A simple,rapid and sensitive liquid chromatography-mass spectrometry(LC-MS)method was developed for the determination of salidroside in rat plasma and study of its pharmacokinetics after oral administration of suspension of Erzhi Wan and Fructus Ligustri lucidi into Wistar rats.Plasma sample of 200 μL was extracted with acetic ether-isopropanol(2∶1)and the extraction was performed on a Kromasil C18 column(150 mm×4.6 mm,5 μm)with the mobile phase of methanol-water(41∶59,v/v)within a run time of 6.0 min.The analyte was monitored with positive electrospray ionization(ESI)by selected ion monitoring(SIM)mode.The target ions were m/z 323.05 for salidroside and m/z 411.05 for internal standard(IS)geniposide.A good linear relationship was obtained over the range of 5.0-500.0 ng/mL and the lower limit of quantification was 5.0 ng/mL.The validated method was successfully applied to the pharmacokinetic study of salidroside in rat plasma after oral administration of suspension of Erzhi Wan and Fructus Ligustri lucidi. 展开更多

OBJECTIVE Erzhi pills is a clas⁃sic prescription of Chinese medicine originated from Fu Shou Jing Fang in Ming dynasty,with the effects of nourishing kidney-Yin and hemosta⁃sis,black hair,strengthening muscles and bones.As a classical prescription for nourishing kidney-Yin,Erzhi pills has been used to treat senile dementia in China for many years.Herein,our study aimed to investigate the protective effects of Erzhi pills in rat models of Alzheimer disease(AD)induced by ovariectomy as well as D-galactose and Aβ1-40 injection and to explore its potential mechanism.METHODS The model of AD rats was established by ovariectomy com⁃bined with D-galactose and Aβ1-40 injection.Ovariectomized rats were randomly divided into four groups:model group,estradiol valerate(0.80 mg·kg-1)group,Erzhi pills high(1.50 mg·kg-1)and low(0.75 mg·kg-1)doses group.In addition,rats of sham operation were selected as the sham operated group.Except for the sham oper⁃ated group,rats were injected intraperitoneally with D-galactose(100 mg·kg-1 per day,for 49 d)on the 8th day,then they were given intracerebro⁃ventricular injection of Aβ1-40(10μg per rat,1 g·L-1)on the 36th day,while the corresponding drugs were given by gastrointestinal administra⁃tion on the 22nd day.In our study,Morris water maze test was used to evaluate the learning and memory abilities,while ELISA kit was used to an⁃alyze serum estrogen level.The morphology of hippocampal neuron cells was observed by HE staining,and Nissl staining was utilized to ob⁃serve the Nissl body in cytoplasm.Then,the ex⁃pression of ERβpositive cells and hippocampal Aβ1-40 and p-Tau404 proteins was determined by immunohistochemistry.In order to further explore the molecular mechanism of Erzhi pills preven⁃tion and treatment of AD,proteomics was used to find potential targets and related pathways,Western blotting was used to verify the expres⁃sion of candidate differential protein.According to the results of proteomics in our experiment,Western blotting was used to detect the protein expression of PI3K,Akt,Bcl-2,Bcl-xl,Bad,14-3-3 and GSK3β.RESULTS The escape latency was significantly shortened,the number of crossing platform was increased,the neuron arrange⁃ment was more orderly and with less nuclear pycnosis in rats of Erzhi pills groups compared to the model group.In rats treated with Erzhi pills,the number of neurons,Nissl bodies,the estro⁃gen levels and ERβpositive cells were increased,while the number of Aβ1-40 and p-Tau404 positive cells was significantly decreased.Proteomics found that there were more than one hundred differentially expressed proteins of rats treated with Erzhi pills,which were involved in 48 signal⁃ing pathways.Among these proteins,five of them were involved in the PI3K/Akt signal path⁃way.As the down-stream protein of PI3K/Akt sig⁃naling pathway,the content of 14-3-3 protein was significantly increased.Western blotting analysis showed that the expression of p-GSK3β/GSK3βand Bad was decreased,while that of p-Akt/Akt,p-PI3K/PI3K,14-3-3,Bcl-xl and Bcl-2 was up-regulated in rats from the Erzhi pills groups compared with the model group.CON⁃CLUSION Erzhi pills can improve estrogen levels,alter proteomics expression of the hippocampus and activate PI3K/Akt pathway in AD rats,reduce Aβaggregation,inhibit the hyperphos⁃phorylation of Tau protein,maintain the morphol⁃ogy of hippocampal neurons and decrease the apoptosis of hippocampal neurons,thereby improving the learning and memory abilities of ovariectomized AD rats induced by D-galactose and Aβ1-40 injection.This study may provide an experimental basis for the clinical treatment of Erzhi pills. 展开更多

