As an important ecological tree species in northern China, Populus simonii plays a crucial role in maintaining ecological balance and promoting environmental sustainability. The academic community has conducted a series of in-depth studies on this species, covering key areas such as genomics, survival mechanisms, and genetic breeding. Through the analysis of the genomic structure and function of P. simonii, we have not only revealed the molecular basis for its adaptation to harsh environments but also identified key genes that promote its growth and resistance to pests and diseases. Furthermore, exploring the survival mechanisms of P. simonii has deepened our understanding of its stress resistance traits, including how it effectively copes with abiotic stresses such as drought, salinization, and heavy metal pollution. In genetic breeding, significant progress has been made through the application of modern biotechnology, improving the growth rate and wood quality of P. simonii and enhancing its environmental adaptability and disease resistance. These research findings have not only enriched our knowledge of the biological characteristics of P. simonii but also provided a solid scientific foundation for its application in ecological restoration, forestry production, and environmental management. 展开更多

Bacterial-based antitumor immunity has become a promising strategy to activate the immune system for fighting cancer.However,the potential application of bacterial therapy is hindered by the presence of instability and susceptibility to infections within bacterial populations.Furthermore,monotherapy is ineffective in completely eliminating complex cancer with multiple contributing factors.In this study,based on our discovery that spore shell(SS)of Bacillus coagulans exhibits excellent tumor-targeting ability and adjuvant activity,we develop a biomimetic spore nanoplatform to boost bacteria-mediated antitumor therapy,chemodynamic therapy and antitumor immunity for synergistic cancer treatment.In detail,SS is separated from probiotic spores and then attached to the surface of liposome(Lipo)that was loaded with hemoglobin(Hb),glucose oxidase(GOx)and JQ1to construct SS@Lipo/Hb/GOx/JQ1.In tumor tissue,highly toxic hydroxyl radicals(·OH)are generated via sequential catalytic reactions:GOx catalyzing glucose into H_(2)O_(2)and Fe^(2+)in Hb decomposing H_(2)O_(2)into·OH.The combination of·OH and SS adjuvant can improve tumor immunogenicity and activate immune system.Meanwhile,JQ1-mediated down-regulation of PD-L1 and Hb-induced hypoxia alleviation synergistically reshape immunosuppressive tumor microenvironment and potentiate immune response.In this manner,SS@Lipo/Hb/GOx/JQ1 significantly suppresses tumor growth and metastasis.To summarize,the nanoplatform represents an optimum strategy to potentiate bacteria-based cancer immunotherapy. 展开更多

At present,the role of many long non-coding RNAs(lncRNAs)as tumor suppressors in the formation and development of cervical cancer(CC)has been studied.However,lncRNA prostate cancer gene expression marker 1(PCGEM1),whose high expression not only aggravates ovarian cancer but also can induce tumorigenesis and endometrial cancer progression,has not been studied in CC.The objective of this study was to investigate the expression and the underlying role of PCGEM1 in CC.The relative expression of PCGEM1 in CC cells was detected by real-time PCR.After the suppression of PCGEM1 expression by shRNA,the changes in the proliferation,migration,and invasion capacities were detected via CCK-8 assay,EdU assay,and colony formation assay wound healing assay.Transwell assay and the changes in expressions of epithelial-to-mesenchymal transition(EMT)markers were determined by western blot and immunofluorescence.The interplay among PCGEM1,miR-642a-5p,and kinesin family member 5B(KIF5B)was confirmed by bioinformatics analyses and luciferase reporter assay.Results showed that PCGEM1 expressions were up-regulated within CC cells.Cell viabilities,migration,and invasion were remarkably reduced after the suppression of PCGEM1 expression by shRNA in Hela and SiHa cells.N-cadherin was silenced,but E-cadherin expression was elevated by sh-PCGEM1.Moreover,by sponging miR-642a-5p in CC,PCGEM1 was verified as a competitive endogenous RNA(ceRNA)that modulates KIF5B levels.MiR-642a-5p down-regulation partially rescued sh-PCGEM1’s inhibitory effects on cell proliferation,migration,invasion,and EMT process.In conclusion,the PCGEM1/miR-642a-5p/KIF5B signaling axis might be a novel therapeutic target in CC.This study provides a research basis and new direction for targeted therapy of CC. 展开更多

