With regard to precision/ultra-precision motion systems,it is important to achieve excellent tracking performance for various trajectory tracking tasks even under uncertain external disturbances.In this paper,to overc...With regard to precision/ultra-precision motion systems,it is important to achieve excellent tracking performance for various trajectory tracking tasks even under uncertain external disturbances.In this paper,to overcome the limitation of robustness to trajectory variations and external disturbances in offline feedforward compensation strategies such as iterative learning control(ILC),a novel real-time iterative compensation(RIC)control framework is proposed for precision motion systems without changing the inner closed-loop controller.Specifically,the RIC method can be divided into two parts,i.e.,accurate model prediction and real-time iterative compensation.An accurate prediction model considering lumped disturbances is firstly established to predict tracking errors at future sampling times.In light of predicted errors,a feedforward compensation term is developed to modify the following reference trajectory by real-time iterative calculation.Both the prediction and compen-sation processes are finished in a real-time motion control sampling period.The stability and convergence of the entire control system after real-time iterative compensation is analyzed for different conditions.Various simulation results consistently demonstrate that the proposed RIC framework possesses satisfactory dynamic regulation capability,which contributes to high tracking accuracy comparable to ILC or even better and strong robustness.展开更多
Introduction In recent years,three-dimensional printing(3DP),an additive manufacturing process,has gained widespread clinical application,and 3DP has been considered as the third industrial revolution.1 In its early i...Introduction In recent years,three-dimensional printing(3DP),an additive manufacturing process,has gained widespread clinical application,and 3DP has been considered as the third industrial revolution.1 In its early introduction in the 1980s,3DP served as a software-controlled technology that converted computer-aided-design(CAD)data into a physical object via a single process.By depositing multiple two-dimensional cross-sections one above the other,3DP can now be used to build arbitrarily complex geometries and patient-specific constructs using the patient’s imaging data.Till date,computed tomography has been the main imaging data source for 3DP owing to its excellent spatial resolution.Furthermore,current 3D printers have enabled bedside on-demand fabrication of medical products in hospitals.New materials including polymers,ceramics,biomaterials,and metals have been developed for such applications over the last few decades.Medical fields that employ 3DP technologies have also expanded,such as tissue engineering,regenerative medicine,pharmaceutics,and medical models and devices.2 The market for additive manufacturing is expected to surpass$20 billion in the global industry by the end of the 2020.3 Although the use of 3DP technology in interventional medicine is still relatively new,advancements are occurring within this discipline at a rapid rate.Different 3DP technologies,materials,and clinical applications relevant to the interventional field are discussed in this article.展开更多
Several studies have demonstrated that Slug,which encodes a zinc finger of the Snail family of transcription factors,is a potential risk factor for neural tube defects.Neural tube defects tend to occur with a high rat...Several studies have demonstrated that Slug,which encodes a zinc finger of the Snail family of transcription factors,is a potential risk factor for neural tube defects.Neural tube defects tend to occur with a high rate in Shanxi province,China.The present case-control study investigated genotypic distributions and allele frequencies of Slug C1548A polymorphisms in DNA samples from59 women with a history of neural tube defect pregnancies and 73 controls during the same period from Shanxi Province,China.Results demonstrated that women with a history of neural tube defect pregnancies had significantly greater genotypic distributions of Slug AA genotypes and A allele frequencies compared with controls,and A allele Slug C1548A was a risk factor for neural tube defects(odds ratio = 3.444;95% confidence interval;2.021-5.868,P 〈 0.05).Three-dimensional structure prediction revealed that Slug C1548A resulted in transition of aspartic acid into glutamate at position 119.This indicated that these mutations could lead to damaged protein structure and function.These findings suggest that Slug C1548A gene polymorphism is closely related to neural tube defects in a population of Han Chinese origin from Shanxi Province,China展开更多
