Meta-analysis of the efficacy and safety of Bushen Huoxue decoction in the treatment of Osteoporosis

Objective:To systematically evaluate the long-term efficacy of Bushen Huoxue Decoction combined with vertebroplasty(PVP or PKP)in the treatment of osteoporotic vertebral compression fractures(OVCF),in order to provide evidence-based reference for clinical application.Methods:To ensure the novelty of research data,a computer search was conducted between 2017 and February 2023 to publicly publish all randomized controlled studies and clinical trials at home and abroad on the treatment of OVCF with Bushen Huoxue Decoction combined with vertebroplasty published in CNKI,Wanfang,Vip,PubMed,CBM,and Cochrane libraries.Two researchers independently conducted literature screening and data extraction,evaluated the quality of randomized controlled trials included one by one according to the Cochrane collaboration network standards,and conducted a meta statistical analysis using RevMan5.3 for studies that met the inclusion criteria.Results:A total of 684 patients were included in 7 randomized controlled trials,including 342 patients in the observation group and 342 patients in the control group,with a ratio of 1:1;The meta-analysis results showed that in the observation group,the overall effective rate[RR=1.30,95%CI(1.14,1.47),P<0.001],visual analog pain(VAS)score[SMD=1.19,95%CI(0.77,1.61),P<0.0001],bone mineral density score[SMD=1.09,95%CI(0.15,2.04),P=0.02],COQOL score[SMD=0.99,95%CI(0.68,1.30),P<0.00001],OPG score[SMD=0.48,95%CI(0.18,0.77),P=0.002]The RANKL score[SMD=1.33,95%CI(1.00,1.65),P<0.0001]was significantly superior to the control group,with statistically significant differences.There was no significant difference in the Oswestry Disability Index(ODI)score[SMD=0.27,95%CI(-0.03,0.57),P=0.08],Cobb score[SMD=1.52,95%CI(-1.05,4.09),P=0.25],and vertebral height score[SMD=0.43,95%CI(-0.14,1.01),P=0.14].Conclusion:The results show that Bushen Huoxue Decoction combined with vertebroplasty has significant advantages in improving bone mineral density and alleviating pain in patients after OVCF,which is significantly superior to using OVCF alone. 展开更多

Effect of Bushen Yiqi Huoxue Recipe on Placental Vasculature in Pregnant Rats with Fetal Growth Restriction Induced by Passive Smoking 被引量:5

Interactions of vascular endothelial growth factor (VEGF) with receptors VEGFR1/Fltl and VEGFR2/Flk1, and those of angiopoietins (Ang-1, Ang-2) with receptor Tie2 play important roles in placental angiogenesis. This study investigated vascular morphology and expression of these angiogenic factors in rat placenta on the day 15, 18, 21 of gestation (D 15, D 18 and D21). The rats were randomly assigned into 3 groups: normal group, model group [fetal growth restriction (FGR) model], and Bushen Tqi Huoxue (BYHR) recipe treatment group (BYHR group, the pregnant rats with FGR were treated with BYHR recipe). Morphological analysis indicated that during initial villous formation, fetal nucle- ated erythrocytes (FNEs) appeared in maternal blood sinus (MBS). Subsequently, FNEs were sur- rounded by endothelial cells to form fetal capillary (FC) and then by trophoblast cells to form villi. As pregnancy proceeded, FC density increased progressively with increasing endothelial identification staining (EIS) in normal and BYHR groups. Whereas, villous formation was suppressed, normal in- crease in FC density was impaired and EIS was weakened in model group. Quantitative PCR analysis showed that VEGF and Flkl mRNA increased over gestation in all groups, indicating that VEGF might play a pivotal role in FC growth during late gestation. VEGF mRNA was increased on D15, while de- creased on D21 in model group as compared with normal group and BYHR group. Immunohistochemi- cally, Ang-2 protein was highly expressed in FNEs, gradually disappeared as villi matured, and decreased over gestation in all groups, indicating that Ang-2 might play a pivotal role in villous formation, which was further supported by decreased Ang-2 mRNA and protein expression in model group on D 15. Ang-1 mRNA, Tie2 mRNA and Ang-1/Ang-2 ratio increased from D15 to D18 in all groups as placenta matured. Ang-1 mRNA, Tie2 mRNA and Ang-1/Ang-2 ratio were decreased on D18 in model group as compared with normal and BYHR groups, indicating delayed maturity of FGR placenta. Alterations in angiogenic factors may result in altered placental vasculature and cause placental insufficiency. BYHR recipe could balance the angiogenic factors to promote the formation and maturation of FGR placental vasculature. 展开更多

