摩腹法对IBS-D模型大鼠肠黏膜紧密连接功能及ZO-1的影响研究

目的验证摩腹法干预腹泻型肠易激综合征(irritable bowel syndrome with predominant diarrhea,IBS-D)模型大鼠效果,阐明腹部推拿对IBS-D模型大鼠肠黏膜紧密连接的影响。方法采用母婴分离法+慢性应激法制备IBS-D大鼠模型。将30只SD雄性... 目的验证摩腹法干预腹泻型肠易激综合征(irritable bowel syndrome with predominant diarrhea,IBS-D)模型大鼠效果,阐明腹部推拿对IBS-D模型大鼠肠黏膜紧密连接的影响。方法采用母婴分离法+慢性应激法制备IBS-D大鼠模型。将30只SD雄性大鼠,随机分为正常组、模型组、摩腹组,每组10只。采用粪便含水量测试、粪便性状评分评价干预效果。利用透射电镜观察结肠黏膜组织状态。应用Western-blot法检测大鼠结肠组织中ZO-1蛋白表达。结果(1)与模型组比较,摩腹组粪便含水率、粪便性状评分明显下降(P<0.05),接近正常组(P>0.05)。(2)透射电镜下观察,模型组肠上皮细胞胞浆内有空泡,线粒体大量减少,分布不均,少见完整细胞核,细胞形态大小不一,细胞间间隙变大,紧密连接结构遭到破坏,肠上皮微绒毛缺如;摩腹干预后,肠上皮细胞质空泡减少得到恢复,线粒体增多,结构清晰,细胞核结构可见,细胞间间隙变窄,紧密连接结构部分恢复,肠上皮微绒毛分布不均部分短缺。(3)与正常组相比,模型组ZO-1蛋白表达明显下调(P<0.05),当予以摩腹干预后,ZO-1蛋白表达明显上调(P<0.05)。结论摩腹法能够通过上调ZO-1蛋白表达对大鼠肠上皮紧密连接产生影响,进而对肠上皮紧密连接和肠上皮通透性产生影响,进而改善IBS-D大鼠肠道症状。 展开更多

BACKGROUND The most common causes of scrotal enlargement in patients include primary tumor of the scrotum,inflammation,hydrocele of the tunica vaginalis,and indirect inguinal hernia;scrotal enlargement caused by external tumors of the scrotum is rare.The patient had both a greater omentum tumor and an inguinal hernia,and the tumor protruded into the scrotum through the hernia sac,which is even rarer.Moreover,omental tumors are mostly metastatic,and primary omental fibroma is rare.CASE SUMMARY Here,we report a rare case of a 25-year-old young man with scrotal enlargement and pain for 3 months.Preoperative examination and multidisciplinary discu-ssions considered intra-abdominal tumor displacement and inguinal hernia,and intraoperative exploration confirmed that the greater omentum tumor protruded into the scrotum.Therefore,tumor resection and tension-free inguinal hernia repair were performed.The final diagnosis was benign fibroma of the greater omentum accompanied by an indirect inguinal hernia.CONCLUSION This unusual presentation of a common inguinal hernia disease illustrates the necessity of performing detailed history taking,physical examination,and imaging before surgery. 展开更多

【目的】分析COIL及BMPR-IB基因与产羔数关联性,探究多浪羊与湖羊(简称多湖杂交)后代产羔性状COIL及BMPR-IB基因的遗传效应。【方法】研究以多浪羊与湖羊级进杂交F_(1)代、F_(2)代、F_(3)代为研究对象,随机选取多湖杂交F_(1)代(228只)、... 【目的】分析COIL及BMPR-IB基因与产羔数关联性,探究多浪羊与湖羊(简称多湖杂交)后代产羔性状COIL及BMPR-IB基因的遗传效应。【方法】研究以多浪羊与湖羊级进杂交F_(1)代、F_(2)代、F_(3)代为研究对象,随机选取多湖杂交F_(1)代(228只)、F_(2)代(254只)、F_(3)代(252只),进行全血DNA提取,COIL基因利用扩增阻滞突变系统PCR(ARMS-PCR)技术基因分型、BMPR-IB基因利用限制性内切酶片段长度多态性技术(PCR-RFLP)基因分型后,与产羔数关联分析。【结果】在多湖杂交F_(1)代中,突变纯合GG型产羔数极显著高于突变杂合CG、野生CC型产羔数(P<0.01),突变杂合CG型产羔数极显著高于野生CC型产羔数(P<0.01);在多湖杂交F_(2)代中突变纯合GG型与突变杂合CG、野生CC型产羔数间均差异极显著(P<0.01);在多湖杂交F_(3)代中,突变纯合GG、突变杂合CG型产羔数极显著高于CC型(P<0.01),GG型产羔数极显著高于CG型(P<0.05)。在多湖杂交F_(1)、F_(2)、F_(3)代中均为突变纯合BB型产羔数极显著高于突变杂合B+型、野生++型产羔数(P<0.01),突变杂合B+型产羔数极显著高于野生++型产羔数(P<0.01)。【结论】COIL基因与BMPR-IB基因均对多湖杂交后代产羔数影响显著,但BMPR-IB基因在多湖杂交后代突变率较低,可能不适合作为筛选多湖杂交双羔群体的关键基因,COIL基因可以作为筛选多湖杂交高繁殖率的新候选基因。 展开更多

