The dominant annual cycle of sea surface temperature(SST)in the tropical Pacific exhibits an antisymmetric mode,which explains 83.4%total variance,and serves as a background of El Niño-Southern Oscillation(ENSO)....The dominant annual cycle of sea surface temperature(SST)in the tropical Pacific exhibits an antisymmetric mode,which explains 83.4%total variance,and serves as a background of El Niño-Southern Oscillation(ENSO).However,there is no consensus yet on its anomalous impacts on the phase and amplitude of ENSO.Based on data during 1982-2022,results show that anomalies of the antisymmetric mode can affect the evolution of ENSO on the interannual scale via Bjerknes feedback,in which the positive(negative)phase of the antisymmetric mode can strengthen El Niño(La Niña)in boreal winter via an earlier(delayed)seasonal cycle transition and larger(smaller)annual mean.The magnitude of the SST anomalies in the equatorial eastern Pacific can reach more than±0.3◦C,regulated by the changes in the antisymmetric mode based on random sensitivity analysis.Results reveal the spatial pattern of the annual cycle associated with the seasonal phase-locking of ENSO evolution and provide new insight into the impact of the annual cycle of background SST on ENSO,which possibly carries important implications for forecasting ENSO.展开更多
Grapes,one of the oldest tree species globally,are rich in vitamins.However,environmental conditions such as low temperature and soil salinization significantly affect grape yield and quality.The glutamate receptor(GLR...Grapes,one of the oldest tree species globally,are rich in vitamins.However,environmental conditions such as low temperature and soil salinization significantly affect grape yield and quality.The glutamate receptor(GLR)family,comprising highly conserved ligand-gated ion channels,regulates plant growth and development in response to stress.In this study,11 members of the VvGLR gene family in grapes were identified using whole-genome sequence analysis.Bioinformatic methods were employed to analyze the basic physical and chemical properties,phylogenetic trees,conserved domains,motifs,expression patterns,and evolutionary relationships.Phylogenetic and collinear analyses revealed that the VvGLRs were divided into three subgroups,showing the high conservation of the grape GLR family.These members exhibited 2 glutamate receptor binding regions(GABAb and GluR)and 3-4 transmembrane regions(M1,M2,M3,and M4).Real-time quantitative PCR analysis demonstrated the sensitivity of all VvGLRs to low temperature and salt stress.Subsequent localization studies in Nicotiana tabacum verified that VvGLR3.1 and VvGLR3.2 proteins were located on the cell membrane and cell nucleus.Additionally,yeast transformation experiments confirmed the functionality of VvGLR3.1 and VvGLR3.2 in response to low temperature and salt stress.Thesefindings highlight the significant role of the GLR family,a highly conserved group of ion channels,in enhancing grape stress resistance.This study offers new insights into the grape GLR gene family,providing fundamental knowledge for further functional analysis and breeding of stress-resistant grapevines.展开更多
PCrNi3MoV steel is a medium-carbon,low-alloy quenched and tempered steel that finds its applications in military gun barrels due to the high wear resistance and ablation resistance.To study the penetration and failure...PCrNi3MoV steel is a medium-carbon,low-alloy quenched and tempered steel that finds its applications in military gun barrels due to the high wear resistance and ablation resistance.To study the penetration and failure modes of PCrNi3MoV plates impacted by tungsten spheres,tungsten spheres of various diameters(5 mm,8 mm,and 10 mm)were used to impact PCrNi3MoV steel plates with thicknesses of 6 mm,9 mm,and 14 mm.The penetration performance of the spheres was analyzed for different velocities,and the ultimate penetration velocity of the plate was obtained.It was found that the primary failure modes of the PCrNi3MoV plate were compression pitting failure and shear failure.Using the dimensional analysis method,a relationship between the bulge height of the steel plate and the fragment velocity,an equation for the ultimate penetration velocity,and a relationship between the target penetration energy and the fragment velocity were obtained.Then,a projectile-target action index was proposed to describe the process of tungsten spheres with different velocities impacting target plates.The results suggested that under the same thickness of the target plate,a larger-diameter fragment required more kinetic energy to obtain the same ultimate penetration effect as a smaller-diameter fragment.The equations obtained through dimensional analysis predicted values that agreed well with experimental values,indicating that these equations can be applied to engineering applications.展开更多
Antarctic coastal polynyas are biological hotspots in the Southern Ocean that support the abundance of hightrophic-level predators and are important for carbon cycling in the high-latitude oceans.In this study,we exam...Antarctic coastal polynyas are biological hotspots in the Southern Ocean that support the abundance of hightrophic-level predators and are important for carbon cycling in the high-latitude oceans.In this study,we examined the interannual variation of summertime phytoplankton biomass in the Marguerite Bay polynya(MBP)in the western Antarctic Peninsula area,and linked such variability to the Southern Annular Mode(SAM)that dominated the southern hemisphere extratropical climate variability.Combining satellite data,atmosphere reanalysis products and numerical simulations,we found that the interannual variation of summer chlorophyll-a(Chl-a)concentration in the MBP is significantly and negatively correlated with the spring SAM index,and weakly correlated with the summer SAM index.The negative relation between summer Chl-a and spring SAM is due to weaker spring vertical mixing under a more positive SAM condition,which would inhibit the supply of iron from deep layers into the surface euphotic layer.The negative relation between spring mixing and spring SAM results from greater precipitation rate over the MBP region in positive SAM phase,which leads to lower salinity in the ocean surface layer.The coupled physical-biological mechanisms between SAM and phytoplankton biomass revealed in this study is important for us to predict the future variations of phytoplankton biomasses in Antarctic polynyas under climate change.展开更多
This studay is to construct a lentiviral vector harbouring an RNA interference (RNAi) sequence that targets the gene encoding the human high-mobility group nucleosomal binding protein 1 (NSBP1); to study its role ...This studay is to construct a lentiviral vector harbouring an RNA interference (RNAi) sequence that targets the gene encoding the human high-mobility group nucleosomal binding protein 1 (NSBP1); to study its role in inducing G2/M phase arrest and apoptosis in prostate cancer (PCa) DU145 cells; and to assess the effect of its knockdown on cell proliferation in vitro and in vivo. RNAi was applied to knock down NSBP1 expression in the PCa cell line DU145 by lentiviral plasmids producing an NSBP1 small hairpin RNA. After NSBP1 knockdown in DU145 cells, the growth rate of cells was analyzed by MTT, and G2/M cell cycle arrest and apoptosis were assessed using a FAC- Scalibur flow cytometer. Tumour growth was assessed in nude mice. The mRNA and protein expression levels of NSBP1, cyclin B1 and Bcl-2 were analysed in vitro and in vivo by reverse-transcriptase polymerase chain reaction and Western blotting. Knockdown of NSBP1 resulted in a 22.6% decrease in the growth rate of cells compared with the PscNC lentivirus control cells at 96 h, decreased tumour growth in nude mice, and the induction of Gz/M cell cycle arrest (8.78%) and apoptosis (2.19-fold). Consistent with the cell cycle arrest and apoptosis, the mRNA and protein expression levels of cyclin B 1 and Bcl-2 were decreased. In conclusion, knockdown of NSBP 1 causes a statistically significant inhibition of the in vitro and in vivo growth of the PCa cell line DU145. Growth suppression is at least partially due to NSBP1 knockdown-induced Gz/M cell cycle arrest and apoptosis. The present data provide the evidence that the NSBP1 knockdown-induced G2/M phase arrest and apoptosis may result from negative regulation of cyclin B 1 and Bcl-2 by NSBP1, with the resulting reduced expression of these proteins.展开更多
It has been suggested that a warm(cold)ENSO event in winter is mostly followed by a late(early)onset of the South China Sea(SCS)summer monsoon(SCSSM)in spring.Our results show this positive relationship,which is mainl...It has been suggested that a warm(cold)ENSO event in winter is mostly followed by a late(early)onset of the South China Sea(SCS)summer monsoon(SCSSM)in spring.Our results show this positive relationship,which is mainly determined by their phase correlation,has been broken under recent rapid global warming since 2011,due to the disturbance of cold tongue(CT)La Niña events.Different from its canonical counterpart,a CT La Niña event is characterized by surface meridional wind divergences in the central-eastern equatorial Pacific,which can delay the SCSSM onset by enhanced convections in the warming Indian Ocean and the western subtropical Pacific.Owing to the increased Indian−western Pacific warming and the prevalent CT La Niña events,empirical seasonal forecasting of SCSSM onset based on ENSO may be challenged in the future.展开更多

