Silencing of the long non-coding RNA LINC00265 triggers autophagy and apoptosis in lung cancer by reducing protein stability of SIN3A oncogene

Background:Long non-coding RNAs are important regulators in cancer biology and function either as tumor suppressors or as oncogenes.Their dysregulation has been closely associated with tumorigenesis.LINC00265 is upregulated in lung adenocarcinoma and is a prognostic biomarker of this cancer.However,the mechanism underlying its function in cancer progression remains poorly understood.Methods:Here,the regulatory role of LINC00265 in lung adenocarcinoma was examined using lung cancer cell lines,clinical samples,and xenografts.Results:We found that high levels of LINC00265 expression were associated with shorter overall survival rate of patients,whereas knockdown of LINC00265 inhibited proliferation of cancer cell lines and tumor growth in xenografts.Western blot andflow cytometry analyses indicated that silencing of LINC00265 induced autophagy and apoptosis.Moreover,we showed that LINC00265 interacted with and stabilized the transcriptional co-repressor Switch-independent 3a(SIN3A),which is a scaffold protein functioning either as a tumor repressor or as an oncogene in a context-dependent manner.Silencing of SIN3A also reduced proliferation of lung cancer cells,which was correlated with the induction of autophagy.These observations raise the possibility that LINC00265 functions to promote the oncogenic activity of SIN3A in lung adenocarcinoma.Conclusions:Ourfindings thus identify SIN3A as a LINC00265-associated protein and should help to understand the mechanism underlying LINC00265-mediated oncogenesis. 展开更多

The Metaverse depicts a parallel digitalized world where virtuality and reality are fused.It has economic and social systems like those in the real world and provides intelligent services and applications.In this paper,we introduce the Metaverse from a new technology perspective,including its essence,corresponding technical framework,and potential technical challenges.Specifically,we analyze the essence of the Metaverse from its etymology and point out breakthroughs promising to be made in time,space,and contents of the Metaverse by citing Maslow's Hierarchy of Needs.Subsequently,we conclude four pillars of the Metaverse,named ubiquitous connections,space convergence,virtuality and reality interaction,and human-centered communication,and establish a corresponding technical framework.Additionally,we envision open issues and challenges of the Metaverse in the technical aspect.The work proposes a new technology perspective of the Metaverse and will provide further guidance for its technology development in the future. 展开更多

Many surgeons are developing their skill but it needs experience and patience for successful outcomes. There are different factors which affect the outcome after total knee arthroplasty, among them;the coronal alignment of lower limb plays a major role. Many studies have supported that neutral mechanical alignment after total knee arthroplasty leads to better function. Hybrid-driven Underwater Glider(HUG)is a new type of underwater vehicle which integrates the functions of an Autonomous Underwater Glider(AUG)and an Autonomous Unmanned Vehicle(AUV).Although HUG has the characteristics of long endurance distance,its maneuverability still has room to be improved.This work introduces a new movement form of the neck of the underwater creature into HUG and proposes a parallel mechanism to adjust the attitude angle and displacement of the HUG’s bow,which can improve the steering maneuverability.Firstly,the influence of bow movement and rotation on the hydrodynamic force and flow field of the whole machine is analyzed by using the Computational Fluid Dynamics(CFD)method.The degree of freedom,attitude control range and movement amount of the Movable Bow Mechanism(MBM)are obtained,and then the design of MBM is completed based on these constraints.Secondly,the kinematic and dynamic models of MBM are established based on the closed vector method and the Lagrange equation,respectively,which are fully verified by comparing the results of simulation in Matlab and Adams software,then a Radial Basis Function(RBF)neural network adaptive sliding mode controller is designed to improve the dynamic response effect of the output parameters of MBM.Finally,a prototype of MBM is manufactured and assembled.The kinematic,dynamics model and controller are verified by experiments,which provides a basis for applying MBM in HUGs. 展开更多

Face views are particularly important in person-to-person communication.Differenes between the camera location and the face orientation can result in undesirable facial appearances of the participants during video conferencing.This phenomenon is particularly noticeable when using devices where the frontfacing camera is placed in unconventional locations such as below the display or within the keyboard.In this paper,we take a video stream from a single RGB camera as input,and generate a video stream that emulates the view from a virtual camera at a designated location.The most challenging issue in this problem is that the corrected view often needs out-of-plane head rotations.To address this challenge,we reconstruct the 3D face shape and re-render it into synthesized frames according to the virtual camera location.To output the corrected video stream with natural appearance in real time,we propose several novel techniques including accurate eyebrow reconstruction,high-quality blending between the corrected face image and background,and template-based 3D reconstruction of glasses.Our system works well for different lighting conditions and skin tones,and can handle users wearing glasses.Extensive experiments and user studies demonstrate that our method provides high-quality results. 展开更多
