小酸模入侵对人工草地植被、放牧黑山羊生长性能、瘤胃发酵参数以及血清指标的影响

试验旨在探究小酸模入侵对人工草地主要植被、放牧黑山羊生长性能、瘤胃发酵参数以及血清指标的影响。在贵州省威宁县选择4块小酸模入侵程度不同的人工草地,分析不同草地主要植被密度和高度;挑选体重(22.69±1.43)kg的黑山羊公羔羊... 试验旨在探究小酸模入侵对人工草地主要植被、放牧黑山羊生长性能、瘤胃发酵参数以及血清指标的影响。在贵州省威宁县选择4块小酸模入侵程度不同的人工草地,分析不同草地主要植被密度和高度;挑选体重(22.69±1.43)kg的黑山羊公羔羊120只,按完全随机区组设计将其分为4组,每组3个重复,每个重复10只羊。各组黑山羊自然放牧3个月后,分析其瘤胃发酵参数和血清指标。结果显示:随着人工草地小酸模密度的增加,鸭茅和白三叶的密度和高度逐渐降低。试验Ⅱ组自然放牧黑山羊的平均日增重显著高于Ⅳ组(P<0.05)。试验Ⅳ组黑山羊瘤胃总挥发性脂肪酸的含量显著低于试验Ⅱ组和Ⅲ组(P<0.05);试验Ⅰ组黑山羊瘤胃的丁酸含量最高,乙酸、丙酸含量低于其他3组。试验Ⅳ组黑山羊血清谷丙转氨酶(ALT)和谷草转氨酶(AST)含量最高,生长激素(GH)含量最低。试验Ⅳ组黑山羊血清免疫球蛋白G(IgG)、免疫球蛋白M(IgM)含量显著低于其他试验组(P<0.05),肿瘤坏死因子-α(TNF-α)和白细胞介素-2(IL-2)含量高于其他3组。试验Ⅰ组黑山羊血清的谷胱甘肽过氧化物酶(GSH-Px)活性显著低于其他3组(P<0.05),丙二醛(MDA)含量显著高于其他3组(P<0.05)。研究表明,当小酸模密度达到560株/m^(2)时,降低了放牧黑山羊瘤胃发酵功能、免疫力及生长性能。 展开更多

小酸模RumexAcetosella为廖科Polygonaceae酸模属Rumex多年生草本植物 ,适应于温暖、半干旱至潮湿的高海拔山区 ,目前在云南滇东北的昭通地区已经成为重要的害草 ,为害面积 1 67万hm2 之多。为了研究小酸模侵占规律 ,2 0 0 3年在昭通... 小酸模RumexAcetosella为廖科Polygonaceae酸模属Rumex多年生草本植物 ,适应于温暖、半干旱至潮湿的高海拔山区 ,目前在云南滇东北的昭通地区已经成为重要的害草 ,为害面积 1 67万hm2 之多。为了研究小酸模侵占规律 ,2 0 0 3年在昭通彝良分别对弃荒地、未耕草地和农田上的小酸模危害情况进行了调查和分析。结果表明 ,入侵农田后 ,小酸模个体生长势旺盛 ,平均株高较其他样地高 1 0cm之多。小酸模侵占弃荒后的样地 ,株高明显降低 ;随着弃荒年限的增加甚至不耕作 ,小酸模生物量逐渐减少 ,生长势明显削弱。种群密度分析结果表明 ,耕地最易于小酸模的繁殖 ,土地耕作次数越多 ,小酸模的有性繁殖能力越强 ,一个稳定植物群落可以有效地抑制小酸模的有性繁殖。在小酸模危害严重的地区 ,尽量减少土地的耕作 ,建立多年生人工草地易于形成稳定的植物群落 ,可以有效的抑制小酸模生长、发育及其繁殖 。 展开更多

AIM: To ascertain the molecule mechanism of nuclear factor-KB (NF-κB) inhibitor curcumin preventive and therapeutic effects in rats' colitis induced by trinitrobenzene sulfonic acid (TNBS). METHODS: Sixty rats with TNBS-induced colitis were treated with 2.0% curcumin in the diet. Thirty positive control rats were treated with 0.5% sulfasalazine (SASP). Thirty negative control rats and thirty model rats were treated with general diet. Changes of body weight together with histological scores were evaluated. Survival rates were also evaluated. Cell nuclear NF-κB activity in colonic mucosa was evaluated by using electrophoretic mobility shift assay. Cytoplasmic IκB protein in colonic mucosa was detected by using Western Blot analysis. Cytokine messenger expression in colonic tissue was assessed by using semiquantitative reverse-transcription polymerase chain reaction. RESULTS: Treatment with curcumin could prevent and treat both wasting and histopathologic signs of rats with TNBS-induced intestinal inflammation. In accordance with these findings, NF-κB activation in colonic mucosa was suppressed in the curcumin-treated groups. Degradations of cytoplasmic IκB protein in colonic mucosa were blocked by curcumin treatment. Proinflammatory cytokine messenger RNA expression in colonic mucosa was also suppressed. CONCLUSION: This study shows that NF-κB inhibitor curcumin could prevent and improve experimental colitis in murine model with inflammatory bowel disease (IBD). The findings suggest that NF-κB inhibitor curcumin could be a potential target for the patients with IBD. 展开更多

