BACKGROUND Tacrolimus(FK506)is a key calcineurin inhibitor used to prevent organ transplant rejection and is effective in improving graft survival.However,it is linked to hyperglycemia and insulin resistance,contributing to new-onset diabetes after transplantation and negatively affecting islet function.AIM To study the effects of tacrolimus on the insulin signaling pathway of hepatocytes.METHODS HL7702 cells were treated with different concentrations of tacrolimus(0.1 mg/L,1 mg/L,5 mg/L)for 24 hours.The proteins involved in insulin signaling were detected by Western blotting.RESULTS Compared with the control group,phosphorylation of insulin receptor substrate(IRS)1 at Ser 307 and Ser 323 were increased significantly when the tacrolimus concentration reached 1 and 5 mg/L.Phosphorylation of IRS1 at Ser 1101 was also increased,although not significantly.However,phosphorylation of Ribosomal protein S6 kinase beta-1 at Thr 389 was decreased significantly.The levels of phosphorylated glycogen synthase kinase 3αSer 21 and Ser 9 were increased.Surprisingly,phosphorylation of glycogen synthase at Ser 641 was increased.There was no significant change in the activity of glycogen phosphorylase.CONCLUSION Tacrolimus has no direct effect on hepatic glucose metabolism,but inhibits IRS1-mediated insulin signaling.This may be one of the underlying mechanisms by which tacrolimus induces insulin resistance. 展开更多

Generation of transgenic chicken through ovarian injection

Background : Traditional DNA microinjection methods used in mammals are difficult to apply to avian species due to their unique reproductive characteristics. Genetic manipulation in chickens, particularly involving immature follicles within living ovaries, has not been extensively explored. This study seeks to establish an efficient method for generating transgenic chickens through ovarian injection, potentially bypassing the challenges associated with primordial germ cell (PGC) manipulation and fertilized egg microinjection. Methods : Hens were anesthetized and underwent a surgical procedure to access the ovary for DNA injection into immature follicles. The study used liposomes to deliver GFP- expressing plasmids at various dosages. After injection, hens recovered, and their eggs were fertilized through artificial insemination. Results : Transgenic chickens were successfully generated in one generation without needing G0 founders. The injection of 20 μg plasmid yielded the highest transgenic efficiency at 12.1%. GFP- positive embryos were confirmed through microscopy, and successful transgene expression was validated at the tissue level using immunostaining. TERT and GFP elements introduced in the G1 generation resulted in 4.1% positive transgene rates, as confirmed by PCR and Southern blotting. Conclusion : This ovarian injection method offers a promising alternative for avian genetic manipulation, bypassing complex PGC procedures and enabling direct generation of G1 transgenic chickens. This technique simplifies the transgenic process for chickens and has the potential to be adapted for other avian species, especially those without established PGCs culture systems. 展开更多

Objective:To observe and study the actual effects of percutaneous pedicle screw minimally invasive surgery in the treatment of spinal fractures and its impact on spinal function.Methods:This study included 48 patients with spinal fractures admitted between May 2023 and May 2024.The patients were divided into a control group and an experimental group based on treatment differences,with 24 patients in each group.The control group underwent open internal fixation surgery,while the experimental group received percutaneous pedicle screw minimally invasive surgery.Clinical index improvements,cervical dysfunction index,Japanese Orthopaedic Association scores,and pain level improvements were compared between the two groups.Results:The intraoperative blood loss,incision length,operation time,and hospitalization duration in the experimental group were(88.63±18.85),(6.32±1.05),(73.42±4.05),and(12.58±2.56),respectively,compared to(279.95±17.32),(12.89±1.36),(89.93±4.79),and(22.41±2.87)in the control group.Significant differences were observed between the groups,with the experimental group showing superior improvements across all metrics(P<0.05).Conclusion:Percutaneous pedicle screw minimally invasive surgery shows more significant effects in treating spinal fractures,particularly in improving cervical and lumbar spine function,enhancing treatment efficacy and safety,reducing pain levels,and shortening recovery time.Clinical application and promotion are recommended. 展开更多