Coal-mining activities give rise to a series of ecological environmental problems,such as ground settlement and groundwater pollution.In fact,they are mainly caused by mining-induced fractures.Hence,it is necessary to...Coal-mining activities give rise to a series of ecological environmental problems,such as ground settlement and groundwater pollution.In fact,they are mainly caused by mining-induced fractures.Hence,it is necessary to study the mining-induced fracture distribution to identify the behavior of rock mass movement.However,the fractures in overburden strata cannot be directly measured owing to the special condition.Therefore,the majority of previous studies are based on experiments or experience.For this reason,this study first used a discrete element method to simulate the shape of mining-induced fractures in overburden strata.Then,a geophysical tool of transient electromagnetic method(TEM)was used to investigate the mining-induced fracture distribution.Based on the low-resistivity anomaly area,the water-rich area in overburden strata was analyzed to be mainly caused by fracture seepage.Through the mutual authentication between numerical simulation and TEM results,the mining-induced fractures in overburden strata were explored.This study can enhance the understanding of mining-induced fracture distribution on the one hand and guarantee the coal mining safety on the other,thus guiding the coordinated development between coal mining and environmental protection.展开更多

However, results obtain...Environmental and genetic factors influence the occurrence of neural tube defects, such as spina bifida. Specific disease expression patterns will help to elucidate the pathogenesis of disease. However, results obtained from animal models, which often exhibit organism specificity, do not fully explain the mechanisms of human spina bifida onset. In the present study, three embryos with a gestational age of approximately 17 weeks and a confirmed diagnosis of spina bifida, as well as 3 age-matched normal embryos, were obtained from abortions. Fetal brain stem tissues were dissected for RNA isolation, and microarray analyses were conducted to examine profiles of gene expression in brain stems of spina bifida and normal embryos using Affymetrix HG-U133A 2.0 GeneChip arrays. Of the 14 500 gene transcripts examined, a total of 182 genes exhibited at least 2.5-fold change in expression, including 140 upregulated and 42 downregulated genes. These genes were placed into 19 main functional categories according to the Gene Ontology Consortium database for biological functions. Of the 182 altered genes, approximately 50% were involved in cellular apoptosis, growth, adhesion, cell cycle, stress, DNA replication and repair, signal transduction, nervous system development, oxidoreduction, immune responses, and regulation of gene transcription. Gene expression in multiple biological pathways was altered in the brain stem of human spina bifida embryos.展开更多
BACKGROUND Fiberoptic bronchoscopy has been widely used in the diagnosis and treatment of respiratory diseases.Numerous major and minor complications have been reported following this procedure.The incidence of major ...BACKGROUND Fiberoptic bronchoscopy has been widely used in the diagnosis and treatment of respiratory diseases.Numerous major and minor complications have been reported following this procedure.The incidence of major postoperative complications is approximately 0.5%and includes respiratory depression,pneumothorax,pulmonary edema,pneumonia,airway obstruction and cardiorespiratory arrest.Minor complications include vasovagal reactions,cardiac arrhythmias,hemorrhage,pneumothorax,aphonia,nausea,vomiting and fever.However,to our knowledge,a case of atrial fibrillation(AF)concomitant with fatal arterial embolism in the upper extremities following diagnostic bronchoscopy has never been reported.CASE SUMMARY A 70-year-old female patient presented with a history of rheumatic heart disease beginning at 10 years of age and an approximately 10-year history of hypertension.The patient was transferred from the cardiology department to the respiratory department due to recurrent coughing,pneumonia,and fever.She underwent fiberoptic bronchoscopy in the respiratory department.Approximately 2 h after completion of bronchoscopy,she complained of left arm numbness and weakness.Physical examination detected cyanosis of the left upper extremity,grade III weakened limb muscle strength,and undetectable left brachial artery pulsation.Auscultation indicated AF.B-mode ultrasound examination of the blood vessels showed hyperechoic material in the left subclavian,axillary and brachial arteries,and parallel veins.As our hospital has no vascular surgery capability,the patient was transferred to a specialized hospital for emergency thrombectomy that day.A tracking investigation found that the patient’s conditions improved after successful thrombectomy.CONCLUSION Thromboembolism following bronchoscopy is rare,and only a few cases of cerebral air embolism after bronchoscopy have been reported.展开更多