Objective To investigate the expression of phosphatase and tension homolog(PTEN) in adipose tissue of KKAy diabetic mice, a mouse model of type 2 diabetes. Methods KKAy diabetic mice were fed with high fat diet for 4 weeks. After blood glucose met the criteria of diabetes(over 16.7 mmol/L), mice were randomly divided into 3 groups: a control group(without any treatment), a rosiglitazone group(treated with rosiglitazone 12.5 mg/kg·d once per day), and a metformin group(treated with metformin 3 g/kg·d twice daily). After 4 weeks, we then determined the expression of PTEN and phosphoserine 473-Akt(pS473-Akt) in the epididymal adipose tissue with Western blots. The mice in each group were further divided into the insulin(-) subgroup and insulin(+) subgroup, which were intraperitoneally injected with saline and insulin(5 mU/g body weight), respectively. Results The expression of PTEN was elevated in the epididymal adipose tissue obtained from KKAy diabetic mice compared with that from the C57BL/6J mice(P<0.001). In accordance with the enhanced expression of PTEN, the level of pS473-Akt stimulated by insulin was decreased in the adipose tissue of KKAy mice compared to the C57BL/6J mice(P<0.001). Treatment with the insulin-sensitizing agents, rosiglitazone and metformin did not inhibit the elevated expression of PTEN in adipose tissue of KKAy diabetic mice. Conclusion PTEN may play an important role in the development of insulin resistance in adipose tissue of type 2 diabetes mice model. 展开更多

Methods Temporal lobe epilepsy model was established by stereotaxically unilateral and intrahip-pocampal injection of kainite acid (KA) in adult male C57BL/6 mice. Latent and chronic epileptogenesis were represented by mice 5 days after KA injection (n=5) and mice 5 weeks after KA injection (n=8), respectively. Control mice (n=5) were injected with saline. Immunohistochemical assays were performed on brain sections of the mice. Results Hippocampus both ipsilateral and contralateral to the KA injection displayed significantly up-regulated pS6 immunoreactivity in dispersed granule cells in 5-day and 5-week model mice. Conclusion The activation of p70S6k is mainly located in the dentate gyrus in KA-induced mouse model of temporal lobe epilepsy, indicating that the activation may be related with the disperse degree and hypertrophy of granule cells. 展开更多

The ingestion of probiotic lactic acid bacteria has been evaluated and noted that it has an effect on the balance of desirable microbiota in the gastrointestinal tract. Lactobacillus gasseri demonstrates good survival in the gastrointestinal tract, and it has been associated with a variety of probiotic activities and roles, including the reduction of fecal mutagenic enzymes, the production of bacteriocins and the stimulation of macrophages immunomodulation. The aim of the study was to evaluate the effects of a pool of L. gasseri strains isolated from the feces of breastfed infants added in the human milk of healthy women. The milk was both pasteurized and unpasteurized, to verify the cell cytotoxicity of macrophages and to quantify the production of immunologic mediators such as IL-4, IL-6, IFN-γ, TNF-α, NO and oxygen intermediary compounds (H2O2). The administration of raw human milk and pasteurized human milk to infants is a regular, encouraged practice in units of intensive therapy (UITs) and our present investigation verified the beneficial effect of addition of a pool of L. gasseri to pasteurized human milk (PHML). Our results show that probiotic supplementation helped to maintain cell viability, reduced IL-6 and IFN-γ production and stimulated TNF-α, NO, H2O2, IL-4 production. Nevertheless, the results indicate that the addition of lactobacillus to human milk was not a determinant in the production of TNF-α. L. gasseri added to breast milk did not present a cytotoxic risk, and the addition ofL. gasseri to pasteurized milk of human milk bank would benefit newborns that depend on milk banks for the colonization of more desirable microbiota. 展开更多

Genomic distribution of the nucleosome, the basic unit of chromatin, contains important epigenetic information. To map nucleosome distribution in structurally and functionally differentiated micronucleus(MIC) and macronucleus(MAC) of the ciliate Tetrahymena thermophila, we have purified MIC and MAC and performed micrococcal nuclease(MNase) digestion as well as hydroxyl radical cleavage. Different factors that may affect MNase digestion were examined, to optimize mono-nucleosome production. Mono-nucleosome purity was further improved by ultracentrifugation in a sucrose gradient. As MNase concentration increased, nucleosomal DNA sizes in MIC and MAC converged on 147 bp, as expected for the nucleosome core particle. Both MNase digestion and hydroxyl radical cleavage consistently showed a nucleosome repeat length of^200 bp in MAC of Tetrahymena, supporting ~50 bp of linker DNA. Our work has systematically tested methods currently available for mapping nucleosome distribution in Tetrahymena, and provided a solid foundation for future epigenetic studies in this ciliated model organism. 展开更多
