Method: SD rat model with acute skin injury were randomly divided into g...Objective: To observe the effect of Zizhu ointment on acute wound healing and its effect on the expression of autophagy proteins LC3 and Becin1. Method: SD rat model with acute skin injury were randomly divided into groups. The experimental group was treated with external application of Zizhu ointment, and the control group was given external saline. The wound healing rate of the two groups was compared at 4d, 7d, 14d and 21d after trauma. The morphology of wounds was observed by HE staining, and the autophagosomes of wounds were observed by transmission electron microscopy. Expression and changes of autophagy marker proteins Beclin-1 and LC3 were detected by immunohistochemistry and WB assay. Result: Compared with the control group, experimental group had a high healing rate, a short healing time, and a faster epithelialization rate. Under the electron microscope, autophagosomes of experimental group were reduced, and the expression of LC3 and Beclin-1 was significantly decreased. Assessment of past-climate simulations of regional climate models(RCMs)is important for understanding the reliability of RCMs when used to project future regional climate.Here,we assess the performance and discuss pos...Assessment of past-climate simulations of regional climate models(RCMs)is important for understanding the reliability of RCMs when used to project future regional climate.Here,we assess the performance and discuss possible causes of biases in a WRF-based RCM with a grid spacing of 50 km,named WRFG,from the North American Regional Climate Change Assessment Program(NARCCAP)in simulating wet season precipitation over the Central United States for a period when observational data are available.The RCM reproduces key features of the precipitation distribution characteristics during late spring to early summer,although it tends to underestimate the magnitude of precipitation.This dry bias is partially due to the model’s lack of skill in simulating nocturnal precipitation related to the lack of eastward propagating convective systems in the simulation.Inaccuracy in reproducing large-scale circulation and environmental conditions is another contributing factor.The too weak simulated pressure gradient between the Rocky Mountains and the Gulf of Mexico results in weaker southerly winds in between,leading to a reduction of warm moist air transport from the Gulf to the Central Great Plains.The simulated low-level horizontal convergence fields are less favorable for upward motion than in the NARR and hence,for the development of moist convection as well.Therefore,a careful examination of an RCM’s deficiencies and the identification of the source of errors are important when using the RCM to project precipitation changes in future climate scenarios.展开更多
BACKGROUND Stem cell therapy has shown great potential for treating diabetic foot(DF).AIM To conduct a bibliometric analysis of studies on the use of stem cell therapy for DF over the past two decades,with the aim of ...BACKGROUND Stem cell therapy has shown great potential for treating diabetic foot(DF).AIM To conduct a bibliometric analysis of studies on the use of stem cell therapy for DF over the past two decades,with the aim of depicting the current global research landscape,identifying the most influential research hotspots,and providing insights for future research directions.METHODS We searched the Web of Science Core Collection database for all relevant studies on the use of stem cell therapy in DF.Bibliometric analysis was carried out using CiteSpace,VOSviewer,and R(4.3.1)to identify the most notable studies.RESULTS A search was conducted to identify publications related to the use of stem cells for DF treatment.A total of 542 articles published from 2000 to 2023 were identified.The United States had published the most papers on this subject.In this field,Iran’s Shahid Beheshti University Medical Sciences demonstrated the highest productivity.Furthermore,Dr.Bayat from the same university has been an outstanding researcher in this field.Stem Cell Research&Therapy is the journal with the highest number of publications in this field.The main keywords were“diabetic foot ulcers,”“wound healing,”and“angiogenesis.”CONCLUSION This study systematically illustrated the advances in the use of stem cell therapy to treat DF over the past 23 years.Current research findings suggested that the hotspots in this field include stem cell dressings,exosomes,wound healing,and adipose-derived stem cells.Future research should also focus on the clinical translation of stem cell therapies for DF.展开更多
We try to use Ho doping combined with band modulation to adjust the thermoelectric properties for BiCuSeO.The results show that Ho doping can increase the carrier concentration and increase the electrical conductivity...We try to use Ho doping combined with band modulation to adjust the thermoelectric properties for BiCuSeO.The results show that Ho doping can increase the carrier concentration and increase the electrical conductivity in the whole temperature range.Although Seebeck coefficient decreases due to the increase of carrier concentration,it still keeps relatively high values,especially in the middle and high temperature range.On this basis,the band-modulation sample can maintain relatively higher carrier concentration while maintaining relatively higher mobility,and further improve the electrical transporting performance.In addition,due to the introduction of a large number of interfaces in the band-modulation samples,the phonon scattering is enhanced effectively and the lattice thermal conductivity is reduced.Finally,the maximal power factor(PF)of 5.18μW·cm^-1K^-2and the dimensionless thermoelectric figure of merits(ZT)of 0.81 are obtained from the 10%Ho modulation doped sample at 873 K.展开更多
To investigate the impact of soil moisture uncertainty on summertime short-range ensemble forecasts(SREFs), a fivemember SREF experiment with perturbed initial soil moisture(ISM) was performed over a northern Chin...To investigate the impact of soil moisture uncertainty on summertime short-range ensemble forecasts(SREFs), a fivemember SREF experiment with perturbed initial soil moisture(ISM) was performed over a northern China domain in summertime from July to August 2014. Five soil moisture analyses from three different operational/research centers were used as the ISM for the ensemble. The ISM perturbation produced notable ensemble spread in near-surface variables and atmospheric variables below 800 h Pa, and produced skillful ensemble-mean 24-h accumulated precipitation(APCP24) forecasts that outperformed any single ensemble member. Compared with a second SREF experiment with mixed microphysics parameterization options, the ISM-perturbed ensemble produced comparable ensemble spread in APCP24 forecasts, and had better Brier scores and resolution in probabilistic APCP24 forecasts for 10-mm, 25-mm and 50-mm thresholds. The ISM-perturbed ensemble produced obviously larger ensemble spread in near-surface variables. It was, however, still under-dispersed, indicating that perturbing ISM alone may not be adequate in representing all the uncertainty at the near-surface level, indicating further SREF studies are needed to better represent the uncertainties in land surface processes and their coupling with the atmosphere.展开更多
The Tibetan Plateau(TP),widely known as the Third Pole,is the highest and largest plateau on Earth.The TP not only plays an important role in influencing the atmospheric circulation,surface energy budget,ecology,weath...The Tibetan Plateau(TP),widely known as the Third Pole,is the highest and largest plateau on Earth.The TP not only plays an important role in influencing the atmospheric circulation,surface energy budget,ecology,weather,and climate but also acts as the Asian Water Tower by holding the largest ice mass outside the polar regions to supply fresh water to over 1.4 billion people downstream in Asia(Qin et al.,2006).The observed historical warming and projected future warming over the TP are much stronger than the corresponding global average(Wang et al.,2008).Glaciers on the TP have been retreating extensively in recent decades,driven by atmospheric warming and circulation changes associated with increasing concentrations of greenhouse gases and aerosols in the atmosphere,light-absorbing particles(e.g.,black carbon,dust)on snow,and other factors(Duan et al.,2012;Qian et al.,2011;Zhao et al.,2020).In addition,due to the thermal and dynam-ical effects of the TP,as well as its proximity to the stratosphere,unique atmospheric boundary layer structures often develop over the TP and in the surrounding areas,which have important impacts on local weather,climate,and mixing and transport of atmospheric constituents(Zhao et al.,2020).The TP is considered a natural laboratory to study multi-sphere inter-actions.A systematic mechanistic understanding of the atmospheric chemical and microphysical processes,climate change,cryospheric variability,and the subsequent environmental impacts is particularly important in the context of the unpreceden-ted warming over the TP(Yao et al.,2019).展开更多