In this study, a focal cerebral ischemia/reperfusion injury model was e...Oxysophoridine, a new alkaloid extracted from Sophora alopecuroides L., has been shown to have a protective effect against ischemic brain damage. In this study, a focal cerebral ischemia/reperfusion injury model was established using middle cerebral artery occlusion in mice. Both 62.5, 125, and 250 mg/kg oxysophoridine, via intraperitoneal injection, and 6 mg/kg nimodipine, via intragastric administration, were administered daily for 7 days before modeling. After 24 hours of reperfusion, mice were tested for neurological deficit, cerebral infarct size was assessed and brain tissue was collected. Results showed that oxysophoridine at 125, 250 mg/kg and 6 mg/kg nimodipine could reduce neurological deficit scores, cerebral infarct size and brain water content in mice. These results provided evidence that oxysophoridine plays a protective role in cerebral ischemia/reperfusion injury. In addition, oxysophoridine at 62.5, 125, and 250 mg/kg and 6 mg/kg nimodipine increased adenosine-triphosphate content, and decreased malondialdehyde and nitric oxide content. These compounds enhanced the activities of glutathione-peroxidase, superoxide dismutase, catalase, and lactate dehydrogenase, and decreased the activity of nitric oxide synthase Protein and mRNA expression levels of N-methyI-D-aspartate receptor subunit NR1 were markedly inhibited in the presence of 250 mg/kg oxysophoridine and 6 mg/kg nimodipine. Our experimental findings indicated that oxysophoridine has a neuroprotective effect against cerebral ischemia/reperfusion injury in mice, and that the effect may be due to its ability to inhibit oxidative stress and expression of the N-methyI-D-aspartate receptor subunit NR1.展开更多

