As one of the most widely distributed and highly conserved transcription factors in eukaryotes,basic leucine zipper proteins(bZIPs)are involved in a variety of biological processes in plants,but they are largely unkno...As one of the most widely distributed and highly conserved transcription factors in eukaryotes,basic leucine zipper proteins(bZIPs)are involved in a variety of biological processes in plants,but they are largely unknown in citrus.In this study,56 bZIP family members were identified genome-wide from an important citrus rootstock,namely trifoliate orange(Poncirus trifoliata L.Raf.),and these putative bZIPs were named PtbZIP1—PtbZIP56.All PtbZIPs were classified into 13 subgroups by phylogenetic comparison with Arabidopsis thaliana bZIPs(AtbZIPs),and they were randomly distributed on nine known(50 genes)chromosomes and one unknown(6 genes)chromosome.Sequence analysis revealed the detailed characteristics of PtPZIPs,including their amino acid length,isoelectric point(pI),molecular weight(MW),predicted subcellular localization,gene structure,and conserved motifs.Prediction of promoter elements suggested the presence of drought,low-temperature,wound,and defense and stress responsive elements,as well as multiple hormone-responsive cis-acting elements.Spatiotemporal expression analysis showed the transcriptional patterns of PtbZIPs in different tissues and under dehydration,high salt,ABA,and IAA treatments.In addition,21 PtbZIPs were predicted to have direct or indirect protein—protein interactions.Among these,PtbZIP49 was experimentally proven to interact with PtbZIP1 or PtbZIP11 by using a yeast two-hybrid assay and bimolecular fluorescence complementation(BiFC).Subcellular localization analysis further revealed that PtbZIP1,PtbZIP11,and PtbZIP49 were localized in the nucleus.Moreover,PtbZIP49 was functionally identified as having an important role in salt stress via ectopic expression in A.thaliana and silenced in trifoliate orange using virus-induced gene silencing(VIGS).This study provided comprehensive information on PtbZIP transcription factors in citrus and highlights their potential functions in abiotic stress.展开更多
Background:The aberrant intraellular expression of a mitochondrial aspartyl tRNA synthetase 2(DARS2)has been reported in human cancers.Nevertheless its critical role and detailed mechanism in lung adenocarcinoma(LUAD)...Background:The aberrant intraellular expression of a mitochondrial aspartyl tRNA synthetase 2(DARS2)has been reported in human cancers.Nevertheless its critical role and detailed mechanism in lung adenocarcinoma(LUAD)remain unexplored.Methods:Initially,The Cancer Genome Atlas(TCGA)based Gene Expression Profiling Interactive Analysis(GEPIA)database (http:/gepia.cancer-pku.cn/)was used to analyze the prognostic relevance of DARS2 expression in LUAD.Further,cell counting kit(CCK)8,immunostaining,and transwell invasion assays in LUAD cell lines in vitro,as well as DARS2 silence on LUAD by tumorigenicity experiments in wivo in nude mice,were performed.Besides,we analyzed the expression levels of p-PI3K(phosphorylated Phosphotylinosital3 kinase),PI3K,AKT(Protein Kinase B),p-AKT(phosphorylated Protein Kinase B),PCNA(proliferating cell nudear antigen),cleaved-caspase 3,E cadherin,and N-cadherin proteins using the Westem blot analysis.Results:LUAD tissues showed higher DARS2 expression compared to normal tissues.Upregulation of DARS2 could be related to Tumor-Node-Metastasis(TNM)stage,high lymph node metastasis,and inferior prognosis.DARS2 silence decreased the proliferation,migration,and invasion abilities of LUAD cells.In addition,the DARS2 downregulation decreased the PCNA and N-cadherin expression and increased cleaved:caspase 3 and E cadherin expressions in LUAD cells,coupled with the inactivation of the PI3K/AKT signaling pathway.Moreover,DARS2 silence impaired the tumonigenicity of LUAD in vivo.Interestingly,let:7b-5p could recognize DARS2 through a complementary sequence.Mechanistically,the increased let 7b 5p expression attenuated the promo oncogenic action of DARS2 during LUAD progression,which were inversely correlated to each other in the LUAD tssues Conclusion:In summary,let 7b-5p,downregulated DARS2 expression,regulating the progression of LUAD cells by the PI3K/AKT signaling pathway.展开更多
Photodynamic therapy(PDT)has limited effects in treating metastatic breast cancer.Immune checkpoints can deplete the function of immune cells;however,the expression of immune checkpoints after PDT is unclear.This stud...Photodynamic therapy(PDT)has limited effects in treating metastatic breast cancer.Immune checkpoints can deplete the function of immune cells;however,the expression of immune checkpoints after PDT is unclear.This study investigates whether the limited e±cacy of PDT is due to upregulated immune checkpoints and tries to combine the PDT and immune checkpoint inhibitor to observe the e±cacy.A metastatic breast cancer model was treated by PDT mediated by hematoporphyrin derivatives(HpD-PDT).The anti-tumor effect of HpD-PDT was observed,as well as CD4þT,CD8þT and calreticulin(CRT)by immunohistochemistry and immunofluorescence.Immune checkpoints on T cells were analyzed byflow cytometry after HpD-PDT.When combining PDT with immune checkpoint inhibitors,the antitumor effect and immune effect were assessed.For HpD-PDT at 100 mW/cm2 and 40,60 and 80 J/cm2,primary tumors were suppressed and CD4þT,CD8þT and CRT were elevated;however,distant tumors couldn't be inhibited and survival could not be prolonged.Immune checkpoints on T cells,especially PD1 and LAG-3 after HpD-PDT,were upregulated,which may explain the reason for the limited HpD-PDT effect.After PDT combined with anti-PD1 antibody,but not with anti-LAG-3 antibody,both the primary and distant tumors were signi-cantly inhibited and the survival time was prolonged,additionally,CD4þT,CD8þT,IFN-þCD4þT and TNF-þCD4þT cells were signi-cantly increased compared with HpD-PDT.HpD-PDT could not combat metastatic breast cancer.PD1 and LAG-3 were upregulated after HpD-PDT.Anti-PD1 antibody,but not anti-LAG-3 antibody,could augment the antitumor effect of HpD-PDT for treating metastatic breast cancer.展开更多
Trade credit,as an effective tool for integrating and coordinating material,information,and financial flows in supply chain management,is becoming increasingly widespread.We explore how a manufacturer can design optim...Trade credit,as an effective tool for integrating and coordinating material,information,and financial flows in supply chain management,is becoming increasingly widespread.We explore how a manufacturer can design optimal trade credit contracts when a risk-averse retailer hides its sales cost information(adverse selection)and selling effort level(moral hazard).We develop incentive models for a risk-averse supply chain when adverse selection and moral hazard coexist,which are then compared with the results under single information asymmetry(moral hazard).Moreover,we analyze the effects of private information and risk-aversion coefficient on contract parameters,selling effort level and the profit or utility of the supply chain.The study shows that when the degree of retailer’s risk aversion is within a certain range,reasonable trade credit contracts designed by the manufacturer can effectively induce the retailer to report its real sales cost and encourage it to exert appropriate effort.Furthermore,we find that the optimal trade credit period,optimal transfer payment,and retailer’s optimal sales effort level under dual information asymmetry are less than those under single information asymmetry.Numerical analysis are conducted to demonstrate the effects of the parameters on decisions and profits.展开更多