The aim of the present study was to explore the effect and mechanism of Bushen Huoxue recipe(BHR) on ovarian reserve in mice with premature ovarian failure(POF). Mice were divided into 3 groups: normal group, model group and BHR group. Intraperitoneal injection of cyclophosphamide was performed to create the POF model. Primordial follicular(PDF) number, ovarian wet weight, ovarian index, and estrous cycle were analyzed to evaluate the effect of BHR on POF. Meanwhile, the m RNA and protein level of Mouse Vasa Homologue(MVH) in the bone marrow, peripheral blood and ovary were detected, to explore the underlying mechanism of the treatment efficacy of BHR on ovarian reserve. By the time of BHR treatment for 28 days, BHR increased the PDF number and shortened the estrous cycle of POF mice. BHR also decreased the m RNA level of MVH in the bone marrow, and increased m RNA and protein level of MVH in the ovary of POF mice. Our results demonstrated a treatment efficacy of BHR on POF mice, and revealed that BHR might repair the dysfunction of germline stem cells in the bone marrow, and thus to improve the ovarian reserve and enhance the ovarian function of POF mice through neo-oogenesis. 展开更多

Objective:To evaluate the effectiveness and safety of Bushen Huoxue prescription in the treatment of recurrent spontaneous abortion(RSA)due to prethrombotic state(PTS).Methods:Databases such as CNKI,WanFang,VIP,CBM,PubMed,Embase,and Cochrane Library were searched for randomized controlled trials on Bushen Huoxue prescription in treating RSA due to PTS from inception to March 2021;meta-analysis was performed by RevMan Version 5.3.0 following quality evaluation.Results:Seven trials were included,with 496 patients;the meta-analysis indicated that Bushen Huoxue prescription has advantages on the improvement of total clinical effective rate[RR=1.22,95%CI(1.10,1.35),Z=3.80(P=0.0001)],embryo survival rate at pregnancy of 12 weeks[RR=1.25,95%CI(1.10,1.41),Z=3.53(P=0.0004)],D-dimer levels[SMD=-1.59,95%CI(-2.20,-0.97),Z=5.07(P<0.00001)],and fibrinogen levels[MD=-1.00,95%CI(-1.29,-0.70),Z=6.61(P<0.00001)],but the statistical heterogeneity was significant;in terms of incidence of adverse reactions,there was no statistical difference between Bushen Huoxue prescription and western medicine.Conclusion:Compared with western medicine alone,Bushen Huoxue prescription alone or in combination with western medicine showed significant advantages in improving the overall clinical efficiency,embryonic survival rate at 12 weeks of pregnancy,and reducing D-dimer values as well as fibrinogen levels,without any significant difference in the incidence of adverse effects;however,the number of included studies is small and there are drawbacks,such as small sample size and low quality;therefore,high-quality clinical studies with large sample size and rigorous trial designs are needed in the future to provide a reliable basis for the effectiveness and safety of TCM in reducing the incidence of RSA due to prethrombotic state. 展开更多

Objective:To evaluate the efficiency of Bushen Huoxue method with ACEI/ARB in treating diabetic kidney disease(stageⅢ).Methods:A total of 8 major electronic databases(CNKI,WanFang,VIP,Sinomed,Pubmed,Embase,Cochrane Liberary,Web of Science)were retrieved since the establishment of the database to October 9,2019.Two reviewers extracted data,and assessed the methodological quality of the included studies.The analysis was made by Stata 15.0 and TSA 0.9 softwares.Results:A total of 10 RCT studies were obtained,including 711 patients with diabetic kidney disease of stageⅢ.Meta-analysis showed that the method of Bushen Huoxue with ACEI/ARB could reduce UAER[WMD=-31.24,95%CI(-42.98,-19.51)],β2-GM[WMD=-92.95,95%CI(-166.05,-19.85)],LDL-C[WMD=-0.19,95%CI(0.19,-0.30)].However,there were no significant effect for HbAlc[WMD=-0.08,95%CI(-0.17,-0.00)],Scr[WMD=-12.96,95%CI(-39.82,13.90)],BUN[WMD=-0.14,95%CI(-0.44,0.17)].The result of TSA indicated that the method of Bushen Huoxue with ACEI/ARB was effective in the treatment of diabetic kidney disease of stageⅢ.Conclusion:The study show that the method of Bushen Huoxue with ACEI/ARB can reduce urinary protein,renal tubular injury and LDL-C in patients with diabetic kidney disease of stageⅢ,and had no effect on HbAlc and renal function.But the conclusion of this study needs further research of high quality. 展开更多

