However, they can generate a great... Purinergic P2X receptors are a family of ligand-gated cationic channels activated by extracellular ATP. P2X subunit protein sequences are highly conserved between vertebrate species. However, they can generate a great diversity of coding splicing variants to fulfill several roles in mammalian physiology. Despite intensive research in P2X expression in both central and peripheral nervous system, there is little information about their homology, genomic structure and other key features that can help to develop selective drugs or regulatory strategies of pharmacological value which are lacking today. In order to obtain clues on mammalian P2X diversity, we have performed a bioinformatics analysis of the coding regions and introns of the seven P2X subunits present in human, simian, dog, mouse, rat and zebrafish. Here we report the arrangements of exon and intron sequences, considering its number, size, phase and placement;proposing some ideas about the gain and loss of exons and retention of introns. Taken together, these evidences show traits that can be used to gain insight into the evolutionary history of vertebrate P2X receptors and better understand the diversity of subunits coding the purinergic signaling in mammals. 展开更多

Background:This study explores the relationship between endoplasmic reticulum(ER)stress and diabetes,particularly focusing on the impact of physical exercise on ER stress mechanisms and identifying potential therapeutic drugs and targets for diabetes-related sepsis.The research also incorporates traditional physical therapy perspectives,emphasizing the genomic insights gained from exercise therapy in disease management and prevention.Methods:Gene analysis was conducted on the GSE168796 and GSE94717 datasets to identify ER stress-related genes.Gene interactions and immune cell correlations were mapped using GeneCard and STRING databases.A screening of 2,456 compounds from the TCMSP database was performed to identify potential therapeutic agents,with a focus on their docking potential.Techniques such as luciferase reporter gene assay and RNA interference were used to examine the interactions between microRNA-149-5p and MMP9.Results:The study identified 2,006 differentially expressed genes and 616 miRNAs.Key genes like MMP9,TNF-α,and IL1B were linked to an immunosuppressive state.Licorice glycoside E demonstrated high affinity for MMP9,suggesting its potential effectiveness in treating diabetes.The constructed miRNA network highlighted the regulatory roles of MMP9,IL1B,IFNG,and TNF-α.Experimental evidence confirmed the binding of microRNA-149-5p to MMP9,impacting apoptosis in diabetic cells.Conclusion:The findings highlight the regulatory role of microRNA-149-5p in managing MMP9,a crucial gene in diabetes pathophysiology.Licorice glycoside E emerges as a promising treatment option for diabetes,especially targeting MMP9 affected by ER stress.The study also underscores the significance of physical exercise in modulating ER stress pathways in diabetes management,bridging traditional physical therapy and modern scientific understanding.Our study has limitations.It focuses on the microRNA-149-5p-MMP9 network in sepsis,using cell-based methods without animal or clinical trials.Despite strong in vitro findings,in vivo studies are needed to confirm licorice glycoside E’s therapeutic potential and understand the microRNA-149-5p-MMP9 dynamics in real conditions. 展开更多

Pasteurellosis is the most prevalent, extremely contagious bacterial disease among domestic rabbits and is considered the leading cause of deaths in rabbits, resulting in enormous economic losses to the rabbit industry. Screening for bacterial agents causing mortalities in rabbits revealed the presence of Enterobacteriacae species in approximately 42% of studied cases, with E. coli the most commonly isolated organism. The present study was designed to evaluate the immune response of rabbits vaccinated with a locally prepared, combined inactivated vaccine of Pasteurella multocida and E. coli, adjuvanated with Montanide ISA70. A total of 370 rabbits, aged 2 - 3 weeks, were divided into four groups: (G1) vaccinated with a polyvalent P. multocida vaccine, (G2) vaccinated with a polyvalent E. coli vaccine, (G3) vaccinated with a combined inactivated Montanide ISA70 vaccine of P. multocida and E. coli, and (G4) kept as a non-vaccinated control group. All rabbits received two doses of 0.5 ml of the prepared vaccines, administered one month apart, and were then challenged with virulent strains of P. multocida and E. coli three weeks after the second vaccination. The prepared vaccines were evaluated by determining humoral immunity using indirect haemagglutination (IHA) test and ELISA. The potency of the vaccines was assessed through challenge and determination of LD50. Experimental findings on the prepared polyvalent combined inactivated P. multocida and E. coli vaccine indicated that it is a potent vaccine, producing the highest antibody titers and a 90% protection rate against challenges with virulent strains of P. multocida type A, D2, and E. coli types O157, O151 and O125. Thus, this vaccine is promising in addressing both P. multocida and E. coli problems in rabbits, farms, providing significant protection, and we recommend its commercial production to help rabbit producers control these two major bacterial infections. 展开更多