To explore differences in leaf morphology between Sapindus mukorossi and Sapindus delavayi,and how the environment might drive these differences,80 germplasm samples from the Sapindus germplasm nursery in Fujian Provi...To explore differences in leaf morphology between Sapindus mukorossi and Sapindus delavayi,and how the environment might drive these differences,80 germplasm samples from the Sapindus germplasm nursery in Fujian Province were selected.The study revealed a wide variation and diversity in 16 germplasm traits,both within and between species grown under the same conditions.On average,the relative contribution of intraspecific variability to total variability was more important(83%) than the relative contribution of interspecific variability(17%).PERMANOVA analysis showed differences in leaflet thickness,length,perimeter,length to width ratio,and leaf hairs or trichome density.Correlation analyses between leaf morphological traits and environmental variables indicated that leaves tended to be larger,longer,and thicker in wetter,warmer,and low-altitude conditions.Our analysis of the relationship between climate and leaf morphology revealed that S.mukorossi had a greater sensitivity to climate variation,particularly in response to mean temperatures of the coldest and warmest seasons,which led to differences in leaf traits and the distribution of the two species.These findings contribute to the understanding of leaf morphology variations in S.mukorossi and S.delavayi,and provide a basis for the collection of Sapindus germplasm resources,their cultivation and use to help address climate change.展开更多
Photodynamic therapy(PDT)not only destroys tumor cells directly but also induced anti-tumor immune response through damage-associated molecular patterns(DAMPs).It is reported that anti-tumor response was associated wi...Photodynamic therapy(PDT)not only destroys tumor cells directly but also induced anti-tumor immune response through damage-associated molecular patterns(DAMPs).It is reported that anti-tumor response was associated with light dose and photosensitizer used in PDT.In this study,4T1 tumor cells were implanted on both the right and left flanks of mice.Only the right tumor was treated by HpD-PDT,while the left tumor was not irradiated.The anti-tumor immune response induced by HpD-PDT was investigated.The expression of DAMPs and costimulatory molecules induced by HpD-PDT were tested by immuno fluorescence and flow cytometry in vivo.Different light doses of PDT were designed to treat 4T1 cells.The killing effect was assessed by CCK-8 kit and apoptosis kit.The expression of DAMPs on 4T1 cells after HpDPDT were evaluated by flow cytometry,western blot and ATP kit.This study showed that CD4^(+)T,CD8^(+)T and the production of IFN-γwere increased significantly on day 10 in righttumor after PDT treatment compared with control group.HpD-PDT enhanced the expression of calreticulin(CRT)on tumor tissue.Importantly,co-stimulatory molecular OX-40 and 4-1BB were elevated on CD8^(+)T cells.In vitro,immunogenic death of 4T1 cells was induced after PDT.Besides,the expression of DAMPs increased with the increasing of energy density.This study indicates that anti-tumor immune effect was induced by HpD-PDT.The knowledge of the involvement of CRT,ATP and co-stimulatory molecules uncovers important mechanistic insight into the anti-tumor immunogenicity.It was the first time that co-stimulatory molecules were investigated and found to elevate after PDT.展开更多