Intervention of Bushen Yizhi formula on cognitive disorder rat and the related mechanism of the protective function on neural network

OBJECTIVE Bushen Yizhi formula,constructed by Prof LIAO Shi-long′research group(Guangzhou University of chinese medicine),is the combination of clinical experience and modern pharmacological research,and mainly focuses on the etiology and pathogenesis of AD for treating both cause and symptoms.In this research,the pharmacodynamic study protocol of Bushen Yizhi formula was designed according to the characteristics and indications function of herbs in the formula,and was conformed to preclinical pharmacological research of traditional Chinese medicine for AD,The Technical Requirements of Pharmacology and Toxicology Research in New Drug Development Enacted by State Food and Drug administration also was guiding principle.METHODS A preliminary randomized double-blind clinical trail with 141 cases of AD showed that the efficiency of Bushen Yizhi formula was 61.9%and the increased mean of MMSE was 3.17,but the specific mechanism remains unclear.This study was aimed to reveal the preventive and therapeutic effect of Bushen Yizhi formula on AD rats and its related mechanism.In this research,forebrain-injected IBO-induced AD rats(cholinergic neuron lesion),senescence accelerated mouse,scopolamineinduced learning and memory deficiency rats(mild cognitive impairment)were all established.The learning and memory ability was tested with Morris water maze.Then formation of age pigment,extent of neuronal loss,activation of astrocytes,content of NTFs and degree of oxidized stress damage were determined by morphology,mmunohistochemical and molecular biology methods.RESULTS As the application of Bushen Yizhi formula,the learning and memory ability in all three groups were significantly improved,the formation of age pigment and the content of ACH in cortex and hippocampus were reduced,the activation of astrocytes and release of inflammatory factor(TNF-αand IL-1β)were inhibited,and the antioxidases(CAT,SOD,GSH-PX)were up-regulated and MDA was down-regulated.CONCLUSION Bushen Yizhi formula can prevent and treat AD rats,which might be achieved by anti-inflammatory,antioxidant and protecting cholinergic neuron. 展开更多

While the Bushen Yizhi Formula can treat Alzheimer’s disease(AD),the yet to be ascertained specific mechanism of action was explored in this work.Methods:Different concentrations of the Bushen Yizhi Formula and amyloid-beta peptide(Aβ)were used to treat rat pheochromocytoma cells(P12)and human neuroblastoma cells(SH-SY5Y).Cell morphological changes were observed to determine the in vitro cell damage.Cell Counting Kit(CCK)-8 assay and flow cytometry were employed to identify cell viability and apoptosis/cell cycle,respectively.Western blotting and immunohistochemistry were employed to measure the expressions of endoplasmic reticulum stress(ERS)-related proteins(GRP78 and CHOP),p-IRE1α,IRE1α,ASK1,p-JNK,JNK,Bax,Bcl-2,XBP-1,and Bim.Fura 2-acetoxymethyl ester(Fura-2/AM)was used to determine the intracellular calcium(Ca^(2+))concentration.Also,an AD model was constructed by injecting Aβinto the CA1 area of the hippocampus in Sprague Dawley rats.AD model rats were gavaged with different concentrations of Bushen Yizhi Formula for 14 consecutive days.The Morris water maze experiment was conducted to test the learning and memory of rats.Hematoxylin&Eosin(H&E)and Terminal-deoxynucleotidyl Transferase(TdT)-mediated dUTP Nick-End Labeling(TUNEL)staining were done to determine histopathological changes in the brain.Results:Bushen Yizhi Formula relieved the Aβ-induced effects including cell injury,decreased viability,increased apoptosis,G0/G1 phase cell cycle arrest,upregulation of GRP78,CHOP,p-IRE1α,p-JNK,Bax,XBP-1 and Bim,as well as down-regulation of Bcl-2.These results were also seen with IRE1αsilencing.While Aβsuppressed the learning and memory abilities of rats,the Bushen Yizhi Formula alleviated these effects of Aβ.Brain nerve cell injury induced by Aβcould also be treated with Bushen Yizhi Formula.Conclusion:Bushen Yizhi Formula could influence ERS through the IRE1αsignaling pathway to achieve its therapeutic effects on AD. 展开更多