Methods: During the period from April 2016 to March 2018, a total of...Objective: To study the curative effect of licorice flavonoid Chinese herbal frond cream combined with Awei A capsule in the treatment of psoriasis. Methods: During the period from April 2016 to March 2018, a total of 116 patients with psoriasis in our hospital were randomly divided into the experimental group (the treatment of licorice flavonoids and Chinese acupoint cream combined with Awei A capsule) and the control group (single use) Avi A capsule treatment). Comparison of blood lipid metabolism indexes (high-density lipoprotein, apolipoprotein, low-density lipoprotein) and serum biochemical indicators (neutrophil elastase, endogenous inhibitor of NE, placental cadherin) statistical difference in content. The statistical differences between PASI score and DLQI score before and after treatment were compared between the two groups. At the end of treatment, the statistical difference between the clinical efficacy and the incidence of adverse reactions was compared between the two groups. Results:The levels of serum NE, Trappin-2 and P-cad in patients before and after treatment showed a significant downward trend. The content of NE, Trappin-2 and P-cad in the experimental group was significantly lower than that in the control group. The content of HDL-C in lipid metabolism index increased significantly before and after treatment, and the content of APOA and LDL-C decreased significantly. The content of HDL-C in the experimental group was significantly higher than that in the control group. The content of APOA and LDL-C was significantly lower than that of the control group. The PASI score and DLQI score of patients before and after treatment decreased significantly. After treatment, the PASI score and DLQI score of the experimental group were significantly lower than those of the control group. The clinical efficacy of the two groups was significantly different. The efficacy of the experimental group was significantly better than that of the control group. The skin of the experimental group was dry, itchy, and the skin desquamation was significantly less than that of the control group. Conclusion: Licorice Flavonoids and Avi A capsules have the advantages of improving blood lipid metabolism in the treatment of psoriasis, while reducing the recurrence rate and reducing the occurrence of common adverse reactions such as skin itching, dryness and abnormal liver function. It has a good clinical application significance.展开更多
Lung adenocarcinoma(LUAD)and squamous carcinoma(LUSC)are two major subtypes of non-small cell lung cancer with distinct pathologic features and treatment paradigms.The heterogeneity can be attributed to genetic,transc...Lung adenocarcinoma(LUAD)and squamous carcinoma(LUSC)are two major subtypes of non-small cell lung cancer with distinct pathologic features and treatment paradigms.The heterogeneity can be attributed to genetic,transcriptional,and epigenetic parameters.Here,we established a multi-omics atlas,integrating 52 single-cell RNA sequencing and 2342 public bulk RNA sequencing.We investigated their differences in genetic amplification,cellular compositions,and expression modules.We revealed that LUAD and LUSC contained amplifications occurring selectively in subclusters of AT2 and basal cells,and had distinct cellular composition modules associated with poor survival of lung cancer.Malignant and stage-specific gene analyses further uncovered critical transcription factors and genes in tumor progression.Moreover,we identified subclusters with proliferating and differentiating properties in AT2 and basal cells.Overexpression assays of ten genes,including sub-cluster markers AQP5 and KPNA2,further indicated their functional roles,providing potential targets for early diagnosis and treatment in lung cancer.展开更多
Background:Functional mitral regurgitation(FMR)is common in critically ill patients and may cause left atrial(LA)pressure elevation.This study aims to explore the prognostic impact of synergistic LA pressure elevation...Background:Functional mitral regurgitation(FMR)is common in critically ill patients and may cause left atrial(LA)pressure elevation.This study aims to explore the prognostic impact of synergistic LA pressure elevation and FMR in patients with shock.Methods:We retrospectively screened 130 consecutive patients of 175 patients with shock from April 2016 to June 2017.The incidence and impact of FMR and early diastolic transmitral velocity to early mitral annulus diastolic velocity ratio(E/e’)≥4 within 6 h of shock on the prognosis of patients were evaluated.Finally,the synergistic effect of FMR and E/e’were assessed by combination,grouping,and