An accurate assessment of host and pathogen gene expression during infection is critical for understanding the molecular aspects of host-pathogen interactions.Often,pathogen-derived transcripts are difficult to ascert...An accurate assessment of host and pathogen gene expression during infection is critical for understanding the molecular aspects of host-pathogen interactions.Often,pathogen-derived transcripts are difficult to ascertain at early infection stages owing to the unfavourable transcript representation compared to the host genes.In this study,we compare two sequencing techniques,RNAseq and enrichment sequencing(RenSeq and PenSeq)of cDNA,to investigate gene expression patterns in the doubled monoploid potato(DM)infected with the late blight pathogen Phytophthora infestans.Our results reveal distinct advantages of cDNA RenSeq and PenSeq over traditional RNAseq in terms of target gene representation and transcriptional quantification at early infection stages.Throughout the infection time course,cDNA enrichment sequencing enables transcriptomic analyses for more targeted host and pathogen genes.For highly expressed genes that were sampled in parallel by both cDNA enrichment and RNAseq,a high level of concordance in expression profiles is observed,indicative of at least semi-quantitative gene expression representation following enrichment.展开更多

Two formulations of sweets based on jujube, baobab fruit powder and sweet potato puree were produced, and their nutritional values and sensory properties such as taste, smell, color and acceptability were assessed. The formulation containing the most jujube powder, baobab fruit and sweet potato puree gave the best nutritional value. The sweets are rich in calcium (259.80 mg/100 g), magnesium (67.98 mg/100 g), potassium (782.04 mg/100 g), vitamin C (32.37 mg/100 g), iron (6.45 g mg/100 g) and zinc (0.97 mg/100 g). Sensory tests on the two formulations showed acceptability scores ranging from 81.26% to 84.58%, well above the minimum acceptable threshold. Candies with jujube, baobab fruit powder and sweet potato puree could be an alternative for preventing micronutrient deficiencies.展开更多

However, some constraints related to climate change limit its production. This study evaluated the agronomic and technological performances of eight varieties of orange-fleshed of sweet potato (Ipomoea batatas (L.) to identify the most promising ones. The results show three groups of varieties with different agronomic performances. Group 1 composed of Irene and Vita is characterized by low production of small and large tuberous roots, low number of productive plants and low yield. Group 2 consisting mainly of Kabode is characterized by high production and weight of large tuberous roots as well as high yield. Group 3, composed of Tacha-2 Ininda, Ejumula Gloria, TIB-440060, CIP-199062-1, Bela bela is characterized by high production of small and large tuberous roots, high number of productive plants and average yield. The TIB-440060 variety has a recovery rate of 100%. The Irene, Kabode, Vita, Kabode varieties were resistant to viral and fungal diseases. In terms of technological quality, the TIB-440060 and CIP-199062-1 varieties have the best organoleptic qualities. Given the agronomic performances of the Kabode, TIB-440060 and CIP-199062-1 varieties, could be selected as the elites varieties and distributed to producers.展开更多
Re-domestication of diploid potato(Solanum tuberosum)into a seed crop is an innovative breeding method to accelerate genetic improvement.Seed propagation would allow hybrid production and mix superior alleles.However,...Re-domestication of diploid potato(Solanum tuberosum)into a seed crop is an innovative breeding method to accelerate genetic improvement.Seed propagation would allow hybrid production and mix superior alleles.However,almost all diploid potatoes in nature are self-incompatible(SI).Gametophytic self-incompatible(GSI)is a widespread SI in Solanaceae and is controlled by the S locus that contains a ribonuclease(S-RNase)and multiple F-box(SLFs);however,the genetic diversity of the S locus in potato is unclear.This study identified 21 S-RNase alleles involved in SI from 194 diploid potato accessions by large-scale transcriptome sequencing.The levels of amino acid similarity among different S-RNase proteins varied from 31.3 to 95.8%.S2 allele is the most widespread in 194 diploid potatoes and is mainly distributed in the S.tuberosum Group Phureja.Based on genomic annotation and expression analysis,we identified 12 potential functional SI male-determinant genes,S-locus F-box(SLFs),encoding F-box proteins in the S2 locus on a genomic region of approximately 13 Mb.Comparative genomics analysis showed that eight SLF genes are relatively conserved among four homozygous S locus.The Ka and Ks analysis suggested that S-RNase and intra-haplotypic SLF genes have co-evolved.These findings help select suitable pollinators,combine more hybrid combinations,and fully use heterosis to accelerate diploid potato breeding.展开更多
The formation of root system architecture(RSA)plays a crucial role in plant growth.OsDRO1 is known to have a function in controlling RSA in rice,however,the role of potato StDRO2,a homolog of rice OsDRO1,in root growt...The formation of root system architecture(RSA)plays a crucial role in plant growth.OsDRO1 is known to have a function in controlling RSA in rice,however,the role of potato StDRO2,a homolog of rice OsDRO1,in root growth remains unclear.In this study,we obtained potato dro2 mutant lines by Clustered Regularly Interspaced Short Palindromic Repeats-CRISPR-Associated 9(CRISPR/Cas9)-mediated genome editing system.The mutant lines were generated from a splicing defect of the StDRO2 intron 1,which causes a nonsense mutation in StDRO2.Furthermore,the secondary structure of StDRO2 mRNA analyzed with RNAfold Web Server was altered in the dro2 mutant.Mutation of StDRO2 conveys potato adaptation through changing the RSA via alteration of auxin transport under drought stress.The potato dro2 lines showed higher plant height,longer root length,smaller root growth angle and increased tuber weight than the wild-type.The alteration of RSA was associated with a disturbance of IAA distribution in the dro2 mutant,and the levels of StPIN7 and StPIN10 detected by using real-time PCR were up-regulated in the roots of potato dro2 lines grown under drought stress.Moreover,the microRNAs(miRNAs)PmiREN024536 and PmiREN024486 targeted the StDRO2 gene,and auxin positively and negatively regulated the expression of StDRO2 and the miRNAs PmiREN024536 and PmiREN024486,respectively,in the potato roots.Our data shows that a regulatory network involving auxin,StDRO2,PmiREN024536 and PmiREN024486 can control RSA to convey potato fitness under drought stress.展开更多
Sweet potato leaf tips have high nutritional value,and exploring the differences in the metabolic profiles of leaf tips among different sweet potato varieties can provide information to improve their qualities.In this...Sweet potato leaf tips have high nutritional value,and exploring the differences in the metabolic profiles of leaf tips among different sweet potato varieties can provide information to improve their qualities.In this study,a UPLC-Q-Exactive Orbitrap/MS-based untargeted metabolomics method was used to evaluate the metabolites in leaf tips of 32 sweet potato varieties.Three varieties with distinct overall metabolic profiles(A01,A02,and A03),two varieties with distinct profiles of phenolic acids(A20 and A18),and three varieties with distinct profiles of flavonoids(A05,A12,and A16)were identified.In addition,a total of 163 and 29 differentially expressed metabolites correlated with the color and leaf shape of sweet potato leaf tips,respectively,were identified through morphological characterization.Group comparison analysis of the phenotypic traits and a metabolite-phenotypic trait correlation analysis indicated that the color differences of sweet potato leaf tips were markedly associated with flavonoids.Also,the level of polyphenols was correlated with the leaf shape of sweet potato leaf tips,with lobed leaf types having higher levels of polyphenols than the entire leaf types.The findings on the metabolic profiles and differentially expressed metabolites associated with the morphology of sweet potato leaf tips can provide useful information for breeding sweet potato varieties with higher nutritional value.展开更多