Objective:To evaluate and analyze the actual efficacy of percutaneous vertebroplasty in the treatment of old unstable osteoporotic spinal fractures.Methods:From March 2023 to March 2024,46 patients with old unstable osteoporotic spinal fractures in our hospital were included in this study.They were divided into the conventional group and the observation group based on treatment differences,with 23 patients in each group.The conventional group received conservative drug therapy,while the observation group underwent percutaneous vertebroplasty.The following indicators were compared and analyzed between the two groups:clinical treatment effect and improvement in physical function indicators.Results:The treatment efficiency of the observation group was 95.65%(22/23),while that of the conventional group was 69.57%(16/23).There was a significant difference between the groups,and the treatment effect of the observation group was significantly better(P<0.05).After treatment,the scores of physical status,daily living ability,functional independence,and life obstacles in the observation group were(89.33±4.08),(88.72±4.08),(90.41±2.89),(72.35±3.22),respectively,while those in the conventional group were(68.54±4.21),(67.42±4.11),(73.48±2.75),(72.35±3.22).There was a significant difference between the groups,and the improvement in physical function indicators in the observation group was more pronounced(P<0.05).Conclusion:For patients with old unstable osteoporotic spinal fractures,the basic value of percutaneous vertebroplasty is significant.It can not only improve clinical efficacy and safety but also promote the gradual recovery of patients'physical function indicators.It is recommended for clinical reference and practical application. 展开更多

Background Rosemary extract(RE)has been reported to exert antioxidant property.However,the application of RE in late-phase laying hens on egg quality,intestinal barrier and microbiota,and oviductal function has not been systematically studied.This study was investigated to detect the potential effects of RE on performance,egg quality,serum parameters,intestinal heath,cecal microbiota and metabolism,and oviductal gene expressions in late-phase laying hens.A total of 21065-week-old“Jing Tint 6”laying hens were randomly allocated into five treatments with six replicates and seven birds per replicate and fed basal diet(CON)or basal diet supplemented with chlortetracycline at 50 mg/kg(CTC)or RE at 50 mg/kg(RE50),100 mg/kg(RE100),and 200 mg/kg(RE200).Results Our results showed that RE200 improved(P<0.05)Haugh unit and n-6/n-3 of egg yolk,serum superoxide dismutase(SOD)compared with CON.No significant differences were observed for Haugh unit and n-6/n-3 of egg yolk among CTC,RE50,RE100 and RE200 groups.Compared with CTC and RE50 groups,RE200 increased serum SOD activity on d 28 and 56.Compared with CON,RE supplementation decreased(P<0.05)total cholesterol(TC)level.CTC,RE100 and RE200 decreased(P<0.05)serum interleukin-6(IL-6)content compared with CON.CTC and RE200 increased jejunal m RNA expression of ZO-1 and Occludin compared with CON.The biomarkers of cecal microbiota and metabolite induced by RE 200,including Firmicutes,Eisenbergiella,Paraprevotella,Papillibacter,and butyrate,were closely associated with Haugh unit,n-6/n-3,SOD,IL-6,and TC.PICRUSt2 analysis indicated that RE altered carbohydrate and amino acid metabolism of cecal microbiota and increased butyrate synthesizing enzymes,including 3-oxoacid Co A-transferase and butyrate-acetoacetate Co A-transferase.Moreover,transcriptomic analysis revealed that RE200 improved gene expressions and functional pathways related to immunity and albumen formation in the oviductal magnum.Conclusions Dietary supplementation with 200 mg/kg RE could increase egg quality of late-phase laying hens via modulating intestinal barrier,cecal microbiota and metabolism,and oviductal function.Overall,RE could be used as a promising feed additive to improve egg quality of laying hens at late stage of production. 展开更多

One-step preparation of efficient cuprous chloride catalyst for direct synthesis of trimethoxysilane