Objective: To test the therapeutic effect of integrated traditional Chinese and western medicine on venous ulcer of lower limbs by Meta analysis method, and then to propose the best clinical treatment scheme. Methods:...Objective: To test the therapeutic effect of integrated traditional Chinese and western medicine on venous ulcer of lower limbs by Meta analysis method, and then to propose the best clinical treatment scheme. Methods: Domestic databases in the past 20 years (January 1999-December 2019) were searched by computer, including China Journal Full-text Database (CNKI), VIP, Wanfang and CBM. Literature was screened according to inclusion and exclusion criteria. Cochrane risk assessment tool was used to evaluate the quality of the included literature. Two-classification method was used to extract data. RevMan 5.3 software was used to make statistical analysis of the data. Results: Eleven articles were finally included, totaling 1,576 patients, 804 cases in the treatment group and 772 cases in the control group. Meta-analysis results show that there is no difference in heterogeneity and bias among the included literatures. The total effective rate (risk ratio [RR]=1.12, 95% confidence interval [CI]=1.09-1.15) of integrated traditional Chinese and western medicine in treating venous ulcer of lower limbs is higher, which is obviously better than that of western medicine alone. Conclusion: Through data analysis, it is concluded that in the study population, the treatment of integrated traditional Chinese and western medicine has a significant effect on venous ulcer of lower limbs. Make up for the deficiency of simple western medicine treatment, improve the cure rate and reduce the recurrence rate. However, there is currently a lack of relevant high-quality literature, so a large sample size, rigorous and standardized experimental design and randomized double-blind clinical study are needed to further test the conclusions.展开更多