Objective:The aim of this study was to investigate the potential ability of medicated serum of Erzhi pills on growth,proliferation and inflammation of the human breast cancer cell line MCF-7 cells.Methods:3-month-old female Sprague–Dawley rats were used to prepared medicated serum,and randomly assigned into four groups:control group(distillated water),low doses(2.25g/kg/d),middle doses(4.5g/kg/d),high does(9.0g/kg/d).The growth curve was meas-ured by MTT method,the proliferation was examined by colony formation assay.Inflam-matory cytokines IL-1β,IL-18 and IL-6 was detected by Quantitative real time polymerase chain reaction(RT-qPCR).Western blot was used to examine the expression of NF-kB signal pathway.Results:The medicated serum of Erzhi pills could inhibit the growth and proliferation of breast cancer cell MCF-7,especially,in the group of middle doses and high does(P<0.05).The protein expression of IKKα,IκBαand p-NF-κB were significantly down regulated.As well as the mRNA expression levels of Inflammatory cytokine,such as IL-1β,IL-18 and IL-6,were decreased.Conclusion:Medicated serum of Erzhi pills played an anti-tumor role in breast cancer via inhibiting the growth and proliferation of MCF-7.Moreover,Erzhi pills could suppress in-flammatory responses from the NFκB signaling pathways,and reduced skeletal muscle pain caused by breast cancer endocrine therapy,and Erzhi pills may be a new target for the treatment of breast cancer. 展开更多

Clinical Analysis of Erzhi Mingmu Decoction in the Treatment of Non-exudative Age-related Macular Degeneration

OBJECTIVE: To study the effect of Erzhi Mingmu Decoction in the treatment of non-exudative age-related macular degeneration. METHODS: A total of 100 patients with non-exudative age-related macular degeneration admitted to our hospital from May 2017 to May 2018 were selected. They were randomly divided into the study group and the control group. The control group was treated with oral vitamin C and vitamin E while study group was treated with Erzhi Mingmu Decoction based on the same treatment of the control group. Both groups were continuously treated for 1 month, the visual improvement, fundus changes, and changes of serum VEGF and CRP levels were compared before and after treatment. RESULTS: The number of patients with visual improvement was higher in the study group than that in the control group(P < 0.05). The number of patients with fundus improvement in the study group was more than that of the control group(P < 0.05). Serum VEGF levels were decreased in the 2 groups after treatment, and they were decreased more in the study group than those in control group(all P < 0.05). Serum CRP levels were decreased in the 2 groups after treatment, and the serum CRP levels were decreased more in the study group than those in control group(all P < 0.05). CONCLUSION: The treatment of non-exudative age-related macular degeneration with Erzhimingmu Decoction can significantly improve the vision of patients, improve the condition of the fundus, and reduce the levels of serum VEGF and CRP, which is worthy of clinical promotion and application. 展开更多