Effects of tree size and organ age on variations in carbon,nitrogen,and phosphorus stoichiometry in Pinus koraiensis

Carbon(C),nitrogen(N),and phosphorus(P)are of fundamental importance for growth and nutrient dynamics within plant organs and deserve more attention at regional to global scales.However,our knowledge of how these nutrients vary with tree size,organ age,or root order at the individual level remains limited.We determined C,N,and P contents and their stoichiometric ratios(i.e.,nutrient traits)in needles,branches,and fine roots at different organ ages(0-3-year-old needles and branches)and root orders(1st-4th order roots)from 64 Pinus koraiensis of varying size(Diameter at breast height ranged from 0.3 to 100 cm)in northeast China.Soil factors were also measured.The results show that nutrient traits were regulated by tree size,organ age,or root order rather than soil factors.At a whole-plant level,nutrient traits decreased in needles and fine roots but increased in branches with tree size.At the organ level,age or root order had a negative effect on C,N,and P and a positive effect on stoichiometric ratios.Our results demonstrate that nutrient variations are closely related to organ-specific functions and ecophysiological processes at an individual level.It is suggested that the nutrient acquisition strategy by younger trees and organ fractions with higher nutrient content is for survival.Conversely,nutrient storage strategy in older trees and organ fractions are mainly for steady growth.Our results clarified the nutrient utilization strategies during tree and organ ontogeny and suggest that tree size and organ age or root order should be simultaneously considered to understand the complexities of nutrient variations. 展开更多

miR-221-5p靶向调控E2F3对结肠癌细胞增殖、迁移的影响

目的探讨miR-221-5p抑制结肠癌细胞增殖、迁移的作用及相关机制。方法稳定培养HT-29、SW480细胞,分别过表达miR-221-5p和(或)敲低E2F转录因子(E2F)3表达,细胞分为miR-NC组、miR-221-5p组、si-NC组、si-E2F3组、miR-NC+si-NC组、miR-NC+s... 目的探讨miR-221-5p抑制结肠癌细胞增殖、迁移的作用及相关机制。方法稳定培养HT-29、SW480细胞,分别过表达miR-221-5p和(或)敲低E2F转录因子(E2F)3表达,细胞分为miR-NC组、miR-221-5p组、si-NC组、si-E2F3组、miR-NC+si-NC组、miR-NC+si-E2F3组、miR-221-5p+si-NC组、miR-221-5p+si-E2F3组,检测HT-29、SW480细胞活性、凋亡、迁移和侵袭能力。将40只裸鼠分为miR-221-5p组与miR-NC组,构建皮下结肠癌移植瘤模型,分别经皮下注射miR-221-5p agomir与NC agomir,检测小鼠各项指标差异。结果与miR-NC组相比,miR-221-5p组SW480细胞培养3 d后的光密度(OD)450值明显降低,HT-29和SW480细胞凋亡率明显升高,迁移和侵袭细胞数明显减少(均P<0.05)。与si-NC组相比,si-E2F3组HT-29和SW480细胞培养3 d后的OD450值明显降低,细胞凋亡率明显升高,迁移和侵袭细胞数明显减少(均P<0.05)。miR-221-5p可直接与E2F3的3′非翻译区(UTR)结合来抑制E2F3表达。与miR-NC组相比,miR-221-5p组肿瘤生长明显抑制,肿瘤组织中E2F3、Ki-67、基质金属蛋白酶(MMP)-2、MMP-9蛋白阳性表达得分明显降低(均P<0.05)。结论miR-221-5p可靶向调控E2F3表达抑制结肠癌细胞的增殖和迁移,从而抑制结肠癌进展。 展开更多