METHODS: A total of 80 male Wistar rats were randomly assigned to control(4, 8, 12 and 16 wk, n =10 in each group) and diabetes mellitus(DM) groups(4, 8, 12 and 16wk, n =10 in each group). A diabetic rat model was established by intraperitoneal injection of streptozotocin(60 mg/kg). After 4, 8, 12 and 16 wk, rats were sacrificed.Retinal layers and retinal neovascularization growth were stained with hematoxylin-eosin and examined under light microscopy. Cell apoptosis in the retina was detected by Td T-mediated d UTP nick end labeling, and NF-κB distribution and expression in the retina was determined using immunohistochemistry. RESULTS: DM model success rate up to 100%.Diabetes model at each time point after the experimental groupcompared with the control group, the blood glucose was significantly increased, decreased body weight, each time point showed significant differences compared with the control group(P 【0.01). After 12 wk other pathological changes in the retina of diabetic rats were observed; after 16 wk, neovascularization were observed. After 1mo, retinal cell apoptosis was observed.Compared with the control group, NF-κB expression in the DM group significantly increased with disease duration.CONCLUSION: With the prolonging of DM progression,the expression NF-κB increases. NF-κB may be related to retinal cell apoptosis and neovascularization.展开更多
Our previous studies indicated that tomato miR482b could negatively regulate the resistance of tomato to Phytophthora infestans and the expression of miR482b was decreased after inoculation with P.infestans.However,th...Our previous studies indicated that tomato miR482b could negatively regulate the resistance of tomato to Phytophthora infestans and the expression of miR482b was decreased after inoculation with P.infestans.However,the mechanism by which the accumulation of miR482b is suppressed remains unclear.In this study,we wrote a program to identify 89 long noncoding RNA(lncRNA)-originated endogenous target mimics(eTMs)for 46 miRNAs from our RNA-Seq data.Three tomato lncRNAs,lncRNA23468,lncRNA01308 and lncRNA13262,contained conserved eTM sites for miR482b.When lncRNA23468 was overexpressed in tomato,miR482b expression was significantly decreased,and the expression of the target genes,NBS-LRRs,was significantly increased,resulting in enhanced resistance to P.infestans.Silencing lncRNA23468 in tomato led to the increased accumulation of miR482b and decreased accumulation of NBS-LRRs,as well as reduced resistance to P.infestans.In addition,the accumulation of both miR482b and NBS-LRRs was not significantly changed in tomato plants that overexpressed lncRNA23468 with a mutated eTM site.Based on the VIGS system,a target gene of miR482b,Solyc02g036270.2,was silenced.The disease symptoms of the VIGS-Solyc02g036270.2 tomato plants were in accordance with those of tomato plants in which lncRNA23468 was silenced after inoculation with P.infestans.More severe disease symptoms were found in the modified plants than in the control plants.Our results demonstrate that lncRNAs functioning as eTMs may modulate the effects of miRNAs in tomato and provide insight into how the lncRNA23468-miR482b-NBS-LRR module regulates tomato resistance to P.infestans.展开更多
Tomato is an important horticultural and economic crop cultivated worldwide.As Phytophthora infestans becomes a huge threat to tomato production,it is necessary to study the resistance mechanisms of tomato against P.i...Tomato is an important horticultural and economic crop cultivated worldwide.As Phytophthora infestans becomes a huge threat to tomato production,it is necessary to study the resistance mechanisms of tomato against P.infestans.Our previous research has found that miR482 might be involved in tomato–P.infestans interaction.In this study,miR482b precursor was cloned from Solanum pimpinellifolium“L3708”and miR482b was shown to decrease in abundance in tomato following P.infestans infection.Compared to wild-type tomato plants,tomato plants that overexpressed miR482b displayed more serious disease symptoms after P.infestans infection,with more necrotic cells,longer lesion diameters,and increased P.infestans abundance.Meanwhile,silencing of miR482b was performed by short tandem target mimic(STTM),resulting in enhancement of tomato resistance to P.infestans.Using miRNA and degradome data sets,NBS–LRR disease-resistance genes targeted by miR482b were validated.Negative correlation between the expression of miR482b and its target genes was found in all miR482b-overexpressing and-silencing tomato plants.Our results provide insight into tomato miR482b involved in the response to P.infestans infection,and demonstrate that miR482b–NBS–LRR is an important component in the network of tomato–P.infestans interaction.展开更多
A novel scheme for generating optical chaos is proposed and experimentally demonstrated,which supports to simultaneously produce two low-correlation chaotic signals with wideband spectrum and suppressed time-delay-sig...A novel scheme for generating optical chaos is proposed and experimentally demonstrated,which supports to simultaneously produce two low-correlation chaotic signals with wideband spectrum and suppressed time-delay-signature(TDS).In the proposed scheme,we use the output of an external-cavity semiconductor laser(ECSL)as the driving signal of a phase modulator to modulate the output of a CW laser.Then the phase-modulated continuous-wave(CW)light is split into two parts,one is injected back into the ECSL that outputs one chaotic signal,while the other part is passed through a dispersion module for generating another chaotic signal simultaneously.The experimental results prove that the proposed scheme has three merits.Firstly,it can improve the bandwidth of ECSL-based chaos by several times,and simultaneously generate another wideband flat-spectrum chaotic signal.Secondly,the undesired TDS characteristics of the simultaneously-generated chaotic signals can be efficiently suppressed to an indistinguishable level within a wide parameter range,as such the complexities of the chaotic signals are considerably high.Thirdly,the correlation coefficient between these two simultaneously-generated chaotic signals is smaller than 0.1.The proposed scheme provides an attractive solution for parallel multiple chaos generation,and shows great potential for multiple channel chaos communications and multiple random bit generations.展开更多