Objective: To investigate the effects of Erzhi Pill (二至丸,,EZP) on nerve cell apoptosis iinn senescence model rats.Methods: The rats model of senescence was established by peritoneal D-galactose injection combined with thymusectomy.Forty SD rats were randomized into four groups,the normal control group,the senescence model group,the EZP treated group,and the vitamins treated group,10 in each group.The rats were made into senescence model except those in the normal group.In the same time of D-galactose injection,the rats were treated respectively with distilled water,EZP 4.32 g/kg,and vitamins E and C 0.06 g/kg daily for 6 weeks via intragastric infusion.The index of main viscera (as brain,testis,etc.),serum levels of superoxide dismutase (SOD) activity,and total anti-oxidation capacity (T-AOC) were measured after a 6-week treatment.Meanwhile,the cerebral cortex neuronal apoptosis proportion and mitochondrial membrane potential (MMP) were detected by flow cytometry.Results: Both EZP and vitamins E and C treatments showed effects on increasing testis index and serum level of T-AOC,reducing the percentage of neuronal apoptosis in the cerebral cortex,and elevating MMP in the aging rats model.Conclusions: EZP could inhibit the cerebral cortex neuron apoptosis and maintain the mitochondrial function in the senescent process of rats induced by peritoneal D-galactose injection combined with thymusectomy.It also shows antioxidation effect to some extents. 展开更多

Objective:Traditional Chinese medicine(TCM)has been widely used throughout China to prevent and cure diseases for thousands of years,and now it is a part of the integrative medicine field that is available in Western societies.To ensure the safety and quality of the herbal medicines that are a major part of the TCM tradition,they must be held to modern pharmaceutical standards.Erzhi pill(EZP)is a Chinese Pharmacopeia-listed herbal preparation that is used in the long-term clinical management of postmenopausal symptoms,osteoporosis and menstrual disorders.Until now,whether the drug release mechanism of EZP is in line with its intended TCM usage has not been studied.Methods:The release of specnuezhenide from three EZPs(self-made,Leiyunshang and Renhe)in simulated gastric fluid(SGF),acetate buffer(p H 4.5 buffer)and simulated intestinal fluid(SIF)was investigated in a dissolution test.The water uptake capacity and erosion extent of the three EZPs were investigated using swelling and erosion studies.The drug release mechanism was further assessed through statistical model fitting,using DDSolver software.Results:The release of specnuezhenide from all three EZPs in SGF was less than 50%within a 4 h period.However,over 70%of the specnuezhenide was released from each EZP in both p H 4.5 buffer and SIF in the same time.Analysis of the swelling and erosion behaviors and the drug release mechanism of the three EZPs confirmed that the release rate from EZP followed a sustained release profile,which was an interactive combination of swelling and erosion.Conclusion:This study showed that the release pattern from the pills was in line with the intended TCM use of EZP.TCM had not only theoretically considered sustained release from the pills,but also formulated them to achieve this release pattern.When establishing quality control standards for pills,the theoretical TCM usage and the actual release patterns need to be considered. 展开更多

基于网络药理学和分子对接探讨二至丸治疗脂溢性脱发的作用机制.通过TCMSP数据库检索和收集二至丸的药物成分信息,通过Disgenet,TTD,OMIM,Genecards,Drugbank等数据库收集脂溢性脱发相关疾病靶点,利用Swiss Target Prediction预测药物... 基于网络药理学和分子对接探讨二至丸治疗脂溢性脱发的作用机制.通过TCMSP数据库检索和收集二至丸的药物成分信息,通过Disgenet,TTD,OMIM,Genecards,Drugbank等数据库收集脂溢性脱发相关疾病靶点,利用Swiss Target Prediction预测药物活性成分靶点,并获取活性药物成分和疾病的共同靶点,运用String数据库、Cytoscape3.7.2软件构建PPI网络图,通过Metas⁃cape数据库进行GO生物注释和KEGG通路富集分析,并用Cytoscape3.7.2进行可视化,构建靶点和通路网络图.收集并筛选到二至丸活性成分15个、靶点170个,与脂溢性脱发相关的靶点533个,共同靶点44个,其中AKT1、VEGFA、IL6、TNF、PPARG、ERBB2、ESR1的相互作用最显著,其参与了RNA聚合酶Ⅱ启动子调控、癌症通路、脂质与动脉粥样硬化、糖尿病并发症中的AGE-RAGE信号通路、PI3K-Akt信号通路、IL-17信号通路、前列腺癌、膀胱癌、肿瘤坏死因子信号通路、癌症中的蛋白聚糖等147条信号通路.二至丸可以通过抑制体内炎症发生,改善毛囊细胞氧化应激反应,调节脂质和滋补肝肾、促进头皮局部血管新生等机制治疗脂溢性脱发,体现出了二至丸在治疗脂溢性脱发上的多成分、多靶点,多通路的药效特点. 展开更多

