Dyssynchronous heart failure models in canines:New insights into electrocardiographic,echocardiographic and histological features

Background:We investigated the similarities and differences between two experimental approaches using tachy-pacing technology to induce desynchronized heart failure in canines.Methods:A total of eight dogs were included in the experiment,four were tachypaced in right ventricle apex (RVAP) and 4 were paced in right atrium after the ablation of left bundle branch to achieve left bundle branch block (RAP+LBBB).Three weeks of follow-up were conducted to observe the changes in cardiac function and myocardial staining was performed at the end of the experiment.Results:Both experimental approaches successfully established heart failure with reduced ejection fraction models,with similar trends in declining cardiac function.The RAP+LBBB group exhibited a prolonged overall ventricular activation time, delayed left ventricular activation,and lesser impact on the right ventricle.The RVAP approach led to a reduction in overall right ventricular compliance and right ve ntricular enlargement.The RAP+LBBB group exhibited significant reductions in left heart compliance (LVGLS,%:RAP+LBBB-12.60±0.12 to-5.93±1.25;RVAP-13.28±0.62to-8.05±0.63, p=0.023;LASct,%:RAP+LBBB-15.75±6.85 to-1.50±1.00;RVAP-15.75±2.87 to-10.05±6.16,p=0.035).Histological examination revealed more pronounced fibrosis in the left ventricular wall and left atrium in the RAP+LBBBgroup while the RVAP group showed more prominent fibrosis in the right ventricular myocardium.Conclusion:Both approaches establish HFrEF models with comparable trends.The RVAP group shows impaired right ventricular function,while the RAP+LBBB group exhibits more severe decreased compliance and fibrosis in left ventricle. 展开更多

Most current crowdsourced logistics aim to minimize systems cost and maximize delivery capacity,but the efforts of crowdsourcers such as drivers are almost ignored.In the delivery process,drivers usually need to take long-distance detours in hitchhiking rides based package deliveries.In this paper,we propose an approach that integrates offline trajectory data mining and online route-and-schedule optimization in the hitchhiking ride scenario to find optimal delivery routes for packages and drivers.Specifically,we propose a two-phase framework for the delivery route planning and scheduling.In the first phase,the historical trajectory data are mined offline to build the package transport network.In the second phase,we model the delivery route planning and package-taxi matching as an integer linear programming problem and solve it with the Gurobi optimizer.After that,taxis are scheduled to deliver packages with optimal delivery paths via a newly designed scheduling strategy.We evaluate our approach with the realworld datasets;the results show that our proposed approach can complete citywide package deliveries with a high success rate and low extra efforts of taxi drivers. 展开更多

The FUT2 loss-of-function mutations are highly prevalent and are associated with inflammatory bowel disease(IBD).To investigate the impact of FUT2 loss-of-function mutation on the gut microbiota in patients with IBD,81 endoscopically confirmed IBD patients were genotyped and divided into 3 groups:homozygous for functional FUT2 genes(SeSe),with one copy of non-functional FUT2 gene(Sese),or homozygous for non-functional FUT2 genes(sese).Escherichia,which attaches to fucosylated glycoconjugates,was the only abundant genus exhibiting decreased abundance in sese patients.Compared with SeSe or Sese patients,sese patients exhibited higher abundance in CD8+inducing Alistipe and Phascolarctobacterium and Th17 inducing Erysipelotrichaceae UCG-003.Counter-intuitively,butyrate-producing bacteria were more abundant in sese patients.Consistently,metabolomics analysis found higher levels of butyrate in sese patients.Our data support the hypothesis that FUT2 loss-of-function mutation participates in the IBD pathogenesis by decreasing binding sites for adherent bacteria and thus altering the gut microbiota.Decreased abundances of adherent bacteria may allow the overgrowth of bacteria that induce inflammatory T cells,leading to intestinal inflammation.As FUT2 loss-of-function mutations are highly prevalent,the identification of T cell inducing bacteria in sese patients could be valuable for the development of personalized microbial intervention for IBD. 展开更多

Atrial fibrillation(AF)is a common and serious disease.Its diagnosis usually requires 12-lead electrocardiogram,which is heavy and inconvenient.At the same time,the venue for diagnosis is also limited to the hospital.With the development of the concept of intelligent medical,a wearable,portable,and reliable diagnostic method is needed to improve the patient’s comfort and alleviate the patient’s pain.Here,we reported a wearable atrial fibrillation prediction wristband(AFPW)which can provide longterm monitoring and AF diagnosis.AFPW uses polyvinylidene fluoride piezoelectric film as sensing material and hydrogel as skin bonding material,of which the structure and design have been optimized and improved.The hydrogel skin bonding layer has good stability and skin affinity,which can greatly improve the user experience.AFPW has enhanced signal,strong signal-tonoise ratio,and wireless transmission function.After a sample library of 385 normal people/patients is analyzed and tested by linear discriminant analysis,the diagnostic success rate of atrial fibrillation is 91%.All these excellent performances demonstrate the great application potential of AFPW in wearable device diagnosis and intelligent medical treatment. 展开更多