Electrochemical detection of 3-methyl-4-nitrophenol (MNP) in direct phenol oxidation occurs at high potentials and generally leads to progressive passivation of the electrochemical sensor. This study describes the use of a carbon fiber microelectrode modified with a tetrasulfonated nickel phthalocyanine complex for the detection of MNP at a lower potential than that of direct phenol oxidation. The MNP voltammogram showed the presence of an anodic peak at -0.11 V vs SCE, corresponding to the oxidation of the hydroxylamine group generated after the reduction of the nitro group. The effect of buffer pH on the peak current and SWV parameters such as frequency, scan increment, and pulse amplitude were studied and optimized to have better electrochemical response of the proposed sensor. With these optimal parameters, the calibration curve shows that the peak current varied linearly as a function of MNP concentration, leading to a limit of detection (LoD) of 1.1 μg/L. These results show an appreciable sensitivity of the sensor for detecting the MNP at relatively low potentials, making it possible to avoid passivation phenomena. 展开更多

基于FEM-IBIEM的河谷坝址地震动输入方法及地震动场模拟研究

地震动输入是大坝抗震安全分析的基础,然而目前地震动输入尚未合理考虑河谷散射效应对大坝动力响应的影响。将河谷场地总运动场分解为平坦半空间自由场和河谷散射场分别求解,采用间接边界积分方程法求解河谷场地散射场,将其与平坦半空... 地震动输入是大坝抗震安全分析的基础,然而目前地震动输入尚未合理考虑河谷散射效应对大坝动力响应的影响。将河谷场地总运动场分解为平坦半空间自由场和河谷散射场分别求解,采用间接边界积分方程法求解河谷场地散射场,将其与平坦半空间自由场叠加获得河谷场地总运动场(地震动场),通过封闭解验证了间接边界积分方程法的精度。进而,采用有限元法模拟河谷场地内部,同时将河谷地基截断边界处总运动场转换为等效地震输入荷载,建立了总运动场结合黏弹性人工边界的波动输入方法。应用基于有限元法-间接边界积分方程法建立地震波动输入方法,数值求解了梯形河谷场地地震动场,分析了自由场波动输入和总运动场波动输入下河谷动力响应的差异。结果表明:自由场波动输入下河谷关键位置位移峰值、波形与精确解存在较大偏差,水平和竖向位移最大误差分别达15.6%、29.3%;总运动场波动输入下位移峰值、波形与精确解拟合良好。该研究建立的波动输入方法计算精度高,合理考虑了河谷散射效应,为更准确地预测河谷上大坝动力响应提供了基础。 展开更多

针对镇域尺度甘肃文县城关镇山体-沉积河谷三维地震动模拟问题,利用边界元法求解无限域弹性波动问题时降维和自动满足弹性波无限远辐射的优势,将间接边界元法拓展到城关镇山间河谷场地三维地震动模拟。根据钻孔资料及高精度地形数据建... 针对镇域尺度甘肃文县城关镇山体-沉积河谷三维地震动模拟问题,利用边界元法求解无限域弹性波动问题时降维和自动满足弹性波无限远辐射的优势,将间接边界元法拓展到城关镇山间河谷场地三维地震动模拟。根据钻孔资料及高精度地形数据建立三维精细化模型,以平面P波和SV波作为输入,在频域内分析了城关镇山间河谷地形对地震动的放大效应。为提高计算效率,在计算耗时严重的散射波场构造和虚拟荷载求解部分采用OpenMP(Open Multi-Processing)并行编程,并通过与半解析解对比验证了方法的准确性。城关镇的模拟结果表明:(1)沉积河谷和山体均会放大地震动,但沉积河谷的放大效应更为明显,在P波入射下,沉积河谷和山体观测点的位移放大系数(地震动位移与入射波位移幅值的比值)最大值分别为19.64和3.66,SV波入射下则为14.13和3.48;(2)两侧山体对地震波的反射会加剧山间河谷的边缘效应,河谷边缘的地震动放大系数最大可达19.64;(3)沉积河谷地震动的空间分布随频率变化明显,随着频率的升高,强震区从河谷中心逐渐往边缘转变。研究可为此类场地的地震区划和抗震设防提供一定参考。 展开更多