Due to the great influences of both climate warming and human activities,permafrost on the Qinghai-Xizang Plateau(QXP) has been undergoing considerable degradation.Continuous degradation of plateau permafrost dramatically modifies the regional water cycle and hydrological processes,affecting the hydrogeological conditions,and ground hydrothermal status in cold regions.Permafrost thawing impacts the ecological environment,engineering facilities,and carbon storage functions,releasing some major greenhouse gases and exacerbating climate change.Despite the utilization of advanced research methodologies to investigate the changing hydrological processes and the corresponding influencing factors in permafrost regions,there still exist knowledge gaps in multivariate data,quantitative analysis of permafrost degradation's impact on various water bodies,and systematic hydrological modeling on the QXP.This review summarizes the main research methods in permafrost hydrology and elaborates on the impacts of permafrost degradation on regional precipitation distribution patterns,changes in surface runoff,expansion of thermokarst lakes/ponds,and groundwater dynamics on the QXP.Then,we discuss the current inadequacies and future research priorities,including multiple methods,observation data,and spatial and temporal scales,to provide a reference for a comprehensive analysis of the hydrological and environmental effects of permafrost degradation on the QXP under a warming climate. 展开更多

Objective:To investigate the regulatory role of cyclic adenosine monophosphate responsive element binding protein(CREB)/brain-derived neurotrophic factor(BDNF)signaling pathway in acute sleep deprivation(SD)-induced anxiety-like behavior mice(SD group)to study the mechanism of anxiety-like behavior better.Methods:The SD chamber was used to deprive the mice of sleep,and the anxiety-like behavior of the mice was verified using an open field test(OFT),elevated plus maze(EPM),forced swim test(FST),and tail suspension test(TST).Finally,proteins were detected by Western blotting.Result:OFT showed that the active distance and the time of stay in the central area were significantly reduced(P<0.05).EPM showed that the time and number of open arms in the SD group were significantly lower than in the control group(P<0.05).The FST showed that the forced swimming immobility time of the SD group was significantly lower than that of the control(P<0.05).Moreover,the TST showed that the immobility time of the tail suspension experiment in the SD group was significantly higher than that in the control group(P<0.05).Conclusion:Acute SD can regulate anxiety-like behavior in mice through the CREB/BDNF signaling pathway. 展开更多

我国的新冠疫情防控政策取得了举世瞩目的成效,为世界各国的防疫提供了宝贵经验。武汉在三个月内成功地应对了"医疗挤兑",实现了新冠患者清零。本文分析了武汉从"医疗挤兑"到"普惠医疗"的抗疫过程,强调... 我国的新冠疫情防控政策取得了举世瞩目的成效,为世界各国的防疫提供了宝贵经验。武汉在三个月内成功地应对了"医疗挤兑",实现了新冠患者清零。本文分析了武汉从"医疗挤兑"到"普惠医疗"的抗疫过程,强调了疫情防控策略转变对救治效果的影响,以及在这种转变过程中政府有效管控的重要性。基于武汉一家新冠收治定点医院的诊疗记录,从病死率和愈后恢复情况,分析了"应收尽收"的普惠医疗对新冠救治的积极影响。研究发现,相比早期的医疗挤兑阶段,武汉逐渐实现普惠医疗后新冠患者死亡概率明显降低;院内的医疗资源稀缺程度改善(联排氧装置启动、医患比例上升),也显著降低了新冠病死率;危重、高龄患者的病死率对医疗资源获得性更为敏感;此外,经历过医疗挤兑的新冠患者在痊愈后心理健康状况相对较差。本文强调了应对公共卫生危机,为避免挤兑的发生,需政府介入在短期内加强稀缺医疗资源供给的重要性。 展开更多

目的:探讨同时性多发性胃癌和(或)高级别上皮内瘤变的临床病理特点,分析病灶之间的关联性。方法:收集2007年11月至2017年12月间于石河子大学医学院第一附属医院接受内镜下黏膜剥脱术或外科手术的56例同时性多发性胃癌和(或)高级别上皮... 目的:探讨同时性多发性胃癌和(或)高级别上皮内瘤变的临床病理特点,分析病灶之间的关联性。方法:收集2007年11月至2017年12月间于石河子大学医学院第一附属医院接受内镜下黏膜剥脱术或外科手术的56例同时性多发性胃癌和(或)高级别上皮内瘤变患者的临床资料,分析其临床病理特点,根据多发性胃癌中对主、副病灶的定义,探究病灶间的关联性。结果:56例同时性多发性胃癌和(或)高级别上皮内瘤变患者平均年龄为(63.82±11.88)岁,男性多见,占75%(42/56),伴有病灶周围黏膜萎缩患者为25例(44.64%)。主、副病灶的浸润深度均以T1期为主。主、副病灶的直径呈正相关(r=0.797,P<0.001),病理类型具有关联性(P=0.007);内镜分型具有较高的一致性(P<0.001);在空间分布方面,主、副病灶也具有关联性,其中垂直位置列联相关系数c=0.484(P=0.002),水平位置列联相关系数c=0.535(P=0.007)。同时性多发性胃癌和(或)高级别上皮瘤变中主病灶在淋巴结转移、TNM分期、是否合并黏膜萎缩等方面与单发性胃癌或高级别上皮内瘤变存在差异。结论:同时性多发性胃癌和(或)高级别上皮内瘤变的高危人群为合并黏膜萎缩的老年男性,在胃镜检查时应注意根据主、副病灶间的关联性仔细观察,避免漏诊。 展开更多