The experiments were carried out at a constant temperature of 523 K, n(H2...The isothermal kinetics of the Fischer-Tropsch synthesis (FTS) over Fe-Cu-K spray-dried catalyst was studied in a spinning basket reactor. The experiments were carried out at a constant temperature of 523 K, n(H2)/n(CO) feed ratios of 0.8 2.0, reactor pressures of 1.1 2.5 MPa, and space velocity of 0.556× 10^-3 Nm^3/kgcat·s. Kinetic model for hydrocarbon formation was derived on the basis of simplified carbide mechanism to reduce the number of parameters. Two individual rate constants for methane and ethene were considered. Furthermore, the model was modified empirically by non-intrinsic effect, such as physisorption and fictitious olefin pressures that were taken into account, and the influences of secondary reaction of α-olefins on product distribution. The simulation results showed that the experimental phenomena of FTS and the deviations from ASF distribution, such as the relatively high yield of methane and low yield of ethene observed experimentally could be depicted basically.展开更多
Sapindus L.species are widely cultivated for biodiesel,biomedical,and biochemical raw materials in southern China.However,yields fuctuate widely due to the lack of high-yielding,high-quality,stable cultivars.Therefore...Sapindus L.species are widely cultivated for biodiesel,biomedical,and biochemical raw materials in southern China.However,yields fuctuate widely due to the lack of high-yielding,high-quality,stable cultivars.Therefore,the objectives of this study were to evaluate three species and one variety among 149 nationwide Sapindus species and screen for elite accessions which would serve as Sapindus breeding materials.Accessions were evaluated on 19 agro-morphological traits by correlation analysis and principal component analysis.These displayed substantial diversity and a broad range of economic traits.In particular,accessions of Sapindus mukorossi and Sapindus rarak had more variation in economic traits than Sapindus delavayi and Sapindus rarak var.velutinus.Increased saponin accumulation may be achieved at the cost of seed oil production.Thirty elite accessions for oil and saponin production,and for comprehensive utilization were screened and accessions no.80,110,and 112 had significant potential to produce high yields.These elite accessions will facilitate the identification of genetic determinants of valuable traits and the effective utilization of trait variability in Sapindus breeding.展开更多