We also examined the molecular mechanisms tha...We investigated the importance of HMGN5, a nuclear protein that binds to nucleosomes, unfolds chromatin, and affects transcription, in the LNCaP prostate cancer cell line. We also examined the molecular mechanisms that promote apoptosis of LNCaP cells after infection with small interfering RNA (siRNA) targeting HMGN5 (siRNA-HMGN5). The androgen-dependent LNCaP human prostate cancer cells were infected with siRNA-HMGN5. Apoptosis was detected using the Annexin V-PE/7-AAD double staining and the terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling (TUNEL) assays. Mitochondrial membrane potential was measured by JC-1 staining. HMGN5and GAPDHmRNA expression were determined using real-time PCR. Bcl-2 and other apoptosis-related protein levels were determined by Western blot analysis. Caspase activity was measured by cleavage of the caspase substrate. Infection with siRNA targeting HMGN5 efficiently and specifically reduced the HMGN5 expression in LNCaP cells. The downregulation of HMGN5 induced remarkable apoptosis of LNCaP cells and resulted in the reduction of mitochondrial membrane potential. The induction of cell apoptosis was accompanied by the upregulation of Bax, the Bax/Bcl-2 ratio and the activation of caspase3. The HMGN5-targeted siRNA was effective in downregulating the expression of HMGN5 in androgen-dependent prostate cancer cells and inducing cell apoptosis via the regulation of a caspase-related mitochondrial pathway and Bcl-2 family proteins. This study suggests that HMGN5 may be a potential molecular target with therapeutic relevance for the treatment of prostate cancer.展开更多
The presence of ethanol has an adverse effect on foam spreading,and ethanol fire is difficult to extinguish with aqueous fire-fighting foams.Thus,it is necessary to explore the foam formulation suitable for ethanol fu...The presence of ethanol has an adverse effect on foam spreading,and ethanol fire is difficult to extinguish with aqueous fire-fighting foams.Thus,it is necessary to explore the foam formulation suitable for ethanol fuels and study the spreading behavior of foam over ethanol surface.In the current work,stable foams based on hydrocarbon surfactant(SDS),fluorocarbon surfactant(FC1157),and polymers(XG)were prepared by using the compressed-air foam system.The spreading behaviors of foam on polar ethanol and non-polar heptane surface were observed and compared.Furthermore,the effects of stabilizer concentrations,foam flow rates and expansion ratios on foam spreading performance were investigated,respectively.The results indicate that aqueous SDS foam can spread on the heptane layer continuously,but it is difficult to cover the ethanol surface.The addition of XG and FC1157 can synergistically improve the spreading performance of aqueous foam over ethanol.Depending on stabilizer concentrations,there are remarkable differences in foam spreading behaviors.Besides,different foam application parameters including expansion ratios and foam flow rates significantly affect the foam spreading rate,despite the same foam formulation.The research methods and results guide the optimal design of foam formulations as well as the practical application of aqueous foam for ethanol fire extinguishment.展开更多

In this work, we adopt the first principles calculations to investigate the pos-sibility of a new two dimensional boron material, named Be, as anode material for Li-ion batteries. Thecalculated results show that the maximum theoretical specific capacity of Bc is 1653mAh g-1 (LiBl.s).Additionally, the energy barriers of Li ion and Li vacancy diffusion are 330 meV and 110 meV, respec-tively, which imply fast charge and discharge ability for B6 as an anode material. The theoretical findingsreported in this work suggest that BG is a potential candidate as anode material of high-energy-density Li-ion batteries.展开更多
BACKGROUND The poor prognosis and rising incidence of esophageal cancer highlight the need for improved therapeutics that are essential prior to treatment.LCL161 is an SMAC(second mitochondrial activator of caspases)m...BACKGROUND The poor prognosis and rising incidence of esophageal cancer highlight the need for improved therapeutics that are essential prior to treatment.LCL161 is an SMAC(second mitochondrial activator of caspases)mimic and inhibitor of apoptosis protein(IAP)antagonist which exhibits anti-tumor effects and improves the chemical sensitivity of many cancers.AIM To ascertain the effects and mechanisms of the SMAC analog LCL161 on esophageal cancer cells.METHODS MTT assay and TUNEL assay were used to detect cell proliferation and apoptosis,respectively.Western blot analysis was used to study the molecular mechanisms of LCL161-induced death of ECA109 cells.RESULTS LCL161 decreased ECA109 cell proliferation in dose-and time-dependent manner and induced apoptosis of ECA109 cells in a dose-dependent manner.Also,LCL161 induced a significant decrease in the expression of the XIAP and significant increase in the expression of Caspase-3.In addition,Bax increased significantly with increasing concentrations of LCL161,and the relative expression of Bax was significantly different between groups.CONCLUSION These findings support the hypothesis that LCL161 can inhibit proliferation and induce apoptosis in esophageal cancer cells by regulating the expression of IAP family members,suggesting that it has potential to be an effective treatment for esophageal squamous cell carcinoma.展开更多
Traditionally,a delayed(early)onset of the South China Sea summer monsoon(SCSSM)has been observed to follow a warm(cold)El Niño-Southern Oscillation(ENSO)event in winter,supporting high seasonal predictability of...Traditionally,a delayed(early)onset of the South China Sea summer monsoon(SCSSM)has been observed to follow a warm(cold)El Niño-Southern Oscillation(ENSO)event in winter,supporting high seasonal predictability of SCSSM onset.However,the empirical seasonal forecasting skill of the SCSSM onset,solely based on ENSO,has deteriorated since 2010.Meanwhile,unexpected delayed onsets of the SCSSM have also occurred in the past decade.We attribute these changes to the Northwest Indian Ocean(NWIO)warming of the sea surface.The NWIO warming has teleconnections related to(1)suppressing the seasonal convection over the South China Sea,which weakens the impacts of ENSO on SCSSM onset and delays the start of SCSSM,and(2)favoring more high-frequency,propagating moist convective activities,which enhances the uncertainty of the seasonal prediction of SCSSM onset date.Our results yield insight into the predictability of the SCSSM onset under the context of uneven ocean warming operating within the larger-scale background state of global climate change.展开更多

