BMPRⅡ^(+)neural precursor cells isolated and characterized from organotypic neurospheres:an in vitro model of human fetal spinal cord development 被引量:1

Roof plate secretion of bone morphogenetic proteins(BMPs)directs the cellular fate of sensory neurons during spinal cord development,including the formation of the ascending sensory columns,though their biology is not well understood.Type-ⅡBMP receptor(BMPRⅡ),the cognate receptor,is expressed by neural precursor cells during embryogenesis;however,an in vitro method of enriching BMPRⅡ^(+)human neural precursor cells(hNPCs)from the fetal spinal cord is absent.Immunofluorescence was undertaken on intact second-trimester human fetal spinal cord using antibodies to BMPRⅡand leukemia inhibitory factor(LIF).Regions of highest BMPRⅡ^(+)immunofluorescence localized to sensory columns.Parenchymal and meningeal-associated BMPRⅡ^(+)vascular cells were identified in both intact fetal spinal cord and cortex by co-positivity with vascular lineage markers,CD34/CD39.LIF immunostaining identified a population of somas concentrated in dorsal and ventral horn interneurons,mirroring the expression of LIF receptor/CD118.A combination of LIF supplementation and high-density culture maintained culture growth beyond 10 passages,while synergistically increasing the proportion of neurospheres with a stratified,cytoarchitecture.These neurospheres were characterized by BMPRⅡ^(+)/MAP2ab^(+/–)/βⅢ-tubulin^(+)/nestin^(–)/vimentin^(–)/GFAP^(–)/NeuN^(–)surface hNPCs surrounding a heterogeneous core ofβⅢ-tubulin^(+)/nestin^(+)/vimentin^(+)/GFAP^(+)/MAP2ab^(–)/NeuN^(–)multipotent precursors.Dissociated cultures from tripotential neurospheres contained neuronal(βⅢ-tubulin^(+)),astrocytic(GFAP+),and oligodendrocytic(O4+)lineage cells.Fluorescence-activated cell sorting-sorted BMPRⅡ^(+)hNPCs were MAP2ab^(+/–)/βⅢ-tubulin^(+)/GFAP^(–)/O4^(–)in culture.This is the first isolation of BMPRⅡ^(+)hNPCs identified and characterized in human fetal spinal cords.Our data show that LIF combines synergistically with high-density reaggregate cultures to support the organotypic reorganization of neurospheres,characterized by surface BMPRⅡ^(+)hNPCs.Our study has provided a new methodology for an in vitro model capable of amplifying human fetal spinal cord cell numbers for>10 passages.Investigations of the role BMPRⅡplays in spinal cord development have primarily relied upon mouse and rat models,with interpolations to human development being derived through inference.Because of significant species differences between murine biology and human,including anatomical dissimilarities in central nervous system(CNS)structure,the findings made in murine models cannot be presumed to apply to human spinal cord development.For these reasons,our human in vitro model offers a novel tool to better understand neurodevelopmental pathways,including BMP signaling,as well as spinal cord injury research and testing drug therapies. 展开更多

Modernization,as a specific stage in the historical development of human society,possesses a multi-layered composite structure,with each layer playing both a promoting and inhibiting role in human development and dignity simultaneously.Chinese modernization is a strategic choice made based on lessons learned from various experiences of modernization processes in other countries,reflecting judgments on the benefits and choices on the values of human development and dignity in modernization.It values the promotion of the free and well-rounded development of all individuals as its standard,demanding the full realization of the positive effects of modernization on human development and the overcoming of its negative effects.Human rights,aiming to promote the free and well-rounded development of every person as the ultimate value goal,are not only an intrinsic structural element of the modernization stage but also a social normative tool that restrains the direction of modernization development,aligning closely with the values pursued by Chinese modernization.Chinese modernization poses dual requirements for human rights protection:on the one hand,it demands the affirmation and promotion of the positive effects of modernization on human development and dignity through human rights standards,and on the other hand,it requires the prevention and mitigation of the negative effects of modernization on human development and dignity through human rights standards.There exists a certain tension between these two dimensions,necessitating an appropriate balance and thereby presenting corresponding requirements for China’s approach to human rights protection. 展开更多

This study explores the complex relationship between climate change and human development. The aim is to understand how climate change affects human development across countries, regions, and the global population. Visual analytics were used to examine the impact of various climate change indicators on different aspects of human development. The study highlights the urgent need for climate change action and encourages policymakers to make decisive moves. Climate change adversely affects numerous aspects of daily life, leading to significant consequences that must be addressed through policy changes and global governance recommendations. Key findings include that regions with higher CO2 emissions experience a significantly higher incidence of life-threatening diseases compared to regions with lower emissions. Additionally, higher CO2 emissions correlate with consistent death rates. Increased pollution exposure is associated with a higher prevalence of life-threatening diseases and higher rates of malnutrition. Moreover, greater mineral depletion is linked to more frequent life-threatening diseases, suggesting that industrialization contributes to adverse health effects. These results provide valuable insights for policy and decision-making aimed at mitigating the impact of climate change on human development. 展开更多