β-Sitosterol-D-glucoside(β-SDG)is a phytosterol compound whose antitumor activity has been confirmed by previous studies.However,its suppression on breast cancer remains unclear.To that purpose,we isolatedβ-SDG fro...β-Sitosterol-D-glucoside(β-SDG)is a phytosterol compound whose antitumor activity has been confirmed by previous studies.However,its suppression on breast cancer remains unclear.To that purpose,we isolatedβ-SDG from sweet potato and investigated the breast-cancer-inhibiting mechanism using proteomic analysis.The sweet potato species S6 with highβ-SDG content were chosen form 36 species andβ-SDG was isolated by HPLC.Afterwards,an in situ animal model of breast cancer was established,andβ-SDG significantly reduced the tumor volume of MCF-7 xenograft mice.Proteomic analysis of tumor tissues revealed that 127 of these proteins were upregulated and 80 were downregulated.Gene ontology and network analysis showed that regulatory proteins were mainly associated with epithelial-mesenchymal transition(EMT),myogenesis,cholesterol homeostasis,oxidative phosphorylation and reactive oxygen pathways,while Vimentin,NDUF,VDAC1,PPP2CA and SNx9 were the most significant 5 node degree genes.Meanwhile,in vitro and in vivo results showed that the protein expression of PPP2CA and Vimentin,which are markers of EMT,were involved in breast cancer cell metastasis and could be reversed byβ-SDG.This work highlightsβ-SDG as a bioactive compound in sweet potato and the potential therapeutic effect ofβ-SDG for the treatment of breast cancer by inhibiting metastasis.展开更多

Four field experiments were conducted in two different locations in Bangladesh to develop integrated nutrient management practices to produce quality potato seed in industrial processing varieties Asterix and Courage. For the inorganic trial, Factorial Randomized Complete Block Design (RCBD) including 2 potato varieties, 5 treatments with 4 replications, and in the organic fertilizer trial, Factorial RCBD including 2 potato varieties, 6 treatments with 4 replications were used. In the inorganic fertilizer trail, the highest yield was obtained in the variety Asterix due to Nitrogen, Phosphorus, Potassium, and Sulfur (NPKS) plus Magnesium treated plot in Domar BADC farm and due to NPKS plus Boron, Zinc, and Magnesium treated plots in Kashimpur Farm. In the case of variety Courage, the highest yield was found in the treatment of NPKS plus Zinc in Domar BADC farm while in Kashimpur farm, NPKS plus Mg treated plots had the highest yield of potato variety-Courage. In the organic fertilizer trail, the highest tuber yield per hill was obtained by applying the government-approved commercial brand Northern organic fertilizer in variety Asterix and by organic fertilizer brand Chook Chook in variety Courage. Parameters such as days to tuber initiation, number of stems per hill, plant height, and number of tubers per hill were found statistically different among the treatments and between the two varieties. Treatments namely Northern organic fertilizer and Cowdung combined with mustard oil cake performed better considering standard grade tuber yield (grade A and B) compared to other treatments. Hence, the combination of NPKS MgZn and either Northern organic or Chook Chook or Cowdung plus mustard oil cake could be used to grow the varieties Asterix and Courage.展开更多
Sweet potato leaves are consumed globally for their nutritional and functional properties,with their taste characteristics significantly influencing their market value and consumer acceptance.However,the metabolic fac...Sweet potato leaves are consumed globally for their nutritional and functional properties,with their taste characteristics significantly influencing their market value and consumer acceptance.However,the metabolic factors determining the taste of sweet potato leaves remain unclear.This study evaluated the taste and metabolic profile of sweet potato leaves cultivated in soil and hydroponic systems,revealing that hydroponic culture improved the taste of the leafy sweet potatoes.Using gas chromatography system(GC)-ToF-MS,200 metabolites were identified,encompassing most plant metabolic pathways.A comparison of good-tasting vs.poor-tasting sweet potato leaves identified 71 metabolites associated with taste quality formation.Poor-tasting leaves exhibited lower levels of amino acid metabolites and higher levels of carbohydrate and secondary metabolites.This research provides novel insights into enhancing the taste of leafy sweet potatoes.展开更多