目的探讨血清可溶性P-选择素(sP-selectin)、可溶性E-选择素(sE-selectin)对原发性高血压(EH)伴2型糖尿病(T2DM)的诊断价值及其与胰岛素抵抗的关系。方法选取2019年9月至2021年9月该院收治的124例EH患者作为研究对象,根据是否伴有T2DM... 目的探讨血清可溶性P-选择素(sP-selectin)、可溶性E-选择素(sE-selectin)对原发性高血压(EH)伴2型糖尿病(T2DM)的诊断价值及其与胰岛素抵抗的关系。方法选取2019年9月至2021年9月该院收治的124例EH患者作为研究对象,根据是否伴有T2DM将其分为非T2DM组66例和T2DM组58例;另选取同期在该院体检的50例健康志愿者作为对照组。收集研究对象的收缩压、舒张压、脉压及甘油三酯、胆固醇、空腹血糖水平等临床资料;采用酶联免疫吸附试验检测血清sP-selectin、sE-selectin水平;采用电化学发光法测定空腹胰岛素水平,并计算胰岛素抵抗指数。采用Pearson相关分析T2DM患者血清sP-selectin、sE-selectin水平与空腹胰岛素水平、胰岛素抵抗指数的相关性;绘制受试者工作特征(ROC)曲线分析血清sP-selectin、sE-selectin对EH伴T2DM的诊断价值。结果与对照组比较,非T2DM组、T2DM组收缩压、舒张压、脉压及甘油三酯、胆固醇、空腹血糖水平均明显升高(P<0.05);与非T2DM组比较,T2DM组甘油三酯、胆固醇、空腹血糖水平均明显升高(P<0.05)。血清sP-selectin、sE-selectin水平及空腹胰岛素水平、胰岛素抵抗指数在对照组<非T2DM组<T2DM组,且两两比较,差异均有统计学意义(P<0.05)。Pearson相关分析结果显示,T2DM患者血清sP-selectin、sE-selectin水平与空腹胰岛素水平、胰岛素抵抗指数均呈正相关(P<0.05)。ROC曲线分析结果显示,血清sP-selectin、sE-selectin联合诊断EH伴T2DM的曲线下面积优于各指标单独诊断(Z二者联合-sP-selectin=2.099、P=0.036;Z二者联合-sE-selectin=2.998、P=0.003)。结论EH伴T2DM患者血清sP-selectin、sE-selectin水平升高,且与空腹胰岛素水平、胰岛素抵抗指数呈正相关,二者联合检测对EH伴T2DM具有较高的诊断价值。 展开更多

血清NT-proBNP联合T_(p)-T_(e)间期对慢性心力衰竭患者发生恶性心律失常的预测价值分析

目的分析血清N末端B型利钠肽原(NT-proBNP)联合T波峰末间期(T_(p)-T_(e))对慢性心力衰竭患者发生恶性心律失常的预测价值。方法选取2023年2-11月潍坊市人民医院收治的101例慢性心力衰竭患者的临床资料,根据患者住院期间是否发生恶性心... 目的分析血清N末端B型利钠肽原(NT-proBNP)联合T波峰末间期(T_(p)-T_(e))对慢性心力衰竭患者发生恶性心律失常的预测价值。方法选取2023年2-11月潍坊市人民医院收治的101例慢性心力衰竭患者的临床资料,根据患者住院期间是否发生恶性心律失常将其分为发生组(n=26)和未发生组(n=75)。收集两组患者的一般资料、血清NT-proBNP、T_(p)-T_(e)间期,并进行单因素分析与多因素Logistic回归分析,以明确慢性心力衰竭患者发生恶性心律失常的影响因素,并绘制受试者工作特征(ROC)曲线,探讨血清NT-proBNP、T_(p)-T_(e)间期及其联合预测对慢性心力衰竭患者发生恶性心律失常的价值。结果101例慢性心力衰竭患者中,26例发生恶性心律失常,发生率为25.74%(26/101)。单因素分析结果显示,两组的血清胱抑素C(Cys-C)、QRS波群时限比较,组间差异有统计学意义(P<0.05)。发生组的血清NT-proBNP、T_(p)-T_(e)间期均高于未发生组,组间差异有统计学意义(P<0.05)。多因素分析结果显示,血清Cys-C、QRS波群时限、血清NT-proBNP、T_(p)-T_(e)间期是慢性心力衰竭患者发生恶性心律失常的危险因素(P<0.05)。ROC曲线结果显示,血清NT-proBNP、T_(p)-T_(e)间期及两者联合预测在慢性心力衰竭发生恶性心律失常的的曲线下面积分别为0.846、0.843、0.908,敏感度分别为76.90、84.60、76.90,特异度分别为85.30、76.00、94.70。血清NT-proBNP、T_(p)-T_(e)间期联合预测慢性心力衰竭患者发生恶性心律失常的曲线下面积高于两者单独预测。结论血清NT-proBNP、T_(p)-T_(e)间期是评估慢性心力衰竭患者发生恶性心律失常的有效指标,且两者联合能够有效增强预测效能。 展开更多