文摘This paper used atomic absorption spectrophotometry to determine the content distribution of Cu and Cd in the soil of Duanzhou District,Zhaoqing City.The single factor index method,Nemerow comprehensive index method,pollution load index method,geoaccumulation index method,and potential ecological hazard index method were used to analyze the content and pollution status of Cu and Cd in the soil of Duanzhou District,providing a basis for understanding the pollution status of Cu and Cd in the soil of Zhaoqing City.

文摘To explore differences in leaf morphology between Sapindus mukorossi and Sapindus delavayi,and how the environment might drive these differences,80 germplasm samples from the Sapindus germplasm nursery in Fujian Province were selected.The study revealed a wide variation and diversity in 16 germplasm traits,both within and between species grown under the same conditions.On average,the relative contribution of intraspecific variability to total variability was more important(83%) than the relative contribution of interspecific variability(17%).PERMANOVA analysis showed differences in leaflet thickness,length,perimeter,length to width ratio,and leaf hairs or trichome density.Correlation analyses between leaf morphological traits and environmental variables indicated that leaves tended to be larger,longer,and thicker in wetter,warmer,and low-altitude conditions.Our analysis of the relationship between climate and leaf morphology revealed that S.mukorossi had a greater sensitivity to climate variation,particularly in response to mean temperatures of the coldest and warmest seasons,which led to differences in leaf traits and the distribution of the two species.These findings contribute to the understanding of leaf morphology variations in S.mukorossi and S.delavayi,and provide a basis for the collection of Sapindus germplasm resources,their cultivation and use to help address climate change.

文摘Photodynamic therapy(PDT)not only destroys tumor cells directly but also induced anti-tumor immune response through damage-associated molecular patterns(DAMPs).It is reported that anti-tumor response was associated with light dose and photosensitizer used in PDT.In this study,4T1 tumor cells were implanted on both the right and left flanks of mice.Only the right tumor was treated by HpD-PDT,while the left tumor was not irradiated.The anti-tumor immune response induced by HpD-PDT was investigated.The expression of DAMPs and costimulatory molecules induced by HpD-PDT were tested by immuno fluorescence and flow cytometry in vivo.Different light doses of PDT were designed to treat 4T1 cells.The killing effect was assessed by CCK-8 kit and apoptosis kit.The expression of DAMPs on 4T1 cells after HpDPDT were evaluated by flow cytometry,western blot and ATP kit.This study showed that CD4^(+)T,CD8^(+)T and the production of IFN-γwere increased significantly on day 10 in righttumor after PDT treatment compared with control group.HpD-PDT enhanced the expression of calreticulin(CRT)on tumor tissue.Importantly,co-stimulatory molecular OX-40 and 4-1BB were elevated on CD8^(+)T cells.In vitro,immunogenic death of 4T1 cells was induced after PDT.Besides,the expression of DAMPs increased with the increasing of energy density.This study indicates that anti-tumor immune effect was induced by HpD-PDT.The knowledge of the involvement of CRT,ATP and co-stimulatory molecules uncovers important mechanistic insight into the anti-tumor immunogenicity.It was the first time that co-stimulatory molecules were investigated and found to elevate after PDT.