Methods: Search the Chinese and English databases of relevant literatures on Traditional Chinese Cutting Therapy, and make statistics and Analysis on the distribution, types, distribution, indications and dominant diseases, the number of cases, clinical efficiency, intervention measures and operation sites of the published literatures. Results: A total of 311 articles were included. There are 40 indications for Traditional Chinese Cutting Therapy at this stage. The dominant diseases are psoriasis, asthma, facial paralysis, rhinitis, neurodermatitis, facial neuritis, chloasma, acne, migraine and insomnia. Traditional Chinese Cutting Therapy is closely related to meridian theory. At present, there are few theoretical and basic studies, the quality of relevant clinical studies is low, and the writing standard of research reports is inadequate. Conclusion: At present, the indications of Traditional Chinese Cutting Therapy are extensive, and the dominant diseases are concentrated, with dermatological diseases and neurological diseases as the main diseases. In the future, we should increase theoretical research and basic research, attach importance to the combination of meridian theory and Traditional Chinese Cutting Therapy, and standardize the implementation of clinical research and the writing of reports.展开更多

Method: 32 SD male rats were randomly divided into three groups including the common ulcer group (N), the diabetic ulcer group (DU) and the model group (M) and Befuji group (B). The stability and practicability of the model were evaluated by HE performed on the local skin tissue of the wounds and ELISA detection detecting the content of High sensitive c-reactive protein (HCRP), basic fibroblast growth factor (bFGF) in rat serum after modeling and HCRP, bFGF, tumor necrosis factor-alpha (TNF-α), advanced glycation end products (AGEs), Interleukin-1β (IL-1β) and Interleukin-6 (IL-6) levels after wound healing. Results: After modeling, the HCRP content in N group was lower than that in the other three groups. There was no significant difference in bFGF content between the groups. After wound healing, the HCRP content in serum of rats in each group showed statistical difference. The level of HCRP in group B was lower than that in group M. There was no difference in bFGF content between the groups. The level of bFGF in group B was higher than that in group N. The contents of AGEs in each group were statistically different. The contents of AGEs in group B were lower than those in group DU and group M. The contents of TNF-α, IL-1βand IL-6 in each group were different. The levels of TNF-α, IL-1βand IL-6 in group B were lower than those in group M. Compared with DU group, the levels of TNF-α, IL-1βin M group were higher . Wound healing time in group M was significantly delayed than that in group N, group DU and group B. Conclusion:This model is more in line with the local environmental characteristics of infected diabetic ulcer and provides a reliable tool for the study of diabetic infections.展开更多

