin(sh)RNA in LPS-treated microglia.Finally,SPP1 depletion or BTX-A treatment reduced the levels of GSDMD-N,NLPRP3,and ASC and suppressed the production of inflammatory factors.CONCLUSION Notably,BTX-A therapy and SPP1 shRNA enhance microglial proliferation and apoptosis and inhibit microglial death.It improves pain perception and inhibits microglial activation in rats with selective nerve pain.
文摘Jute mallow is a nutritious leafy vegetable. The leaves are rich in proteins, vitamins and essential amino acids. Molecular characterization of Jute mallow with focus on improvement of leaf yield is scarcely reported. In the present study, inter sequence simple repeats (ISSR) molecular markers were employed to assess genetic diversity and relationships of 83 accessions of Jute mallow from different parts of Africa and Asia conserved at the World Vegetable Center East and Southern Africa. A total of 89 bands were amplified by 8 ISSR primers. Number of polymorphic bands per primer ranged from 2 to 6 with an average of 2.75 bands per primer. Polymorphic information content (PIC) values ranged from 0.390 to 0.760 with average of 0.53. Average Nei’s gene diversity (h) and Shannon’s information index (I) were 0.335 and 0.494 respectively. The highest pairwise genetic distance was 0.431 observed in a population from East Africa accessions. PC1 and PC2 axis explained 21.69% and 11.66% of the total variation respectively. UPGMA cluster analysis grouped the accessions into six main clusters at genetic similarity coefficient of 0.53 as standard value for classification. These results have important implications for jute mallow breeding and conservation.

文摘Natural polysaccharides named PEP-0.1-1,PEP-0-1 and PEP-0-2 from edible mushroom species Pleurotus eryngii were obtained in the present study.Results showed that molecular weights of these polysaccharides were 3235,2041 and 23933 Da,respectively.Further,structural characterization revealed that PEP-0.1-1 had a→4-α-D-Glcp-1→backbone and contained→4)-α-D-Glcp and→4)-β-D-Glcp reducing end groups.PEP-0-1 backbone contained→4-α-D-Glcp-1→and→6-α-3-O-Me-D-Galp-1→,and the side chains containedα-D-Glcp,β-D-Manp-1→andα-D-Glcp-3→.However,PEP-0-2 backbone consisted of→4-α-DGlcp-1→and→6-α-3-O-Me-D-Galp-(1→6)-α-D-Galp-1→while the side chains containedα-D-Glcp andβ-D-Manp-1→.Biological activity analysis was then carried out and found that all these polysaccharides could significantly suppress the relative mRNA expression of toll-like receptor 4,nitric oxide(NO),tumor necrosis factor-α,interleukin(IL)-1βand IL-6 in lipopolysaccharide(LPS)-induced inflammation of RAW264.7 cells,as well as the over secretion of the above cell cytokines.Moreover,Western blotting analysis revealed that all these purified fractions displayed significant inhibition effects on the expression of c-Jun N-terminal kinases protein induced by LPS in mitogen activated protein kinase pathway,along with the relieving on the inhibition effect of LPS on IκB-αprotein expression.In summary,the information generated by the present study could provide a theoretical basis for the exploration of novel healthy food materials from edible mushroom with antiinflammation activities.