CuCl-based catalysts are the most commonly used catalysts for the“direct synthesis”of trimethoxysilane(M3).CuCl species are sensitive to air and water,and are prone to oxidation deactivation.When CuCl is directly used as a catalyst,it needs to be purified before the utilization,and the operating conditions for the catalyst preparation are relatively harsh,requiring the inert gas environment.Considering a high-temperature activation step required for CuCl-based catalysts used for catalyzing synthesis of M3 to form active phase Cu–Si alloys(Cu_(x)Si)with Si powder,in this work,a series of catalysts for the“direct synthesis”of M3 were obtained by a one-step high-temperature activation of the mixture of stable CuCl_(2) precursors,activated carbon-reducing agent,and Si powder,simultaneously achieving the reduction of CuCl_(2) to CuCl and the formation of active phase Cu_(x)Si alloys of CuCl with Si powder.The prepared samples were characterized through various characterization techniques,and investigated for the catalytic performance for the“direct synthesis”of M3.Moreover,the operation conditions were optimized,including the activation temperature,catalyst dosage,Si powder particle size,and reaction temperature.The characterization results indicate that during the one-step activation process,the CuCl_(2) precursor is reduced to CuCl,and the resulting CuCl simultaneously reacts with Si powder to form active phases Cu3Si and Cu15Si4 alloys.The optimal catalyst Sacm(250,0.8:10)exhibits a good catalytic activity with selectivity of 95%and yield of 77%for M3,and shows a good universality for various alcohol substrates.Furthermore,the catalytic mechanism of the prepared catalyst for the“direct synthesis”of M3 was discussed. 展开更多

It is of vital significance to investigate mass transfer enhancements for chemical engineering processes.This work focuses on investigating the coupling influence of embedding wire mesh and adding solid particles on bubble motion and gas-liquid mass transfer process in a bubble column.Particle image velocimetry(PIV)technology was employed to analyze the flow field and bubble motion behavior,and dynamic oxygen absorption technology was used to measure the gas-liquid volumetric mass transfer coefficient(kLa).The effect of embedding wire mesh,adding solid particles,and wire mesh coupling solid particles on the flow characteristic and kLa were analyzed and compared.The results show that the gas-liquid interface area increases by 33%-72%when using the wire mesh coupling solid particles strategy compared to the gas-liquid two-phase flow,which is superior to the other two strengthening methods.Compared with the system without reinforcement,kLa in the bubble column increased by 0.5-1.8 times with wire mesh coupling solid particles method,which is higher than the sum of kLa increases with inserting wire mesh and adding particles,and the coupling reinforcement mechanism for affecting gas-liquid mass transfer process was discussed to provide a new idea for enhancing gas-liquid mass transfer. 展开更多

Power transfer efficiency in an air-breathing radio frequency ion thruster

Due to a series of challenges such as low-orbit maintenance of satellites, the air-breathing electric propulsion has got widespread attention. Commonly, the radio frequency ion thruster is favored by low-orbit missions due to its high specific impulse and efficiency. In this paper, the power transfer efficiency of the radio frequency ion thruster with different gas compositions is studied experimentally, which is obtained by measuring the radio frequency power and current of the antenna coil with and without discharge operation. The results show that increasing the turns of antenna coils can effectively improve the radio frequency power transfer efficiency, which is due to the improvement of Q factor. In pure N_2 discharge,with the increase of radio frequency power, the radio frequency power transfer efficiency first rises rapidly and then exhibits a less steep increasing trend. The radio frequency power transfer efficiency increases with the increase of gas pressure at relatively high power, while declines rapidly at relatively low power. In N_(2)/O_(2) discharge, increasing the N_(2) content at high power can improve the radio frequency power transfer efficiency, but the opposite was observed at low power. In order to give a better understanding of these trends, an analytic solution in limit cases is utilized, and a Langmuir probe was employed to measure the electron density. It is found that the evolution of radio frequency power transfer efficiency can be well explained by the variation of plasma resistance, which is related to the electron density and the effective electron collision frequency. 展开更多

