CHO细胞高密度流加工艺参数优化

[目的]优化细胞接种密度、培养基、细胞培养温度等参数,提高生产细胞株PCSK9蛋白表达滴度。[方法]试验分四步进行,(1)探讨几款市售培养基优化组合后对CHO细胞株蛋白表达滴度的影响;(2)探讨10.0×10^(6)、15.0×10^(6)、50.0×10^(6)cells/mL高密度接种流加过程培养基、降温时间等对CHO细胞蛋白滴度的影响;(3)在(2)实验数据基础上继续优化,通过更换培养基继续探讨高密度流加工艺的可行性;(4)在反应器对实验数据进行工艺验证。[结果](1)CHO细胞PCSK9蛋白滴度提升至2.6 g/L,蛋白滴度成倍增长;(2)15.0×10^(6)cells/mL接种,30.0×10^(6)cells/mL降温至34℃,继续优化培养基1^(#)、2^(#)配比,蛋白滴度提升至4.5 g/L,反应器工艺验证,细胞生长状态稳定,蛋白滴度突破3.8 g/L;(3)继续筛选市售培养基,成功实现80.0×10^(6)cells/mL高密度流加接种,蛋白滴度提升至7.5 g/L。[结论]以贝谙基Eden-B600、Eden-400S培养基能够实现CHO细胞80.0×10^(6)cells/mL的高密度接种,流加培养D15蛋白滴度突破7.5 g/L。在细胞流加培养过程可以通过培养基成分优化、提高接种细胞密度等改善高密度流加工艺细胞株生长状态,提高目标蛋白产量。以国产培养基替换进口Sigma EX-CELL Advanced CHO Fed-Batch Medium极大地降低了生产成本。 展开更多

The electrolysis of water powered by renewable energy sources offers a promising method of"green hydrogen"production,which is considered to be at the heart of future carbon-neutral energy systems.In the past decades,researchers have reported a number of hydrogen evolution reaction(HER)electrocatalysts with activity comparable to that of commercial Pt/C,but most of them are tested within a small current density range,typically no more than 500 mA cm^(-2).To realize the industrial application of hydrogen production from water electrolysis,it is essential to develop high-efficiency HER electrocatalysts at high current density(HCD≥500 mA cm^(-2)).Nevertheless,it remains challenging and significant to rational design HCD electrocatalysts for HER.In this paper,the design strategy of HCD electrocatalysts is discussed,and some HCD electrocatalysts for HER are reviewed in seven categories(alloy,metal oxide,metal hydroxide,metal sulfide/selenide,metal nitride,metal phosphide and other derived electrocatalysts).At the end of this article,we also pro-pose some viewpoints and prospects for the future development and research directions of HCD electrocatalysts for HER. 展开更多

With the bluff internals, the extremely non-uniform radial profiles of solid fraction and particle velocity become flat and the dense downflow layer near the wall disappears, indicating the significant enhancement of solid turbulence introduced by the internals. The fluctuation velocity and solid fraction transient signal analysis indicates a significant increase in fluctuation intensity near the wall region. The length influenced by the internals on the flow structure is about 1 meter. The lateral gas dispersion coefficient increases significantly as the bluff internals exist in the riser. 展开更多

Electrochemical impedance spectroscopy (EIS) is widely used in fuel cell impedance analysis. However, for ohmic resistance (R Ω), EIS has some disadvantages such as long test period and complex data analysis with equivalent circuits. Therefore, the current interruption method is explored to measure the value of RΩ in direct methanol fuel cells (DMFC) at different temperatures and current densities. It is found that RΩ decreases as temperature increase, and decreases initially and then increases as current density increases. These results are consistent with those measured by the EIS technique. In most cases, the ohmic resistances with current interruption (R iR ) are larger than those with EIS (R EIS ), but the difference is small, in the range from –0.848% to 5.337%. The errors of R iR at high current densities are less than those of R EIS . Our results show that the R iR data are reliable and easy to obtain in the measurement of ohmic resistance in DMFC. 展开更多

