Myoglobin produced by fermentation using engineered Komagataella phaffii is an important color additive in meat analogue products,but its allergenicity is poorly understood.Here,we initially searched the Allergen Online database and did not find any allergic or cross-reactive proteins in porcine myoglobin(PM).In vitro simulated digestion demonstrated that PM did not exhibit notable acid-base resistance or anti-digestion capabilities.However,sensitization was observed in BALB/c mice,including a significant increase in specific antibodies and biomarkers for allergic reactions,as well as alterations in gut microbiome and serum metabolome.Interestingly,the intensity of sensitization exhibited a negative correlation with the purity of PM.60%and 88%purities showed weaker sensitization compared to the ovalbumin control group.These allergic reactions were likely due to the non-myoglobin protein portion,highlighting the importance of purification processes and the urgent need to assess the allergenicity of this portion. 展开更多

Insufficient sleep can produce a multitude of deleterious repercussions on various domains of human well-being.Concomitantly,the walnut(Juglans mandshurica)confers numerous salutary biological activities pertaining to sleep.Nevertheless,the sedative and hypnotic capacities of walnut's functional constituents remain obscure.In this investigation,we analyzed the sedative and hypnotic components of the walnut Diaphragma juglandis fructus and innovatively discovered a compound,defined as 3-hydroxy-4-iminobutyric acid(HIBA),which disrupts motor activity and enhances sleep duration by regulating the neurotransmitters(GABA,DA,etc.)within the brain and serum of mice.Subsequently,a metabolomics approach of the serum,basal ganglia,hypothalamus,and hippocampus as well as the gut microbiota was undertaken to unravel the underlying molecular mechanisms of sleep promotion.Our data reveal that HIBA can regulate the metabolism of basal ganglia(sphingolipids,acylcarnitines,etc.),possibly in relation to HIBA's influence on the gut microbiome(Muribaculum,Bacteroides,Lactobacillus,etc.).Therefore,we introduce a novel natural product,HIBA,and explicate the modulation of sleep promotion in mice based on the microbiota-gut-brain axis.This study contributes fresh insights toward natural product-based sleep research. 展开更多

Carbon dots/graphite carbon nitride(CDs/g-C_(3)N_(4)),a novel composite photocatalyst,has shown great po-tential for applications in energy regeneration and environmental remediation owing to its following advantages:metal-free,low cost,easily tunable,and excellent photocatalytic performance.Hence,we reviewed the development of synthetic strategies,photocatalytic enhancement mechanisms,and pho-tocatalytic applications of CDs/g-C_(3)N_(4) in this study.First,the three composite strategies of CDs and g-C_(3)N_(4)-self-assembly,solvothermal,and calcination polymerization are outlined,and their advantages and disadvantages are described in detail.Moreover,the photocatalytic enhancement mechanism of the com-posite strategies was elucidated according to the variation trends of CDs/g-C_(3)N_(4) band structure,electronic properties,light absorption range,and interfacial charge transfer.Then,the applications of CDs/g-C_(3)N_(4) in hydrogen evolution,pollutant degradation,CO_(2) reduction,and bacterial disinfection in recent years are reviewed systematically.Finally,the current obstacles and future research directions of CDs/g-C_(3)N_(4) are discussed from the perspective of preparation technology and practical applications,respectively. 展开更多

Fumonisin B1(FB1)is a mycotoxin that is harmful to the nervous system;however,there is currently a lack of studies on plant compounds that alleviate the neurotoxicity of FB1.Polygonatum sibiricum polysaccharides exhibit neuroprotective effects,but it is unclear whether they protect against FB1-induced neurotoxicity.Here,Caenorhabditis elegans(C.elegans)was used to study the alleviation effects of P.sibiricum polysaccharide extract(PSPE)preprotection on FB1-induced neurotoxicity.PSPE pretreatment improved the behavioral deficits and neuronal damage in FB1-induced C.elegans.In addition,20-200μg/mL PSPE alleviated the oxidative damage in FB1-induced C.elegans by regulating the levels of reactive oxygen species,antioxidant enzyme activities and gene expression.Further studies have shown that PSPE pretreatment induced mitochondrial expression,increased ATP levels and mitochondrial membrane potential,and regulated mitochondrial oxidative phosphorylation and dynamics.Together,the protective effects of PSPE against FB1-induced neurotoxicity might be closely related with the reduction of oxidative stress and the regulation of mitochondria. 展开更多

Isolation and characterization of uric acid-lowering functional components from Polygonum cuspidatum

Polygonum cuspidatum(P.cuspidatum)is an herbaceous perennial plant known as Asian knotweed that is used in Chinese medicine due to its active components.Recent studies of the uric acid-lowering activity of P.cuspidatum extracts have mainly focused on analysis of its crude extract or specific chemical components,with limited research about its uric acid-lowering activity in vivo.In present study,monomeric compounds with higher uric acid-lowering ability were isolated from P.cuspidatum extract by column chromatography and thin-layer chromatography(TLC)and tested in a hyperuricemia mouse model and xanthine oxidase(XOD)inhibition assay in vitro.Structural characterization of the isolated components was performed by nuclear magnetic resonance(NMR)spectroscopy and the potential mechanism of decreasing uric acid was analyzed using untargeted metabolomics.Two main novel compounds were isolated and identified,1-(4-hydroxy-2-methoxyphenyl)-2-(4-hydroxy-3,5-dimethylphenyl)butane-1,2,3-triol(compound 1)and 1-(4-hydroxy-2-methoxyphenyl)-2-(4-hydroxy-3,5-dimethylphenyl)-3-methylbutane-1,2-diol(compound 2),and compounds 1 was proved more significant inhibitory effect on the activity of XOD(p<0.01)with inhibition of 49.80%.Metabolomics analysis indicated that the fractions(compound 1 and compound 2)affected hyperuricemic mice mainly through changes in galactose metabolism(p=5.74E-4)and the effects were closely related to metabolic pathways of organic acid,carbohydrate,and lipid metabolism.These results contribute to providing theoretical support for the utilization of P.cuspidatum as a food and medicinal material,which can increase its use as an alternative treatment for hyperuricemia. 展开更多