MicroRNAs(miRNAs)are versatile regulators of gene expression at both the transcription and posttranscription levels.The microRNA miR396 plays vital roles in growth,development,and resistance to abiotic stresses in many plant species.However,the roles and functions of miR396 in soybeans are not well understood.Here,we report that Gm-miR396a influences soybean development and salinity tolerance.We found that soybean miR396a was responsive to salt stress.Gm-miR396a gene-edited lines(miR396a-GEs),created using CRISPR/Cas9,exhibited more branches,higher grain yields,and greater salinity tolerance than control plants.The transcripts in lines with altered abundance of miR396a-GE were significantly enriched for biological processes related to hormone regulation.Overexpression of the Gm-miR396a precursor(pre-miR396a-OE)resulted in developmental deficiencies including dwarfness,abnormal inflorescences and flowers,smaller and fewer seeds,and small leaves with larger and more numerous stomata.Transcriptome analysis indicated photosynthesis-related genes were downregulated in pre-miR396a-OE plants.These results contribute valuable insights into the function of GmmiR396a in soybeans and hold promise for enhancing soybean yield and salinity tolerance through germplasm innovation. 展开更多

The dual cylindrical inductively coupled plasma source,compared to the conventional structure of inductively coupled plasma source,can significantly improve the uniformity of plasma.It has an enhanced potential for application in processes,such as etching and ashing.A uniform plasma can be obtained by allowing the remote plasma from the upper chamber modulate the main plasma generated in the lower chamber.In this study,a fluid model was employed to investigate a dual cylindrical inductively coupled Ar/O_(2)discharge.The effects of external parameters on electron density,electron temperature,O atomic density,and plasma uniformity in the main chamber were studied,and the reasons were analyzed.The results of this study show that remote power can control the plasma uniformity and increase the plasma density in the main chamber.As the remote power increased,plasma uniformity improved initially and then deteriorated.The main power affected the plasma density at the edge of the main chamber and can modulate the plasma density in the main chamber.The gas pressure affected both the uniformity and density of the plasma.As the gas pressure increased,the plasma uniformity deteriorated,but the free radical density improved. 展开更多

The electrical parameters of H_(2)/Ar plasma in a cylindrical inductive discharge with an expansion region are investigated by a Langmuir probe,where Ar fractions range from 0%to 100%.The influence of gas composition and pressure on electron density,the effective electron temperature and the electron energy probability functions(EEPFs)at different spatial positions are present.In driver region,with the introduction of a small amount of Ar at 0.3 Pa,there is a rapid increase in electron density accompanied by a decrease in the effective electron temperature.Additionally,the shape of the EEPF transitions from a three-temperature distribution to a bi-Maxwellian distribution due to an increase in electron-electron collision.However,this phenomenon resulting from the changes in gas composition vanishes at 5 Pa due to the prior depletion of energetic electrons caused by the increase in pressure during hydrogen discharge.The EEPFs for the total energy in expansion region is coincident to these in the driver region at 0.3 Pa,as do the patterns of electron density variation between these two regions for differing Ar fractions.At 5 Pa,as the discharge transitions from H_(2)to Ar,the EEPFs evolved from a bi-Maxwellian distribution with pronounced low energy electrons to a Maxwellian distribution in expansion region.This evolve may be attributed to a reduction in molecular vibrational excitation reactions of electrons during transport and the transition from localized electron dynamics in hydrogen discharge to non-localized electron dynamics in argon discharge.In order to validate the experimental results,we use the COMSOL simulation software to calculate electrical parameters under the same conditions.The evolution and spatial distribution of the electrical parameters of the simulation results agree well with the trend of the experimental results. 展开更多

Inhibition of sympathetic tone via hypothalamic descending pathway propagates glucocorticoid-induced endothelial impairment and osteonecrosis of the femoral head