BACKGROUND Shifting from the inflammatory to the proliferative phase represents a pivotal step during managing diabetic foot ulcers(DFUs);however,existing medical interventions remain insufficient.MicroRNAs(miRs)highl...BACKGROUND Shifting from the inflammatory to the proliferative phase represents a pivotal step during managing diabetic foot ulcers(DFUs);however,existing medical interventions remain insufficient.MicroRNAs(miRs)highlight notable capacity for accelerating the repair process of DFUs.Previous research has demonstrated which miR-122-5p regulates matrix metalloproteinases under diabetic conditions,thereby influencing extracellular matrix dynamics.AIM To investigate the impact of miR-122-5p on the transition from the inflammatory to the proliferative stage in DFU.METHODS Analysis for miR-122-5p expression in skin tissues from diabetic ulcer patients and mice was analyzed using quantitative real-time polymerase chain reaction(qRT-PCR).A diabetic wound healing model induced by streptozotocin was used,with mice receiving intradermal injections of adeno-associated virus-DJ encoding empty vector or miR-122.Skin tissues were retrieved at 3,7,and 14 days after injury for gene expression analysis,histology,immunohistochemistry,and network studies.The study explored miR-122-5p’s role in macrophage-fibroblast interactions and its effect on transitioning from inflammation to proliferation in DFU healing.RESULTS High-throughput sequencing revealed miR-122-5p as crucial for DFU healing.qRT-PCR showed significant upregulation of miR-122-5p within diabetic skin among DFU individuals and mice.Western blot,along with immunohistochemical and enzyme-linked immunosorbent assay,demonstrating the upregulation of inflammatory mediators(hypoxia inducible factor-1α,matrix metalloproteinase 9,tumor necrosis factor-α)and reduced fibrosis markers(fibronectin 1,α-smooth muscle actin)by targeting vascular endothelial growth factor.Fluorescence in situ hybridization indicated its expression localized to epidermal keratinocytes and fibroblasts in diabetic mice.Immunofluorescence revealed enhanced increased presence of M1 macrophages and reduced M2 polarization,highlighting its role in inflammation.MiR-122-5p elevated inflammatory cytokine levels while suppressing fibrotic activity from fibroblasts exposed to macrophage-derived media,highlighting its pivotal role in regulating DFU healing.CONCLUSION MiR-122-5p impedes cutaneous healing of diabetic mice via enhancing inflammation and inhibiting fibrosis,offering insights into miR roles in human skin wound repair.展开更多

文摘BACKGROUND Stem cell therapy has shown great potential for treating diabetic foot(DF).AIM To conduct a bibliometric analysis of studies on the use of stem cell therapy for DF over the past two decades,with the aim of depicting the current global research landscape,identifying the most influential research hotspots,and providing insights for future research directions.METHODS We searched the Web of Science Core Collection database for all relevant studies on the use of stem cell therapy in DF.Bibliometric analysis was carried out using CiteSpace,VOSviewer,and R(4.3.1)to identify the most notable studies.RESULTS A search was conducted to identify publications related to the use of stem cells for DF treatment.A total of 542 articles published from 2000 to 2023 were identified.The United States had published the most papers on this subject.In this field,Iran’s Shahid Beheshti University Medical Sciences demonstrated the highest productivity.Furthermore,Dr.Bayat from the same university has been an outstanding researcher in this field.Stem Cell Research&Therapy is the journal with the highest number of publications in this field.The main keywords were“diabetic foot ulcers,”“wound healing,”and“angiogenesis.”CONCLUSION This study systematically illustrated the advances in the use of stem cell therapy to treat DF over the past 23 years.Current research findings suggested that the hotspots in this field include stem cell dressings,exosomes,wound healing,and adipose-derived stem cells.Future research should also focus on the clinical translation of stem cell therapies for DF.

