Based on its morphology, physiology, biochemical analysis and phylogenetic characteristics, the isolate was identified as Diaphorobacter sp. PD-7. Biodegradation tests of phenol showed that the maximum phenol degradation occurred at the late phase of exponential growth stages, with 1400 mg·L^-1 phenol completely degraded within 85 h. Diaphorobacter sp. PD-7 accumulated a vast quantity of phenol hydroxylase in this physiological phase, ensuring that the cells quickly utilize phenol as a sole carbon and energy source. The kinetic behavior ofDiaphorobacter sp. PD-7 in batch cultures was investigated over a wide range of initial phenol concentrations (0-1400mg·L^-1) by using the Haldane model, which adequately describes the dynamic behavior of phenol biodegradation by strain Diaphombacter sp. PD-7. At initial phenol concentration of 1400mg· L^-l, batch experiments (0.25 L flask) of nitrogen removal under aerobic condition gave almost entirely removal of 120.69mg· L^- 1 ammonium nitrogen within 75 h, while nitrate nitrogen removal reached 91% within 65 h. Moreover, hydroxylamine oxidase, periplasmic nitrate reductase and nitrite reductase were successfully expressed in the isolate.展开更多

To examine the adsorption-desorption characteristics of HCB, equilibrium batch experiments were carried out using two soils (red soil and paddy soil) with different initial HCB concentrations (0.25, 0.50, 0.75, 1.00, 1.50, 2.50, 3.50, and 5.00 mg L-1 ) by using 0.01 mol L-1 calcium chloride as the background solution. The successive desorption experiments (48, 96, 144, 192, and 240 h) were conducted after each adsorption equilibrium experiment. The results revealed that adsorption and desorption isotherms of HCB on two soils were nonlinear, which can be best described by the Freundlich equation with the square of the correlation coefficient (r 2 ) ranging from 0.97 to 0.99. Desorption of HCB from the two soils exhibited hysteresis at all HCB concentrations because the Freundlich desorption coefficients were always higher than the Freundlich adsorption coefficients. The hysteretic effect was enhanced with increasing initial HCB concentration, and positive hysteresis was observed at different concentrations.展开更多

In addition, changes in the initial concentration of CO2 used in the closed- box model were made by adjusting the CO2 supply system, and a CD-7 multi-function parameter instrument was employed to monitor and record the changes of O2 and CO2 concentration in the closed-box model. Results indicate that the oxygen generation rate of KO2 oxygen plates, the oxygen generation efficiency, and the carbon dioxide absorption rate decrease when there is an increase in the pressure used to mold the oxygen plates, but those values increase when the initial CO2 concentration increases. When the initial concentration of CO2 in the dosed-box model is 3.5% and the forming pressure is 10 kN, the average oxygen generation rate of 15 g KO2 oxygen plate is 11.88 ×10^-3 L/min, the oxygen generation efficiency is 80.3%, and the average CO2 absorption rate is 11.0 × 10^-3 L/min. Compared with the condition where the initial CO2 concentration is 1.5%, the results show that average oxygen genera- tion rate of oxygen plates increases by 88,9%, the oxygen generation efficiency increases by 88.9%, and the CO2 absorption rate increases by 100%.展开更多

Experiments were conducted at various operating parameters. The operating parameters were amount of catalyst (50 rag, 100 mg and 150 mg), initial concentration of dye (30 mg/L, 50 mg/L and 70 mg/L), the pH of solution (2, 4, 6, 8, l0 and 12) and the UV light intensity (6 watt and 12 watt). In this study, lipase production of the crude enzymatic extracts obtained by Aspergillus niger using cassava peel as substrate and sunflower oil as an inductor was investigated. The optimized cultivation temperature and concentration of inductor were determined using the response surface methodology. The two variables studied exercised influence in the production of lipase in the 95% level of confidence. The response surface obtained indicated that the conditions that maximize lipase activity production were 30.5 ~C and initial concentration of sunflower oil was 2.5% (w/w). Through this analysis, it is evident that extremes in temperature and concentration of inductor tend to decrease lipase production, since low temperatures decrease metabolism and high temperatures may inactivate the lipase. Optimum lipase yield was 59.8 U/g of dry peel which was fermented for 60 h. Lipase production presents a peak of 61.3 U/g, at 72 h of fermentation. However, this value is statistically equal (p 〉 0.05) of the value of lipase activity obtained for 60 h and 84 h of fermentation.展开更多

Among nitrophenols, p-nitrophenol (PNP) is a priority pollutant that has been widely used as pesticide. PNP is a toxic compound that enters the environment during manufacturing and processing of a variety of industrial products. This situation generalized its presence in multiple natural ecosystems: rivers, wastewaters, subterranean waters, soil treated by pesticides and urban atmosphere. This study aims to test the ability of wastewater microflora to degrade high concentration of PNP (500 rag/L) aerobically. An identification of the dominant microorganisms involved in the biodegradation is also carried. The cultures are performed using a minimum medium, where PNP is the sole source of carbon, energy and nitrogen. The kinetic of biodegradation is followed for more than 30 days of incubation at 30 ~C on a shaker (150 tours/min). The obtained results show that more than 90% of PNP initial concentration is decomposed at the end of incubation. The isolation of microorganisms degrading PNP gave two bacterial colonies with different macroscopic aspects. Sequence analysis of 16S ribosomal DNA indicated that the PNP degrading isolates were closely related to members of the species: Pseudomonas aeruginosa and Bacillus cereus.展开更多

However, this value is statistically equal (p 〉 0.05) of the value of lipase activity obtained for 60 h and 84 h of fermentation.
文摘Many nitrophenols tend to persist in the environment and they may become public health hazards. Sequence analysis of 16S ribosomal DNA indicated that the PNP degrading isolates were closely related to members of the species: Pseudomonas aeruginosa and Bacillus cereus.
