Coxsackie virus A16(CA16) is commonly recognized as one of the main human pathogens of hand-foot-mouth disease(HFMD). The clinical manifestations of HFMD include vesicles of hand, foot and mouth in young children and severe inflammatory CNS lesions. In this study, experimentally CA16 infected tree shrews(Tupaia belangeri) were used to investigate CA16 pathogenesis. The results showed that both the body temperature and the percentages of blood neutrophilic granulocytes / monocytes of CA16 infected tree shrews increased at 4-7 days post infection. Dynamic distributions of CA16 in different tissues and stools were found at different infection stages. Moreover, the pathological changes in CNS and other organs were also observed. These findings indicate that tree shrews can be used as a viable animal model to study CA16 infection. 展开更多

The Sindbis-like virus was first discovered in China in 1986. Its complete genomic sequence consists of more than 11 000 bp encoding more than 3 700 amino acids. It contains a 5' non-transcriptional region (5'-NTR... The Sindbis-like virus was first discovered in China in 1986. Its complete genomic sequence consists of more than 11 000 bp encoding more than 3 700 amino acids. It contains a 5' non-transcriptional region (5'-NTR) in a non-structural region, four non-structural proteins (nsP1, nsP2, nsP3, nsP4) regions, capsids in conserved and non-conserved regions and structural El, E2, E3, 6K regions and a 3' non-transcriptional region (3'-NTR). The Sindbis-IMB was isolated from the blood of a patient suspected to have encephalitis, and was followed by identification and passage. The virus RNA was extracted from virus supernatant in infected cells and the whole genome was divided into 12 fragments; RT-PCR was then performed to amplify the 12 fragments for complete sequencing. The results showed that the whole genomic sequence of Sindbis-IMB consists of 11 717 bp encoding 3 773 amino acids. Homology comparison with other Sindbis-like isolates demonstrated the highest similarity was the YN87448 with a variation of 1% strain isolated in Yunnan Province and the second highest to the SAAR86 strain with a variation of -1.2%. The nucleotide sequence variations were present in non-structural regions, resulting in amino acids K, E, N, R, H, and L in protein sequences in positions 230, 231,443,781, 1 582, and 1746 in the new isolation respectively. Furthermore, three additional amino acids-glutamic acid, serine and alanine-were noted in nsp4 terminus as compared to the YN87448 isolate 展开更多

This paper presents a case history study on the geologic investigation and numerical modeling of areactivated landslide in the County of Santa Clara, California to identify the failure mechanism. Thelandslide occurred on an approximately 18.3-m high, north-facing slope during March 2011. The landslidemeasured about 33.5 m in width and about 51.8 m in length. Along the toe of the slope, a residentialstructure with a swimming pool was built on a cut and fill pad and there are several other structurespresent along the western side of the pad. The landslide occurred immediately to the south of theresidential building and moved northward between the County Road A and the house's side yard. Themovement of the landslide resulted in damaging the west-bound traffic lane of County Road A andencroached onto the paved driveway for the residential property. An investigation was performed toidentify the failure mechanism of the landslide to conclude whether Road A re-alignment by the Countyor prominent cutting performed along the lower portion of the slope by the homeowner during 2000through 2004 contributed to the reactivation of the old landslide deposit. The investigation included sitereconnaissance, reviewing available published geologic information, reviewing site-specific geologic andgeotechnical data developed by other consultants, and performing numerical modeling. The outcomes ofthe investigation indicate that the primary causation for the reactivation and failure of the subject preexistinglandslide is the prominent cutting performed along the lower portion of the slope during 2000through 2004 and water tank cut bench. It is urgent to design and prepare functionalized materials for efficient radionuclides separation. Porous materials are considered excellent candidates for the separation of radionuclides under complex conditions due to their high specific surface areas, tunable pore structures and controllable functionalities. In this review, we summarized the design, preparation and functionalization of porous materials and their application for separation of radionuclides in the past five years, discussed the separation performance and analyzed the structure-activity relationship between various radionuclides and porous materials, and systematically clarified their characterization and mechanism of different type porous materials. We also introduced the detection, irradiation and chemical toxicity of different reflective radionuclides. 展开更多

The tacticity of vinyl polymers is a key factor affecting the properties of materials.Recently,organic Brønsted acids have been demonstrated as effective catalysts for the development of highly stereoselective cationic reversible addition-fragmentation chain transfer(RAFT)polymerizations of vinyl ethers,in which the use of RAFT agents could allow the control the molecular weight and tacticity of polymer products simultaneously.However,the effect of RAFT agents on the tacticity-regulation remains elusive and lacks of investigation.In this study,we synthesized four types of RAFT agents and evaluated their influence in the stereoselective cationic polymerization of isobutyl vinyl ether in the presence of PADI as a Brønsted acid catalyst,which unveils that the Z group of RAFT agents could not only affect the polydispersity of the products,but also exert a profound effect on the stereoselectivity.After extensive screening of the RAFT agents,high stereoregularity(isotacticity,90%m)was obtained when using dithiocarbonate ester-type RAFT agents with a benzyloxy Z group. 展开更多