文摘We try to use Ho doping combined with band modulation to adjust the thermoelectric properties for BiCuSeO.The results show that Ho doping can increase the carrier concentration and increase the electrical conductivity in the whole temperature range.Although Seebeck coefficient decreases due to the increase of carrier concentration,it still keeps relatively high values,especially in the middle and high temperature range.On this basis,the band-modulation sample can maintain relatively higher carrier concentration while maintaining relatively higher mobility,and further improve the electrical transporting performance.In addition,due to the introduction of a large number of interfaces in the band-modulation samples,the phonon scattering is enhanced effectively and the lattice thermal conductivity is reduced.Finally,the maximal power factor(PF)of 5.18μW·cm^-1K^-2and the dimensionless thermoelectric figure of merits(ZT)of 0.81 are obtained from the 10%Ho modulation doped sample at 873 K.

文摘To investigate the impact of soil moisture uncertainty on summertime short-range ensemble forecasts(SREFs), a fivemember SREF experiment with perturbed initial soil moisture(ISM) was performed over a northern China domain in summertime from July to August 2014. Five soil moisture analyses from three different operational/research centers were used as the ISM for the ensemble. The ISM perturbation produced notable ensemble spread in near-surface variables and atmospheric variables below 800 h Pa, and produced skillful ensemble-mean 24-h accumulated precipitation(APCP24) forecasts that outperformed any single ensemble member. Compared with a second SREF experiment with mixed microphysics parameterization options, the ISM-perturbed ensemble produced comparable ensemble spread in APCP24 forecasts, and had better Brier scores and resolution in probabilistic APCP24 forecasts for 10-mm, 25-mm and 50-mm thresholds. The ISM-perturbed ensemble produced obviously larger ensemble spread in near-surface variables. It was, however, still under-dispersed, indicating that perturbing ISM alone may not be adequate in representing all the uncertainty at the near-surface level, indicating further SREF studies are needed to better represent the uncertainties in land surface processes and their coupling with the atmosphere.

文摘The Tibetan Plateau(TP),widely known as the Third Pole,is the highest and largest plateau on Earth.The TP not only plays an important role in influencing the atmospheric circulation,surface energy budget,ecology,weather,and climate but also acts as the Asian Water Tower by holding the largest ice mass outside the polar regions to supply fresh water to over 1.4 billion people downstream in Asia(Qin et al.,2006).The observed historical warming and projected future warming over the TP are much stronger than the corresponding global average(Wang et al.,2008).Glaciers on the TP have been retreating extensively in recent decades,driven by atmospheric warming and circulation changes associated with increasing concentrations of greenhouse gases and aerosols in the atmosphere,light-absorbing particles(e.g.,black carbon,dust)on snow,and other factors(Duan et al.,2012;Qian et al.,2011;Zhao et al.,2020).In addition,due to the thermal and dynam-ical effects of the TP,as well as its proximity to the stratosphere,unique atmospheric boundary layer structures often develop over the TP and in the surrounding areas,which have important impacts on local weather,climate,and mixing and transport of atmospheric constituents(Zhao et al.,2020).The TP is considered a natural laboratory to study multi-sphere inter-actions.A systematic mechanistic understanding of the atmospheric chemical and microphysical processes,climate change,cryospheric variability,and the subsequent environmental impacts is particularly important in the context of the unpreceden-ted warming over the TP(Yao et al.,2019).

文摘The incidence of diabetic ulcer is high, the disability and mortality are also high, and its treatment is difficult, and the healing mechanism is not clear. The main reason for the delayed healing of DU is that the chronic inflammation window period is too long, and macrophages play a key role in its healing process. The polarization of macrophages controls the length of the inflammatory phase, and autophagy is thought to be closely related to the tendency of macrophage polarization. Autophagy regulators have limited clinical application. Traditional Chinese medicine may become an ideal autophagy inducer and provide new ideas and strategies for clinical treatment of diabetic ulcers. This article summarizes the role of macrophage autophagic polarization in the healing of diabetic ulcers and the prospects of traditional Chinese medicine.