Small-scale farmers in Ghana who produce low-technology, subsistence crops, such as sweet potatoes, are more food secure than those who do not. This study was initiated to investigate the effect of chicken manure, compost, and cow dung on the growth and yield of “apomuden”, “SARI-Nyoriberigu”, “SARI-Nan” and “kufour” sweet potato under the Guinea Savannah agroecological zone of Ghana. Organic fertilizer increased leaf chlorophyll content and leaf area index. The application of cow dung, chicken manure and compost in 2015 significantly increased total storage root yield by 38%, 55% and 98%, 62%, 45% and 37%, 52%, 61% and 44%, and 33%, 36% and 28% for SARI-Nyoriberigu, Kufour, SARI-Nan and Apomuden, respectively, when compared to the untreated check. In 2016, and in comparison with the untreated check, the application of cow dung, chicken manure and compost increased total storage root yield by 42%, 61% and 93%, 69%, 49% and 41%, 57%, 67% and 48%, and 36%, 39% and 30% for SARI-Nyoriberigu, Kufour, SARI-Nan and Apomuden, respectively. Hence, the application of organic fertilizers will increase sweet potato yield, give higher returns to resource-poor smallholder farmers and contribute to enhancing food and nutrition security.展开更多
The whole cold-chain for exporting sweet potato(native variety“Abees”),to foreign market included immediate curing operation directly after harvest helped in healing skin texture,however,in order to reduce postharve...The whole cold-chain for exporting sweet potato(native variety“Abees”),to foreign market included immediate curing operation directly after harvest helped in healing skin texture,however,in order to reduce postharvest soft rot(Rhizopus stolonifer)incidence following trimming,and washing,ultraviolet light(UV-C)treatment was used as a main sanitizer for eliminating the soft rot.Exposure of the roots to UV-C(254 nm)was applied in a UV-C room on freshly harvested and cured sweet potato while rolling up on a movable line at 20 cm distance for 1,2,and 3 hr.As combining UV-C treatment with chlorine(200 ppm)on roots,marked and significant reduction of the total microbial load and Rhizopus potential was achieved on root surfaces respectively compared with chlorine alone.It also reduced soft rot percentage to almost 0%infection.After 3 months of cold-storage,quality assessment of sweet potato showed that root characteristics were markedly maintained.The ability of UV-C light to induce phenylalanine ammonia lyase(PAL)enzyme activity in root tissue and maintain the activities of peroxidase and polyphenol oxidase,however with slight increase,was detected.UV-C caused an increase of phenol content in sweet potato tissue that made an activation of defense reaction against the rot causal pathogen.As the exposure time to UV-C light increased,a higher content of phenols occurred.Moreover,UV-C application caused decrease in sugar content of root tissue that is flavored by soft rot-causal pathogen.展开更多

The sizes of the cubic samples are 0.5, 1, 1.5, 1.75, 2, 2.5 and 3 cm edge with a respective initial water content of 2.7, 3.76, 3.48, 2.68, 3.28, 2.17 and 2.29 kg/kgms. For cylindrical samples, the radius is set at 0.5 cm and sample heights are 1, 1.5, 2, 2.5, 3, 3.5 and 4 cm with respective water contents of 2.2, 3.19, 2.85, 2.1, 2.17, 2.39 and 2.03 kg/kgms. The effective diffusion coefficients of cubic samples are of the order of 10−10 and 10−9 m2∙s−1 grew with sample edge. As for the cylindrical samples, the effective diffusion coefficients were of the order of 10−9 m2∙s−1 and there was no linear correlation between cylinder height and their effective diffusion coefficient. As for the examination of the initial water content on the effective diffusion coefficient, it turned out that the initial water content had no influence on the effective diffusion coefficient of the sweet potato samples.展开更多

Potatoes are a significant crop and are primarily used in the world. It is a staple food in many countries, where millions of tonnes are produced annually. It is an essential source of many nutrients, such as proteins, carbohydrates, vitamins, and beta-carotene. In addition, potatoes are being used as therapeutic agents against cancer and other human diseases as well. Potatoes are on the third list after wheat and rice. To overcome food shortages and malnutrition, there are two methods used for producing potatoes: the first is sexual, which is seed propagation, and the second is asexual, which is plant tissue culture propagation. Conventional potato breeding is a uniform method, but it is unsafe because there is a risk of pathogen attack. In a laboratory setting, the tissue culture of potatoes produced millions of plants with nutrient-rich medium under controlled environmental conditions that prevent pest attacks. Some environmental stresses, such as salinity and water scarcity, affect potato yield and production;however, applying nanoparticles like organic, inorganic, and silicon dioxide enhances potato quality and combats stress. Biotechnology has proven to be helpful in addressing all these issues. This review discusses the significance of potatoes, their production through the tissue culture technique, and the application of nanoparticles to improve the growth, and impact of potatoes on human health.展开更多

