Objective The aim of the study was to investigate the effects of Chinese herbal formula Weichang’an(WCA)on the proliferation and mitochondria-mediated apoptosis of human gastric cancer cells.Methods Cell Counting Kit...Objective The aim of the study was to investigate the effects of Chinese herbal formula Weichang’an(WCA)on the proliferation and mitochondria-mediated apoptosis of human gastric cancer cells.Methods Cell Counting Kit-8(CCK8)was used to evaluate the antiproliferative activity ofWCA on MKN45 cells;Giemsa staining was used to investigate cell colony formation;flow cytometry was used to analyze cell cycle,apoptosis rate,and caspases activation;and Hoechst staining was used to analyze the morphology of cell nuclei.The mitochondrial membrane potential was analyzed by both flow cytometric measurements and fluorescence microscopy.Western blot was used to analyze the protein levels of pro-caspase-3,Bcl-2,Bax,and Bcl-X.MKN45 cells were subcutaneously injected into the right forelimb of 16 nude mice to establish the tumor xenograft model,and a total of 12 nude mouse tumor xenograft models were successfully created.The nude mice were divided into the control group and the WCA-treated group.The two groups received normal saline and WCA treatment at 35.49 g/kg by a single daily oral gavage for 21 days.The body weight and tumor size of the nude mice were measured twice a week.The ultrastructural changes of subcutaneous tumors were observed by a transmission electron microscope.Results WCA can suppress cell proliferation and colony formation.It can also induce changes in the mitochondrial membrane potential and increase the activities of caspases-3,caspases-8,and caspases-9 in MKN45 cells.WCA induced S-phase arrest and apoptosis in MKN45 cells,and decreased the expression levels of antiapoptotic proteins Bcl-2 and Bcl-X in MKN45 cells,while increasing the expression levels of the proapoptotic proteins Bax and pro-caspase-3 compared with the control group.WCA inhibited the growth of a xenografted MKN45 tumor in nude mice,and the protein levels of Bax and pro-caspase-3 were significantly increased,while Bcl-X and Bcl-2 were reduced compared with the control group.The differences are statistically significant(p<0.05).Conclusion WCA could suppress the proliferation of gastric cancer cells via mitochondrial apoptosis.展开更多
Purpose:Patients undergoing maintenance hemodialysis(MHD)have a higher prevalence of cognitive impairment and inferior cognitive performance than the general population,and those with cognitive impairment are at highe...Purpose:Patients undergoing maintenance hemodialysis(MHD)have a higher prevalence of cognitive impairment and inferior cognitive performance than the general population,and those with cognitive impairment are at higher risk of death than those without cognitive impairment.Having diabetes has been associated with an increased risk of cognitive decline in end-stage kidney disease patients treated with peritoneal dialysis or kidney transplant.However,these findings may not extend to the hemodialysis population.Thus,we aim to investigate the relationship between having diabetes and cognitive function in MHD patients.Methods:This was a cross-sectional study.A total of 203 patients treated with MHD from two blood purification centers were enrolled as subjects.The Chinese version of the Montreal Cognitive Assessment(MoCA)was utilized to assess cognitive function.Results:MHD patients with diabetes had a significantly higher prevalence of global cognitive impairment and inferior performance in global cognition,visuospatial/executive function,naming,language,abstraction and orientation tasks compared with those without diabetes.According to the multiple linear analyses,having diabetes was significantly associated with lower global cognitive function,naming,and language scores,withβcoefficients and 95%CIs of-1.30[-2.59,-0.01],-0.25[-0.47,-0.02],and-0.32[-0.58,-0.07],respectively(all P<0.05).Having diabetes could not independently predict an increased risk of global cognitive impairment.Conclusions:In MHD patients,having diabetes is significantly associated with lower cognitive function scores.Medical staff should evaluate early and focus on the decline of cognitive function in MHD patients with diabetes,in order to achieve early diagnosis and earlv intervention.展开更多
Objective The aim of the study was to screen key proteins involved in the treatment of human gastric cancer subcutaneous xenografts in nude mice by the traditional Chinese medicine formula Weichang’an and to explore ...Objective The aim of the study was to screen key proteins involved in the treatment of human gastric cancer subcutaneous xenografts in nude mice by the traditional Chinese medicine formula Weichang’an and to explore its mechanism of action in treating gastric cancer.Methods Sixteen 7-to 8-week-old female BALB/C nude mice were used to establish a human gastric cancer subcutaneous xenograft model by bilateral axillary injection of MKN45 cells.Mice with successfully established tumors were randomly divided into the Weichang’an group and the model group,with eight mice in each group.Mice in the Weichang’an group were orally administered 0.5mL of Weichang’an decoction,while mice in the model group were given 0.5 mL of normal saline by gavage once a day for 21 consecutive days.On day 28,the animals were sacrificed by cervical dislocation,and tumors were excised to measure tumor weight and assess the tumor suppression rate.Tandem mass tags(TMT)quantitative proteomics was used to analyze the tumor samples,identify differentially expressed proteins,and perform Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis.Results Compared with the model group,the tumor weight in the Weichang’an