The first coupling mode features the interaction between the Mongolian cyclone over North Asia and the South Asian high(SAH) anomalies over the Tibetan Plateau at 200 hPa. The second mode presents the coupling between the anomalous low-level western Pacific anticyclone and upperlevel SAH via the meridional flow over Southeast Asia. These two modes are responsible for the summer rainfall anomalies over China in 24 and 7 out of 37 years, respectively. However, the dominant SST anomalies in the tropical Pacific, the Indian Ocean, and the North Atlantic Ocean fail to account for the first coupling wheel's interannual variability, illustrating the challenges in forecasting summer rainfall over China.展开更多

文摘Grapes,one of the oldest tree species globally,are rich in vitamins.However,environmental conditions such as low temperature and soil salinization significantly affect grape yield and quality.The glutamate receptor(GLR)family,comprising highly conserved ligand-gated ion channels,regulates plant growth and development in response to stress.In this study,11 members of the VvGLR gene family in grapes were identified using whole-genome sequence analysis.Bioinformatic methods were employed to analyze the basic physical and chemical properties,phylogenetic trees,conserved domains,motifs,expression patterns,and evolutionary relationships.Phylogenetic and collinear analyses revealed that the VvGLRs were divided into three subgroups,showing the high conservation of the grape GLR family.These members exhibited 2 glutamate receptor binding regions(GABAb and GluR)and 3-4 transmembrane regions(M1,M2,M3,and M4).Real-time quantitative PCR analysis demonstrated the sensitivity of all VvGLRs to low temperature and salt stress.Subsequent localization studies in Nicotiana tabacum verified that VvGLR3.1 and VvGLR3.2 proteins were located on the cell membrane and cell nucleus.Additionally,yeast transformation experiments confirmed the functionality of VvGLR3.1 and VvGLR3.2 in response to low temperature and salt stress.Thesefindings highlight the significant role of the GLR family,a highly conserved group of ion channels,in enhancing grape stress resistance.This study offers new insights into the grape GLR gene family,providing fundamental knowledge for further functional analysis and breeding of stress-resistant grapevines.
文摘PCrNi3MoV steel is a medium-carbon,low-alloy quenched and tempered steel that finds its applications in military gun barrels due to the high wear resistance and ablation resistance.To study the penetration and failure modes of PCrNi3MoV plates impacted by tungsten spheres,tungsten spheres of various diameters(5 mm,8 mm,and 10 mm)were used to impact PCrNi3MoV steel plates with thicknesses of 6 mm,9 mm,and 14 mm.The penetration performance of the spheres was analyzed for different velocities,and the ultimate penetration velocity of the plate was obtained.It was found that the primary failure modes of the PCrNi3MoV plate were compression pitting failure and shear failure.Using the dimensional analysis method,a relationship between the bulge height of the steel plate and the fragment velocity,an equation for the ultimate penetration velocity,and a relationship between the target penetration energy and the fragment velocity were obtained.Then,a projectile-target action index was proposed to describe the process of tungsten spheres with different velocities impacting target plates.The results suggested that under the same thickness of the target plate,a larger-diameter fragment required more kinetic energy to obtain the same ultimate penetration effect as a smaller-diameter fragment.The equations obtained through dimensional analysis predicted values that agreed well with experimental values,indicating that these equations can be applied to engineering applications.

文摘Antarctic coastal polynyas are biological hotspots in the Southern Ocean that support the abundance of hightrophic-level predators and are important for carbon cycling in the high-latitude oceans.In this study,we examined the interannual variation of summertime phytoplankton biomass in the Marguerite Bay polynya(MBP)in the western Antarctic Peninsula area,and linked such variability to the Southern Annular Mode(SAM)that dominated the southern hemisphere extratropical climate variability.Combining satellite data,atmosphere reanalysis products and numerical simulations,we found that the interannual variation of summer chlorophyll-a(Chl-a)concentration in the MBP is significantly and negatively correlated with the spring SAM index,and weakly correlated with the summer SAM index.The negative relation between summer Chl-a and spring SAM is due to weaker spring vertical mixing under a more positive SAM condition,which would inhibit the supply of iron from deep layers into the surface euphotic layer.The negative relation between spring mixing and spring SAM results from greater precipitation rate over the MBP region in positive SAM phase,which leads to lower salinity in the ocean surface layer.The coupled physical-biological mechanisms between SAM and phytoplankton biomass revealed in this study is important for us to predict the future variations of phytoplankton biomasses in Antarctic polynyas under climate change.