Cooperative communication is regarded as a promising technique for improving the reliability of wireless communication links and enhancing the radio coverage simultaneously. Unlike the conventional half-duplex(HD) mode relaying techniques,the full-duplex based two-way relaying(FD-TWR) enables data exchanges between two nodes to be completed within a single time-slot,thus resulting in a significant improvement in the spectrum efficiency. In this paper,the channel model of the FD-TWR is first given out,followed by deriving the critical performance metrics,including the received signal-to-interference-plus-noise ratio(SINR),the upper bound of the ergodic capacity and the closedform solution of the proposed FD-TWR under amplify-and-forward(AF) mode. Furthermore,taking the limit of sum-transmit-power into account,we formulate the objective function of the optimal power allocation of FD-TWR as an extreme-value problem by deriving the optimal transmit power for both the source nodes and the relay node. As long as the self-interference(SI) signal in the FD-TWR nodes can be sufficiently suppressed,the proposed scheme is shown to outperform the conventional HD mode in terms of both the ergodic capacity and the outage probability. In addition,regardless of the practical SI power,the proposedFD-TWR is always capable of achieving its best performance with an aid of the proposed optimal power allocation scheme. 展开更多

Objective:As an important part of metabolomics analysis,untargeted metabolomics has become a powerful tool in the study of tumor mechanisms and the discovery of metabolic markers with high-throughput spectrometric data which also poses great challenges to data analysis,from the extraction of raw data to the identification of differential metabolites.To date,a large number of analytical tools and processes have been developed and constructed to serve untargeted metabolomics research.The different selection of analytical tools and parameter settings lead to varied results of untargeted metabolomics data.Our goal is to establish an easily operated platform and obtain a repeatable analysis result.Methods:We used the R language basic environment to construct the preprocessing system of the original data and the LAMP(Linux+Apache+MySQL+PHP)architecture to build a cloud mass spectrum data analysis system.Results:An open-source analysis software for untargeted metabolomics data(openNAU)was constructed.It includes the extraction of raw mass data and quality control for the identification of differential metabolic ion peaks.A reference metabolomics database based on public databases was also constructed.Conclusions:A complete analysis system platform for untargeted metabolomics was established.This platform provides a complete template interface for the addition and updating of the analysis process,so we can finish complex analyses of untargeted metabolomics with simple human-computer interactions.The source code can be downloaded from https://github.com/zjuRong/openNAU. 展开更多

Mechanochemical synthesis has been applied for many novel material preparations and gained more and more attention due to green and high-efficiency recently. In order to explore the influences of iron precursors on structure and performance of iron molybdate catalyst prepared by mechanochemical route, three typical and cheap iron precursors have been used in preparation of iron molybdate catalyst. Many characterization methods have been employed to obtain the physical and chemical properties of iron molybdate catalyst. Results indicate that iron precursors have the significant impact on the phase composition, crystal morphology and catalytic performance in the conversion of methanol to formaldehyde. It is hard to regulate the phase composition by changing Mo/Fe mole ratios for Fe2(SO4)3 as iron precursor. In addition, as for Fe2(SO4)3, the formaldehyde yield is lower than that from iron molybdate catalyst prepared with Fe(NO3)3·9H2O due to the reduction in Fe2(MoO4)3 phase as active phase. Based on mechanochemical and coprecipitation method, the solvent water could be a key factor for the formation of MoO3 and Fe2(MoO4) for FeCl3·6H2O and Fe2(SO4)3 as precursors. Iron molybdate catalyst prepared with Fe(NO3)3·9H2O by mechanochemical route, shows the best methanol conversion and formaldehyde yield in this reaction. 展开更多