【目的】基于网络药理学、分子对接及细胞实验验证探讨二至丸治疗多发性骨髓瘤(MM)的潜在作用机制。【方法】应用网络药理学、分子对接筛选药物-疾病-靶标-通路关键指标。培养多发性骨髓瘤细胞株RPMI-8226,给予二至丸干预,采用膜联蛋白V... 【目的】基于网络药理学、分子对接及细胞实验验证探讨二至丸治疗多发性骨髓瘤(MM)的潜在作用机制。【方法】应用网络药理学、分子对接筛选药物-疾病-靶标-通路关键指标。培养多发性骨髓瘤细胞株RPMI-8226,给予二至丸干预,采用膜联蛋白V(Annexin V)-异硫氰酸荧光素(FITC)染色法检测细胞凋亡,Transwell实验测定细胞侵袭能力,实时定量聚合酶链反应(qRT-PCR)法检测蛋白激酶B(Akt)、细胞周期蛋白D1(CCND1)、糖原合酶激酶3β(GSK-3β)、c-MycmRNA表达,Western Blot法检测细胞Akt、磷酸化蛋白激酶B(p-Akt)、CCND1、c-Myc、GSK-3β、磷酸化糖原合酶激酶3β(p-GSK-3β)蛋白表达。【结果】获得二至丸14个有效成分。二至丸治疗MM涉及靶点142个,通路富集分析结果显示关键靶点可能主要集中在癌症通路、脂质与动脉粥样硬化等。分子对接结果显示,木樨草素和槲皮素与GSK-3β有较好的结合活性及稳定性。进一步细胞实验验证发现,与空白组比较,二至丸低、中、高剂量组细胞凋亡率显著升高,细胞侵袭数减少,GSK-3β的mRNA和蛋白水平显著升高,CCND1、Akt和c-Myc的mRNA和蛋白水平显著降低,呈剂量依赖性,其中,中、高剂量组差异均有统计学意义(P<0.05或P<0.01)。【结论】二至丸对MM的治疗作用可能是通过木犀草素和槲皮素等关键活性成分,促进GSK-3β表达,并抑制Akt/GSK-3β/CCND1/c-Myc信号通路来实现的。 展开更多

目的系统性评价二至天癸方改善高龄女性体外受精-胚胎移植(IVF-ET)结局的有效性和安全性,为高龄女性IVF-ET的临床用药提供循证医学依据。方法检索中国知网、万方、维普、PubMed、Web of Science、Cochrane Library数据库中有关二至天癸... 目的系统性评价二至天癸方改善高龄女性体外受精-胚胎移植(IVF-ET)结局的有效性和安全性,为高龄女性IVF-ET的临床用药提供循证医学依据。方法检索中国知网、万方、维普、PubMed、Web of Science、Cochrane Library数据库中有关二至天癸方改善高龄女性IVF-ET结局的随机对照试验,检索时间为建库至2022年5月1日。采用RevMan 5.3软件对纳入文献研究进行Meta分析。结果最终纳入符合标准的文献共9篇,共677例患者。Meta分析结果显示,二至天癸方显著改善了行IVF-ET高龄女性的临床妊娠率、优质胚胎率、获卵数,且显著减少了促性腺激素(Gn)使用剂量(P<0.05)。试验组的2PN受精率虽优于对照组,但差异无统计学意义(P=0.06)。结论二至天癸方在改善高龄女性IVF-ET结局方面效果良好,是高龄女性IVF-ET临床用药中较有前景的中药方剂。 展开更多