Mechanism of treating recurrent abortion with “Peiyuan Bushen Antai formula” based on network pharmacology and molecular docking

Objective:To study the effective compound action target signal pathway of Peiyuan bushenan taifang(PYBSATF)has achieved good clinical efficacy in the treatment of recurrent spontaneous abortion(RSA),but its mechanism has not been clarified because of its complex components.In this study,network pharmacology is applied to study the effective compounds,targets and signal pathways of PYBSATF in the treatment of RSA,so as to reveal its pharmacological mechanism of action in the treatment of recurrent spontaneous abortion.Methods:The Traditional Chinese Medicine Systems Pharmacology database(TCMSP)and CNKI are used to obtain the main compounds and drug action targets of PYBSATF.Genecards and the Online Mendelian Inheritance of Man databases(OMIM)are searched to collect the known genes related to RSA,so as to construct compound-target network and screen out the common target proteins and main active compounds.We also use string database to construct a visual protein-protein interaction network(PPI).Cluster Profiler and R software were used to analyze the common targets of drugs and diseases for GO function analysis and KEGG pathway enrichment analysis.Finally,the compound and the protein sequences were conducted according to the node parameters,so that the core protein and core compounds are used to perform molecular docking.Results:186 potential active components and 65 predicted action targets of PYBSATF were screened out.At the same time,1658 genes were also screened out to be closely related to RSA,among which 65genes overlaped with PYBSATF targets and were considered to be related to way of treatment.PPI network showed that VEGFA,IL6,EGFR,MAPK8 and ESR1were the core targets of PYBSATF for the treatment of RSA.GO and KEGG enrichment analysis obtained 93 biological processes of cells(P<0.01)and 87 signaling pathways(P<0.01).PYBSATF played a pharmacological role through a variety of pathways including anti-inflammatory,anti-apoptosis,promoting proliferation and angiogenesis.Molecular docking showed that most active components and key targets of PYBSATF had strong efficiency.Conclusion:Through the study of network pharmacology,it predicted that PYBSATF might treat RSA through multiple targets and multiple signal pathways.Significantly,the predictive targets may be potential targets for treatment of RSA. 展开更多

Objective:To investigate the effect of Qihuang Bushen Xiezhuo Formula on the expression of the nuclear factor E2 related factor 2(Nrf2)/antioxidant response element(ARE)signaling pathway of human glomerular mesangial cells(HMC)in high glucose medium and its protective effect on oxidative stress.Methods:The HMC were cultured in vitro to prepare normal rat serum.The rats were intragastrically administered with irbesartan and Qihuang Bushen Xiezhuo Formula.The serum containing the drugs was prepared after the blood concentration was reached.The rats were divided into the control group,the high glucose group,the irbesartan group and the Qihuang Bushen Xiezhuo Formula group.The HMC of the control group was cultured with normal rat serum(10%serum concentration)medium,while that of the high glucose group was cultured with high glucose medium of rat serum(10%serum concentration).The irbesartan group and the Qihuang Bushen Xiezhuo Formula group were respectively treated with 10%irbesartan and 10%Qihuang Bushen Xiezhuo Formula in serum high glucose medium.After 48 h of culture,the relevant indicators were collected and detected.The mRNA expression levels of Nrf2,gamma-glutamylcysteine synthetase(γ-GCS)and superoxide dismutase(SOD)in each group were detected by real-time PCR.The expressions ofγ-GCS and SOD were detected by immunohistochemistry.The protein expressions ofγ-GCS and SOD in HMC of each group were observed by Western Blot.Results:The expressions of Nrf2,γ-GCS,SOD mRNA and protein in experimental cells of each group were low in the control group,while those in the high glucose group were decreased as compared with the control group(P<0.05).After interference of irbesartan and Qihuang Bushen Xiezhuo Formula the expressions were increased,and the increase in Qihuang Bushen Xiezhuo Formula group was more significant(P<0.05).Conclusion:Qihuang Bushen Xiezhuo Formula can improve HMC oxidative stress injury in high glucose culture,and its mechanism may be achieved by activating Nrf2 and its related downstream proteinsγ-GCS and SOD. 展开更多