文摘The isothermal kinetics of the Fischer-Tropsch synthesis (FTS) over Fe-Cu-K spray-dried catalyst was studied in a spinning basket reactor. The experiments were carried out at a constant temperature of 523 K, n(H2)/n(CO) feed ratios of 0.8 2.0, reactor pressures of 1.1 2.5 MPa, and space velocity of 0.556× 10^-3 Nm^3/kgcat·s. Kinetic model for hydrocarbon formation was derived on the basis of simplified carbide mechanism to reduce the number of parameters. Two individual rate constants for methane and ethene were considered. Furthermore, the model was modified empirically by non-intrinsic effect, such as physisorption and fictitious olefin pressures that were taken into account, and the influences of secondary reaction of α-olefins on product distribution. The simulation results showed that the experimental phenomena of FTS and the deviations from ASF distribution, such as the relatively high yield of methane and low yield of ethene observed experimentally could be depicted basically.

文摘Sapindus L.species are widely cultivated for biodiesel,biomedical,and biochemical raw materials in southern China.However,yields fuctuate widely due to the lack of high-yielding,high-quality,stable cultivars.Therefore,the objectives of this study were to evaluate three species and one variety among 149 nationwide Sapindus species and screen for elite accessions which would serve as Sapindus breeding materials.Accessions were evaluated on 19 agro-morphological traits by correlation analysis and principal component analysis.These displayed substantial diversity and a broad range of economic traits.In particular,accessions of Sapindus mukorossi and Sapindus rarak had more variation in economic traits than Sapindus delavayi and Sapindus rarak var.velutinus.Increased saponin accumulation may be achieved at the cost of seed oil production.Thirty elite accessions for oil and saponin production,and for comprehensive utilization were screened and accessions no.80,110,and 112 had significant potential to produce high yields.These elite accessions will facilitate the identification of genetic determinants of valuable traits and the effective utilization of trait variability in Sapindus breeding.
文摘Introduction Solitary fibrous tumor (SFT) of the kidney is a rare spindle cell neoplasm and all reported SFTs of the kidney were diagnosed through pathological examination and immunohistochemical study after open nephrectomy or open radical nephrectomy. We present a case of SFT of the kidney diagnosed through fine needle core biopsy preoperatively in a 50-year-old female and managed with laparoscopic partial nephrectomy. Due to the difficulty in discriminating between malignant and benign growth pattern of this tumor entity, a regular follow-up after conservative treatment is mandatory.

文摘The soil heavy metals(Hg and As)in Duanzhou District of Zhaoqing City were determined by AFS200 T atomic fluorescence spectropho-tometer,and the soil environment in Duanzhou District was evaluated by several evaluation methods of soil heavy metal pollution,such as single factor index method,pollution load index method and Nemerow comprehensive pollution index method.Finally,according to the data and conclu-sions,the soil pollution situation in Duanzhou District of Zhaoqing was analyzed.The results will let more people pay attention to the changes of the environment and realize the harm of the environment,and the government can formulate a new plan conducive to the coordinated development of the environment and economy.

文摘With the emergence of ambient sensing technologies which combine mobile crowdsensing and Internet of Things,large amount of people-centric data can be obtained and utilized to build people-centric services.Note that the service quality is highly related to the privacy level of the data.In this paper,we investigate the problem of privacy-aware service subscription in people-centric sensing.An efficient resource allocation framework using a combinatorial auction(CA)model is provided.Specifically,the resource allocation problem that maximizes the social welfare in view of varying requirements of multiple users is formulated,and it is solved by a proposed computationally tractable solution algorithm.Furthermore,the prices of allocated resources that winners need to pay are figured out by a designed scheme.Numerical results demonstrate the effectiveness of the proposed scheme.

文摘The family Limoniidae includes a large number of species,including agricultural pests,pollinators,and environmental indicators.Here,the genus Ormosia Rondani,1856,a large genus in the familyLimoniidae,is recorded from Shandong,China for the first time.The species O.(O.)kunyushana Xu,Yang&Kang sp.nov.is described and illustrated as new to science.This species is distinguished fromits congenersmainly by body color and male genitalia.A key to known Chinese Ormosia s.str.species is also provided.
