AIM: To investigate whether DNA-dependent activator of interferon-regulatory factors (DAI) inhibits hepatitis B virus (HBV) replication and what the mechanism is. METHODS: After the human hepatoma cell line Huh7 was cotransfected with DAI and HBV expressing plas- mid, viral protein (HBV surface antigen and HBV e an- tigen) secretion was detected by enzyme-linked immu- nosorbent assay, and HBV RNA was analyzed by real- time polymerase chain reaction and Northern blotting, and viral DNA replicative intermediates were examined by Southern blotting. Interferon regulatory factor 3 (IRF3) phosphorylation and nuclear translocation were analyzed via Western blotting and immunofluorescence staining respectively. Nuclear factor-KB (NF-KB) activity induced by DAI was detected by immunofluorescence staining of P65 and dual luciferase reporter assay. Tran- swell co-culture experiment was performed in order to investigate whether the antiviral effects of DAI were dependent on the secreted cytokines. RESULTS: Viral protein secretion was significantly re- duced by 57% (P 〈 0.05), and the level of total HBV RNA was reduced by 67% (P 〈 0.05). The viral core particle-associated DNA was also dramatically down- regulated in DAI-expressing Huh7 cells. Analysis of involved signaling pathways revealed that activation of NF-KB signaling was essential for DAI to elicit antivi- ral response in Huh7 cells. When the NF-KB signaling pathway was blocked by a NF-KB signaling suppressor (I~:B^-SR), the anti-HBV activity of DAI was remarkably abrogated. The inhibitory effect of DAI was indepen- dent of IRF3 signaling and secreted cytokines. CONCLUSION: This study demonstrates that DAI can inhibit HBV replication and the inhibitory effect is asso- ciated with activation of NF-KB but independent of IRF3 and secreted cytokines. 展开更多

A water-soluble metallo-supramolecular polymer MSP-f-6Np,which possesses a regular pore aperture of 1.4 nm,has been assembled from a structurally flexible naphthalene-appended[Ru(bipy)3]^2+complex and cucurbit[8]uril.As the first periodic metallo-supramolecular polymer formed by a flexible building block,MSP-f-6Np exhibits a hydrodynamic diameter of 122 and 164 nm at 0.1 and 2.0 mM of the monomer concentrations.Synchrotron small angle X-ray scattering experiments confirm that MSP-f-6Np possesses porosity periodicity in both the solution and solid states.Compared with a control,the new highly ordered porous system displays enhanced photocatalytic activity for the degradation of organic dyes. 展开更多

Photodynamic therapy(PDT) has been gaining popularity in both scientific research and clinic applications due to its non-invasiveness and spatiotemporal targeting properties. Nevertheless, the local hypoxic microenvironment in tumor tissue impedes PDT universality. To overcome this drawback, a 2-pyridonebearing BODIPY photosensitizer was synthesized rationally and introduced to polyethyleneglycol-bpoly(aspartic acid) to form a photosensitizer-^(1)O_(2) generation, storage/release agent dual-loading system(PEG-b-PAsp-BODIPY). The investigation of the PDT effect at different illumination conditions in vitro and in vivo revealed that the system tremendously inhibited tumor proliferation, indicating that this new PEG-b-PAsp-BODIPY could act as a potentially effective photo therapeutic system for cancer therapeutics. 展开更多

Here we use nor-seco-cucurbit[10]uril(ns-CB[10]) based ternary complexation to construct [5]rotaxane,linear supramolecular dynamic rotaxane polymers and cubic 3D supramolecular organic framework.A [5]rotaxane is constructed by ns-CB[10], TMe CB[6] and short linear derivatives of 4,4'-bipyridinium(M2). ns-CB[10], CB[7] and long linear derivatives of 4,4'-bipyridinium(M3) self-assemble into a linear supramolecular dynamic rotaxane polymer. ns-CB[10] and tetracationic tetrahedral monomer selfassemble and form a three-dimensional supramolecular organic framework. The above results demonstrate that ns-CB[10]-based ternary complexation is a versatile platform to build various supramolecular systems. 展开更多

A porous supramolecular framework has been for the first time revealed to undergo interpenetration in crystal and noninterpenetration in solution.A new supramolecular organic framework Bu-SOF has been constructed from the co-assembly of a tetracationic tetrahedral monomer and cucurbit[8]uril(CB[8])in water through the encapsulation of two anti-parallel n-butyl chains by CB[8].X-ray diffraction analysis reveals that Bu-SOF forms 3-fold interpenetrated networks in crystals grown by evaporation of its solution in water.1 H NMR,dynamic light scattering and isothermal titration calorimetric experiments confirm that Bu-SOF is also formed in water.Solid samples,prepared by lyophilizing the aqueous solution of Bu-SOF,can adsorb nanoscaled organic dyes,supporting the porosity of the framework and thus non-interpenetration in solution.The avoidance of interpenetration of Bu-SOF in solution has been attributed to the filling of water inside the porous framework as well as the electrostatic repulsion of the appended bipyridinium units of the tetrahedral building block. 展开更多