trend analyses.Results:Forty-four patients(33.8%)had FMR,and 15 patients(11.5%)had E/e’elevation.A multivariate analysis revealed FMR and E/e’as independent correlated factors for 28-day mortality(P=0.043 and 0.028,respectively).The Kaplan-Meier survival analysis revealed a significant difference in survival between patients with and without FMR(χ2=7.672,P=0.006)and between the E/e’≥14 and E/e’<14 groups(χ2=19.351,P<0.010).Twenty-eight-day mortality was significantly different among the four groups(χ2=30.141,P<0.010).The risk of 28-day mortality was significantly higher in group 4(E/e’≥14 with FMR)compared with groups 1(E/e’<14 without FMR)and 2(E/e’<14 with FMR)(P=0.001 and 0.046,respectively).Conclusions:Patients with shock can be identified by the presence of FMR.FMR and E/e’are independent risk factors for a poor prognosis in these patients,and prognosis is worst when FMR and E/e’≥14 are present.It may be possible to improve prognosis by reducing LA pressure and E/e’.Trial Registration:ClinicalTrials.gov,NCT03082326.展开更多
With the rapid development of urban underground space,the construction of shield-driven cross-river twin tunnels is increasing,and the complex hydro-mechanical coupling effects and twin-tunnel interactions bring huge ...With the rapid development of urban underground space,the construction of shield-driven cross-river twin tunnels is increasing,and the complex hydro-mechanical coupling effects and twin-tunnel interactions bring huge construction risks to such projects,which have attracted more and more attention.This study aims to understand the excavation effects induced by shield driving of cross-river twin tunnels through numerical simulation.A refined three-dimensional numerical model based on the fully coupled hydro-mechanical theory is established.The model considers the main components of the slurry pressure balance shield(SPBS)machine,including support force,jacking thrust,grouting pressure,shield-rock interaction and lining-grouting interaction,as well as the detailed construction process.The purpose is to examine the excavation effects during construction,including rock deformation around tunnels,the change in pore pressure,and the response of the lining.The results show the influence range of twin-tunnel excavation on rock deformation and pore pressure,as well as the modes of lining response.In addition,this study also systematically investigates the effects of water level fluctuation and burial depth on twin-tunnel excavation.The results indicate that the increase of water level or burial depth will enhance the excavation effects and strengthen the twin-tunnel interactions.These results provide useful insights for estimating the construction impact range and degree of twin tunnels,and serve as basic references for the design of cross-river twin tunnels.展开更多

Prefabricated pavement is increasingly applied worldwide due to the rapid construction on-site.This paper presents a state-of-the-art review of the precast systems and assembly connection of concrete slabs,as well as ...Prefabricated pavement is increasingly applied worldwide due to the rapid construction on-site.This paper presents a state-of-the-art review of the precast systems and assembly connection of concrete slabs,as well as the precast pavement features.The previous research indicates that:(1)both super-slab and Michigan systems are recommended with the satisfied road performance close to the cast-in-place;(2)flexible base material is suggested for the fabricated pavement for the satisfied leveling and stress distribution;(3)to prevent the voiding phenomenon of the fabricated pavement,the sand cushion course,dry mixed mortar or self-leveling mortar,and other flowable materials could be used for the secondary leveling of the base after the pavement splicing;(4)two-direction dowel bars are recommended for slab connections of the fabricated pavement,which helps to improve the load transfer capacity of the joints and enhance the durability of the fabricated pavement structure;(5)the sealing treatment of precast slab joints needs strengthening to reduce the impact of surface runoff on the base course;(6)the further research focuses are designing with functional,composite,mechanized,intelligent,lightweight,and flexible pavement slabs.Besides,pavement mechanical properties induced by temperature overlapping traffic loads need to be revealed.展开更多