Leaves from tobacco (Nicotiana tabacum L. cv. K326) were transformed with these four plant expression plasmids by Agrobacterium-mediated transformation, respectively. Southern and Western blotting analyses showed that these four mutants were integrated into tobacco genomic DNA and could express the corresponding proteins in most of die transgenic plants. The challenge of transgenic plants with potato X potexvirus (PVX) revealed that the expression products of PVY-C HC-Pro mutants in transgenic plants greatly abolished functions of HC-Pro in enhancing the accumulation and pathogenicity of PVX, indicating that CCCT and PTK motifs of HC-Pro were required for PVX/PVY synergism. Meanwhile, the results demonstrated that PVY-C HC-Pro had a function in accelerating the long-distance movement of PVX in these transgenic plants for the first time.展开更多

[Method] The starch-oriented Jishu No.21 and raw-eating oriented Jishu No.22 were studied to explore effects of planting density on yield and sink and source characteristics of sweet potato. [IRe- suit] Leaf area index of Jishu No.21 and Jishu No.22 were increasing upon planting density. Leaf area index of the same planting density showed a single-peak curve. Specifically, leaf area index grew fast during the 40th-80th d after planting, and reached the peak on the 80th d after planting, followed by decreasing. What's more, ventilation and sunshine transmission both declined upon planting density, as well as the number of leaf, the number of branch, the length of vine, dry and fresh weights of stem and leaf. When planting density exceeded 75 000 plants/hm2, the yield of sweet potato dropped dramatically. Besides, the optimal planting density tended to be volatile upon cultivars. For example, the range of 45 000-60 000 plants/hm2 is the optimal planting density of Jishu No.21 and the range of 60 000-75 000 plants/hm2 is the optimal planting density of Jishu No.22. [Conclusion] It can be concluded that rational planting densities would well coordinate sweet potato growth of ground parts and underground parts to get a high yield by providing a rational group structure. Considering the optimal planting density differs upon cultivars, it is necessary to take genotype, environment, soil fertility and planting density into consideration in determining planting density.展开更多

[ Method ] The extraction and storage conditions for free radicals scavenging substances from sweet potato leaves were optimized by orthogonal test and the bioactive components in extracts were investigated by correlation analysis. [ Result] Sweet potato leaves contain the bioactive substances scavenging DPPH free radical and hydroxyl free radical. Extracting solvent species is the most important factor that influencing extraction yield. The optimal extraction and storage conditions are as following: water as solvent, pH 8.0 of extracting liquid, storage at 25 ℃. There is a good positive linear relationship between the content of total phenols in sweet potato leaves and corresponding scavenging rates against both the DPPH free radical and hydroxyl free radical. For the content of total flavones in sweet potato leaves, just a correlation with scavenging rate against hydroxyl free radical shown in test. [ Conclusion] The phenols in ex- tracts could effectively scavenge both the DPPH free radical and hydroxyl free radical, whereas the flavones in extracts can only function on the hydroxyl free radical.展开更多
A complete cDNA of potato Phytophthora infestans-induced hypersensitive response-related protein gene (POTHR-1) was cloned using rapid amplification of cDNA ends (RACE) strategy according to a fragment sequence which ...A complete cDNA of potato Phytophthora infestans-induced hypersensitive response-related protein gene (POTHR-1) was cloned using rapid amplification of cDNA ends (RACE) strategy according to a fragment sequence which we had cloned using suppression subtractive hybridization (SSH) technique. The potato POTHR-1 gene encodes a protein of 225 amino acids, which shares 81% identity with tobacco hin1 gene-enoded protein (harpin-induced protein). Southern blot revealed that there are two to three copies of POTHR-1 in potato genome. The POTHR-1 gene expression in potato leaves showed that its transcripts accumulated remarkably in leaves after 36 h inoculation with P. infestans. Mechanical wounding and jasmonic acid (JA) could induce the POTHR-1 gene expression and osmotic stress just induce a slight accumulation of POTHR-1 gene mRNA, while salicylic acid (SA) had no detectable function on the induction accumulation of POTHR-1 gene transcripts. The potato POTHR-1 gene may preferentially associate with hypersensitive response (HR) or biotic cell death during interaction between host and pathogen.展开更多