Tendon heterotopic ossification(HO)is characterized by bone formation inside tendon tissue,which severely debilitates people in their daily life.Current therapies fail to promote functional tissue repair largely due to our limited understanding of HO pathogenesis.Here,we investigate the pathological mechanism and propose a potential treatment method for HO.Immunofluorescence assays showed that the Mohawk(MKX)expression level was decreased in human tendon HO tissue,coinciding with spontaneous HO and the upregulated expression of osteochondrogenic and angiogenic genes in the tendons of Mkx^(−/−)mice.Single-cell RNA sequencing analyses of wild-type and Mkx^(−/−)tendons identified three cell types and revealed the excessive activation of osteochondrogenic genes during the tenogenesis of Mkx^(−/−)tendon cells.Single-cell analysis revealed that the gene expression program of angiogenesis,which is strongly associated with bone formation,was activated in all cell types during HO.Moreover,inhibition of angiogenesis by the small-molecule inhibitor BIBF1120 attenuated bone formation and angiogenesis in the Achilles tendons of both Mkx mutant mice and a rat traumatic model of HO.These findings provide new insights into the cellular mechanisms of tendon HO and highlight the inhibition of angiogenesis with BIBF1120 as a potential treatment strategy for HO. 展开更多

However, most aEEG devices require that these margins be measured semi-subjectively. This paper proposes a step-by-step signal-processing method to calculate a compact aEEG tracing and the upper/lower margin using raw EEG data. The high accuracy of the algorithm was verified by comparison with a recognized commercial aEEG device based on a representative testing dataset composed of 72 aEEG data. The introduced digital algorithm achieved compact aEEG tracing with a small data size. Moreover, the algorithm precisely represented the upper and lower margins in the tracing for objective data interpretation. The described method should facilitate aEEG signal processing and further establish the clinical and experimental application of aEEG methods. 展开更多

Objective This study was designed to determine the methylation profile of four CpGs and the genotypes of two CpG-SNPs located in promoter region of DIO2 in patients with Kashin-Beck disease(KBD).We also analyzed the interaction between the CpGs methylations and CpG-SNPs.Methods Whole blood specimens were collected from 16 KBD patients and 16 healthy subjects.Four CpGs and two CpG-SNPs in the promoter regions of DIO2 were detected using matrix-assisted laser desorption ionization time of flight mass spectrometry(MALDI-TOF-MS).The CpGs methylation levels were compared between samples from KBD patients and healthy subjects.The methylation levels were also analyzed in KBD patients with different CpG-SNP genotypes.Results The mRNA expression of DIO2 in whole blood of KBD patients was significnatly lower than in healthy controls(P<0.05).The methylation levels of DIO2-1_CpG_3 in KBD patients were significantly higher than those in healthy controls(P<0.05).The methylation levels of four CpGs were not significantly different between KBD patients and healthy controls.The methylation level of DIO2-1_CpG_3 in the promoter region of DIO2 in KBD patients with GA/AA genotype was significantly higher than that of KBD patients with GG genotype(P<0.05).Conclusion The methylation level of DIO2 increases in KBD patients.Similar trends exist in KBD carriers of variant genotypes of CpG-SNPs DIO2 rs955849187. 展开更多

However, for large DNA sequences, these algorithms require a long time computationally. Objective: Here we will propose a new numerical method to characterize and compare DNA sequences quickly. Method: Based on a new 2-dimensional (2D) graphical representation of DNA sequences, we can obtain an 8-dimensional vector using two basic concepts of probability, the mean and the variance. Results: We perform similarity/dissimilarity analyses among two real DNA data sets, the coding sequences of the first exon of beta-globin gene of 11 species and 31 mammalian mitochondrial genomes, respectively. Conclusion: Our results are in agreement with the existing analyses in our literatures. We also compare our approach with other methods and find that ours is more effective. 展开更多

Greenway is a green linear corridor connecting scenic spots, residential areas, nature reserves, road traffic, etc. Rational greenway route selection can not only attract tourists to increase the income of local villages, but also arouse people’s awareness of the protection of traditional villages. This paper took Xingtai County, Hebei Province, China as an example to practice greenway route selection in the county territory. Eight factors were selected for greenway route planning, namely elevation, water body, mountain, ecological protection redline, cultural relics protection units, population density, road traffic network, and important transportation hub. These eight factors were assigned in the GIS platform, and the suitability evaluation system of each factor was constructed. The analytic hierarchy process (AHP) and entropy weight method were combined to calculate the weight of these eight factors. The raster calculator was used to weight and overlay the suitability evaluation system of each factor, and the comprehensive suitability evaluation map of greenway route selection was obtained. Based on an important transport hub, along with the water body and the road traffic, the final selection greenway route of traditional villages in Xingtai county was formed. The results of the research can provide a certain reference for the greenway route selection at the county scale. 展开更多