Against the backdrop of aggravating resource and environment restriction,it is an inevitable choice to adopt the sustainable development pattern to maximize social well-being within the ecological threshold.From the perspective of ecological well-being performance,ecological footprint per capita and human development index were selected to construct the evaluation index system for sustainable urban development,and DEA applied to calculate the input-output efficiency,so as to analyze the sustainable development level of Shanghai from 1999 to 2012.The results showed that①human development index,ecological footprint per capita of Shanghai grew in step with economic development,but ecological footprint per capita level exceeded the ecological carrying capacity,thus the overall ecological conditions remained in the state of worsening "deficit";②ecological well-being performance of Shanghai in most years remained in the DEA ineffective state,and the scale return had decreased gradually since 2005.In addition,the paper gave pertinent suggestions. 展开更多

Organic carbon isotope and pollen evidence for mangrove development and response to human activity in Guangxi(Southwest China) over the last 140 years 被引量:4

Mangrove degradation must reduce carbon sequestration in recent years, thereby aggravating global warming.Thus, short-term impacts of human activity on mangrove ecosystems are cause for concern from local governments and scientists. Mangroves sediments can provide detailed records of mangrove species variation in the last one hundred years, based on detailed 210 Pb data. The study traced the history of mangrove development and its response to environmental change over the last 140 years in two mangrove swamps of Guangxi, Southwest China. Average sedimentation rates were calculated to be 0.48 cm/a and 0.56 cm/a in the Yingluo Bay and the Maowei Sea, respectively. Chemical indicators(δ13Corg and C:N) were utilized to trace the contribution of mangrove-derived organic matter(MOM) using a ternary mixing model. Simultaneous use of mangrove pollen can help to supplement some of these limitations in diagenetic/overlap of isotopic signatures. We found that vertical distribution of MOM was consistent with mangrove pollen, which could provide similar information for tracing mangrove ecosystems. Therefore, mangrove development was reconstructed and divided into three stages: flourishing, degradation and re-flourishing/re-degradation period. The significant degradation, found in the period of 1968–1998 and 1907–2007 in the Yingluo Bay and the Maowei Sea, respectively, corresponding to a rapid increase of reclamation area and seawall length, rather than climate change as recorded in the region. 展开更多

Artificial Intelligence (AI) experienced significant advancements in recent years, and its potential power is already recognized across various industries. Yet, the rise of AI has led to a growing concern about its impact on meeting the Sustainable Development Goals (SDGs). The aim of this paper was to evaluate contributions and the potential influence of AI to sustainable development in the society domain. Furthermore, the study analyzed GPT-3 responses, as one of the largest language models developed by OpenAI, descriptively. We conducted a set of queries on the SDGs to gather information on GPT-3’s perceptions of AI impact on sustainable development. Analysis of GPT-3’s contribution potential towards the SDGs showcased its broad range of capabilities for contributing to the SDGs in areas such as education, health, and communication. The study findings provide valuable insights into the contributions of AI to sustainable development in the society domain and highlight the importance of proper regulations to promote the responsible use of AI for sustainable development. We highlighted the potential for improvement in neural language processing skills of GPT-3 by avoiding imitating weak human writing styles with more mistakes in longer texts. 展开更多

Objective This study was to examine the relationship between socioeconomic status and the incidence and mortality of non-Hodgkin lymphoma(NHL).Methods We compared the age-standardized incidence rate(ASIR),age-standardized mortality rate(ASMR),and the ASMR to ASIR ratio(MIR)at national and regional levels and studied the correlation between the MIR and the human development index(HDI)in 2012 and 2018.Results The highest ASIR was in North America in 2012 and in Australia in 2018,and the lowest ASIR was in Central and South Asia in both 2012 and 2018.The highest ASMR was in North Africa in both 2012 and 2018,and the lowest ASMR was in Eastern Asia and South-Central Asia in 2012 and in South-Central Asia in 2018.The lowest MIR was in Australia in both 2012 and 2018,and the highest MIR was in Western Africa in both 2012 and 2018.HDI was strongly negatively correlated with MIR(r:−0.8810,P<0.0001,2012;r:−0.8895,P<0.0001,2018).Compared to the 2012 data,the MIR in the intermediate HDI countries significantly deceased and the HDI in low and high HDI countries significantly increased in 2018.Conclusion The MIR is negatively correlated with HDI.Increasing the HDI in low and intermediate HDI countries may reduce the MIR and increase the survival of patients with NHL. 展开更多