Osteonecrosis of the femoral head(ONFH)is a common complication of glucocorticoid(GC)therapy.Recent advances demonstrate that sympathetic nerves regulate bone homeostasis,and GCs lower the sympathetic tone.Here,we show that the dramatically decreased sympathetic tone is closely associated with the pathogenesis of GC-induced ONFH.GCs activate the glucocorticoid receptor(GR)but hinder the activation of the mineralocorticoid receptor(MR)on neurons in the hypothalamic paraventricular nucleus(PVN).This disrupts the balance of corticosteroid receptors(GR/MR)and subsequently reduces the sympathetic outflow in the PVN.Vascular endothelial cells rapidly react to inhibition of sympathetic tone by provoking endothelial apoptosis in adult male mice treated with methylprednisolone(MPS)daily for 3 days,and we find substantially reduced H-type vessels in the femoral heads of MPS-treated ONFH mice.Importantly,treatment with a GR inhibitor(RU486)in the PVN promotes the activation of MR and rebalances the ratio of GR and MR,thus effectively boosting sympathetic outflow,as shown by an increase in tyrosine hydroxylase expression in both the PVN and the sympathetic postganglionic neurons and an increase in norepinephrine levels in both the serum and bone marrow of the femoral head of MPS-treated mice.Rebalancing the corticosteroid receptors mitigates GC-induced endothelial impairment and ONFH and promotes angiogenesis coupled with osteogenesis in the femoral head,while these effects are abolished by chemical sympathectomy with 6-OHDA or adrenergic receptor-β2(Adrb2)knockout.Furthermore,activating Adrb2 signaling in vivo is sufficient to rescue the GC-induced ONFH phenotype.Mechanistically,norepinephrine increases the expression of the key glycolytic gene 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase 3(PFKFB3)via Adrb2-cyclic AMP response element-binding protein(CREB)signaling.Endothelial-specific overexpression of PFKFB3 attenuates endothelial impairment and prevents severe osteonecrosis in MPS-treated Adrb2 knockout mice.Thus,GC inhibits sympathetic tone via the hypothalamic descending pathway,which,in turn,acts as a mediator of GC-induced ONFH. 展开更多

Aiming at the problem of low accuracy of interpolation error calculation of existing NURBS curves, an approximate method for the distance between a point and a NURBS interpolation curve is proposed while satisfying the accuracy of the solution. Firstly, the minimum parameter interval of the node vector corresponding to the data point under test in the original data point sequence is determined, and the parameter interval is subdivided according to the corresponding step size, and the corresponding parameter value is obtained. Secondly, the distance from the measured point to the NURBS curve is calculated, and the nearest distance is found out. The node interval is subdivided again on one side of the nearest distance. Finally, the distance between the data point to be measured and each subdivision point is calculated again, and the minimum distance is taken as the interpolation error between the point and the NURBS curve. The simulation results of actual tool position data show that this method can more accurately obtain the error of spatial NURBS interpolation curve. 展开更多

This study aimed to determine the effectiveness of comprehensive rehabilitation for patients with aneurysmal subarachnoid hemorrhage (aSAH) and to explore the factors influencing the prognosis of rehabilitation. This was a retrospective study. Twenty-five patients with aSAH were treated with physical therapy, occupational therapy, speech therapy, cognitive therapy, music therapy, Chinese acupuncture, hyperbaric oxygen, and transcranial magnetic stimulation. The general data of all patients were collected, and the functional scores at admission were compared with those at discharge. The Mini Mental State Examination, Fugl-Meyer Assessment Scale (FMAS) for motor and balance assessment, Holden Functional Ambulation Classification (FAC), modified Rankin Scale, National Institute of Health Stroke Scale, Modified Barthel Index for activities of daily living (ADL), and Glasgow Outcome Scale were significantly improved among 25 patients with aSAH after 1 month of comprehensive rehabilitation training. Hydrocephalus was an independent factor of the ability to perform ADLs (odds ratio, 0.29;95% confidence interval, 2.03. 3.15;p = 0.000). The improvement of ADLs in aSAH patients was not related to sex, surgical method, aneurysm location, age, or smoking status. Comprehensive and professional rehabilitation is effective for the cognition, movement, walking, ADLs, and functional prognosis of patients with aSAH, while early hydrocephalus may be a risk factor for poor ADLs. 展开更多

睡眠剥夺是一种能够实现个体睡眠部分或完全丧失的技术手段,由睡眠干扰所引发的逐渐积累的睡眠压力能导致多种生理方面的变化,甚至是个体的死亡。在本次研究中,我们使用旋转滚动式睡眠剥夺仪对大鼠进行了长达14天的睡眠剥夺,同时我们制... 睡眠剥夺是一种能够实现个体睡眠部分或完全丧失的技术手段,由睡眠干扰所引发的逐渐积累的睡眠压力能导致多种生理方面的变化,甚至是个体的死亡。在本次研究中,我们使用旋转滚动式睡眠剥夺仪对大鼠进行了长达14天的睡眠剥夺,同时我们制作了一种16通道的微电极阵列并将其植入到大鼠海马区进行实时电生理信号检测。结果显示睡眠剥夺之后,大鼠海马区内的椎体神经元和中间神经元动作电位幅值提升,两种神经元动作电位的发放频率也显著增大。同时,场电位的波动也更加剧烈。神经细胞在睡眠剥夺后的快速发放模式表明长期清醒状态下神经细胞兴奋性的提升。此外,场电位在0–50 Hz频段的平均功率计算结果显示,睡眠剥夺之后各个频段的功率均有所提升,且在δ频段的变化最为明显。场电位在低频段的功率改变表明了睡眠剥夺所致的睡眠压力增大,此改变还将会进一步损伤大脑的相关功能。 展开更多