We previously constructed a herpes simplex virus 1(HSV-1) UL7 mutant virus(M1) and showed that a partial deletion mutation of the UL7 gene led to a lower proliferative rate and an attenuated phenotype. Using the M1 mutant, we further modified the UL41 gene, which encodes another tegument protein, and the latency-associated transcript(LAT) gene. Observations of the resulting mutants with modified UL7 and UL41(M2) or UL7, UL41 and LAT(M3) genes indicated attenuated phenotypes, with lower proliferative ratios in various cells, non-lethal infections in mice and lower viral loads in nervous tissues compared with the wild-type strain. Furthermore, no LAT stable intron could be detected in the trigeminal ganglion of M3-infected animals. The results obtained with the three HSV-1 mutants indicate that the M3 mutant is an attenuated strain with low pathogenicity during both acute and latent infections. Together, the results support the use of the M3 mutant as a candidate for the development of an HSV-1 vaccine. 展开更多

It is important to eliminate lipopolysaccharide(LPS)along with killing bacteria in periprosthetic joint infection(PJI)therapy for promoting bone repair due to its effect to regulate macrophages response.Although natural antimicrobial peptides(AMPs)offer a good solution,the unknown toxicity,high cost and exogenetic immune response hamper their applications in clinic.In this work,we fabricated a nanowire-like composite material,named P@C,by combining chitosan and puerarin via solid-phase reaction,which can finely mimic the bio-functions of AMPs.Chitosan,serving as the bacteria membrane puncture agent,and puerarin,serving as the LPS target agent,synergistically destroy the bacterial membrane structure and inhibit its recovery,thus endowing P@C with good antibacterial property.In addition,P@C possesses good osteoimmunomodulation due to its ability of LPS elimination and macrophage differentiation modulation.The in vivo results show that P@C can inhibit the LPS induced bone destruction in the Escherichia coli infected rat.P@C exhibits superior bone regeneration in Escherichia coli infected rat due to the comprehensive functions of its superior antibacterial property,and its ability of LPS elimination and immunomodulation.P@C can well mimic the functions of AMPs,which provides a novel and effective method for treating the PJI in clinic. 展开更多

Wound healing is one of the major global health concerns in patients with diabetes.Overactivation of pro-inflammatory M1 macrophages is associated with delayed wound healing in diabetes.miR-29ab1 plays a critical role in diabetes-related macrophage inflammation.Hence,inhibition of inflammation and regulation of miR-29 expression have been implicated as new points for skin wound healing.In this study,the traditional Chinese medicine,puerarin,was introduced to construct an injectable and self-healing chitosan@puerarin(C@P)hydrogel.The C@P hydrogel promoted diabetic wound healing and accelerated angiogenesis,which were related to the inhibition of the miR-29 mediated inflammation response.Compared to healthy subjects,miR-29a and miR-29b1 were ectopically increased in the skin wound of the diabetic model,accompanied by upregulated M1-polarization,and elevated levels of IL-1βand TNF-α.Further evaluations by miR-29ab1 knockout mice exhibited superior wound healing and attenuated inflammation.The present results suggested that miR-29ab1 is essential for diabetic wound healing by regulating the inflammatory response.Suppression of miR-29ab1 by the C@P hydrogel has the potential for improving medical approaches for wound repair. 展开更多

Herpes simplex virus 1(HSV-1),a member of a herpesviruses,shows a high infectivity rate of 30%-60%in populations of various ages.Some herpes simplex(HSV)vaccine candidates evaluated during the past 20 years have not shown protective efficacy against viral infection.An improved understanding of the immune profile of infected individuals and the associated mechanism is needed.HSV uses an immune evasion strategy during viral replication,and various virus-encoded proteins,such as ICP47 and Vhs,participate in this process through limiting the ability of CD8+cytotoxic T lymphocytes to recognize target cells.Other proteins,e.g.,Us3 and Us5,also play a role in viral immune evasion via interfering with cellular apoptosis.In this work,to study the mechanism by which HSV-1 strain attenuation interferes with the viral immune evasion strategy,we constructed a mutant strain,M5,with deletions in the Us3 and Us5 genes.M5 was shown to induce higher neutralizing antibody titers and a stronger cellular immune response than our previously reported M3 strain,and to prevent virus infection more effectively than the M3 strain in an in vivo mouse challenge test. 展开更多

Polyetheretherketone(PEEK)has been used as an implant material because it has similar mechani-cal properties to natural bone.However,inferior osseointegration and bioinertness hamper the clinical application of PEEK.In this study,the surfaces of sulfonated three-dimensional(3D)PEEK porous structures were loaded with different concentrations of strontium ranelate,a compound commonly used in the treatment or prevention of osteoporosis by promoting bone formation and inhibiting bone resorption.Field-emission scanning electron microscopy was used to characterize the topography of the structures,elemental carbon,oxygen and strontium contents were mea-sured by X-ray photoelectron spectroscopy,and surface zeta potentials and water-contact angle were also measured.The results indicated that strontium ranelate was successfully loaded onto the 3D porous structures.In vitro cellular results showed that strontium ranelate-treated sulfonated PEEK(SP-SR)strengthened the adhesion of MC3T3-E1 cells.The activity of alkaline phosphatase,collagen secretion and extracellular matrix mineralization deposition of MC3T3-E1 cells were also improved on the surface of SP-SR.These results indicate that SP-SR could serve a new implant candidate for surgical treatment. 展开更多