The effect of a storage system on drying time and estimation of drying parameters of cocoa beans using an indirect solar dryer with a sensible heat energy storage system (stones which act as both absorber and heat storage media) is the main subject of this article. This dryer, which uses stones as storage material and is made of wood and plywood, was used to dry a quantity of 5 kg of fermented cocoa beans. The drying parameters for the drying curves and the drying efficiency of cocoa beans were established and studied. The drying curves were modelled from semi-empirical models. The results showed that the moisture of cocoa beans decreased from 60% to 7% in wet basis. With a solar collector and drying efficiency of 40% and 34%, respectively. And this with a maximum average difference temperature between the drying air and the ambient temperature of 13.25˚C day or night. The best concordances are obtained with R2 values of 0.9983, 0.9843, 0.9813 and 0.9837 respectively from the models of Hii, Jena and Das, Demir et al. and Alibas. 展开更多

In this editorial,the authors of this paper comment on the article by Bokov et al published in the recent issue of World Journal of Orthopedics.We reviewed a general overview of oblique lumbar interbody fusions(OLIF)and lateral lumbar interbody fusions(LLIF),their indications and complications as an increasingly popular minimally invasive technique to address several lumbar pathologies.This editorial thoroughly discusses and reviews the literature regarding factors affecting outcomes of indirect decompression utilized through OLIF and LLIF procedures.Several parameters play a critical role in patient outcomes including restoration of disc height,foraminal height,central canal squared,and foraminal area.The indirect decompression allows for unbuckling of the ligamentum flavum which can significantly decompress the neural elements as well as aid in reduction of spondylolisthesis.However,the authors further highlight the limitations of indirect decompression and factors that may predict unsuccessful outcomes including bony foraminal stenosis,severe central canal stenosis,and osteoporosis.As a result,failure of indirect decompression can lead to persistent pain,radiculopathy and unsatisfied patients.Spinal surgeons may be left to reimage patients and consider additional procedures with direct decompression. 展开更多

太阳能热发电换热管主要材料渗铝321钢的损伤会导致换热管的寿命缩短甚至断裂,因此必须进行损伤检测。采用声发射方法对渗铝321钢的损伤特性进行分析,实现对换热管性能的在线动态监测。通过采用声发射Ib值特征来表征渗铝321钢的损伤程度... 太阳能热发电换热管主要材料渗铝321钢的损伤会导致换热管的寿命缩短甚至断裂,因此必须进行损伤检测。采用声发射方法对渗铝321钢的损伤特性进行分析,实现对换热管性能的在线动态监测。通过采用声发射Ib值特征来表征渗铝321钢的损伤程度,并运用自组织映射(SOM)神经网络算法进行声发射特征参数聚类,以分析材料的损伤模式。结果表明,力学塑性阶段的声发射事件数量剧增,能量和振铃计数的峰值标志着试件的断裂。此外,在试件失效前,Ib值显著降低且密度变密集,表明Ib值的变化特征可以作为材料临界失效的预警信号。通过SOM算法对特征参数进行聚类分析得到4个簇及其对应的特征频率,并使用扫描电子显微镜(SEM)观察试件的断口形貌,得出4个簇分别对应于孔洞生长与汇合、微裂纹成核、宏观裂纹扩展和纤维状断裂4类损伤模式。这项研究旨在探索金属管材的损伤演化行为,并为管材的损伤分析和健康监测提供依据。 展开更多