文摘An accurate assessment of host and pathogen gene expression during infection is critical for understanding the molecular aspects of host-pathogen interactions.Often,pathogen-derived transcripts are difficult to ascertain at early infection stages owing to the unfavourable transcript representation compared to the host genes.In this study,we compare two sequencing techniques,RNAseq and enrichment sequencing(RenSeq and PenSeq)of cDNA,to investigate gene expression patterns in the doubled monoploid potato(DM)infected with the late blight pathogen Phytophthora infestans.Our results reveal distinct advantages of cDNA RenSeq and PenSeq over traditional RNAseq in terms of target gene representation and transcriptional quantification at early infection stages.Throughout the infection time course,cDNA enrichment sequencing enables transcriptomic analyses for more targeted host and pathogen genes.For highly expressed genes that were sampled in parallel by both cDNA enrichment and RNAseq,a high level of concordance in expression profiles is observed,indicative of at least semi-quantitative gene expression representation following enrichment.
文摘Increasing access to locally produced, safe, nutritious and affordable complementary foods is essential to combat micronutrient deficiencies in young children in low- and middle-income countries. Two formulations of sweets based on jujube, baobab fruit powder and sweet potato puree were produced, and their nutritional values and sensory properties such as taste, smell, color and acceptability were assessed. The formulation containing the most jujube powder, baobab fruit and sweet potato puree gave the best nutritional value. The sweets are rich in calcium (259.80 mg/100 g), magnesium (67.98 mg/100 g), potassium (782.04 mg/100 g), vitamin C (32.37 mg/100 g), iron (6.45 g mg/100 g) and zinc (0.97 mg/100 g). Sensory tests on the two formulations showed acceptability scores ranging from 81.26% to 84.58%, well above the minimum acceptable threshold. Candies with jujube, baobab fruit powder and sweet potato puree could be an alternative for preventing micronutrient deficiencies.
文摘Sweet potato is one of the most important crops in Côte d’Ivoire. However, some constraints related to climate change limit its production. This study evaluated the agronomic and technological performances of eight varieties of orange-fleshed of sweet potato (Ipomoea batatas (L.) to identify the most promising ones. The results show three groups of varieties with different agronomic performances. Group 1 composed of Irene and Vita is characterized by low production of small and large tuberous roots, low number of productive plants and low yield. Group 2 consisting mainly of Kabode is characterized by high production and weight of large tuberous roots as well as high yield. Group 3, composed of Tacha-2 Ininda, Ejumula Gloria, TIB-440060, CIP-199062-1, Bela bela is characterized by high production of small and large tuberous roots, high number of productive plants and average yield. The TIB-440060 variety has a recovery rate of 100%. The Irene, Kabode, Vita, Kabode varieties were resistant to viral and fungal diseases. In terms of technological quality, the TIB-440060 and CIP-199062-1 varieties have the best organoleptic qualities. Given the agronomic performances of the Kabode, TIB-440060 and CIP-199062-1 varieties, could be selected as the elites varieties and distributed to producers.

文摘Re-domestication of diploid potato(Solanum tuberosum)into a seed crop is an innovative breeding method to accelerate genetic improvement.Seed propagation would allow hybrid production and mix superior alleles.However,almost all diploid potatoes in nature are self-incompatible(SI).Gametophytic self-incompatible(GSI)is a widespread SI in Solanaceae and is controlled by the S locus that contains a ribonuclease(S-RNase)and multiple F-box(SLFs);however,the genetic diversity of the S locus in potato is unclear.This study identified 21 S-RNase alleles involved in SI from 194 diploid potato accessions by large-scale transcriptome sequencing.The levels of amino acid similarity among different S-RNase proteins varied from 31.3 to 95.8%.S2 allele is the most widespread in 194 diploid potatoes and is mainly distributed in the S.tuberosum Group Phureja.Based on genomic annotation and expression analysis,we identified 12 potential functional SI male-determinant genes,S-locus F-box(SLFs),encoding F-box proteins in the S2 locus on a genomic region of approximately 13 Mb.Comparative genomics analysis showed that eight SLF genes are relatively conserved among four homozygous S locus.The Ka and Ks analysis suggested that S-RNase and intra-haplotypic SLF genes have co-evolved.These findings help select suitable pollinators,combine more hybrid combinations,and fully use heterosis to accelerate diploid potato breeding.

文摘The formation of root system architecture(RSA)plays a crucial role in plant growth.OsDRO1 is known to have a function in controlling RSA in rice,however,the role of potato StDRO2,a homolog of rice OsDRO1,in root growth remains unclear.In this study,we obtained potato dro2 mutant lines by Clustered Regularly Interspaced Short Palindromic Repeats-CRISPR-Associated 9(CRISPR/Cas9)-mediated genome editing system.The mutant lines were generated from a splicing defect of the StDRO2 intron 1,which causes a nonsense mutation in StDRO2.Furthermore,the secondary structure of StDRO2 mRNA analyzed with RNAfold Web Server was altered in the dro2 mutant.Mutation of StDRO2 conveys potato adaptation through changing the RSA via alteration of auxin transport under drought stress.The potato dro2 lines showed higher plant height,longer root length,smaller root growth angle and increased tuber weight than the wild-type.The alteration of RSA was associated with a disturbance of IAA distribution in the dro2 mutant,and the levels of StPIN7 and StPIN10 detected by using real-time PCR were up-regulated in the roots of potato dro2 lines grown under drought stress.Moreover,the microRNAs(miRNAs)PmiREN024536 and PmiREN024486 targeted the StDRO2 gene,and auxin positively and negatively regulated the expression of StDRO2 and the miRNAs PmiREN024536 and PmiREN024486,respectively,in the potato roots.Our data shows that a regulatory network involving auxin,StDRO2,PmiREN024536 and PmiREN024486 can control RSA to convey potato fitness under drought stress.

