AIM:To further investigate the important role of store-operated calcium channels (SOCs) in rat hepatocytes and to explore the effects of SOC blockers on hepatic ischemia-reperfusion injury (HIRI).METHODS:Using freshly...AIM:To further investigate the important role of store-operated calcium channels (SOCs) in rat hepatocytes and to explore the effects of SOC blockers on hepatic ischemia-reperfusion injury (HIRI).METHODS:Using freshly isolated hepatocytes from a rat model of HIRI (and controls),we measured cyto-solic free Ca 2+ concentration (by calcium imaging),net Ca 2+ fluxes (by a non-invasive micro-test technique),the SOC current (I SOC ;by whole-cell patch-clamp record-ing),and taurocholate secretion [by high-performance liquid chromatography and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assays].RESULTS:Ca 2+ oscillations and net Ca 2+ fluxes medi-ated by Ca 2+ entry via SOCs were observed in rat he-patocytes.I SOC was significantly higher in HIRI groups than in controls (57.0 ± 7.5 pA vs 31.6 ± 2.7 pA,P <0.05) and was inhibited by La 3+.Taurocholate secretion by hepatocytes into culture supernatant was distinctly lower in HIRI hepatocytes than in controls,an effect reversed by SOC blockers.CONCLUSION:SOCs are pivotal in HIRI.SOC blockers protected against HIRI and assisted the recovery of se-cretory function in hepatocytes.Thus,they are likely to become a novel class of effective drugs for prevention or therapy of HIRI patients in the future.展开更多

However, evidence in support of the biological mechanisms of this effect is still limited. Findings from both rodent and human studies suggest brain-derived neu- rotrophic factor (BDNF) as a potential mechanism of the effect of acute exercise on memory. The molecular properties of BDNF allow this protein to be assessed in the periphery (pBDNF) (i.e., blood serum, blood plasma), making measurements of acute exercise-induced changes in BDNF concentration relatively accessible. Studies exploring the acute exercise--pBDNF--cognitive performance relationship have had mixed findings, but this may be more reflective of methodological differences between studies than it is a statement about the role of BDNE For example, significant associations have been observed between acute exercise-induced changes in pBDNF concentration and cognitive performance in studies assessing memory, and non-significant associations have been found in studies assessing non-memory cognitive domains. Three suggestions are made for future research aimed at understanding the role of BDNF as a biological mechanism of this relationship: 1) Assessments of cognitive performance may benefit from a focus on various types of memory (e.g., relational, spatial, long-term); 2) More finegrained measurements of pBDNF will allow for the assessment of concentrations of specific isoforms of the BDNF protein (i.e., immature, mature); 3) Statistical techniques designed to test the mediating role of pBDNF in the acute exercise-cognitive performance relationship should be utilized in order to make causal inferences.展开更多

As a core part of circuit breakers, the operating mechanisms have a trend to be hydraulic-style in high voltage power grid. Compared with other hydranlic systems, the hydraulic operating mechanisms have the characteristics of high hydraulic pressure, high speed, high power and long-term waiting etc., and it is because of the characteristics that the hydraulic operating mechanisms become dif- ficult to be developed and have been arousing significant study interest from more and more researchers as well as their promising applications. Therefore, it is significant to summarize the hydraulic operating mechanisms on their development, characteristics, and key technologies etc. In this review, the evolution process and recent studies of hydraulic operating mechanisms at home and abroad are viewed. The review then focuses on the characteristics and key technologies of hydraulic operating mechanisms, especially on time and velocity characteristics, high-speed cylinder cushioning, fast response and great flow rate control valve, temperature compensation, system monitoring and fault diagnosis, intelligent operation, energy storage module, etc. In the end, the future trends of this field are presented.展开更多