目的:探讨加味二至丸(MEP)对动脉粥样硬化(AS)巨噬细胞铁死亡的作用及机制。方法:(1)50只载脂蛋白E基因敲除(ApoE^(-/-))小鼠构建AS模型,随机分为模型组,辛伐他汀组,以及低、中、高剂量MEP组,每组10只,以10只同背景的C57BL/6J为空白对... 目的:探讨加味二至丸(MEP)对动脉粥样硬化(AS)巨噬细胞铁死亡的作用及机制。方法:(1)50只载脂蛋白E基因敲除(ApoE^(-/-))小鼠构建AS模型,随机分为模型组,辛伐他汀组,以及低、中、高剂量MEP组,每组10只,以10只同背景的C57BL/6J为空白对照组。按照分组干预10周后,HE染色观察小鼠主动脉窦病理学改变,RT-qPCR法检测各组小鼠主动脉组织DJ-1和谷胱甘肽过氧化物酶4(GPX4)的mRNA表达。(2)将40只SD大鼠随机分为空白对照组和MEP组,制备MEP含药血清。体外培养RAW264.7小鼠巨噬细胞,将细胞分为空白对照血清组、氧化型低密度脂蛋白(ox-LDL)诱导的模型组、铁死亡抑制剂(ferrostatin-1)组、MEP含药血清组和MEP含药血清+铁死亡诱导剂(erastin)组共5组。MTT实验检测不同浓度MEP含药血清对细胞活力的影响,油红O染色观察各组细胞脂质沉积,ELISA检测各组细胞活性氧(ROS)、丙二醛(MDA)、总超氧化物歧化酶(SOD)、还原型谷胱甘肽(GSH)及铁离子水平,透射电镜观察各组细胞线粒体结构和形态,RT-qPCR法和Western blot法分别检测检测各组细胞DJ-1、GPX4、溶质载体家族7成员11(SLC7A11)、铁蛋白重链1(FTH1)、p53、前列腺素-内过氧化物合酶2/环加氧酶2(PTGS2/COX2)和NADPH氧化酶1(NOX1)的mRNA和蛋白表达。结果:(1)动物实验结果提示,MEP可以减轻小鼠主动脉窦根部斑块形成,显著升高主动脉DJ-1和GPX4的mRNA表达。(2)细胞实验结果提示,ox-LDL诱导下的RAW264.7细胞脂质沉积显著增多,ROS、MDA和总SOD水平显著升高,GSH水平显著降低,p53、PTGS2/COX2和NOX1的mRNA及蛋白表达显著上调,DJ-1、GPX4、SLC7A11和FTH1的mRNA及蛋白表达显著下调;与ox-LDL组比较,MEP含药血清组细胞脂质沉积显著减少,ROS、MDA和总SOD水平显著降低,GSH水平显著升高,p53、PTGS2/COX2和NOX1的mRNA及蛋白表达显著下调,DJ-1、GPX4、SLC7A11和FTH1的mRNA及蛋白表达显著上调。结论:MEP可以抑制AS巨噬细胞铁死亡,从而延缓AS进展,其机制与DJ-1/GPX4信号通路有关。 展开更多

目的:探讨六味地黄丸合二至丸组方联合恩格列净片、缬沙坦片对早期糖尿病肾病(肝肾阴虚证)患者的疗效及对血糖指标、肾功能、血清单核细胞趋化蛋白-1(MCP-1)、转化生长因子-β1(TGF-β1)和炎症因子的影响。方法:选取2020年9月至2023年2... 目的:探讨六味地黄丸合二至丸组方联合恩格列净片、缬沙坦片对早期糖尿病肾病(肝肾阴虚证)患者的疗效及对血糖指标、肾功能、血清单核细胞趋化蛋白-1(MCP-1)、转化生长因子-β1(TGF-β1)和炎症因子的影响。方法:选取2020年9月至2023年2月该院收治的早期糖尿病肾病(肝肾阴虚证)患者128例,按随机数字表法分为对照组与观察组,各64例。两组患者均给予健康教育和饮食指导,在此基础上,对照组患者口服恩格列净片和缬沙坦片,观察组患者在对照组的基础上联合六味地黄丸合二至丸组方治疗,共治疗8周。评估临床疗效和不良反应发生情况,检测治疗前后两组患者的血糖指标[空腹血糖(FBG)、糖化血红蛋白(HbA_(1)c)]、肾功能指标[尿素氮(BUN)、血肌酐(SCr)和尿白蛋白排泄率(UAER)]、血清MCP-1、TGF-β1和炎症因子[肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)]水平。结果:观察组患者的总有效率为93.75%(60/64),较对照组(81.25%,52/64)更高,差异有统计学意义(P<0.05)。治疗后,两组患者的FBG、HbA_(1)c、BUN、SCr、UAER、MCP-1、TGF-β1、TNF-α和IL-1β水平均较治疗前显著降低,且观察组患者的FBG、HbA_(1)c、BUN、SCr、UAER、MCP-1、TGF-β1、TNF-α和IL-1β水平较对照组更低,差异均有统计学意义(P<0.05)。对照组、观察组患者的总不良反应发生率分别为12.50%(8/64)、14.06%(9/64),差异无统计学意义(P>0.05)。结论:六味地黄丸合二至丸组方联合恩格列净片、缬沙坦片可有效改善早期糖尿病肾病(肝肾阴虚证)患者的血糖、肾功能指标、MCP-1、TGF-β1及炎症因子水平,提高临床疗效,且安全性较好。 展开更多