文摘Urinary tract infections (UTIs) caused by uropathogens are a significant public health problem, and their treatment primarily relies on antibiotic therapy. However, the increasing global development of antibiotic resistance necessitates updating diagnostic techniques to ensure higher sensitivity and specificity, especially with advancements in science and medicine. This study aimed to evaluate the prevalence of UTIs and antibiotic resistance profiles through urine culture, as well as to identify Klebsiella pneumoniae, Klebsiella oxytoca, and Acinetobacter spp. in urine samples using a molecular approach with multiplex real-time PCR. From May 3 to July 25, 2023, at the Pietro Annigoni Biomolecular Research Center (CERBA) and Saint Camille Hospital of Ouagadougou (HOSCO), 209 urine samples collected from patients with suspected UTIs were analyzed using both urine culture and multiplex real-time PCR. Among the 209 patients, 52.15% were male and 47.85% female, with an average age of 46.87 ± 21.33 years. Urine cultures revealed an overall UTI prevalence of 23.44%, with a prevalence of 8.13% in men versus 15.31% in women (P = 0.023). The bacterial prevalence rates were as follows: Escherichia coli (12.92%), Klebsiella spp. (7.18%), Enterobacter cloacae (1.44%), Staphylococcus aureus (0.96%), and other bacteria. Klebsiella spp. demonstrated 100% resistance to Amoxicillin and Amoxicillin/Clavulanic Acid, while Escherichia coli showed 96.2% and 65.4% resistance to Amoxicillin and Amoxicillin/Clavulanic Acid, respectively. PCR analysis of the target bacteria revealed mono-infection prevalence rates of Klebsiella pneumoniae (10.39%), Klebsiella oxytoca (7.79%), and Acinetobacter spp. (7.79%), along with a co-infection prevalence rate of Klebsiella pneumoniae/Acinetobacter spp. (1.30%). This study demonstrated that PCR, with its high sensitivity and specificity, could effectively distinguish Klebsiella pneumoniae from Klebsiella oxytoca and detect Acinetobacter spp. in less than 24 hours—something urine culture alone could not achieve. The relative ease of automating urine PCR testing, combined with its diagnostic accuracy and rapid turnaround time, makes it a valuable addition to modern medical practice for the laboratory diagnosis of UTIs.

文摘Understan ding the mechanism of harmful algal bloom formation is vital for effectively preventing algal bloom outbreaks in coastal environments.Karenia spp.blooms in the East China Sea show a significant correlation with nutrient regimes.However,the impact of key components of nutrients,especially dissolved organic nitrogen(DON),on the blooms of Karenia spp.is not clear.Quantitative research is still lacking.In this study,the cruise observations,field mesocosm-flask culture experiments,and a multinitrogen-tri-phytoplankton-detritus model(NTPD) are combined to reveal the quantitative influence of nutrient regimes on the shift of Prorocentrum donghaiense and Karenia spp.in the East China Sea.It has a synchronism rhythm of diatom-P.donghaienseKarenia spp.-diatom loop in the field culture experiment,which is consistent with the results of the cruise observation.The results showed that the processes of terrigenous DON(TeDON) and dissolved inorganic nitrogen(DIN:NO_(3)^(-)-N,NH_(4)^(+)-N) absorption promoted P.donghaiense to become the dominant algae in the community;whereas the processes of DON from P.donghaiense absorption promoted Karenia spp.to become the dominant algae in ambient DIN exhaustion.In addition,the three-dimensional fluorescence components of humus C,tyrosine and fulvic acid can indicate the processes of growth and extinction of P.donghaiense and Karenia spp.,respectively.This study infers that P.donghaiense and Karenia spp.regime shift mechanism associated with the nutrient regime in coastal waters,which provides a scientific basis for the environmental management of coastal eco system health.
文摘Strawberry (Fragaria spp.) is one of the most important fruits classified as exotic fruits imported into Cameroon. To have an inventory of its cultivation in Cameroon, a survey study was carried out among eight farms of Fragaria spp. from January 2021 to February 2022. The plant was introduced in Cameroon in 2018. There are 13 varieties of Fragaria spp. currently cultivated. Among these 13 varieties, eleven are hybrids of Fragaria x ananassa (“Amiga”, “Amine”, “Camarosa”, “Chandler”, “Charlotte”, “Elsanta”, “Gariguette”, “Madame Moutot”, “Ostara”, “Ruby gem” and “San Andreas”), and two of the hybrids of Fragaria vesca (“Maestro” and “Mara des bois”). The cropping system, irrigation system, and type of fertilizers applied differ from one strawberry farm to another. Biofertilizers (such as mycorrhizal), inorganic and organic fertilizers are actually used to improve production. The potential annual production of strawberries from January 2021 to February 2022, estimated based on the survey data, was 21.216 tons for all growers. Among these eight production farms, the Lolodorf BIO Farm presents 6000 kg (six tons) of strawberries and 100,000 stolons (seedlings) produced, from seven varieties of Fragaria spp. cultivated, with 6 varieties which are hybrids variety Fragaria x ananassa (“Amiga”, “Amine”, “Chandler”, “Gariguette”, “Madame Moutot”, and “Ruby gem”), and one which is a hybrid of Fragaria vesca (“Mara des bois”). Certain diseases were also observed and recorded depending on the growing areas.