文摘Sweet potato leaf tips have high nutritional value,and exploring the differences in the metabolic profiles of leaf tips among different sweet potato varieties can provide information to improve their qualities.In this study,a UPLC-Q-Exactive Orbitrap/MS-based untargeted metabolomics method was used to evaluate the metabolites in leaf tips of 32 sweet potato varieties.Three varieties with distinct overall metabolic profiles(A01,A02,and A03),two varieties with distinct profiles of phenolic acids(A20 and A18),and three varieties with distinct profiles of flavonoids(A05,A12,and A16)were identified.In addition,a total of 163 and 29 differentially expressed metabolites correlated with the color and leaf shape of sweet potato leaf tips,respectively,were identified through morphological characterization.Group comparison analysis of the phenotypic traits and a metabolite-phenotypic trait correlation analysis indicated that the color differences of sweet potato leaf tips were markedly associated with flavonoids.Also,the level of polyphenols was correlated with the leaf shape of sweet potato leaf tips,with lobed leaf types having higher levels of polyphenols than the entire leaf types.The findings on the metabolic profiles and differentially expressed metabolites associated with the morphology of sweet potato leaf tips can provide useful information for breeding sweet potato varieties with higher nutritional value.
文摘[Objective]The aim was to establish the multiplex PCR method for three virus of potato:PVA(potato virus A),TMV(Tobacco mosaic virus)and PVY(potato virus Y).[Method]According to the PVA,TMV and PVY sequences available in GenBank,pairs of primer were designed for establishing a multiplex PCR method,and constructing recombinant plasmid of target genes by PCR amplified of three viruses as reference standard simple to be used in sensitivity test;PVX(Potato virus X),PVM(Potato virus M),PVS(Potato virus S),PVV(Potato virus V)and CMV(Cucumber mosaic virus)were used to carry out the specificity test and detection of 11 samples which were suspected of virus infected.[Result]The detection limit for PVA,TMV and PVY was 14,14 and 14 copies/ml,respectively.No cross-reactivity was observed with other viruses.Seven of 11 samples were infected by three viruses.[Conclusion]The multiplex PCR for PVA,TMV,PVY three viruses of potato was established successfully,which had provided basis for the detection technology of potato virus.

文摘β-Sitosterol-D-glucoside(β-SDG)is a phytosterol compound whose antitumor activity has been confirmed by previous studies.However,its suppression on breast cancer remains unclear.To that purpose,we isolatedβ-SDG from sweet potato and investigated the breast-cancer-inhibiting mechanism using proteomic analysis.The sweet potato species S6 with highβ-SDG content were chosen form 36 species andβ-SDG was isolated by HPLC.Afterwards,an in situ animal model of breast cancer was established,andβ-SDG significantly reduced the tumor volume of MCF-7 xenograft mice.Proteomic analysis of tumor tissues revealed that 127 of these proteins were upregulated and 80 were downregulated.Gene ontology and network analysis showed that regulatory proteins were mainly associated with epithelial-mesenchymal transition(EMT),myogenesis,cholesterol homeostasis,oxidative phosphorylation and reactive oxygen pathways,while Vimentin,NDUF,VDAC1,PPP2CA and SNx9 were the most significant 5 node degree genes.Meanwhile,in vitro and in vivo results showed that the protein expression of PPP2CA and Vimentin,which are markers of EMT,were involved in breast cancer cell metastasis and could be reversed byβ-SDG.This work highlightsβ-SDG as a bioactive compound in sweet potato and the potential therapeutic effect ofβ-SDG for the treatment of breast cancer by inhibiting metastasis.
文摘Potato is one of the most important vegetable crops, which contributes more than half of the total vegetable production in Bangladesh. Four field experiments were conducted in two different locations in Bangladesh to develop integrated nutrient management practices to produce quality potato seed in industrial processing varieties Asterix and Courage. For the inorganic trial, Factorial Randomized Complete Block Design (RCBD) including 2 potato varieties, 5 treatments with 4 replications, and in the organic fertilizer trial, Factorial RCBD including 2 potato varieties, 6 treatments with 4 replications were used. In the inorganic fertilizer trail, the highest yield was obtained in the variety Asterix due to Nitrogen, Phosphorus, Potassium, and Sulfur (NPKS) plus Magnesium treated plot in Domar BADC farm and due to NPKS plus Boron, Zinc, and Magnesium treated plots in Kashimpur Farm. In the case of variety Courage, the highest yield was found in the treatment of NPKS plus Zinc in Domar BADC farm while in Kashimpur farm, NPKS plus Mg treated plots had the highest yield of potato variety-Courage. In the organic fertilizer trail, the highest tuber yield per hill was obtained by applying the government-approved commercial brand Northern organic fertilizer in variety Asterix and by organic fertilizer brand Chook Chook in variety Courage. Parameters such as days to tuber initiation, number of stems per hill, plant height, and number of tubers per hill were found statistically different among the treatments and between the two varieties. Treatments namely Northern organic fertilizer and Cowdung combined with mustard oil cake performed better considering standard grade tuber yield (grade A and B) compared to other treatments. Hence, the combination of NPKS MgZn and either Northern organic or Chook Chook or Cowdung plus mustard oil cake could be used to grow the varieties Asterix and Courage.