文摘Objective: To test the therapeutic effect of integrated traditional Chinese and western medicine on venous ulcer of lower limbs by Meta analysis method, and then to propose the best clinical treatment scheme. Methods: Domestic databases in the past 20 years (January 1999-December 2019) were searched by computer, including China Journal Full-text Database (CNKI), VIP, Wanfang and CBM. Literature was screened according to inclusion and exclusion criteria. Cochrane risk assessment tool was used to evaluate the quality of the included literature. Two-classification method was used to extract data. RevMan 5.3 software was used to make statistical analysis of the data. Results: Eleven articles were finally included, totaling 1,576 patients, 804 cases in the treatment group and 772 cases in the control group. Meta-analysis results show that there is no difference in heterogeneity and bias among the included literatures. The total effective rate (risk ratio [RR]=1.12, 95% confidence interval [CI]=1.09-1.15) of integrated traditional Chinese and western medicine in treating venous ulcer of lower limbs is higher, which is obviously better than that of western medicine alone. Conclusion: Through data analysis, it is concluded that in the study population, the treatment of integrated traditional Chinese and western medicine has a significant effect on venous ulcer of lower limbs. Make up for the deficiency of simple western medicine treatment, improve the cure rate and reduce the recurrence rate. However, there is currently a lack of relevant high-quality literature, so a large sample size, rigorous and standardized experimental design and randomized double-blind clinical study are needed to further test the conclusions.

文摘Objective:To to investigate the correlation between the level of immune function and diabetic foot in type 2 diabetic patients.Methods:76 diabetic patients in our hospital from January 2017 to November 2019 were collected as the observation group(the observation group was divided into two subgroups:38 cases in DM group and 38 cases in DM+DF group)and 76 healthy people in the same period as the healthy control group.The level of cellular immunity in serum of the observation group and the healthy control group were compared,and the relationship between cellular immunity and diabetic foot was further explored Department.Results:there were significant differences in glycosylated hemoglobin and fasting blood glucose between the two groups(P<0.001);there were significant differences in CD8,CD25 and CD28 between the two groups(P<0.001).There were statistical differences in the course and age of diabetes between the two subgroups in the observation group(P<0.05);there were statistical differences in CD2,CD8,CD25 and CD28 between the two subgroups(P<0.05).Logistic regression analysis showed that CD8[or95%CI 0.920(0.850,0.995)]and CD25[or95%CI 0.764(0.619,0.943)]were independent influencing factors and risk factors of diabetic foot.Conclusion:the increased levels of CD8 and CD25 may increase the risk of diabetic foot.

文摘Objective: To analyze the advantages of Traditional Chinese Cutting Therapy at the present stage and the characteristics of clinical research and application, so as to provide a basis for clinical treatment and research. Methods: Search the Chinese and English databases of relevant literatures on Traditional Chinese Cutting Therapy, and make statistics and Analysis on the distribution, types, distribution, indications and dominant diseases, the number of cases, clinical efficiency, intervention measures and operation sites of the published literatures. Results: A total of 311 articles were included. There are 40 indications for Traditional Chinese Cutting Therapy at this stage. The dominant diseases are psoriasis, asthma, facial paralysis, rhinitis, neurodermatitis, facial neuritis, chloasma, acne, migraine and insomnia. Traditional Chinese Cutting Therapy is closely related to meridian theory. At present, there are few theoretical and basic studies, the quality of relevant clinical studies is low, and the writing standard of research reports is inadequate. Conclusion: At present, the indications of Traditional Chinese Cutting Therapy are extensive, and the dominant diseases are concentrated, with dermatological diseases and neurological diseases as the main diseases. In the future, we should increase theoretical research and basic research, attach importance to the combination of meridian theory and Traditional Chinese Cutting Therapy, and standardize the implementation of clinical research and the writing of reports.