Background:Shenque(CV8)acupoint is located on the navel and has been therapeutically used for more than 2000 years in Traditional Chinese Medicine(TCM).However,clinical research on the underlying therapeutic molecular...Background:Shenque(CV8)acupoint is located on the navel and has been therapeutically used for more than 2000 years in Traditional Chinese Medicine(TCM).However,clinical research on the underlying therapeutic molecular mechanisms of the CV8 acupoint lags far behind.This study aimed to study the mechanisms of umbilical acupoint therapy by using stem cells.Methods:The morphological characteristics of CV8 acupoint were detected under a stereomicroscope using hematoxylin and eosin(H&E)staining.Oil Red,Masson,and immunohistochemical staining on multi-layered slices were used to identify the type of cells at the CV8 acupoint.Cell proliferation was measured by a cell counting kit-8(CCK-8)method.Flow cytometry and immunohistochemistry were used for cell identification.Induced differentiation was used to compare the differentiation of cells derived from CV8 acupoint and non-acupoint somatic stem cells into other cell types,such as osteogenic,adipogenic,and neural stem cell-like cells.Results:Morphological observations showed that adipose tissues at the linea alba of the CV8 acupoint in mice had a mass-like distribution.Immunohistochemical staining confirmed the distribution of stem cell antigen-1(Sca-1)positive cells in the multi-layered slices of CV8 acupoint tissues.Cells isolated from adipose tissues at the CV8 acupoint exhibited high expression of Sca-1 and CD44 and low expression of CD31 and CD34,and these cells possessed osteogenic,adipogenic,and neurogenic stem cell-like cell differentiation ability.The cell proliferation(day 4:0.5138±0.0111 vs.0.4107±0.0180,t=8.447,P=0.0011;day 5:0.6890±0.0070 vs.0.5520±0.0118,t=17.310,P<0.0001;day 6:0.7320±0.0090 vs.0.6157±0.0123,t=13.190,P=0.0002;and day 7:0.7550±0.0050 vs.0.6313±0.0051,t=42.560,P<0.0001),adipogenic([9.224±0.345]%vs.[3.933±1.800]%,t=5.000,P=0.0075),and neurogenic stem cell-like cell differentiation(diameter<50 mm:7.2000±1.3040 vs.2.6000±0.5477,t=7.273,P<0.0001;diameter 50–100 mm:2.6000±0.5477 vs.1.0000±0.7071,t=4.000,P=0.0039;and diameter>100 mm:2.6000±0.5477 vs.0.8000±0.8367,t=4.025,P=0.0038)were significantly enhanced in somatic stem cells derived from the CV8 acupoint compared to somatic stem cells from the groin non-acupoint.However,cells possessed significantly weaker osteogenicity([2.697±0.627]%vs.[7.254±0.958]%,t=6.893,P=0.0023)in the CV8 acupoint group.Conclusions:Our study showed that CV8 acupoint was rich with adipose tissues that contained abundant somatic stem cells.The biological examination of somatic stem cells derived from the CV8 acupoint provided novel insights for future research on the mechanisms of umbilical therapy.展开更多

文摘Several studies have demonstrated that Slug,which encodes a zinc finger of the Snail family of transcription factors,is a potential risk factor for neural tube defects.Neural tube defects tend to occur with a high rate in Shanxi province,China.The present case-control study investigated genotypic distributions and allele frequencies of Slug C1548A polymorphisms in DNA samples from59 women with a history of neural tube defect pregnancies and 73 controls during the same period from Shanxi Province,China.Results demonstrated that women with a history of neural tube defect pregnancies had significantly greater genotypic distributions of Slug AA genotypes and A allele frequencies compared with controls,and A allele Slug C1548A was a risk factor for neural tube defects(odds ratio = 3.444;95% confidence interval;2.021-5.868,P 〈 0.05).Three-dimensional structure prediction revealed that Slug C1548A resulted in transition of aspartic acid into glutamate at position 119.This indicated that these mutations could lead to damaged protein structure and function.These findings suggest that Slug C1548A gene polymorphism is closely related to neural tube defects in a population of Han Chinese origin from Shanxi Province,China

文摘Coal-mining activities give rise to a series of ecological environmental problems,such as ground settlement and groundwater pollution.In fact,they are mainly caused by mining-induced fractures.Hence,it is necessary to study the mining-induced fracture distribution to identify the behavior of rock mass movement.However,the fractures in overburden strata cannot be directly measured owing to the special condition.Therefore,the majority of previous studies are based on experiments or experience.For this reason,this study first used a discrete element method to simulate the shape of mining-induced fractures in overburden strata.Then,a geophysical tool of transient electromagnetic method(TEM)was used to investigate the mining-induced fracture distribution.Based on the low-resistivity anomaly area,the water-rich area in overburden strata was analyzed to be mainly caused by fracture seepage.Through the mutual authentication between numerical simulation and TEM results,the mining-induced fractures in overburden strata were explored.This study can enhance the understanding of mining-induced fracture distribution on the one hand and guarantee the coal mining safety on the other,thus guiding the coordinated development between coal mining and environmental protection.

