In this study, rat hippocampal neural pr...The traditional Chinese medicine Jiaweisinisan has antidepressant effects, and can inhibit hypothalamus-pituitary-adrenal gland axis hyperactivity in stress-induced depression. In this study, rat hippocampal neural precursor cells were cultured in serum-free medium in vitro and a stress damage model was established with 120 IJM corticosterone. Cells were treated with 10% (v/v) Jiaweisinisan drug-containing serum and the corticosterone antagonist RU38486. Results of the 3-(4,5-dimethylthiazol-2-yl)-3,5-di-phenytetrazoliumromide assay showed that both Jiaweisinisan drug-containing serum and RU38486 promoted the proliferation of neural precursor cells after corticosterone exposure. Immunofluorescence detection showed that after Jiaweisinisan drug-containing serum and RU38486 treatment, the 5-bromo-2-deoxyuridine/terminal deoxynucleotidyl transferase dUTP nick end labeling ratio in hippocampal neural precursor cells significantly increased, and the apoptotic rates of glial cells reduced, and neuron-like cell differentiation from neural precursor cells significantly increased. Our experimental findings indicate that Jiaweisinisan promotes hippocampal neurogenesis after stress damage.展开更多
Huanglongbing(HLB),which is mainly associated with“Candidatus Liberibacter asiaticus”(CLas),is currently threatening citrus production around the world.This destructive century-old disease results in callose deposit...Huanglongbing(HLB),which is mainly associated with“Candidatus Liberibacter asiaticus”(CLas),is currently threatening citrus production around the world.This destructive century-old disease results in callose deposition in phloem plasmodesmata and starch overaccumulation in leaf chloroplasts.Heat treatment with high humidity has proven to be effective in eliminating the CLas titers in potted citrus trees.This study explored the curative mechanisms and impact of heat treatment(from 32 to 48°C with 75%±5%relative humidity,14h in light and 10h in dark for four days)on mandarin trees(Citrus reticulata Blanco)that were severely infected by CLas using Solexa/Illumina’s digital gene expression(DGE)profiling.A total of 838 differentially expressed genes(DEGs)were identified,of which 510 were downregulated.Proteinprotein interaction analysis of the DEGs indicated that the thermotherapy of HLB trees downregulated the circadian clock related genes in chloroplasts and upregulated the RNA-editing process in the mitochondria,characterized by 141 genes encoding pentatricopeptide repeatcontaining proteins(PPR)being all upregulated.Furthermore,the expression of phenylpropanoid and flavonoid biosynthesis genes,and auxininduced genes were suppressed.The defense response pathway of the thermotherapy treated diseased trees was apparently disrupted,as indicated by the downregulation of LRR receptor-like serine/threonine-protein kinase FLS2 gene.This study improves our understanding of the curative mechanisms of heat treatment on HLB trees and CLas biological regulation.Importantly,this provides new insights on the involvement of RNA modification by PPR genes and changes in the circadian clock after the heat stress treatment of CLas-infected citrus.展开更多

To improve the controllability of the wall-wetting process after the fuel spray-wall impingement in internal combustion engines,the methods of laser etching,chemical etching and surface free energy modification are us...To improve the controllability of the wall-wetting process after the fuel spray-wall impingement in internal combustion engines,the methods of laser etching,chemical etching and surface free energy modification are used to prepare biomimetic structured surfaces with different wettability.The impingement processes of diesel and n-butanol sprays on the walls under different conditions are experimentally investigated.As the surface oleophilicity increases,the spreading radius of wall-impinging sprays decreases.At about 5 s after the fuel injections,the fuel spray droplets hit the walls for the first time,and the secondary breakup and rebound occur.The mixture concentrations of different fuels hitting the various walls reach the peak value.Under a higher surface temperature,the peak value of the mixture concentration is mainly related to the heat flux to the fuel droplets in different boiling regimes from the metal surfaces.The concentration of the air–fuel mixture in the near wall region increases with increasing surface oleophilicity,increasing wall temperature and decreasing ambient pressure.Compared with diesel,n-butanol presents a higher air–fuel mixture concentration in the near wall region.展开更多
Non-damaging ultraviolet B(UV-B)light promotes photomorphogenic development and stress acclimation through UV-B-specific signal transduction in Arabidopsis.UV-B irradiation induces monomerization and nuclear transloca...Non-damaging ultraviolet B(UV-B)light promotes photomorphogenic development and stress acclimation through UV-B-specific signal transduction in Arabidopsis.UV-B irradiation induces monomerization and nuclear translocation of the UV-B photoreceptor UV RESISTANCE LOCUS 8(UVR8).However,it is not clear how the nuclear localization of UVR8 leads to changes in global gene expression.Here,we reveal that nuclear UVR8 governs UV-B-responsive transcriptional networks in concert with several previously known transcription factors,including ELONGATED HYPOCOTYL 5(HY5)and PHYTOCHROME INTERACTING FACTOR 4(PIF4).Based on the transcriptomic analysis,we identify MYB13 as a novel positive regulator in UV-B-induced cotyledon expansion and stress acclimation.MYB13 is UV-B inducible and is predominantly expressed in the cotyledons.Our results demonstrate that MYB13 protein functions as a transcription factor to regulate the expression of genes involved in auxin response and flavonoid biosynthesis through direct binding with their promoters.In addition,photoactivated UVR8 interacts with MYB13 in a UV-B-dependent manner and differentially modulates the affinity of MYB13 with its targets.Taken together,our results elucidate the cooperative function of the UV-B photoreceptor UVR8 with various transcription factors in the nucleus to orchestrate the expression of specific sets of downstream genes and,ultimately,mediate plant responses to UV-B light.展开更多

In DEM generation using interferometric synthetic aperture radar(InSAR),the ground control points(GCPs)for refinement and reflattening are usually selected by manual selection,field surveying,GPS points and existing b...In DEM generation using interferometric synthetic aperture radar(InSAR),the ground control points(GCPs)for refinement and reflattening are usually selected by manual selection,field surveying,GPS points and existing basemaps,which may not be completely suitable for consequent processes.We proposed a new method(auto-PS-GCP)of GCP selection based on permanent scatterers,which automatically defines the thresholds for the coherence,amplitude,and amplitude dispersion index to select permanent scatterer as the GCPs.The GCP thinning(auto-PS-GCP-Thin)was further conducted considering the point density,distances among points and terrain conditions.We used a three-stage assessment that includes:(1)phase stability and intensity of the GCPs,(2)RMSEs of the elevations between GCPs and homonymous points in the reference DEM,and(3)generated DEM accuracy.Three areas respectively in the plain,hilly and mountainous regions were selected to verify the proposed methods.The assessment using both SRTM DEM andICESat-2 points shows that the DEM accuracy of auto-PS-GCP-Thin was improved by 20%∼30%for different areas compared to the manual,where the best DEM accuracy of 4.71 m was found in the plain area.It is concluded that the proposed methods are effective and reliable in various areas with different terrain conditions.展开更多

文摘Super-enhancers are regions of the mammalian genome comprising multiple enhancers in close proximity and often enriched at cancer genes.^(1)Recent studies suggest the emerging roles of super-enhancer-associated genes in un-derlying the cell identity of neuroblastoma(NB)2 which rises from neural crest-derived cells and is one of the most common malignant solid tumors in children.
