棉花人工杂交制种方法评介

利用杂交棉是提高棉花单产的有效途径之一。生产杂交棉的方法很多,有些方法目前还不能大量采用。1980年我国从美国引进棉花不育系、保持系,恢复系,其中以恢复系存在的问题最大。

以海岛棉海1为供体亲本,中221、中棉所36(中36)两个陆地棉品种(系)为轮回亲本配制了两套杂交回交的组合:中36×海1组合和中221×海1组合。通过对这2套组合的不同世代群体杂交铃、自交铃纤维品质性状的分析,结果:以分离群体BC1F... 以海岛棉海1为供体亲本,中221、中棉所36(中36)两个陆地棉品种(系)为轮回亲本配制了两套杂交回交的组合:中36×海1组合和中221×海1组合。通过对这2套组合的不同世代群体杂交铃、自交铃纤维品质性状的分析,结果:以分离群体BC1F1为父本,轮回亲本中221、中36分别为母本时,母体植株上的杂交铃(BC2F0)的纤维品质性状与父本BC1F1群体植株上自交铃的纤维品质性状之间在平均值、极差和变异系数上存在显著差异,相关性不显著,而与母本(轮回亲本)值接近;又以分离群体BC2F1为母本,轮回亲本中221、中36分别为父本,母体植株上的杂交铃(BC3F0)纤维品质与母本BC2F1群体植株上自交铃的纤维品质性状在平均值、极差和变异系数上接近,除整齐度以外,纤维长度、比强度、麦克隆值和伸长率都达到了极显著的正相关。这些结果表明,杂交铃的纤维品质偏向母体植株上的自交铃的纤维品质,父本花粉的基因型对杂交当代种子上的纤维品质没有显著的影响,杂交铃种子上的纤维品质主要由当代种子的表皮细胞基因型即母体植株基因型决定,而当代种子胚的基因型作用不明显。 展开更多

Measuring fitness is essential for understanding the potential impacts of hybridisation between transgenic crops and their wild relatives. Transgenic CrylAc cotton Gossypium hirsutum and G. barbadense hybridise, the latter being valued as backyard cotton and an important genetic resource. G. barbadense was outcrossed to G. hirsutum containing the gene CrylAc and the isoline and the offspring evaluated for fitness as a measure of resistance to common pests and other growth parameters, Resistance to Pectinophora gossypiella and Alabama argillacea were greater in transgenic plants. Number of seeds per plant was higher in the F1 hybrids and G. hirsutum, F2 populations were intermediary; G. barbadense produced the least seeds per plant during the period of analysis. Anthesis of the first flower and opening of the first boll occurred earlier for G. hirsutum and Ft hybrids than F2; latest for G. barbadense. The observed-performance of resistance to the worms in the hybrids suggests that among FI and F2 populations transgenic hybrids may exhibit a competitive advantage over non-transgenic plants as a result of gene flow. 展开更多

Pepino mosaic virus (PepMV), monopartite RNA virus, 6,500 pb, belonging to Flexiviridae and Potexvirus group, is highly infectious and easily transmissible. Its economic impact is major for the tomato producer's countries. Prevention, based on early virus detection is the only effective control measure. Monoclonal antibodies appeared to be very useful tool. The authors used for the production of monoclonal antibodies hybridomas technique, by fusing spleen cells of immunized BALB/c mice to PepMV and SP2/O cancerous cells. The aim of this work is to produce hybridomas producers of Mab that could be used for ELISA in Morocco. At the same time, these efforts will serve to decrease expenses of producers concerning phytosanitory control. We obtained 16 hybridomas lines producers of Mab specific for PepMV. They were tested for efficiencies in ELISA and two lines were retained for production of Mab on large scale (1B 11-G 10 and 5A l-G5). Isotyping of these two lines showed that they are belonging to IgG 1 class and easily purified by affinity chromatography in agarose column by protein A. The conjugation of these two antibodies to alkaline phosphatase has been verified by DAS-ELISA. These antibodies will enable to diagnose the disease from infected tomato plants, integrating several serological tests to control it and target the actions of struggles. 展开更多