右美托咪定预处理通过调控p62/Keap1/Nrf2途径减轻脂多糖诱导的大鼠肝脏氧化应激、炎症和铁死亡

目的研究右美托咪定(Dex)对脂多糖(LPS)诱导的肝脏氧化应激、炎症及铁死亡的影响及其可能的机制。方法2022年8月至2023年6月,腹腔注射LPS制备大鼠肝损伤模型。50只雄性Wistar大鼠按随机数字表法分为五组:对照组、Dex组、LPS组、LPS+Dex... 目的研究右美托咪定(Dex)对脂多糖(LPS)诱导的肝脏氧化应激、炎症及铁死亡的影响及其可能的机制。方法2022年8月至2023年6月,腹腔注射LPS制备大鼠肝损伤模型。50只雄性Wistar大鼠按随机数字表法分为五组:对照组、Dex组、LPS组、LPS+Dex组和LPS+Dex+p62活化抑制剂(XRK3F2)组,每组各10只。Kaplan‒Meier法进行生存分析;酶联免疫吸附测定(ELISA)大鼠血清中天冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)、肿瘤坏死因子α(TNF-α)、白细胞介素(IL)-1β和IL-6含量;苏木素-伊红(HE)染色观察大鼠肝组织的形态学变化;流式细胞术检测肝脏组织中活性氧簇含量;丙二醛、谷胱甘肽和总谷胱甘肽(T-GSH)含量,超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)活性及谷胱甘肽/氧化型谷胱甘肽(GSH/GSSG)比值采用试剂盒进行检测;实时荧光定量PCR(RT-qPCR)检测炎症因子、铁死亡及信号通路相关分子mRNA的表达;大鼠肝组织中Fe^(2+)含量用组织铁测定试剂盒检测;透射电镜观察大鼠肝组织细胞中线粒体形态;蛋白质印迹法检测铁死亡及信号通路相关分子蛋白的表达。结果与对照组相比,LPS组大鼠生存率和体质量明显降低,肝组织湿/干比及AST和ALT酶活力显著增加,肝组织形态发生损坏、炎症和坏死细胞增加;相较于LPS组,LPS+Dex组大鼠生存率和体质量明显增加,肝水肿、肝功能受损及肝组织病理学损伤明显减轻。与对照组相比,LPS可导致大鼠肝组织中活性氧簇和丙二醛含量增加,SOD、CAT和GSH-Px酶活性降低,谷胱甘肽和T-GSH含量和GSH/GSSG比值减少;Dex预处理则减弱了LPS诱导的氧化应激参数的改变。与对照组相比,LPS组大鼠血清中TNF-α、IL-1β和IL-6含量和肝组织中TNF-α、IL-1β和IL-6 mRNA表达升高;Dex则降低了LPS诱导的炎症因子的表达和释放。与对照组相比,LPS组大鼠肝组织细胞线粒体皱缩、嵴减少、膜密度增加,Fe^(2+)含量增加,谷胱甘肽过氧化物酶4(GPX4)、溶质载体家族7成员11(SLC7A11)和铁蛋白重链(l FTH1)的mRNA和蛋白表达明显降低,乙酰辅酶A合成酶长链家族成员4(ACSL4)和转铁蛋白受体1(TfR1)的mRNA和蛋白表达水平显著升高;Dex预处理显著改善线粒体受损并抑制铁死亡。与对照组相比,LPS组大鼠肝组织中p62表达(0.44±0.02比1.00±0.05)减少,Kelch样ECH2相关蛋白1(Keap1)(2.98±0.01比1.00±0.04)和核因子E2相关因子2(Nrf2)(2.50±0.06比1.00±0.03)表达升高;Dex预处理则增加了p62和核内Nrf2的表达,减少了Keap1和胞质内Nrf2表达。在用Dex预处理的基础上使用p62抑制剂(XRK3F2)后,与LPS+Dex组相比,Dex的抗氧化应激、抗炎症反应及抗铁死亡作用均明显降低。结论Dex对LPS诱导的大鼠肝损伤的保护作用与其抗氧化应激、抗炎症反应及抗铁死亡效应有关,其作用机制可能是通过调控p62/Keap1/Nrf2途径来实现的。 展开更多