文摘This studay is to construct a lentiviral vector harbouring an RNA interference (RNAi) sequence that targets the gene encoding the human high-mobility group nucleosomal binding protein 1 (NSBP1); to study its role in inducing G2/M phase arrest and apoptosis in prostate cancer (PCa) DU145 cells; and to assess the effect of its knockdown on cell proliferation in vitro and in vivo. RNAi was applied to knock down NSBP1 expression in the PCa cell line DU145 by lentiviral plasmids producing an NSBP1 small hairpin RNA. After NSBP1 knockdown in DU145 cells, the growth rate of cells was analyzed by MTT, and G2/M cell cycle arrest and apoptosis were assessed using a FAC- Scalibur flow cytometer. Tumour growth was assessed in nude mice. The mRNA and protein expression levels of NSBP1, cyclin B1 and Bcl-2 were analysed in vitro and in vivo by reverse-transcriptase polymerase chain reaction and Western blotting. Knockdown of NSBP1 resulted in a 22.6% decrease in the growth rate of cells compared with the PscNC lentivirus control cells at 96 h, decreased tumour growth in nude mice, and the induction of Gz/M cell cycle arrest (8.78%) and apoptosis (2.19-fold). Consistent with the cell cycle arrest and apoptosis, the mRNA and protein expression levels of cyclin B 1 and Bcl-2 were decreased. In conclusion, knockdown of NSBP 1 causes a statistically significant inhibition of the in vitro and in vivo growth of the PCa cell line DU145. Growth suppression is at least partially due to NSBP1 knockdown-induced Gz/M cell cycle arrest and apoptosis. The present data provide the evidence that the NSBP1 knockdown-induced G2/M phase arrest and apoptosis may result from negative regulation of cyclin B 1 and Bcl-2 by NSBP1, with the resulting reduced expression of these proteins.

文摘It has been suggested that a warm(cold)ENSO event in winter is mostly followed by a late(early)onset of the South China Sea(SCS)summer monsoon(SCSSM)in spring.Our results show this positive relationship,which is mainly determined by their phase correlation,has been broken under recent rapid global warming since 2011,due to the disturbance of cold tongue(CT)La Niña events.Different from its canonical counterpart,a CT La Niña event is characterized by surface meridional wind divergences in the central-eastern equatorial Pacific,which can delay the SCSSM onset by enhanced convections in the warming Indian Ocean and the western subtropical Pacific.Owing to the increased Indian−western Pacific warming and the prevalent CT La Niña events,empirical seasonal forecasting of SCSSM onset based on ENSO may be challenged in the future.

文摘Oxysophoridine, a new alkaloid extracted from Sophora alopecuroides L., has been shown to have a protective effect against ischemic brain damage. In this study, a focal cerebral ischemia/reperfusion injury model was established using middle cerebral artery occlusion in mice. Both 62.5, 125, and 250 mg/kg oxysophoridine, via intraperitoneal injection, and 6 mg/kg nimodipine, via intragastric administration, were administered daily for 7 days before modeling. After 24 hours of reperfusion, mice were tested for neurological deficit, cerebral infarct size was assessed and brain tissue was collected. Results showed that oxysophoridine at 125, 250 mg/kg and 6 mg/kg nimodipine could reduce neurological deficit scores, cerebral infarct size and brain water content in mice. These results provided evidence that oxysophoridine plays a protective role in cerebral ischemia/reperfusion injury. In addition, oxysophoridine at 62.5, 125, and 250 mg/kg and 6 mg/kg nimodipine increased adenosine-triphosphate content, and decreased malondialdehyde and nitric oxide content. These compounds enhanced the activities of glutathione-peroxidase, superoxide dismutase, catalase, and lactate dehydrogenase, and decreased the activity of nitric oxide synthase Protein and mRNA expression levels of N-methyI-D-aspartate receptor subunit NR1 were markedly inhibited in the presence of 250 mg/kg oxysophoridine and 6 mg/kg nimodipine. Our experimental findings indicated that oxysophoridine has a neuroprotective effect against cerebral ischemia/reperfusion injury in mice, and that the effect may be due to its ability to inhibit oxidative stress and expression of the N-methyI-D-aspartate receptor subunit NR1.

文摘AIM: To investigate the expression and role of nuclear factor κB(NF-κB) in diabetic retinopathy(DR) and its relationship with neovascularization and retinal cell apoptosis. METHODS: A total of 80 male Wistar rats were randomly assigned to control(4, 8, 12 and 16 wk, n =10 in each group) and diabetes mellitus(DM) groups(4, 8, 12 and 16wk, n =10 in each group). A diabetic rat model was established by intraperitoneal injection of streptozotocin(60 mg/kg). After 4, 8, 12 and 16 wk, rats were sacrificed.Retinal layers and retinal neovascularization growth were stained with hematoxylin-eosin and examined under light microscopy. Cell apoptosis in the retina was detected by Td T-mediated d UTP nick end labeling, and NF-κB distribution and expression in the retina was determined using immunohistochemistry. RESULTS: DM model success rate up to 100%.Diabetes model at each time point after the experimental groupcompared with the control group, the blood glucose was significantly increased, decreased body weight, each time point showed significant differences compared with the control group(P 【0.01). After 12 wk other pathological changes in the retina of diabetic rats were observed; after 16 wk, neovascularization were observed. After 1mo, retinal cell apoptosis was observed.Compared with the control group, NF-κB expression in the DM group significantly increased with disease duration.CONCLUSION: With the prolonging of DM progression,the expression NF-κB increases. NF-κB may be related to retinal cell apoptosis and neovascularization.