目的:分析腹腔镜辅助胃癌根治术后出现切口愈合不良的危险因素。方法:回顾性选取2017年3月至2020年7月郑州大学第二附属医院296例行腹腔镜辅助胃癌根治术的患者作为研究对象,统计分析切口愈合不良的发生与性别、年龄、BMI、糖尿病、高... 目的:分析腹腔镜辅助胃癌根治术后出现切口愈合不良的危险因素。方法:回顾性选取2017年3月至2020年7月郑州大学第二附属医院296例行腹腔镜辅助胃癌根治术的患者作为研究对象,统计分析切口愈合不良的发生与性别、年龄、BMI、糖尿病、高血压、术前血红蛋白和血清白蛋白水平、术前使用抗凝药、新辅助化疗、手术时间、术中出血量、肿瘤大小以及术后行腹腔热灌注化疗术的关系。并行多因素Logistic回归分析引起切口愈合不良的独立危险因素。结果:296例患者中17例(5.7%)出现切口愈合不良,其中切口感染11例,脂肪液化6例。单因素分析结果显示切口愈合良好组与切口愈合不良组患者的年龄、BMI、糖尿病史、血红蛋白水平、血清白蛋白水平的差异具有统计学意义(P<0.05)。进一步行多因素Logistic回归分析,结果显示年龄>65岁(OR=3.434,95%CI:1.035~11.394;P=0.044),BMI(OR=0.629,95%CI:1.365~2.579;P<0.001),血清白蛋白≤35 g/L(OR=1.368,95%CI:1.166~13.232;P=0.027)是腹腔镜辅助胃癌根治术后出现切口愈合不良的独立危险因素。结论:高龄、肥胖、低蛋白血症是腹腔镜辅助胃癌根治术后出现切口愈合不良的独立危险因素,具有上述危险因素的患者应注意预防术后切口愈合不良的出现。 展开更多

研究了三元乙丙橡胶(EPDM)绝热材料配方中的阻燃体系和交联剂对其性能的影响。结果发现,引入次磷酸铝复合物(AHP)、聚磷酸铵(APP)阻燃体系能降低EPDM绝热材料的线性烧蚀率,次磷酸铝复合物阻燃剂能够有效地提高EPDM的耐烧蚀性能和隔热性... 研究了三元乙丙橡胶(EPDM)绝热材料配方中的阻燃体系和交联剂对其性能的影响。结果发现,引入次磷酸铝复合物(AHP)、聚磷酸铵(APP)阻燃体系能降低EPDM绝热材料的线性烧蚀率,次磷酸铝复合物阻燃剂能够有效地提高EPDM的耐烧蚀性能和隔热性能。扫描电镜显示,AHP还可以促进EPDM形成隔热性能更好的炭层。随着烧蚀时间的延长,添加了次磷酸铝复合物的EPDM烧蚀率相比添加了APP阻燃剂的更低,在60 s时为0.046 mm/s,绝热层隔热性能最佳,仅为46.4℃。采用力学性能测试、热重分析、隔热性能测试、热裂解气相色谱/质谱联用(Py-GC/MS)测试分别对含AHP体系,交联剂分别为过氧化二异丙苯(DCP)和二-(叔丁基过氧化异丙基)苯(BIPB)的EPDM及其裂解产物进行了分析。发现添加BIPB的EPDM的力学性能虽然下降约30%,背面温度上升17℃,但其热稳定性几乎不受影响,有害的热裂解产物种类明显减少。 展开更多