目的探讨补肾活血方(BSHX)抑制骨关节炎大鼠软骨细胞凋亡的作用机制。方法将CHON-001细胞随机分为空白组、LPS组(100 ng/mL)、BSHX组(100 ng/mL LPS+20%BSHX含药血清)、BSHX+DMSO组(100 ng/mL LPS+20%BSHX含药血清+0.1%DMSO)、BSHX+Deri... 目的探讨补肾活血方(BSHX)抑制骨关节炎大鼠软骨细胞凋亡的作用机制。方法将CHON-001细胞随机分为空白组、LPS组(100 ng/mL)、BSHX组(100 ng/mL LPS+20%BSHX含药血清)、BSHX+DMSO组(100 ng/mL LPS+20%BSHX含药血清+0.1%DMSO)、BSHX+Derivative 83组(100 ng/mL LPS+20%BSHX含药血清+120μmol/L Derivative 83)。分析各组细胞增殖、凋亡以及上清液中肿瘤坏死因子(TNF)-α、白细胞介素(IL)-6、IL-1β水平;大鼠随机分为假手术组、模型组、BSHX组,检测血清中一氧化氮合成酶(INOS)、IL-6、环氧合酶2(COX2)水平以及软骨组织中CollageⅡ、Aggrecan、ADAMTS-5 mRNA表达水平;并对软骨组织及细胞中Cleaved caspase-3、Bax、Bcl-2及Wnt3a/β-catenin蛋白表达水平进行分析。结果与空白组相比,LPS组细胞凋亡率、TNF-α、IL-6、IL-1β水平、Cleaved caspase-3、Bax、Wnt3a、β-catenin、p-NF-kB p65/NF-kB p65蛋白表达显著增加,24 h、48 h OD450值以及EdU阳性率、Bcl-2表达明显降低(P<0.05);BSHX改善了LPS诱导的CHON-001细胞上述指标(P<0.05),但Derivative 83逆转了BSHX对LPS诱导的CHON-001细胞的保护作用(P<0.05)。动物实验结果显示,与假手术组相比,模型组INOS、IL-6、COX2水平、ADAMTS-5 mRNA、Cleaved caspase-3、Bax、Wnt3a、β-catenin、p-NF-kB p65/NF-kB p65蛋白表达显著增加,CollageⅡmRNA、Aggrecan mRNA、Bcl-2表达明显降低(P<0.05);与模型组相比,BSHX组INOS、IL-6、COX2水平、ADAMTS-5 mRNA、Cleaved caspase-3、Bax、Wnt3a、β-catenin、p-NF-kB p65/NF-kB p65蛋白表达显著降低,CollageⅡmRNA、Aggrecan mRNA、Bcl-2表达明显增加(P<0.05)。结论BSHX抑制骨关节炎大鼠软骨细胞凋亡。 展开更多