过敏性紫癜性肾炎常反复发作,迁延难愈。中医认为本病与伏邪密切相关。笔者根据长期临证经验,基于六经辨证及伏邪理论,认为过敏性紫癜性肾炎病机要点在于邪伏厥阴,阴亏热伏,每感新邪易复发,并从厥阴经络、阴阳、脏腑三个方面论述过敏性... 过敏性紫癜性肾炎常反复发作,迁延难愈。中医认为本病与伏邪密切相关。笔者根据长期临证经验,基于六经辨证及伏邪理论,认为过敏性紫癜性肾炎病机要点在于邪伏厥阴,阴亏热伏,每感新邪易复发,并从厥阴经络、阴阳、脏腑三个方面论述过敏性紫癜性肾炎的病机特点:厥阴经络为邪之通路,厥阴“一阴”之性致邪伏,邪伏厥阴脏腑生风化火。厥阴伏邪成巢,易滞易瘀、易入难出、易积成形,易于发作。然少阳含生发之气,可托伏邪外出,引邪至此才有出路,故以祛邪扶正、化解透托为原则,补少阴之不足,清厥阴之邪,使邪出少阳而解为法。过敏煎为厥阴、少阳两经之方,可将邪托出少阳,边托边清邪,抽丝剥茧,直至托无所托。临床可以二至地黄汤合过敏煎为主方加减治疗,急性发作期,多因新感风热毒邪引触伏邪而发,可稍减滋阴之品,合升降散、银翘散等以增其清热散邪之力;临床缓解期,余毒瘀阻肾络,可稍加化瘀之品以止血,如茜草、紫草、赤芍、桃仁等;恢复期,应以扶正为要,病程日久,其气亦损,可合黄芪、党参、白术等补虚固表,以摄其血、御外风。临证病多复杂,常兼见他证,需灵活辨证加减。文末列验案一则,以期为临床过敏性紫癜性肾炎提供新的诊疗思路。 展开更多

二至丸改善自然衰老大鼠卵母细胞线粒体功能的机制研究

目的:研究二至丸对自然衰老大鼠卵母细胞线粒体改善的效应与机制。方法:选用SPF级SD雌性大鼠,随机分为8周龄青年对照组、20周龄老年对照组、20周龄二至丸用药低剂量组和高剂量组。用药30 d后,进行腹腔注射孕马血清促性腺激素(20 IU/只),... 目的:研究二至丸对自然衰老大鼠卵母细胞线粒体改善的效应与机制。方法:选用SPF级SD雌性大鼠,随机分为8周龄青年对照组、20周龄老年对照组、20周龄二至丸用药低剂量组和高剂量组。用药30 d后,进行腹腔注射孕马血清促性腺激素(20 IU/只),2 d后给予人绒毛膜促性腺激素(10 IU/只),干预12 h后观察卵母细胞腺苷三磷酸(ATP)、活性氧(ROS)含量及线粒体的数目、形态,检测线粒体动态调控蛋白过氧化物酶体增殖物激活受体γ辅助活化因子1α(PGC-1α)、视神经萎缩蛋白1(OPA1)、PTEN诱导蛋白激酶1(PINK1)的蛋白表达水平。结果:和青年对照组相比,老年对照组卵巢卵母细胞ATP的含量减少(P<0.05),细胞内线粒体数量下降,结构紊乱,部分呈空泡化,PGC-1α、OPA1及PINK1蛋白水平下降(P<0.05);二至丸低、高剂量组可提高老年大鼠卵母细胞ATP的浓度(P<0.05),增加细胞内线粒体含量、改善结构及减少ROS的生成(P<0.05),促进PGC-1α、OPA1和PINK1的表达(P<0.05)。结论:二至丸可以改善衰老卵巢内的卵母细胞线粒体的结构和功能,提高线粒体自我更新以达到修复自然衰老卵母细胞的作用。 展开更多

