A rapid and sensitive method for analyzing trace b-blockers in complex biological samples,which involved magnetic solid-phase extraction(MSPE)coupled with Fourier transform ion cyclotron resonance mass spectrometry(FT...A rapid and sensitive method for analyzing trace b-blockers in complex biological samples,which involved magnetic solid-phase extraction(MSPE)coupled with Fourier transform ion cyclotron resonance mass spectrometry(FTICR-MS),was developed.Novel nanosilver-functionalized magnetic nanoparticles with an interlayer of poly(3,4-dihydroxyphenylalanine)(polyDOPA@Ag-MNPs)were synthesized and used as MSPE adsorbents to extract trace b-blockers from biological samples.After extraction,the analytes loaded on the polyDOPA@Ag-MNPs were desorbed using an organic solvent and analyzed by FTICR-MS.The method was rapid and sensitive,with a total detection procedure of less than 10 min as well as limits of detection and quantification in the ranges of 3.5-6.8 pg/mL and 11.7-22.8 pg/mL,respectively.The accuracy of the method was also desirable,with recoveries ranging from 80.9%to 91.0%following the detection of analytes in human blood samples.All the experimental results demonstrated that the developed MSPE-FTICR-MS method was suitable for the rapid and sensitive analysis of trace b-blockers in complex biological samples.展开更多

Vector beams have attracted widespread attention because of their unique optical properties;in particular,their combination with tight focusing can produce many interesting phenomena.The rise of 3D printing technology...Vector beams have attracted widespread attention because of their unique optical properties;in particular,their combination with tight focusing can produce many interesting phenomena.The rise of 3D printing technology provides more possibilities for exploration.In this work,a cascading method involving a metamaterial and a metalens is used to generate a tightly focused field of vector beams in the terahertz band,which is prepared via 3D printing.As a proof-of-concept demonstration,a series of metamaterial modules capable of generating states of different orbital angular momentum are proposed by cascading with a metalens.The experimental results are in good agreement with the simulation results,fully verifying the feasibility of the scheme.The proposed design and fabrication strategy provides a new idea for the tight focusing of terahertz vector beams.展开更多

Tumor necrosis factor a (TNF-a) is a proinflammatory cytokine extensively involved in immune system regulation, cell proliferation and cell survival. However, the effects and mechanisms for the role of TNF-a in laryngeal cancer remain unclear. Here, we demonstrated using western blot analyses and intracellular Ca〉 concentration measurements that TNF-a treatment suppressed both TRPP2 expression and ATP-induced Ca2+ release in a laryngeal cancer cell line (Hep-2). Knockdown of TRPP2 by a specific siRNA significantly decreased ATP-induced Ca2+ release and abolished the effect of TNF-a on the ATP-induced Ca2+ release. TNF-a treatment also enhanced Hep-2 cell proliferation and growth, as determined using cell counting and flow cytometry cell cycle assays. Moreover, TNF-a treatment down-regulated phosphorylated protein kinase R-like endoplasmic reticulum kinase (p-PERK) and phosphorylated eukaryotic translation initiation factor (p-elF2c0 expression levels, without affecting PERK and elF2ct expression levels in Hep-2 cells. We concluded that suppressing TRPP2 expression and TRPP2-mediated Ca2+ signaling may be one mechanism underlying TNF〈t-enhanced Hep-2 cell proliferation. These results offer new insights into the mechanisms of TNF-a-mediated laryngeal cancer cell proliferation, and provide evidences showing a potential role of TNF-a in the development of laryngeal cancer.展开更多