Association between the M98K variant of the OPTN gene and the risk for primary open angle glaucoma:an updated meta-analysis

Background:The association between optineurin(OPTN)M98K variant and primary open angle glaucoma(POAG)has been widely investigated.However,the results remain controversial among published meta-analyses.Therefore,we conducted an updated meta-analysis to further explore the association between M98K and POAG and its subgroups.Methods:PubMed,Embase,Web of Science,and China National Knowledge Infrastructure(CNKI)databases were searched to find all articles describing the relationships between the M98K variant and POAG,which were published from the inception to 31 December,2019.The associations between M98K and overall POAG,normal tension glaucoma(NTG),high tension glaucoma(HTG),POAG in Asian and non-Asian populations,juvenile open angle glaucoma(JOAG)and adult-onset POAG were evaluated by calculating the pooled odds ratio(OR)and 95%confidence interval(CI).The Bonferroni correction was used to determine the statistically significant genetic models.Results:A total of 34 eligible articles involving 7,310 POAG patients and 5,173 controls were identified in the present meta-analysis.The articles achieved an average of 6.21 stars for quality assessment by the Newcastle-Ottawa Scale(NOS).Evidence from the pooled results indicated significant association between M98K and overall POAG susceptibility under the dominant model(OR=1.30,95%CI,1.12-1.52;P<0.001).In the subgroup analyses,no significant associations were found between M98K and the risks of NTG,HTG,JOAG,adult-onset POAG,Asian POAG or non-Asian POAG.Conclusions:The updated meta-analysis revealed that OPTN M98K was significantly associated with the susceptibility to overall POAG. 展开更多

Objective:To explore the characteristics of audiology examination of Noise-Induced Hidden Hearing Loss(NIHHL),and then explore its valuable diagnostic methods.Methods:A total of 80 young men aged between 19 and 35 were selected.They were classified into the experimental group with noise exposure history and the control group without noise exposure history,with 40 men in each group.We carried out extended high-frequency audiometry test for good ears;conducted speech in noise measurement and distortion product emission for good ears;DPOAE test,compare and analyze the results of each test.Results:The extended high frequency hearing threshold test results showed that the hearing threshold of the experimental group was larger than that of the control group at the frequency above 8 kHz,with significant statistical difference(p<.01).The results of speech audiometry under noise showed that the signal-to-noise ratio loss in the experimental group was greater than that in the control group,with a significant statistical difference(p<.01).The results of otoacoustic emission showed that the signal-to-noise ratio of the experimental group was lower than that of the control group at the frequency of 6 kHz and above,with a statistically significant difference(p<.01).There is a correlation between pure tone audiometry and DPOAE test results at high frequencies(6 kHz:p<.0001,r=-.478;8 kHz:p<.0001,r=-.491);There was a correlation between speech audiometry and DPOAE test results under noise(p=.031,r=-.299).Conclusions:Compared with the control group,the extended high frequency pure tone hearing threshold increased or could not be elicited in the experimental group,speech recognition ability decreased significantly under noise,DPOAE was normally elicited but signal-to-noise ratio decreased,and there was a certain correlation between them.The above three audiological examination methods have a certain reference value for early recognition of hidden hearing loss. 展开更多

Bone metastasis,a life-threatening complication of advanced breast cancer,is often accompanied by debilitating pain(cancer-induced bone pain,CIBP)that severely impairs life quality and survival.The concurrent treatment of bone metastases and CIBP remains a clinical challenge because the therapeutic options are limited.In this study,we construct a near-infrared light-activated nano-therapeutic system to meet this conundrum.In detail,sorafenib(SRF)and photosensitizer(chlorin e6,Ce6)are encapsulated into mesoporous hydroxyapatite nanoparticles(HANPs),which are further functionalized with hyaluronic acid(HA)to obtain HA-SRF/Ce6@HANPs system.The designed nanoplatform destroys tumor cells in vitro and in vivo via the synergism of SRF(interrupting the exchange of cystine/glutamate by inhibiting SLC7A11)and photodynamic therapy(PDT,inducing reactive oxygen species generation).The decrease in tumor burden and reduction of extracellular glutamate significantly attenuate CIBP in mice model with developing bone cancer.Moreover,the combination of HA-SRF/Ce6@HANPs and PDT inhibit osteoclasts activation,promote osteoblast differentiation and accelerate bone repair.Overall,the nanoagent with good biocompatibility may provide an effective therapy method for the concurrent treatment of breast cancer bone metastasis and CIBP. 展开更多