Full-length nucleoproteins from Ebola and Marburg viruses were expressed as His-tagged recombinant proteins in Escherichia coli and nucleoprotein-based enzyme-linked immunosorbent assays(ELISAs) were established for the detection of antibodies specific to Ebola and Marburg viruses. The ELISAs were evaluated by testing antisera collected from rabbit immunized with Ebola and Marburg virus nucleoproteins. Although little cross-reactivity of antibodies was observed in antiEbola virus nucleoprotein rabbit antisera, the highest reactions to immunoglobulin G(Ig G) were uniformly detected against the nucleoprotein antigens of homologous viruses. We further evaluated the ELISA's ability to detect antibodies to Ebola and Marburg viruses using human sera samples collected from individuals passing through the Guangdong port of entry. With a threshold set at the mean plus three standard deviations of average optical densities of sera tested, the ELISA systems using these two recombinant nucleoproteins have good sensitivity and specificity. These results demonstrate the usefulness of ELISA for diagnostics as well as ecological and serosurvey studies of Ebola and Marburg virus infection. 展开更多

Porcine reproductive and respiratory syndrome is caused by the PRRS virus (PRRSV),which has six structural proteins (GP2,GP3,GP4,GP5,M and N). GP5 and N protein are important targets for serological detection by enzyme-linked immunosorbent assay (ELISA) and other methods. Toward this goal,we developed an indirect ELISA with recombinant GP5 antigens and this method was validated by comparison to the LSI PRRSV-Ab ELISA kit. The results indicated that the optimal concentration of coated recombinant antigen was 0.2 μg/well for a serum dilution of 1:40. The rate of agreement with the LSI PRRSV-Ab kit was 88.7% (266/300). These results support the potential use of recombinant GP5 as an antigen for indirect ELISA to detect PRRSV antibodies in pigs. 展开更多

基于平面波势函数,采用间接边界积分方程法(indirect boundary integral equation method, IBIEM)研究了双孔隙流体饱和介质中弹性波入射下二维孔洞的散射特性。推导得到了双孔隙介质中全空间二维线源动力格林函数,并给出了各散射波的... 基于平面波势函数,采用间接边界积分方程法(indirect boundary integral equation method, IBIEM)研究了双孔隙流体饱和介质中弹性波入射下二维孔洞的散射特性。推导得到了双孔隙介质中全空间二维线源动力格林函数,并给出了各散射波的位移场和应力场。在数值精度验证的基础上,以双孔隙二维饱和全空间中孔洞为例,解决了平面P、SV波入射下的地震波散射问题。数值结果表明:双重孔隙介质中的位移幅值、环向应力幅值、孔隙压力变化规律与不同入射波形,入射频率,孔隙率和边界排水条件密切相关,位移幅值在低频(无量纲频率η≤2)入射时出现峰值。环向应力幅值与干土条件相比更为复杂,基质孔压与裂缝孔压的存在增大了双重孔隙饱和介质的能量效应,总体震动趋势大于干土条件,环向应力放大可达62%。 展开更多

[Objective]The aim was to establish an indirect ELISA for detection on antibody of Hps. [Method] The optimal conditions of indirect ELISA were selected and determined based on heat-resistant serotype 4 and 5 of Hps; specific,repeating and sensitive tests were conducted and 200 serums were detected. [Result]The optimal conditions were as follows: coating concentration of antigen at 10 μg /ml,and coating for 2 h at 37 °C; PBST containing 20 g /L of skim milk powder as blocking fluid for 30 min; serum dilution at 1∶ 80; reaction time of antigen for 45 min; dilution of secondary antibody at 1∶ 12 000 and effecting for 30 min; color development reaction for 15 min. [Conclusion] The established indirect ELISA is good in specificity and repetitiveness with higher sensitivity than that of indirect hemagglutination test; the results of clinic samples ( negative /positive serums) were in consistent with those detected with foreign ELISA kits. The established method can be made use of in serum antibody of Hps de- tection and seroepidemiology study. 展开更多

A fast and indirect fluorescent antibody assay for the Vibrio alginolyticus and V. parahaemolyticus infecting the large yellow croaker has been developed. The specific antisera for the two strains of vibrio were prepared with New Zealand rabbit and the antiserum and cross-reactive efficacy was tested by coagulation in tube. It showed that the goat anti-rabbit IgG had been labeled by fluorescence isothiocyanate (FITC). The results showed that positive reactions were 100% for the large yellow croaker Pseudosciaena crocea with typical symptom of vibrio infection, while the positive reaction to the pathogen in healthy yellow croakers reached 40%, but seemed negative for aquaculture water. The results demonstrated that this fast and indirect fluorescent antibody assay can be used not only to test the vibrio pathogen in diseased yellow croaker but also in infected animals with no symptom. 展开更多