文摘Environmental and genetic factors influence the occurrence of neural tube defects, such as spina bifida. Specific disease expression patterns will help to elucidate the pathogenesis of disease. However, results obtained from animal models, which often exhibit organism specificity, do not fully explain the mechanisms of human spina bifida onset. In the present study, three embryos with a gestational age of approximately 17 weeks and a confirmed diagnosis of spina bifida, as well as 3 age-matched normal embryos, were obtained from abortions. Fetal brain stem tissues were dissected for RNA isolation, and microarray analyses were conducted to examine profiles of gene expression in brain stems of spina bifida and normal embryos using Affymetrix HG-U133A 2.0 GeneChip arrays. Of the 14 500 gene transcripts examined, a total of 182 genes exhibited at least 2.5-fold change in expression, including 140 upregulated and 42 downregulated genes. These genes were placed into 19 main functional categories according to the Gene Ontology Consortium database for biological functions. Of the 182 altered genes, approximately 50% were involved in cellular apoptosis, growth, adhesion, cell cycle, stress, DNA replication and repair, signal transduction, nervous system development, oxidoreduction, immune responses, and regulation of gene transcription. Gene expression in multiple biological pathways was altered in the brain stem of human spina bifida embryos.
文摘BACKGROUND Fiberoptic bronchoscopy has been widely used in the diagnosis and treatment of respiratory diseases.Numerous major and minor complications have been reported following this procedure.The incidence of major postoperative complications is approximately 0.5%and includes respiratory depression,pneumothorax,pulmonary edema,pneumonia,airway obstruction and cardiorespiratory arrest.Minor complications include vasovagal reactions,cardiac arrhythmias,hemorrhage,pneumothorax,aphonia,nausea,vomiting and fever.However,to our knowledge,a case of atrial fibrillation(AF)concomitant with fatal arterial embolism in the upper extremities following diagnostic bronchoscopy has never been reported.CASE SUMMARY A 70-year-old female patient presented with a history of rheumatic heart disease beginning at 10 years of age and an approximately 10-year history of hypertension.The patient was transferred from the cardiology department to the respiratory department due to recurrent coughing,pneumonia,and fever.She underwent fiberoptic bronchoscopy in the respiratory department.Approximately 2 h after completion of bronchoscopy,she complained of left arm numbness and weakness.Physical examination detected cyanosis of the left upper extremity,grade III weakened limb muscle strength,and undetectable left brachial artery pulsation.Auscultation indicated AF.B-mode ultrasound examination of the blood vessels showed hyperechoic material in the left subclavian,axillary and brachial arteries,and parallel veins.As our hospital has no vascular surgery capability,the patient was transferred to a specialized hospital for emergency thrombectomy that day.A tracking investigation found that the patient’s conditions improved after successful thrombectomy.CONCLUSION Thromboembolism following bronchoscopy is rare,and only a few cases of cerebral air embolism after bronchoscopy have been reported.
文摘Objective: To study the curative effect of licorice flavonoid Chinese herbal frond cream combined with Awei A capsule in the treatment of psoriasis. Methods: During the period from April 2016 to March 2018, a total of 116 patients with psoriasis in our hospital were randomly divided into the experimental group (the treatment of licorice flavonoids and Chinese acupoint cream combined with Awei A capsule) and the control group (single use) Avi A capsule treatment). Comparison of blood lipid metabolism indexes (high-density lipoprotein, apolipoprotein, low-density lipoprotein) and serum biochemical indicators (neutrophil elastase, endogenous inhibitor of NE, placental cadherin) statistical difference in content. The statistical differences between PASI score and DLQI score before and after treatment were compared between the two groups. At the end of treatment, the statistical difference between the clinical efficacy and the incidence of adverse reactions was compared between the two groups. Results:The levels of serum NE, Trappin-2 and P-cad in patients before and after treatment showed a significant downward trend. The content of NE, Trappin-2 and P-cad in the experimental group was significantly lower than that in the control group. The content of HDL-C in lipid metabolism index increased significantly before and after treatment, and the content of APOA and LDL-C decreased significantly. The content of HDL-C in the experimental group was significantly higher than that in the control group. The content of APOA and LDL-C was significantly lower than that of the control group. The PASI score and DLQI score of patients before and after treatment decreased significantly. After treatment, the PASI score and DLQI score of the experimental group were significantly lower than those of the control group. The clinical efficacy of the two groups was significantly different. The efficacy of the experimental group was significantly better than that of the control group. The skin of the experimental group was dry, itchy, and the skin desquamation was significantly less than that of the control group. Conclusion: Licorice Flavonoids and Avi A capsules have the advantages of improving blood lipid metabolism in the treatment of psoriasis, while reducing the recurrence rate and reducing the occurrence of common adverse reactions such as skin itching, dryness and abnormal liver function. It has a good clinical application significance.