文摘Sweet potato leaves are consumed globally for their nutritional and functional properties,with their taste characteristics significantly influencing their market value and consumer acceptance.However,the metabolic factors determining the taste of sweet potato leaves remain unclear.This study evaluated the taste and metabolic profile of sweet potato leaves cultivated in soil and hydroponic systems,revealing that hydroponic culture improved the taste of the leafy sweet potatoes.Using gas chromatography system(GC)-ToF-MS,200 metabolites were identified,encompassing most plant metabolic pathways.A comparison of good-tasting vs.poor-tasting sweet potato leaves identified 71 metabolites associated with taste quality formation.Poor-tasting leaves exhibited lower levels of amino acid metabolites and higher levels of carbohydrate and secondary metabolites.This research provides novel insights into enhancing the taste of leafy sweet potatoes.
文摘Twelve percent (12%) of Ghanaians are food insecure, and climate-smart crops like sweet potatoes are required to help end poverty. Small-scale farmers in Ghana who produce low-technology, subsistence crops, such as sweet potatoes, are more food secure than those who do not. This study was initiated to investigate the effect of chicken manure, compost, and cow dung on the growth and yield of “apomuden”, “SARI-Nyoriberigu”, “SARI-Nan” and “kufour” sweet potato under the Guinea Savannah agroecological zone of Ghana. Organic fertilizer increased leaf chlorophyll content and leaf area index. The application of cow dung, chicken manure and compost in 2015 significantly increased total storage root yield by 38%, 55% and 98%, 62%, 45% and 37%, 52%, 61% and 44%, and 33%, 36% and 28% for SARI-Nyoriberigu, Kufour, SARI-Nan and Apomuden, respectively, when compared to the untreated check. In 2016, and in comparison with the untreated check, the application of cow dung, chicken manure and compost increased total storage root yield by 42%, 61% and 93%, 69%, 49% and 41%, 57%, 67% and 48%, and 36%, 39% and 30% for SARI-Nyoriberigu, Kufour, SARI-Nan and Apomuden, respectively. Hence, the application of organic fertilizers will increase sweet potato yield, give higher returns to resource-poor smallholder farmers and contribute to enhancing food and nutrition security.
文摘The whole cold-chain for exporting sweet potato(native variety“Abees”),to foreign market included immediate curing operation directly after harvest helped in healing skin texture,however,in order to reduce postharvest soft rot(Rhizopus stolonifer)incidence following trimming,and washing,ultraviolet light(UV-C)treatment was used as a main sanitizer for eliminating the soft rot.Exposure of the roots to UV-C(254 nm)was applied in a UV-C room on freshly harvested and cured sweet potato while rolling up on a movable line at 20 cm distance for 1,2,and 3 hr.As combining UV-C treatment with chlorine(200 ppm)on roots,marked and significant reduction of the total microbial load and Rhizopus potential was achieved on root surfaces respectively compared with chlorine alone.It also reduced soft rot percentage to almost 0%infection.After 3 months of cold-storage,quality assessment of sweet potato showed that root characteristics were markedly maintained.The ability of UV-C light to induce phenylalanine ammonia lyase(PAL)enzyme activity in root tissue and maintain the activities of peroxidase and polyphenol oxidase,however with slight increase,was detected.UV-C caused an increase of phenol content in sweet potato tissue that made an activation of defense reaction against the rot causal pathogen.As the exposure time to UV-C light increased,a higher content of phenols occurred.Moreover,UV-C application caused decrease in sugar content of root tissue that is flavored by soft rot-causal pathogen.
文摘In this article, we investigated the influence of size and initial water content on the effective diffusion coefficient of sweet potatoes samples cut into cubic and cylindrical shapes. The sizes of the cubic samples are 0.5, 1, 1.5, 1.75, 2, 2.5 and 3 cm edge with a respective initial water content of 2.7, 3.76, 3.48, 2.68, 3.28, 2.17 and 2.29 kg/kgms. For cylindrical samples, the radius is set at 0.5 cm and sample heights are 1, 1.5, 2, 2.5, 3, 3.5 and 4 cm with respective water contents of 2.2, 3.19, 2.85, 2.1, 2.17, 2.39 and 2.03 kg/kgms. The effective diffusion coefficients of cubic samples are of the order of 10−10 and 10−9 m2∙s−1 grew with sample edge. As for the cylindrical samples, the effective diffusion coefficients were of the order of 10−9 m2∙s−1 and there was no linear correlation between cylinder height and their effective diffusion coefficient. As for the examination of the initial water content on the effective diffusion coefficient, it turned out that the initial water content had no influence on the effective diffusion coefficient of the sweet potato samples.
文摘Plant tissue culture is a technique that enhances the quality and quantity of potatoes. Potatoes are a significant crop and are primarily used in the world. It is a staple food in many countries, where millions of tonnes are produced annually. It is an essential source of many nutrients, such as proteins, carbohydrates, vitamins, and beta-carotene. In addition, potatoes are being used as therapeutic agents against cancer and other human diseases as well. Potatoes are on the third list after wheat and rice. To overcome food shortages and malnutrition, there are two methods used for producing potatoes: the first is sexual, which is seed propagation, and the second is asexual, which is plant tissue culture propagation. Conventional potato breeding is a uniform method, but it is unsafe because there is a risk of pathogen attack. In a laboratory setting, the tissue culture of potatoes produced millions of plants with nutrient-rich medium under controlled environmental conditions that prevent pest attacks. Some environmental stresses, such as salinity and water scarcity, affect potato yield and production;however, applying nanoparticles like organic, inorganic, and silicon dioxide enhances potato quality and combats stress. Biotechnology has proven to be helpful in addressing all these issues. This review discusses the significance of potatoes, their production through the tissue culture technique, and the application of nanoparticles to improve the growth, and impact of potatoes on human health.
文摘To mutagenize two conserved CCCT and PTK motifs in the central domain of Chinese strain of potato Y potyvirus (PVY-C) helper component proteinase (HC-Pro), four mutants of HC-Pro gene were obtained by PCR and site-directed mutagenesis, and then were inserted into the constitutive expression vector pBin438. Leaves from tobacco (Nicotiana tabacum L. cv. K326) were transformed with these four plant expression plasmids by Agrobacterium-mediated transformation, respectively. Southern and Western blotting analyses showed that these four mutants were integrated into tobacco genomic DNA and could express the corresponding proteins in most of die transgenic plants. The challenge of transgenic plants with potato X potexvirus (PVX) revealed that the expression products of PVY-C HC-Pro mutants in transgenic plants greatly abolished functions of HC-Pro in enhancing the accumulation and pathogenicity of PVX, indicating that CCCT and PTK motifs of HC-Pro were required for PVX/PVY synergism. Meanwhile, the results demonstrated that PVY-C HC-Pro had a function in accelerating the long-distance movement of PVX in these transgenic plants for the first time.