文摘Our previous studies indicated that tomato miR482b could negatively regulate the resistance of tomato to Phytophthora infestans and the expression of miR482b was decreased after inoculation with P.infestans.However,the mechanism by which the accumulation of miR482b is suppressed remains unclear.In this study,we wrote a program to identify 89 long noncoding RNA(lncRNA)-originated endogenous target mimics(eTMs)for 46 miRNAs from our RNA-Seq data.Three tomato lncRNAs,lncRNA23468,lncRNA01308 and lncRNA13262,contained conserved eTM sites for miR482b.When lncRNA23468 was overexpressed in tomato,miR482b expression was significantly decreased,and the expression of the target genes,NBS-LRRs,was significantly increased,resulting in enhanced resistance to P.infestans.Silencing lncRNA23468 in tomato led to the increased accumulation of miR482b and decreased accumulation of NBS-LRRs,as well as reduced resistance to P.infestans.In addition,the accumulation of both miR482b and NBS-LRRs was not significantly changed in tomato plants that overexpressed lncRNA23468 with a mutated eTM site.Based on the VIGS system,a target gene of miR482b,Solyc02g036270.2,was silenced.The disease symptoms of the VIGS-Solyc02g036270.2 tomato plants were in accordance with those of tomato plants in which lncRNA23468 was silenced after inoculation with P.infestans.More severe disease symptoms were found in the modified plants than in the control plants.Our results demonstrate that lncRNAs functioning as eTMs may modulate the effects of miRNAs in tomato and provide insight into how the lncRNA23468-miR482b-NBS-LRR module regulates tomato resistance to P.infestans.

文摘Tomato is an important horticultural and economic crop cultivated worldwide.As Phytophthora infestans becomes a huge threat to tomato production,it is necessary to study the resistance mechanisms of tomato against P.infestans.Our previous research has found that miR482 might be involved in tomato–P.infestans interaction.In this study,miR482b precursor was cloned from Solanum pimpinellifolium“L3708”and miR482b was shown to decrease in abundance in tomato following P.infestans infection.Compared to wild-type tomato plants,tomato plants that overexpressed miR482b displayed more serious disease symptoms after P.infestans infection,with more necrotic cells,longer lesion diameters,and increased P.infestans abundance.Meanwhile,silencing of miR482b was performed by short tandem target mimic(STTM),resulting in enhancement of tomato resistance to P.infestans.Using miRNA and degradome data sets,NBS–LRR disease-resistance genes targeted by miR482b were validated.Negative correlation between the expression of miR482b and its target genes was found in all miR482b-overexpressing and-silencing tomato plants.Our results provide insight into tomato miR482b involved in the response to P.infestans infection,and demonstrate that miR482b–NBS–LRR is an important component in the network of tomato–P.infestans interaction.

文摘A novel scheme for generating optical chaos is proposed and experimentally demonstrated,which supports to simultaneously produce two low-correlation chaotic signals with wideband spectrum and suppressed time-delay-signature(TDS).In the proposed scheme,we use the output of an external-cavity semiconductor laser(ECSL)as the driving signal of a phase modulator to modulate the output of a CW laser.Then the phase-modulated continuous-wave(CW)light is split into two parts,one is injected back into the ECSL that outputs one chaotic signal,while the other part is passed through a dispersion module for generating another chaotic signal simultaneously.The experimental results prove that the proposed scheme has three merits.Firstly,it can improve the bandwidth of ECSL-based chaos by several times,and simultaneously generate another wideband flat-spectrum chaotic signal.Secondly,the undesired TDS characteristics of the simultaneously-generated chaotic signals can be efficiently suppressed to an indistinguishable level within a wide parameter range,as such the complexities of the chaotic signals are considerably high.Thirdly,the correlation coefficient between these two simultaneously-generated chaotic signals is smaller than 0.1.The proposed scheme provides an attractive solution for parallel multiple chaos generation,and shows great potential for multiple channel chaos communications and multiple random bit generations.
文摘We investigated the importance of HMGN5, a nuclear protein that binds to nucleosomes, unfolds chromatin, and affects transcription, in the LNCaP prostate cancer cell line. We also examined the molecular mechanisms that promote apoptosis of LNCaP cells after infection with small interfering RNA (siRNA) targeting HMGN5 (siRNA-HMGN5). The androgen-dependent LNCaP human prostate cancer cells were infected with siRNA-HMGN5. Apoptosis was detected using the Annexin V-PE/7-AAD double staining and the terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling (TUNEL) assays. Mitochondrial membrane potential was measured by JC-1 staining. HMGN5and GAPDHmRNA expression were determined using real-time PCR. Bcl-2 and other apoptosis-related protein levels were determined by Western blot analysis. Caspase activity was measured by cleavage of the caspase substrate. Infection with siRNA targeting HMGN5 efficiently and specifically reduced the HMGN5 expression in LNCaP cells. The downregulation of HMGN5 induced remarkable apoptosis of LNCaP cells and resulted in the reduction of mitochondrial membrane potential. The induction of cell apoptosis was accompanied by the upregulation of Bax, the Bax/Bcl-2 ratio and the activation of caspase3. The HMGN5-targeted siRNA was effective in downregulating the expression of HMGN5 in androgen-dependent prostate cancer cells and inducing cell apoptosis via the regulation of a caspase-related mitochondrial pathway and Bcl-2 family proteins. This study suggests that HMGN5 may be a potential molecular target with therapeutic relevance for the treatment of prostate cancer.