文摘Lung adenocarcinoma(LUAD)and squamous carcinoma(LUSC)are two major subtypes of non-small cell lung cancer with distinct pathologic features and treatment paradigms.The heterogeneity can be attributed to genetic,transcriptional,and epigenetic parameters.Here,we established a multi-omics atlas,integrating 52 single-cell RNA sequencing and 2342 public bulk RNA sequencing.We investigated their differences in genetic amplification,cellular compositions,and expression modules.We revealed that LUAD and LUSC contained amplifications occurring selectively in subclusters of AT2 and basal cells,and had distinct cellular composition modules associated with poor survival of lung cancer.Malignant and stage-specific gene analyses further uncovered critical transcription factors and genes in tumor progression.Moreover,we identified subclusters with proliferating and differentiating properties in AT2 and basal cells.Overexpression assays of ten genes,including sub-cluster markers AQP5 and KPNA2,further indicated their functional roles,providing potential targets for early diagnosis and treatment in lung cancer.

文摘Background:Functional mitral regurgitation(FMR)is common in critically ill patients and may cause left atrial(LA)pressure elevation.This study aims to explore the prognostic impact of synergistic LA pressure elevation and FMR in patients with shock.Methods:We retrospectively screened 130 consecutive patients of 175 patients with shock from April 2016 to June 2017.The incidence and impact of FMR and early diastolic transmitral velocity to early mitral annulus diastolic velocity ratio(E/e’)≥4 within 6 h of shock on the prognosis of patients were evaluated.Finally,the synergistic effect of FMR and E/e’were assessed by combination,grouping,and trend analyses.Results:Forty-four patients(33.8%)had FMR,and 15 patients(11.5%)had E/e’elevation.A multivariate analysis revealed FMR and E/e’as independent correlated factors for 28-day mortality(P=0.043 and 0.028,respectively).The Kaplan-Meier survival analysis revealed a significant difference in survival between patients with and without FMR(χ2=7.672,P=0.006)and between the E/e’≥14 and E/e’<14 groups(χ2=19.351,P<0.010).Twenty-eight-day mortality was significantly different among the four groups(χ2=30.141,P<0.010).The risk of 28-day mortality was significantly higher in group 4(E/e’≥14 with FMR)compared with groups 1(E/e’<14 without FMR)and 2(E/e’<14 with FMR)(P=0.001 and 0.046,respectively).Conclusions:Patients with shock can be identified by the presence of FMR.FMR and E/e’are independent risk factors for a poor prognosis in these patients,and prognosis is worst when FMR and E/e’≥14 are present.It may be possible to improve prognosis by reducing LA pressure and E/e’.Trial Registration:ClinicalTrials.gov,NCT03082326.

文摘With the rapid development of urban underground space,the construction of shield-driven cross-river twin tunnels is increasing,and the complex hydro-mechanical coupling effects and twin-tunnel interactions bring huge construction risks to such projects,which have attracted more and more attention.This study aims to understand the excavation effects induced by shield driving of cross-river twin tunnels through numerical simulation.A refined three-dimensional numerical model based on the fully coupled hydro-mechanical theory is established.The model considers the main components of the slurry pressure balance shield(SPBS)machine,including support force,jacking thrust,grouting pressure,shield-rock interaction and lining-grouting interaction,as well as the detailed construction process.The purpose is to examine the excavation effects during construction,including rock deformation around tunnels,the change in pore pressure,and the response of the lining.The results show the influence range of twin-tunnel excavation on rock deformation and pore pressure,as well as the modes of lining response.In addition,this study also systematically investigates the effects of water level fluctuation and burial depth on twin-tunnel excavation.The results indicate that the increase of water level or burial depth will enhance the excavation effects and strengthen the twin-tunnel interactions.These results provide useful insights for estimating the construction impact range and degree of twin tunnels,and serve as basic references for the design of cross-river twin tunnels.