目的:观察补肾活血方对抗磷脂抗体阳性反复种植失败(RIF)患者妊娠结局的影响。方法:选取188例抗磷脂抗体阳性RIF患者,按随机数字表法分为补肾活血方组和对照组,两组各94例。两组均给予冻融胚胎移植方案治疗,对照组同时口服拜阿司匹林治... 目的:观察补肾活血方对抗磷脂抗体阳性反复种植失败(RIF)患者妊娠结局的影响。方法:选取188例抗磷脂抗体阳性RIF患者,按随机数字表法分为补肾活血方组和对照组,两组各94例。两组均给予冻融胚胎移植方案治疗,对照组同时口服拜阿司匹林治疗,治疗组在对照组基础上给予补肾活血方治疗。比较两组子宫内膜容受性超声检测指标、子宫内膜容受性分子标志物指标、中医证候积分及临床疗效、安全性。结果:两组基线时子宫内膜厚度及血流参数比较,无统计学差异(P>0.05)。两组胚胎移植前1天子宫内膜厚度及子宫内膜收缩期峰值速度/舒张末期流速(S/D)较基线时升高,子宫内膜搏动指数(PI)、阻力指数(RI)较基线时下降(P<0.05)。治疗组胚胎移植前1天子宫内膜厚度及子宫内膜S/D高于对照组,子宫内膜PI、RI低于对照组(P<0.05)。治疗组种植窗期整合素β3低于对照组,整合素β1、同源框基因11(HoxA11)高于对照组(P<0.05)。两组治疗后中医证候积分较治疗前降低,治疗组更低(P<0.05)。治疗组临床妊娠率62.22%、活产率55.56%高于对照组,胚胎种植率40.27%,流产率10.71%,与对照组比较,无统计学差异(P>0.05)。结论:补肾活血方可改善抗磷脂抗体阳性RIF患者子宫内膜容受性,提高临床妊娠率,降低早期流产率。 展开更多

目的 探讨补肾化浊活血汤联合体外冲击波治疗早中期股骨头坏死的效果。方法 选取2022年1月—2023年1月吉林省吉林中西医结合医院收治的60例早中期股骨头坏死患者进行研究。依据随机数字表法分为对照组(n=30,口服仙灵骨葆胶囊治疗)与治疗... 目的 探讨补肾化浊活血汤联合体外冲击波治疗早中期股骨头坏死的效果。方法 选取2022年1月—2023年1月吉林省吉林中西医结合医院收治的60例早中期股骨头坏死患者进行研究。依据随机数字表法分为对照组(n=30,口服仙灵骨葆胶囊治疗)与治疗组(n=30,补肾化浊活血汤联合体外发散式冲击波治疗)。对比2组患者治疗效果、髋关节功能优良率、股骨头形态及髋关节功能评分。结果 治疗组患者治疗总有效率为93.33%,高于对照组的66.67%,差异有统计学意义(P <0.05)。治疗组患者髋关节功能优良率为90.00%,高于对照组的66.67%,差异有统计学意义(P <0.05)。治疗组治疗后股骨头形态改善有效率为96.67%,高于对照组的70.00%,差异有统计学意义(P <0.05)。治疗后1、6个月,治疗组髋关节功能量表评分(Harris hip score,HHS)分别为(85.66±8.04)分、(83.73±7.93)分,均高于对照组的(77.31±9.99)分、(71.31±8.23)分,差异有统计学意义(P<0.001)。结论 补肾化浊活血汤联合体外冲击波对早中期股骨头坏死治疗效果显著,利于髋关节功能改善。 展开更多

目的探讨补肾活血方通过调节Keap1/Nrf2/ARE通路减轻LPS诱导流产小鼠氧化应激的作用机制。方法将32只ICR孕鼠随机分为空白组、空白+补肾活血方组、模型组和模型+补肾活血方组。采用尾静脉注射LPS法诱导流产模型,同时进行补肾活血方灌胃... 目的探讨补肾活血方通过调节Keap1/Nrf2/ARE通路减轻LPS诱导流产小鼠氧化应激的作用机制。方法将32只ICR孕鼠随机分为空白组、空白+补肾活血方组、模型组和模型+补肾活血方组。采用尾静脉注射LPS法诱导流产模型,同时进行补肾活血方灌胃干预。于妊娠第13天麻醉小鼠后解剖子宫,计算胚胎吸收率。采用HE染色观察蜕膜组织显微结构,ELISA检测外周血SOD和MDA含量,Western blot和RT-qPCR检测蜕膜组织Keap1、Nrf2、HO-1、NQO1和γ-GCS的表达。结果空白组与空白+补肾活血方组各项指标无明显差异。与空白组相比,模型组胚胎吸收率升高(P<0.05),蜕膜组织结构破坏,MDA、Keap1表达上升(P<0.05),SOD、Nrf2、HO-1和NQO1表达下降(P<0.05),γ-GCS表达无显著变化。与模型组相比,补肾活血方干预后胚胎吸收率降低(P<0.05),蜕膜组织结构改善,MDA、Keap1表达水平下降(P<0.05),SOD、Nrf2、HO-1和NQO1表达上升(P<0.05),γ-GCS表达无明显差异。结论补肾活血方可能通过激活Keap1/Nrf2/ARE信号通路减缓过度氧化应激反应,达到治疗RSA的作用。 展开更多