Based on an investigation of the meaning of development, the neo-classical economic approach to development, and the post-welfarist theory of development, this paper proposes a conceptual framework for understanding human development potentials, while undertaking empirical analysis using cross-sectional and time series data on human development. Human development is associated with basic necessities for subsistence, the quality of life, and political and civil rights, in addition to income indicators. Our analysis suggests that the concept of human development potentials has two dimensions: the rights of development and limits to human development. Both are largely ignored in the neoclassical theory of development. However, human development is not unbounded, which approaches to a relatively fixed constant at given economic, technological and institutional conditions. The aim of this research was to value, using a multiple regression model, the role of knowledge to guarantee the development in rural areas of European Union countries over 10 years. The main question was to find out relationships among some variable, as the percentage of national Gross Domestic Product (GDP) used to improve the high training, and rural development in terms of agricultural labour units. The results underlined in 2001 as an high value of rural development, in terms of working force in agriculture, was identified in some countries of European Union characterised by a low value both in high training investments and also by a low value of Human Development Index, according to the definition of The Economist. The results in 2010 pointed out an inverse correlation among the dependent variable development in rural areas and the independent variables per capita GDP and national expenditure in advanced training, in percentage of national GDP. The learning by doing and by using, the introduction of advanced training in agriculture, using Long Life Learning measures of European Union, are important to improve the development of European rural areas but, sometimes, these actions are not perceived as something of useful. 展开更多

Mining activity in Brazil has significantly contributed to the country development.However,this contribution is not always fully noticed by society.This study aims to bring more evidence to this mining activity contribution,based on highly regarded development indicators,such as the HDI(Human Development Index),created by the UNDP(United Nations Development Programme).The HD1 was traditionally designed as an instrument to evaluate the degree of countries development and was subsequently deployed to states and municipalities(IDHM(Municipal Human Development Index)for Brazilian municipalities).In addition to lDHM released by the UNDP,FIRJAN(Federation of the Industries of the State of Rio de Janeiro)developed its own IFDM(FIR JAN Municipal Development Index).The statistical analysis shows that the average of the municipalities with mining activities has superior development indices than those with non-mining activities,especially in the two major mining states:Minas Gerais and Parfi. 展开更多

Since reform and opening up in 1978, China has persisted in the central task of economic development and put development in the first place. Such challenges are unique to each country context. The overarching challenge, all above other challenges is, to seek appropriate knowledge as a tool and mean to resolve such challenges. A critical such knowledge to be accumulated in all days is on how to develop people or human resources. However, with the existing knowledge claims in this regard, countries have been unable to resolve such challenges. Human Development, one such knowledge claim, is too broader, and therefore, no guided protocol is suggested in finding solutions. Human Resource Development, as another such knowledge claim is too limited in scope to address broader level challenges. National Human Resource Development (NHRD) has emerged to bridge this gap, emphasising people oriented solutions to resolve country level challenges. The NHRD literature encourages NHRD country case studies, and that, justifications for starting NHRD country case studies is needed. This study justifies such a research on NHRD practices in Malaysia's country context, using content analysis. As a result, this study has shown higher level country challenges, derived a need of people based approach in resolving such challenges, highlighted NHRD as capable as possible in guiding to establish people development knowledge claim, and finally justified an NHRD research to be done within Malaysia's context to see how Malaysia practices NHRD. 展开更多

Climate is one of the environmental conditions on which people live.Climate conditions impact every aspect of people's daily life and production activities,even the survival of humankind.In recent years,human activities have adversely changed the climate.The emergence of global warming and extreme weather has prompted people to pay attention to the impact of climate on life.The adverse climate issues include reduced quality of air and food,facilitated spread of diseases and increased mortality of the population.Being more orientated to climate changes in the past,research has been less alerted to the negative impacts that climate changes could bring to human health.This could be ascribed to that after living in a region for a long time,the human body has"adapted"to climate.And after adaptation,routine health outcomes such as disease incidence and hospitalization rate become less obvious,making the habitants blind from the undesirable impacts of climate on the health status.This review article summarizes the impacts of cold climate on human development,including maternal/fetal factors,lifestyle factors,etc.,and delineates the relationship between cold climate and human health. 展开更多

The response to the COVID-19 pandemic has compelled Institut Pasteur de Cote d’Ivoire (IPCI) to set up strategies for an outstanding mobilisation of human resources to fight against it. Among these strategies, we can quote the screening of people requesting COVID-19 tests at IPCI for emergency travel after the lifting of flight restrictions. Newly recruited Young researchers at IPCI, as well as Ph.D. students and trainees, were mobilised. This document describes firstly, the benefits, knowledge acquired, and difficulties encountered, and secondly, recommendations to be followed for a future large-scale epidemic. This study helped to outline some achievements, such as upgrading the talents of young researchers, strong collaboration between young researchers from different fields, and between IPCI and other institutions. The lessons learned are numerous, including mood and stress management. The difficulties encountered during this study are mainly misunderstandings, increased workload, and insufficient awareness of COVID-19 travel test procedures. These results are relevant for a possible pandemic management in our country, as well as in other African countries. 展开更多