文摘Prefabricated pavement is increasingly applied worldwide due to the rapid construction on-site.This paper presents a state-of-the-art review of the precast systems and assembly connection of concrete slabs,as well as the precast pavement features.The previous research indicates that:(1)both super-slab and Michigan systems are recommended with the satisfied road performance close to the cast-in-place;(2)flexible base material is suggested for the fabricated pavement for the satisfied leveling and stress distribution;(3)to prevent the voiding phenomenon of the fabricated pavement,the sand cushion course,dry mixed mortar or self-leveling mortar,and other flowable materials could be used for the secondary leveling of the base after the pavement splicing;(4)two-direction dowel bars are recommended for slab connections of the fabricated pavement,which helps to improve the load transfer capacity of the joints and enhance the durability of the fabricated pavement structure;(5)the sealing treatment of precast slab joints needs strengthening to reduce the impact of surface runoff on the base course;(6)the further research focuses are designing with functional,composite,mechanized,intelligent,lightweight,and flexible pavement slabs.Besides,pavement mechanical properties induced by temperature overlapping traffic loads need to be revealed.

文摘Background:Shenque(CV8)acupoint is located on the navel and has been therapeutically used for more than 2000 years in Traditional Chinese Medicine(TCM).However,clinical research on the underlying therapeutic molecular mechanisms of the CV8 acupoint lags far behind.This study aimed to study the mechanisms of umbilical acupoint therapy by using stem cells.Methods:The morphological characteristics of CV8 acupoint were detected under a stereomicroscope using hematoxylin and eosin(H&E)staining.Oil Red,Masson,and immunohistochemical staining on multi-layered slices were used to identify the type of cells at the CV8 acupoint.Cell proliferation was measured by a cell counting kit-8(CCK-8)method.Flow cytometry and immunohistochemistry were used for cell identification.Induced differentiation was used to compare the differentiation of cells derived from CV8 acupoint and non-acupoint somatic stem cells into other cell types,such as osteogenic,adipogenic,and neural stem cell-like cells.Results:Morphological observations showed that adipose tissues at the linea alba of the CV8 acupoint in mice had a mass-like distribution.Immunohistochemical staining confirmed the distribution of stem cell antigen-1(Sca-1)positive cells in the multi-layered slices of CV8 acupoint tissues.Cells isolated from adipose tissues at the CV8 acupoint exhibited high expression of Sca-1 and CD44 and low expression of CD31 and CD34,and these cells possessed osteogenic,adipogenic,and neurogenic stem cell-like cell differentiation ability.The cell proliferation(day 4:0.5138±0.0111 vs.0.4107±0.0180,t=8.447,P=0.0011;day 5:0.6890±0.0070 vs.0.5520±0.0118,t=17.310,P<0.0001;day 6:0.7320±0.0090 vs.0.6157±0.0123,t=13.190,P=0.0002;and day 7:0.7550±0.0050 vs.0.6313±0.0051,t=42.560,P<0.0001),adipogenic([9.224±0.345]%vs.[3.933±1.800]%,t=5.000,P=0.0075),and neurogenic stem cell-like cell differentiation(diameter<50 mm:7.2000±1.3040 vs.2.6000±0.5477,t=7.273,P<0.0001;diameter 50–100 mm:2.6000±0.5477 vs.1.0000±0.7071,t=4.000,P=0.0039;and diameter>100 mm:2.6000±0.5477 vs.0.8000±0.8367,t=4.025,P=0.0038)were significantly enhanced in somatic stem cells derived from the CV8 acupoint compared to somatic stem cells from the groin non-acupoint.However,cells possessed significantly weaker osteogenicity([2.697±0.627]%vs.[7.254±0.958]%,t=6.893,P=0.0023)in the CV8 acupoint group.Conclusions:Our study showed that CV8 acupoint was rich with adipose tissues that contained abundant somatic stem cells.The biological examination of somatic stem cells derived from the CV8 acupoint provided novel insights for future research on the mechanisms of umbilical therapy.