背景:芳香化酶抑制剂尽管显著提高了激素受体阳性乳腺癌患者的临床获益,但其相关的不良事件——骨质疏松严重影响了患者的生活质量,黄芪补肾活血汤能有效预防芳香化酶抑制剂所致骨质疏松的发生,但是其作用机制尚不清楚。目的:探究黄芪... 背景:芳香化酶抑制剂尽管显著提高了激素受体阳性乳腺癌患者的临床获益,但其相关的不良事件——骨质疏松严重影响了患者的生活质量,黄芪补肾活血汤能有效预防芳香化酶抑制剂所致骨质疏松的发生,但是其作用机制尚不清楚。目的:探究黄芪补肾活血汤对芳香化酶抑制剂所致骨质疏松模型小鼠破骨细胞活性的影响及机制。方法:选取60只8周龄C57BL/6J雌性小鼠随机分为假手术组、模型组、黄芪补肾活血汤高、中、低剂量组、阳性对照组各10只,除假手术组外,其余组小鼠均切除双侧卵巢联合皮下注射来曲唑构建绝经后芳香化酶抑制剂所致骨质疏松模型,黄芪补肾活血汤高、中、低剂量组分别给予19.24,9.62,4.81 g/(kg·d)黄芪补肾活血汤进行灌胃(1次/d),阳性对照组给予阿仑膦酸钠5 mg/kg灌胃(1次/周)。给药3个月后,Micro-CT检测胫骨骨密度和骨微结构,对股骨进行苏木精-伊红染色、抗酒石酸酸性磷酸酶染色及免疫组化检测核因子κB受体活化因子配体、骨保护素蛋白表达,ELISA检测血清中Ⅰ型胶原交联羧基端肽、抗酒石酸酸性磷酸酶5b水平。结果与结论:①与假手术组相比,模型组小鼠骨密度显著下降、骨小梁形态疏松断裂、血清中Ⅰ型胶原交联羧基端肽、抗酒石酸酸性磷酸酶5b水平显著上升,表明芳香化酶抑制剂所致骨质疏松模型构建成功;②与模型组相比,黄芪补肾活血汤高、中、低剂量组和阳性对照组小鼠骨密度、骨微结构显著改善,骨小梁形态增粗致密,血清中Ⅰ型胶原交联羧基端肽、抗酒石酸酸性磷酸酶5b水平显著下降,破骨细胞数量减少,核因子κB受体活化因子配体蛋白表达下降,骨保护素蛋白表达升高。结果表明,黄芪补肾活血汤可能调控核因子κB受体活化因子配体/核因子κB受体活化因子/骨保护素信号通路抑制破骨细胞活性,改善骨小梁形态和骨微结构,提高骨密度,进而预防芳香化酶抑制剂所致骨质疏松模型的发生发展。 展开更多