文摘The presence of ethanol has an adverse effect on foam spreading,and ethanol fire is difficult to extinguish with aqueous fire-fighting foams.Thus,it is necessary to explore the foam formulation suitable for ethanol fuels and study the spreading behavior of foam over ethanol surface.In the current work,stable foams based on hydrocarbon surfactant(SDS),fluorocarbon surfactant(FC1157),and polymers(XG)were prepared by using the compressed-air foam system.The spreading behaviors of foam on polar ethanol and non-polar heptane surface were observed and compared.Furthermore,the effects of stabilizer concentrations,foam flow rates and expansion ratios on foam spreading performance were investigated,respectively.The results indicate that aqueous SDS foam can spread on the heptane layer continuously,but it is difficult to cover the ethanol surface.The addition of XG and FC1157 can synergistically improve the spreading performance of aqueous foam over ethanol.Depending on stabilizer concentrations,there are remarkable differences in foam spreading behaviors.Besides,different foam application parameters including expansion ratios and foam flow rates significantly affect the foam spreading rate,despite the same foam formulation.The research methods and results guide the optimal design of foam formulations as well as the practical application of aqueous foam for ethanol fire extinguishment.

文摘To design the high-energy-density Li-ion batteries, the anode materials with high specific capacity haveattracted much attention. In this work, we adopt the first principles calculations to investigate the pos-sibility of a new two dimensional boron material, named Be, as anode material for Li-ion batteries. Thecalculated results show that the maximum theoretical specific capacity of Bc is 1653mAh g-1 (LiBl.s).Additionally, the energy barriers of Li ion and Li vacancy diffusion are 330 meV and 110 meV, respec-tively, which imply fast charge and discharge ability for B6 as an anode material. The theoretical findingsreported in this work suggest that BG is a potential candidate as anode material of high-energy-density Li-ion batteries.
文摘BACKGROUND The poor prognosis and rising incidence of esophageal cancer highlight the need for improved therapeutics that are essential prior to treatment.LCL161 is an SMAC(second mitochondrial activator of caspases)mimic and inhibitor of apoptosis protein(IAP)antagonist which exhibits anti-tumor effects and improves the chemical sensitivity of many cancers.AIM To ascertain the effects and mechanisms of the SMAC analog LCL161 on esophageal cancer cells.METHODS MTT assay and TUNEL assay were used to detect cell proliferation and apoptosis,respectively.Western blot analysis was used to study the molecular mechanisms of LCL161-induced death of ECA109 cells.RESULTS LCL161 decreased ECA109 cell proliferation in dose-and time-dependent manner and induced apoptosis of ECA109 cells in a dose-dependent manner.Also,LCL161 induced a significant decrease in the expression of the XIAP and significant increase in the expression of Caspase-3.In addition,Bax increased significantly with increasing concentrations of LCL161,and the relative expression of Bax was significantly different between groups.CONCLUSION These findings support the hypothesis that LCL161 can inhibit proliferation and induce apoptosis in esophageal cancer cells by regulating the expression of IAP family members,suggesting that it has potential to be an effective treatment for esophageal squamous cell carcinoma.

文摘Traditionally,a delayed(early)onset of the South China Sea summer monsoon(SCSSM)has been observed to follow a warm(cold)El Niño-Southern Oscillation(ENSO)event in winter,supporting high seasonal predictability of SCSSM onset.However,the empirical seasonal forecasting skill of the SCSSM onset,solely based on ENSO,has deteriorated since 2010.Meanwhile,unexpected delayed onsets of the SCSSM have also occurred in the past decade.We attribute these changes to the Northwest Indian Ocean(NWIO)warming of the sea surface.The NWIO warming has teleconnections related to(1)suppressing the seasonal convection over the South China Sea,which weakens the impacts of ENSO on SCSSM onset and delays the start of SCSSM,and(2)favoring more high-frequency,propagating moist convective activities,which enhances the uncertainty of the seasonal prediction of SCSSM onset date.Our results yield insight into the predictability of the SCSSM onset under the context of uneven ocean warming operating within the larger-scale background state of global climate change.

文摘Two types of three-dimensional circulation of the East Asian summer monsoon(EASM) act as the coupling wheels determining the seasonal rainfall anomalies in China during 1979–2015. The first coupling mode features the interaction between the Mongolian cyclone over North Asia and the South Asian high(SAH) anomalies over the Tibetan Plateau at 200 hPa. The second mode presents the coupling between the anomalous low-level western Pacific anticyclone and upperlevel SAH via the meridional flow over Southeast Asia. These two modes are responsible for the summer rainfall anomalies over China in 24 and 7 out of 37 years, respectively. However, the dominant SST anomalies in the tropical Pacific, the Indian Ocean, and the North Atlantic Ocean fail to account for the first coupling wheel's interannual variability, illustrating the challenges in forecasting summer rainfall over China.