The Prognostic Value of Pathological and Molecular Margins Marked by p53 and eIF4E in Laryngeal Carcinoma

Objective: To study the prognostic value of the pathological margin and molecular margin marked by eIF4E and P53 protein in laryngeal carcinoma. Methods: The prognostic value of pathological and molecular margin was studied in 253 cases and 67 cases respectively, the latter were pathological negative margin chosen from the former. Immunohistochemisty was used to detect the expression of eIF4E and p53 proteins. Results: The rate of pathological, p53 and eIF4E positive margins was 20.2%, 19.4% and 32.8% respectively. The recurrent rate of those with positive margins was higher than that of negative margins, which including pathological margin (70.6% vs 35.1%, P =0.0000), p53 margin (69.2% vs 33.3%, P =0.018) and eIF4E margin (63.6% vs 28.9%, P =0.018); The survival rate of those with negative margins was higher than those with positive margins, including pathological margin (the 5-year cumulative survival rate was 37.52% and 64.37% respectively, P =0.0023), p53 margin (the 5-year cumulative survival rate was 24.62% and 75.69% respectively, P =0.0012) and eIF4E margin (the 5-year cumulative survival rate was 43.31% and 77.52% respectively, P =0.0006). Conclusion: The prognosis of those with both pathological and molecular positive margins was worse than that of the negative margins; Both the eIF4E and p53 were useful markers to pick out the poor prognostic patients from those with pathological negative margin, and the former seemed to be more potential. 展开更多

CYP2C19、ApoE基因多态性检测指导用药对急性脑梗死患者疗效及认知功能的影响

目的:观察细胞色素P450家族成员2C19(CYP2C19)、载脂蛋白E(ApoE)基因多态性检测指导用药对急性脑梗死(ACI)患者疗效及认知功能的影响。方法:选取2022年9月—2024年2月萍乡市人民医院收治的80例ACI患者,依据随机数字表法分为两组,各40例... 目的:观察细胞色素P450家族成员2C19(CYP2C19)、载脂蛋白E(ApoE)基因多态性检测指导用药对急性脑梗死(ACI)患者疗效及认知功能的影响。方法:选取2022年9月—2024年2月萍乡市人民医院收治的80例ACI患者,依据随机数字表法分为两组,各40例。对照组予以常规治疗,试验组予以CYP2C19、ApoE基因多态性检测指导用药。比较两组治疗前后血脂指标[低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)及总胆固醇(TC)]、蒙特利尔认知评估量表(MoCA)、改良Rankin量表(MRS)及美国国立卫生研究院卒中量表(NIHSS)评分,以及疗效及不良反应。结果:试验组总有效率高于对照组,差异有统计学意义(P<0.05);试验组治疗后LDL-C、TC均低于对照组,HDL-C高于对照组,差异均有统计学意义(P<0.05);试验组治疗后MoCA评分高于对照组,MRS、NIHSS评分均低于对照组,差异均有统计学意义(P<0.05);两组不良反应发生率比较,差异无统计学意义(P>0.05)。结论:CYP2C19、ApoE基因多态性检测指导用药有利于更好地改善ACI患者血脂水平及认知功能,提高临床疗效。 展开更多