group was significantly reduced(p<0.001),and the tumor suppression rate was 41.40%,indicating that Weichang’an can inhibit the growth of human gastric cancer subcutaneous xenografts in nude mice.TMT quantitative proteomics identified a total of 2,856 proteins,with 13 proteins showing downregulated expression and 25 proteins showing upregulated expression in the Weichang’an group.GO enrichment analysis revealed that differentially expressed proteins were mainly enriched in cellular components such as the cell membrane,extracellular matrix,and envelope,and participated in biological processes like negative regulation of cell adhesion hematopoiesis,and skeletal system development,with functions in cell adhesion molecule binding and molecular sensor activity.The KEGG pathway enrichment analysis identified the autophagy-lysosome pathway.Conclusion Weichang’an may exert its therapeutic effect on gastric cancer by regulating the expression of various proteins and modulating the autophagy-lysosome pathway.展开更多
Purpose:Our institution developed the subspecialized medical team(SMT)mode in radiology reporting workflows in July 2022 to strengthen profes-sionalism in radiology residency training.This study aimed to investigate w...Purpose:Our institution developed the subspecialized medical team(SMT)mode in radiology reporting workflows in July 2022 to strengthen profes-sionalism in radiology residency training.This study aimed to investigate whether adopting the SMT mode facilitated radiology resident training.Method:This observational study was conducted in October 2022.An online questionnaire was distributed among first-to third-year radiology residents to determine their levels of perceived stress,anxiety,competency in image interpretation,and learning effectiveness in using the SMT mode and other approaches.Responses were on 4 or 5 point Likert scales,and multiple-choice responses were used for semi-open questions.The turnaround time for radi-ology reports was measured for both residents and senior radiologists.Sta-tistical significance(p<0.05)was determined using chi-square or Fisher's exact tests,one-way analysis of variance,pairwise t-tests,and the Kruskal-Wallis H test.Results:In total,71 residents completed the questionnaire survey.The turnaround times for radiology reports were collected for these residents and 23 senior radiologists.Residents'stress and anxiety scores,perceived levels of competency in image interpretation,and time spent writing reports were significantly increased in the SMT mode.In contrast,the time senior radiol-ogists spent reviewing a single report was decreased for the majority of report types after 1 year of implementing the SMT mode.Conclusion:These findings suggest adopting the SMT mode could facilitate the training of radiology residents.However,residents'mental health and well-being should also be considered.展开更多
Hepatocellular carcinoma (HCC) is the sixth-most common cancer and the third leading cause of cancer-related death in the world. However, 40–70% patients eventually suffer from postoperative recurrence within 5 years...Hepatocellular carcinoma (HCC) is the sixth-most common cancer and the third leading cause of cancer-related death in the world. However, 40–70% patients eventually suffer from postoperative recurrence within 5 years. HCC recurrence after surgery severely affects prognosis of the patients. Nevertheless, there is an opportunity to improve patients' prognosis if doctors and researchers can recognize the importance of a standardized perioperative management and study it in clinical and pre-clinical settings. Hence, based on our own experience and published studies from other researchers, we develop this consensus regarding multidisciplinary management of locally recurrent and metastatic hepatocellular carcinoma after resection. This consensus consists of the entire course of recurrent hepatocellular carcinoma (RHCC) management, including prediction of recurrence, prevention, diagnosis, treatment and surveillance of RHCC. Consensus recommendations are presented with grades of evidences (Ia, Ib, IIa, IIb, III and IV), and strength ofrecommendations (A, B, C, D and E). We also develop a decision-making path for RHCC treatment, which can intuitively demonstrate the management for RHCC. It is hoped that we may make some effort to standardize the management of RHCC and ultimately understand how to improve outcomes.展开更多
With the increase in consumer demand,wheat grain quality improvement has become a focus in China and worldwide.Transcriptome analysis is a powerful approach to research grain traits and elucidate their genetic regulat...With the increase in consumer demand,wheat grain quality improvement has become a focus in China and worldwide.Transcriptome analysis is a powerful approach to research grain traits and elucidate their genetic regulation.In this study,two cDNA libraries from the developing grain and leaf-stem components of bread wheat cultivar,Nongda211,were sequenced using Roche/454 technology.There were 1061274 and 1516564 clean reads generated from grain and leaf-stem,respectively.A total of 61393 high-quality unigenes were obtained with an average length of 1456 bp after de novo assembly.The analysis of the 61393 unigenes involved in the biological processes of the grain showed that there were 7355 differentially expressed genes upregulated in the grain library.Gene ontology enrichment and the Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis showed that many transcription products and transcription factors associated with carbohydrate and protein metabolism were abundantly expressed in the grain.These results contribute to excavate genes associated with wheat quality and further study how they interact.展开更多