文摘[Objective] This study aimed to investigate the salt-tolerance mechanism of sweet potato.[Method]Two sweet potato varieties of Xu 25-2(salt-tolerant cultivar)and Triumph 100(salt-sensitive cultivar)were treated by sodium chloride with the concentration of 0 mmol/L and 100 mmol/L.After 20 days,Na+ content and Na+/K+ ratio in the roots,shoots and leave were determined by the flame photometer,while dry weight and fresh weight of roots,shoots and leave in different varieties were also studied.[Result]The growth of two sweet potato varieties was inhibited under salt stress,so the plant became shorter,leaf and root became fewer,dry weight of roots and leave decreased,but seedlings of Xu 25-2 were inhibited slightly.Furthermore,Na+ content and Na+/K+ ratio in roots,shoots and leaves of two sweet potato varieties increased.Na+ content of salt-tolerant Xu 25-2 was low in roots,shoots and leaves,while Na+ content of salt-sensitive Triumph 100 was high in shoots and leave of seedlings,but the change range of Xu 25-2 was less than that of Triumph 100.[Conclusion]The lower Na+ content and Na+/K+ ratio in leaves under salt stress were the most important characteristics for salt-tolerance of sweet potato varieties.

文摘[Objective] The aim was to resolve the issue of sparsely planting (37 500-40 500 plants/hm2) of sweet potato in hilly areas. [Method] The starch-oriented Jishu No.21 and raw-eating oriented Jishu No.22 were studied to explore effects of planting density on yield and sink and source characteristics of sweet potato. [IRe- suit] Leaf area index of Jishu No.21 and Jishu No.22 were increasing upon planting density. Leaf area index of the same planting density showed a single-peak curve. Specifically, leaf area index grew fast during the 40th-80th d after planting, and reached the peak on the 80th d after planting, followed by decreasing. What's more, ventilation and sunshine transmission both declined upon planting density, as well as the number of leaf, the number of branch, the length of vine, dry and fresh weights of stem and leaf. When planting density exceeded 75 000 plants/hm2, the yield of sweet potato dropped dramatically. Besides, the optimal planting density tended to be volatile upon cultivars. For example, the range of 45 000-60 000 plants/hm2 is the optimal planting density of Jishu No.21 and the range of 60 000-75 000 plants/hm2 is the optimal planting density of Jishu No.22. [Conclusion] It can be concluded that rational planting densities would well coordinate sweet potato growth of ground parts and underground parts to get a high yield by providing a rational group structure. Considering the optimal planting density differs upon cultivars, it is necessary to take genotype, environment, soil fertility and planting density into consideration in determining planting density.

[ Method ] The extraction and storage conditions for free radicals scavenging substances from sweet potato leaves were optimized by orthogonal test and the bioactive components in extracts were investigated by correlation analysis. [ Result] Sweet potato leaves contain the bioactive substances scavenging DPPH free radical and hydroxyl free radical. Extracting solvent species is the most important factor that influencing extraction yield. The optimal extraction and storage conditions are as following: water as solvent, pH 8.0 of extracting liquid, storage at 25 ℃. There is a good positive linear relationship between the content of total phenols in sweet potato leaves and corresponding scavenging rates against both the DPPH free radical and hydroxyl free radical. For the content of total flavones in sweet potato leaves, just a correlation with scavenging rate against hydroxyl free radical shown in test. [ Conclusion] The phenols in ex- tracts could effectively scavenge both the DPPH free radical and hydroxyl free radical, whereas the flavones in extracts can only function on the hydroxyl free radical.
文摘A complete cDNA of potato Phytophthora infestans-induced hypersensitive response-related protein gene (POTHR-1) was cloned using rapid amplification of cDNA ends (RACE) strategy according to a fragment sequence which we had cloned using suppression subtractive hybridization (SSH) technique. The potato POTHR-1 gene encodes a protein of 225 amino acids, which shares 81% identity with tobacco hin1 gene-enoded protein (harpin-induced protein). Southern blot revealed that there are two to three copies of POTHR-1 in potato genome. The POTHR-1 gene expression in potato leaves showed that its transcripts accumulated remarkably in leaves after 36 h inoculation with P. infestans. Mechanical wounding and jasmonic acid (JA) could induce the POTHR-1 gene expression and osmotic stress just induce a slight accumulation of POTHR-1 gene mRNA, while salicylic acid (SA) had no detectable function on the induction accumulation of POTHR-1 gene transcripts. The potato POTHR-1 gene may preferentially associate with hypersensitive response (HR) or biotic cell death during interaction between host and pathogen.