目的基于网络药理学方法探讨二至丸治疗早发性卵巢功能不全(POI)的可能分子作用机制。方法使用网络药理学平台(TCMSP)采集二至丸的有效成分和相关的基因;通过基因组注释数据平台(GeneCards)、人类孟德尔遗传数据库(OMIM)数据库获取POI... 目的基于网络药理学方法探讨二至丸治疗早发性卵巢功能不全(POI)的可能分子作用机制。方法使用网络药理学平台(TCMSP)采集二至丸的有效成分和相关的基因;通过基因组注释数据平台(GeneCards)、人类孟德尔遗传数据库(OMIM)数据库获取POI疾病靶点;用蛋白互作网络数据库(STRING)构建蛋白质相互作用网络(PPI),通过CytoHubba筛选核心靶点。使用生物学信息注释数据库(DAVID)平台进行富集分析,采用Cytoscape 3.10.0软件进行拓扑学分析,利用AutoDockTools进行分子对接。结果二至丸内最主要的有效成分为槲皮素、木犀草素、山柰酚、刺槐素等,其中的AKT1、TP53等是关键靶点,包含的通路主要为PI3K-Akt信号通路、流体剪切应力和动脉粥样硬化信号通路等。分子对接验证大部分靶点与成分能进行有效结合。结论揭示二至丸能够经多种成分、靶点及多条信号通路对POI发挥协同治疗作用。 展开更多

二至丸合丹参饮治疗阴虚血瘀型慢性胃炎合并老年睡眠障碍的临床研究

目的观察二至丸合丹参饮治疗阴虚血瘀型慢性胃炎合并老年睡眠障碍的临床疗效。方法选取2022年6月至2024年1月于淄博市中医医院诊断为慢性胃炎合并老年睡眠障碍属“阴虚血瘀”型的患者90例,按照随机数表法分为对照组和观察组,每组各45例... 目的观察二至丸合丹参饮治疗阴虚血瘀型慢性胃炎合并老年睡眠障碍的临床疗效。方法选取2022年6月至2024年1月于淄博市中医医院诊断为慢性胃炎合并老年睡眠障碍属“阴虚血瘀”型的患者90例,按照随机数表法分为对照组和观察组,每组各45例,对照组用“四联疗法”联合阿普唑仑治疗,观察组在对照组基础上加用二至丸合丹参饮汤药治疗,两组患者均治疗4周,观察两组患者治疗前后胃镜结果,幽门螺杆菌转阴率,匹兹堡睡眠质量指数量表(PSQI),失眠严重程度指数量表(ISI),中医证候评分的指标,并比较总有效率。结果治疗后,观察组总有效率为64.44%,高于对照组的31.11%,差异有统计学意义(P<0.05);治疗后两组患者胃镜结果,幽门螺杆菌阳性率、PSQI睡眠量表评分、ISI失眠严重程度量表评分、中医证候评分部分项目均较治疗前改善,差异有统计学意义(P<0.05),且观察组在各比较项目改善情况均优于同期对照组,差异有统计学意义(P<0.05)。结论二至丸合丹参饮治疗阴虚血瘀型慢性胃炎合并老年睡眠障碍疗效确切,改善患者慢性胃炎情况,提高老年睡眠障碍患者睡眠质量,值得进一步研究和应用。 展开更多