African swine fever(ASF),caused by the African swine fever virus(ASFV),is a devastating disease of domestic and wild pigs.There is no effective vaccine,and the control of the disease relies mainly on surveillance and early detection of infected pigs.Previously,serological assays,such as ELISA,have been developed mainly based on recombinant structural viral proteins of ASFV,including p72,p54,and p30.However,the antibodies against these proteins do not provide efficient protection against ASFV infection in pigs.Therefore,new serological assays that can be applied for clinical diagnosis and evaluating serological immune response in vaccinated pigs are still required.In this study,we expressed and purified a recombinant p B602 L protein.The purified p B602 L protein was then used as an antigen to develop an indirect ELISA assay.This assay has no cross-reaction with the anti-sera against the 15 most common pig pathogens in China,such as classical swine fever virus,pseudorabies virus,and porcine parvovirus.This assay and a commercial ELISA kit were then used to detect 60 field pig serum samples,including an unknown number of antiASFV sera.The coincidence of the two assays was 95%.Furthermore,the p B602 L-based ELISA was employed to test the antibody responses to the seven-gene-deleted ASFV strain HLJ/18-7 GD in pigs.The results showed that the antibody levels in all vaccinated pigs,starting from the 10 th day post-inoculation,have increased continuously during the observation period of 45 days.Our results indicate that this p B602 L-based indirect ELISA assay can be employed potentially in the field of ASFV diagnosis. 展开更多

本文利用六面顶压机,在5.7 GPa、1560~1600 K的压力温度条件下,分别采用ϕ15 mm和ϕ30 mm两种尺寸合成腔体,系统开展了Ib型金刚石单晶的晶体生长工作,并借助于有限元法对两种尺寸合成腔体的温度场分布进行了研究。首先,借助于有限元法分... 本文利用六面顶压机,在5.7 GPa、1560~1600 K的压力温度条件下,分别采用ϕ15 mm和ϕ30 mm两种尺寸合成腔体,系统开展了Ib型金刚石单晶的晶体生长工作,并借助于有限元法对两种尺寸合成腔体的温度场分布进行了研究。首先,借助于有限元法分别对两种尺寸合成腔体的触媒温度场分布进行了模拟。模拟结果揭示了ϕ30 mm合成腔体温度场的均匀性要明显优于ϕ15 mm合成腔体。其次,金刚石晶体生长实验结果表明,采用ϕ15 mm合成腔体很难实现质量超过1.2 ct(1 ct=0.2 g)的优质Ib型金刚石单晶的生长,而ϕ30 mm合成腔体更适合用于生长大尺寸优质Ib型金刚石单晶。再次,扫描电子显微镜表面形貌测试结果表明,合成腔体尺寸的扩大不会对Ib型金刚石单晶的表面结晶质量产生显著影响。最后,拉曼光谱的测试结果表明,除了多晶金刚石的结晶质量较差,本研究两种腔体合成的其他金刚石单晶测试样品均具有较好的结晶质量。本研究对宝石级金刚石单晶大尺寸合成腔体的设计、大尺寸金刚石单晶的生长,以及多晶种法金刚石单晶合成技术的完善均具有一定的学术参考价值。 展开更多

针对传统SURF算法(speeded up robust features)在拼接高分辨率无人机航拍图像时运行速度慢、特征匹配率低的特点,提出了一种基于IB-SURF(image block-SURF)技术的无人机图像拼接算法。结合无人机定位定姿系统(position and orientation... 针对传统SURF算法(speeded up robust features)在拼接高分辨率无人机航拍图像时运行速度慢、特征匹配率低的特点,提出了一种基于IB-SURF(image block-SURF)技术的无人机图像拼接算法。结合无人机定位定姿系统(position and orientation system,POS)求取图像重叠区域;构造掩模在无人机图像重叠区域检测特征点,减少特征提取时间;借助图像分块(image block,IB)的思想对图像划分网格,精简筛选特征点;引入Neighborhood-KNN(neighborhood-K nearest neighbors)进行特征点匹配,提高图像匹配效率。实验结果表明,IB-SURF算法有较快的运行速度和较高的特征匹配率,平均特征匹配率达到84.3%,特征匹配正确率超过95.1%,为图像高质量拼接提供了技术基础。 展开更多