文摘Objective: It is expected to establish a diabetic ulcer rat model infected with Escherichia coli to provide an appropriate model for the experimental study of diabetic foot ulcer. Method: 32 SD male rats were randomly divided into three groups including the common ulcer group (N), the diabetic ulcer group (DU) and the model group (M) and Befuji group (B). The stability and practicability of the model were evaluated by HE performed on the local skin tissue of the wounds and ELISA detection detecting the content of High sensitive c-reactive protein (HCRP), basic fibroblast growth factor (bFGF) in rat serum after modeling and HCRP, bFGF, tumor necrosis factor-alpha (TNF-α), advanced glycation end products (AGEs), Interleukin-1β (IL-1β) and Interleukin-6 (IL-6) levels after wound healing. Results: After modeling, the HCRP content in N group was lower than that in the other three groups. There was no significant difference in bFGF content between the groups. After wound healing, the HCRP content in serum of rats in each group showed statistical difference. The level of HCRP in group B was lower than that in group M. There was no difference in bFGF content between the groups. The level of bFGF in group B was higher than that in group N. The contents of AGEs in each group were statistically different. The contents of AGEs in group B were lower than those in group DU and group M. The contents of TNF-α, IL-1βand IL-6 in each group were different. The levels of TNF-α, IL-1βand IL-6 in group B were lower than those in group M. Compared with DU group, the levels of TNF-α, IL-1βin M group were higher . Wound healing time in group M was significantly delayed than that in group N, group DU and group B. Conclusion:This model is more in line with the local environmental characteristics of infected diabetic ulcer and provides a reliable tool for the study of diabetic infections.

文摘Diabetic foot is one of the serious complications of diabetes, which formed foot ulcer in the later stage of diabetes mellitus. The treatment of diabetic foot ulcer is difficult, greatly increasing the risk of amputation, which will seriously affect the quality of life of patients. Promoting the healing of ulcer in the early stage has become the guiding ideology for the treatment of diabetic foot ulcer. Macrophages, as important immune cells, play a regulatory role throughout the healing process of diabetic foot ulcers. Therefore, we can provide a new direction for the treatment of diabetic foot ulcers by deeply understanding the mechanism of macrophages and the healing of diabetic foot ulcers.

文摘BACKGROUND Shifting from the inflammatory to the proliferative phase represents a pivotal step during managing diabetic foot ulcers(DFUs);however,existing medical interventions remain insufficient.MicroRNAs(miRs)highlight notable capacity for accelerating the repair process of DFUs.Previous research has demonstrated which miR-122-5p regulates matrix metalloproteinases under diabetic conditions,thereby influencing extracellular matrix dynamics.AIM To investigate the impact of miR-122-5p on the transition from the inflammatory to the proliferative stage in DFU.METHODS Analysis for miR-122-5p expression in skin tissues from diabetic ulcer patients and mice was analyzed using quantitative real-time polymerase chain reaction(qRT-PCR).A diabetic wound healing model induced by streptozotocin was used,with mice receiving intradermal injections of adeno-associated virus-DJ encoding empty vector or miR-122.Skin tissues were retrieved at 3,7,and 14 days after injury for gene expression analysis,histology,immunohistochemistry,and network studies.The study explored miR-122-5p’s role in macrophage-fibroblast interactions and its effect on transitioning from inflammation to proliferation in DFU healing.RESULTS High-throughput sequencing revealed miR-122-5p as crucial for DFU healing.qRT-PCR showed significant upregulation of miR-122-5p within diabetic skin among DFU individuals and mice.Western blot,along with immunohistochemical and enzyme-linked immunosorbent assay,demonstrating the upregulation of inflammatory mediators(hypoxia inducible factor-1α,matrix metalloproteinase 9,tumor necrosis factor-α)and reduced fibrosis markers(fibronectin 1,α-smooth muscle actin)by targeting vascular endothelial growth factor.Fluorescence in situ hybridization indicated its expression localized to epidermal keratinocytes and fibroblasts in diabetic mice.Immunofluorescence revealed enhanced increased presence of M1 macrophages and reduced M2 polarization,highlighting its role in inflammation.MiR-122-5p elevated inflammatory cytokine levels while suppressing fibrotic activity from fibroblasts exposed to macrophage-derived media,highlighting its pivotal role in regulating DFU healing.CONCLUSION MiR-122-5p impedes cutaneous healing of diabetic mice via enhancing inflammation and inhibiting fibrosis,offering insights into miR roles in human skin wound repair.
