Objective:To investigate the impact of the preinduced intestinal heat shock protein 70(HSP70)on the visceral hypersensitivity and abnormal intestinal motility in a post-infectious irritable bowel syndrome(PI-IBS) mous...Objective:To investigate the impact of the preinduced intestinal heat shock protein 70(HSP70)on the visceral hypersensitivity and abnormal intestinal motility in a post-infectious irritable bowel syndrome(PI-IBS) mouse model.Methods:Eighty-four female C57BL/6 mice were randomly assigned to four groups:control group(n=21) and induction+PI-IBS group(n=21),PI-IBS group(n=21) and induction group(n=21).The mice in PI-IBS group were infected in vivo with trichinella spiralis by oral administration.The visceral hypersensitivity and intestinal motility were evaluated respectively with abdominal withdrawal reflex and colon transportation test.The intestinal HSP70 protein and mRNA level was measured by Western blot and realtime PCR.Meanwhile,the intestinal proinflammatory cytokines IL-10 and TNF-α level was detected by ELISA.Results:Compared with their counterparts in PI-IBS group,the animals in the Induction+PI-IBS group show significantly increased intestinal level of HSP70 and obviously ameliorative clinical tigurcs.including abdominal withdrawal reflex score,intestine transportation time and Bristol scores(P<0.05).Meanwhile,the intestinal post-inflammatory cytokines remarkably changed,including increased IL-10 level and decreased TNF-αlevel(P<0.05).Conclusions:Intestinal IISP70 may play a potential protective role through improving the imbalance between the intestinal post-inflammatory and anti-inflammatory cytokines in PI-IBS.展开更多

Baill.var.hainanensis Tsiang on an experimental murine colitis model.Methods:Experimental colitis was induced by dextran sulfate sodium(DSS),and mice were divided into 4 groups:control.DSS alone.DSS plus SASP,DSS plus pectic polysaccharides.The disease activity index(DAI) and histological score were observed.The tumor necrosis factor(TNF)-α and interleukin(IL)-17 levels were measured by enzyme-linked immunosorbent assay.I κ B and NF-κB p65 expression were assessed by western blot analysis.Myeloperoxidase(MPO) activity was determined by using MPO assay kit.Re.sults:Administration of pectic polysaccharides significantly reduced the severity of DSS-induced colitis as assessed by DAT and histological score,and resulted in down regulation of MPO activity and NF-κB p65 expression and subsequent degradation of IκB protein,strikingly reduced the production of TNF-α and IL-17.Conclusions:Pectic polysaccharides extracted from Rauvolfia verticillata(Lour.)Baill.var.hainanensis Tsiang exerts beneficial effects in experimental colitis and may therefore provide a useful therapeutic approach for the treatment of UC.展开更多
Objective:To evaluate the expression levels and correlations among the expressions of transforming growth factor-β1(TGF-β1),Kunx3 and CD83 in colonic mucosal specimens from IBS patients.Methods:A total of 40 patie...Objective:To evaluate the expression levels and correlations among the expressions of transforming growth factor-β1(TGF-β1),Kunx3 and CD83 in colonic mucosal specimens from IBS patients.Methods:A total of 40 patients were selected,who were confirmed as IBS by Rome III standard and 40 healthy volunteers served as control.Colonic mucosal specimens of each subject were collected from colon sigmoideum with biopsy forceps.Runx3,TGF-β1?and CD83(the marker for immunecompetent mature dendritic cells(DCs)mRNA in the sigmoid colon tissue were measured by real-time fluorescence quantitative PCR.Results:Compared with the control group,CD83 mRNA expressions were higher in patients with IBS than in healthy controls(P【0.05)and were associated with runt-related transcription factor 3(Runx3)mRNA levels(r=-0.361,P【0.05).Meanwhile,Runx3 mRNA levels were associated with TGF-β1,mRNA expressions in irritable bowel syndrome(IBS)patients(r=0.402,P【0.05).However,there was no correlation between the mRNA expressions of TGF-β1 and CD83(P】0.05).Conclusions:The increase of abnormal dendritic cells might influence the occurrence and development of IBS.TGF-β1 signal pathway might not be involved in Runx 3-regulated maturation of dendritic cells in IBS.展开更多
Objective:To investigate the effects of mechanical stretching and lipopolysaccharide(LPS) on the early apoptosis and IL-8 production of alveolar epithelial type II cells A549.Methods:The experimental matrix consisted ...Objective:To investigate the effects of mechanical stretching and lipopolysaccharide(LPS) on the early apoptosis and IL-8 production of alveolar epithelial type II cells A549.Methods:The experimental matrix consisted of three integrated studies.In the first study,A549 cells were subjected to different stretching strain frequency and duration time to see the effects on the early apoptosis.In the second study,A549 cells were subjected to mechanical stretch(13%4 h, 0.3 Hz) and LPS(1 or 100 ng/mL) to see whether mechanical strain and LPS also have an addictive effect on the early apoptosis.In the third study to investigate whether this addictive effect could be induced by LPS and mechanical stretch on IL-8 production,A549 cells were subjected to LPS(100 ng/mL) and mechanical strain(13%.0.3 Hz,4 h).Real time PCR and enzyme linked immunosorbent assay were used to measure mRN A and protein level of IL-8.The early apoptosis was detected by flow cytometry.Results:Mechanical stretch induced the early apoptosis in a force and frequency and time-dependent manner.In the presence of LPS,mechanical stretch enhanced LPS-induced early apoptosis,especially in 100 ng/mL IPS group compared with 1 ng/ mL LPS and the control group.Mechanical stretch increased IL-8 production and enhanced LPS-induced IL-8 screation both in mRNA and protein levels.Conclusions:Mechanical stretch can induce the early apoptosis and IL-8 secretion.Mechanical stretch and LPS have an addictive effect on the early apoptosis and IL-8 production in alveolar type 2 cells,which is one of the mechanisms of ventilator-induced lung injury.展开更多