A high-density, high-flux circulating fluidized bed (CFB) riser (100 mm in ID and 10.614 m in height) was ap- plied in a wide range of operating conditions (with solid fluxes up to 400 kg/m2s and superficial gas velocities up to 12 m/s) to examine its radial non-uniformity dynamics. The solids holdup was determined through the use of a fiber-optic probe at 11 axial levels. The results indicated that under all operating conditions, the high superficial gas velocity and low solid flux- es maintained a low radial non-uniformity index (RNI). The high-density/flux CFB riser had several unique characteristics, so that the peak of the radial solids holdup profile occurred at a position with r/R=0.8. The RNI and solids holdup at the cross-sectional position had a good logarithmic relationship at the low-density condition (with a mean solids holdup of 〈0.2), and the RNI decreased when the mean solids holdup exceeded 0.2. Investigation of the dynamics of stratified injec- tion revealed that the feed ratio had an important effect on G, and on solids holdup distribution. A novel "〈" shaped axial solids holdup profile was found. Gs decreased sharply when the up-flow feed ratio exceeded 0.5, and RNI was lowest when the up-flow feed ratio was 1. 展开更多

The hydrodynamic behavior in a high-density downer reactor was studied. A two-fluid model based on the kinetic theory of granular flow with a k-ε turbulent model was developed to simulate the flow behavior in the system. This simulation achieved an averaged solid fraction in the bed as high as 18% in this operating regime. The flow development in high-density downer consists of 3 regions, which are first acceleration, second acceleration, and fully developed regions. In the fully developed region, the lateral distribution of the solid volume fraction is low and almost uniform in the center region with a high density peak near the wall region. Gas and solid velocities gradually increase toward the wall and form a peak near the wall region. In addition, the solid volume fraction, gas and solid velocities increase with solid circulation rate. 展开更多

Knowledge on size distribution of bubbles in a flotation system is highly important. In this study, bubble distributions in different reagent concentrations, electrolyte concentrations, cathode apertures, and current densities in electroflotation are determined using a high-speed camera. Average bubble sizes under different conditions are calculated using Image-Pro@ Plus (Media Cybernetics@, MD, USA) and SigmaScan@ Pro (Systat Software, CA, USA) software. Results indicate that the average sizes of bubbles, which were generated through 38, 50, 74, 150, 250, 420, and 1000 μm cathode apertures, are 20.2, 29.5, 44.6, 59.2, 68.7, 78.5, and 88.8 μm, respectively. The optimal current density in electroflotation is 20 A/m2. Reagent and electrolyte concentrations, current density, and cathode aperture are important factors in controlling bubble size and nucleation. It causes serious damage and renders difficulty of water resource management in Taiwan. In this study, the quantity of wood debris and variation of migratory wood debris during flood events were examined. The downstream of Gaoshan Creek and Qijiawan Creek, located at Central Taiwan, was selected as the study area. The distribution and dynamic of wood debris in a high gradient headwater catchment were quantified using field surveys. A formula of critical depth for wood debris entrainment was used to evaluate the wood debris migration during three flooding events. In the study area, wood abundance and unit volume increased downstream, and wood density decreased downstream within a channel network. The channel morphology, riparian vegetation, and wood debris characteristics were found to influence the wood storage. As a result, the wood debris has an irregular accumulative distribution in the steep stream, and it migrates easily in the stream because of a high flow discharge. Strong relationships between the channel width and wood debris variables are discovered. Moreover, wood debris has a tendency to accumulate at sites with low stream power and wood debris dams, topographical notches, and unique geological structures. Our findings assist in the understanding of the effects of channel characteristics on distributions of wood debris in steep stream systems. 展开更多

In order to quickly explore the quality of cut-off wall in dams, a new method of high-density seismic image was adopted and estimated by model and in-situ wall tests.The vibration exciter was employed and several parameters such as hypocentral distance, length of signal record and sampling space in signal collection were determined, which are 8 m, 0.25 ms and 128 ms respectively. Through time and frequency field signal analyses, it is concluded that, the smaller arrival times of reflected longitudinal and surface waves, and the higher their main frequencies, the higher the strength of the wall, vice versa. Accordingly the construction quality of the wall can be evaluated quickly by high-density seismic image. 展开更多

GaN (gallium nitride) buck-rectifier has been proposed to realize high power density ISOP (input series and output parallel)-IPOS (input parallel and output series) converter-based dc distribution system. The ultra-low loss bi-directional switch can be developed by the GaN power device because of the low on-resistance, the high-speed switching behavior and its own device structure. The buck-rectifier using the GaN bi-directional switches has the potential to achieve higher power density than the commonly utilized boost-rectifier. Availability of the GaN-HEMT (high electron mobility transistor) for the buck rectifier has been verified taking the theoretical limit of the on-resistance and the switching loss energy into account. Design consideration for a high power density buck-rectifier has been also conducted and the application effect of the GaN bidirectional switches has been evaluated quantitatively. The ISOP-IPOS converter-based dc (direct current) distribution system takes full advantage of the buck-rectifier and the rectifier using GaN devices contributes to realizing higher power density dc distribution system. 展开更多