健脾补肾方对肌萎缩侧索硬化小鼠的干预效果及作用机制

目的探索健脾补肾方对肌萎缩侧索硬化(ALS)模型小鼠运动功能的影响及其机制。方法将12只采取转基因法繁育的保留部分运动功能的ADAR2flox/flox/VAChT-Cre小鼠(AR2小鼠)随机分为模型组和治疗组,每组6只;以6只同批次野生型(WT)小鼠为对照... 目的探索健脾补肾方对肌萎缩侧索硬化(ALS)模型小鼠运动功能的影响及其机制。方法将12只采取转基因法繁育的保留部分运动功能的ADAR2flox/flox/VAChT-Cre小鼠(AR2小鼠)随机分为模型组和治疗组,每组6只;以6只同批次野生型(WT)小鼠为对照组。治疗组小鼠灌胃给予健脾补肾方[30 g/(kg·d)],对照组和模型组小鼠灌胃给予0.2 mL氯化钠溶液(9 g/L),每日1次,连续干预8周。每周观察小鼠活动状况、体质量以及抓力、抗疲劳能力(滚轮测试)等行为学功能。第24周后取样,采用苏木精-伊红(HE)染色法、尼氏染色法观察各组小鼠脊髓前角组织病理变化,采用Western blot法检测各组小鼠脊髓组织中作用于RNA的腺苷脱氨酶2(ADAR2)、转录反应DNA结合蛋白-43(TDP-43)的蛋白表达量,透射电镜观察小鼠脊髓组织超微结构。结果与对照组比较,模型组小鼠体质量随周龄逐渐增长;与模型组比较,治疗组小鼠体质量降低。与对照组比较,模型组小鼠与治疗组小鼠抗疲劳程度均有所下降,但是治疗组小鼠的抗疲劳能力下降相对模型组有所延缓。与对照组比较,模型组小鼠抓力下降;与模型组比较,治疗组抓力下降相对减缓。与对照组相比,模型组TDP-43表达量显著降低(P<0.001);与模型组相比,治疗组TDP-43表达量显著升高(P<0.05)。与对照组相比,模型组ADAR2表达量显著降低(P<0.01),治疗组与模型组之间ADAR2表达量没有显著差异(P>0.05)。与对照组相比,模型组脊髓前角运动神经元损伤严重,神经元结构不完整;与模型组相比,治疗组神经细胞凋亡数量减少。透射电镜下,模型组与对照组比较出现明显的线粒体肿胀凋亡、髓鞘脱散现象;治疗组与对照组相比较存在部分髓鞘脱散、线粒体肿胀,相对于模型组,存在肿胀凋亡现象的神经细胞数量有所减少。结论健脾补肾方能延缓ALS模型小鼠运动功能的减退,减少脊髓运动神经元的凋亡和损伤,阻止散发性ALS(sALS)病理异常标志物TDP-43的生成,保护脊髓神经元有髓神经纤维与线粒体,有效延缓ALS小鼠疾病的进展与功能的丧失。 展开更多

目的观察补肾化瘀方对宫腔粘连(IUA)大鼠子宫内膜形态学、纤维化程度和纤维化标志物(TGF-β1、Smad2、Smad3和Smad7)表达的影响,探讨其改善IUA大鼠纤维化的作用机制。方法将42只SPF级雌性SD大鼠随机分成空白对照组(9只)和造模组(33只),... 目的观察补肾化瘀方对宫腔粘连(IUA)大鼠子宫内膜形态学、纤维化程度和纤维化标志物(TGF-β1、Smad2、Smad3和Smad7)表达的影响,探讨其改善IUA大鼠纤维化的作用机制。方法将42只SPF级雌性SD大鼠随机分成空白对照组(9只)和造模组(33只),造模组采用双重损伤法建立并验证IUA大鼠模型,将造模成功的大鼠随机均分为4组,即模型对照组和药物组(补肾化瘀方低、中、高剂量组),每组8只。相应药物干预8周后,取大鼠子宫组织,肉眼观察子宫大体形态,HE染色法观察子宫内膜形态学变化,记录子宫内膜厚度及腺体数量,Masson染色法检测子宫内膜胶原纤维面积比;Western Blot及q-PCR法检测子宫内膜组织TGF-β1、Smad2、Smad3、Smad7蛋白及mRNA表达水平。结果与空白对照组比较,模型对照组大鼠子宫形态不规则,色泽暗淡,出现一定程度的迂曲、僵直和水肿,局部可见子宫积液;子宫内膜厚度下降(P<0.01)且厚薄不均,上皮细胞排列紊乱,腺体稀少(P<0.01)且结构变形,胶原纤维面积比明显增加(P<0.01);子宫组织中TGF-β1、Smad2、Smad3蛋白及mRNA相对表达显著升高,Smad7蛋白及mRNA相对表达显著降低(P<0.01)。与模型对照组比较,补肾化瘀方中、高剂量组大鼠子宫形态、色泽明显好转,子宫内膜厚度和腺体计数增加,胶原纤维面积比下降(P<0.01);子宫组织中TGF-β1、Smad2、Smad3蛋白及mRNA相对表达显著降低,Smad7蛋白和mRNA相对表达显著升高(P<0.01)。结论补肾化瘀方一定程度上可降低纤维化标志物TGF-β1、Smad2、Smad3的表达量,提高Smad7的表达量。补肾化瘀方可能通过TGF-β1/Smads信号通路来改善IUA大鼠纤维化损伤程度,促进IUA大鼠子宫内膜的修复。 展开更多