Here, we r...Heavy electron-doped FeSe-derived materials have attracted attention due to their uncommon electronic structures with only ‘electron pockets’, and they are different from other iron-based superconductors. Here, we report the crystal structures, superconductivities and normal state properties of two new Li-doped FeSe-based materials, i.e.,Li0.15(C3H10N2)0.32 FeSe(P-4) and Lix(C3H10N2)0.32 FeSe(P4/nmm, 0.25 < x < 0.4) with superconducting transition temperatures ranging from 40 K to 46 K. The determined crystal structures reveal a coupling between Li concentration and the orientation of 1,3-diaminopropane molecules within the largely expanded FeSe layers. Superconducting fluctuations appear in the resistivity of the two superconductors and are fitted in terms of the quasi two-dimensional(2 D) Lawrence–Doniach model. The existence of a crossing point and scaling behavior in the T-dependence of diamagnetic response also suggests that the two superconductors belong to the quasi-2 D system. Interestingly, with the increase of temperature, a sign of Hall coefficient(RH) reversing from negative to positive is observed at ~185 K in both phases, suggesting that‘hole pockets’ emerge in these electron-doped FeSe materials. First principle calculations indicate that the increase in FeSe layer distance will lift up a ‘hole band’ associated with dx2-y2 character and increase the hole carriers. Our findings suggest that the increase in two dimensionalities may lead to the sign-reversal Hall resistivity in Lix(C3H10N2)0.32 FeSe at high temperature.展开更多
Objective:To investigate the mechanism of Yiqi Huoxue traditional Chinese medicine affecting the osteopontin(OPN)level in myocardial tissue and improving ventricular remodeling in heart failure rats after myocardial i...Objective:To investigate the mechanism of Yiqi Huoxue traditional Chinese medicine affecting the osteopontin(OPN)level in myocardial tissue and improving ventricular remodeling in heart failure rats after myocardial infarction.Methods:Forty SPF-grade SD male rats were prepared,and 10 rats were reserved as blank controls.The remaining rats were prepared by anterior descending coronary artery ligation combined with reduced diet and exhausted swimming to prepare a rat model of heart failure after myocardial infarction.The rats in the blank group and the model group were orally administered with distilled water,the Chinese medicine group was administered with Yiqi Huoxue Chinese medicine decoction,and the western medicine group was administered with captopril.After 6 weeks of treatment,small animal ultrasound was used to detect changes in ventricular structure and function in rats.Kill all the rats,and take myocardial tissue,observe the morphological changes of myocardial tissu under a light microscope.Use real-time quantitative PCR to detect the expression of OPN mRNA in rat myocardial tissue,and use immunohistochemical method to detect the expression of OPN protein in myocardial tissue.Results:Compared with the blank group,the left ventricular ejection fraction(EF),left ventricular short axis shortening fraction(FS)of the model group were significantly reduced,and the left ventricular end-diastolic diameter(LVEDD)and left ventricular end-systolic diameter(LVESD)were significantly increased,OPN mRNA and protein expression were significantly up-regulated(P<0.01),and myocardial structure disorder was seen under light microscope.Compared with the model group,the EF and FS of the Chinese medicine group and the Western medicine group were both significantly increased,the LVEDD and LVESD were significantly reduced,the expressions of OPN mRNA and protein were significantly reduced(P<0.01),and the myocardial structure was significantly improved under light microscopy.There was no statistically significant difference between the western medicine group and the traditional Chinese medicine group(P>0.05).Conclusion:Yiqi Huoxue Chinese medicine may reduce the expression of OPN in myocardial tissue,improve ventricular remodeling,improve cardiac function and prevent heart failure after myocardial infarction.展开更多

Before and after the treatment, the patients were scanned with 3.0 T MRE after small intestinal preparation. The imaging characteristic was analyzed and the relevance analysis between the imaging performance and the clinical score were investigated.Results: (1) Before IFX treatment, MRE could clearly detect the intestinal wall and extra luminal lesion in active CD, including intestinal wall multiple segmental thickening and abnormal enhancement, irregular signaling, comb sign, and intestinal stricture or dilation;(2) After IFX treatment, MRE show that intestinal wall thinning and the dilated bowel lumen were improved, and the comb sign disappeared;(3) During artery phase, the signal of the intestinal lesion was improved from high signal or equal signal before the IFX treatment to equal signal or low signal after treatment (P<0.05). During vein phase, there was no significant change of lesion signal before and after treatment (P>0.05).4. the changes of imaging performance were consistant with that of the clinical score (P>0.05).
