Tumor cell apoptosis was examined by TUNEL method.RESULTS: Immunohistochemical analysis showed that survivin expression was positive in 61 of 80 patients (76%) with gastric carcinoma. In contrast, no expression of survivin in adjacent normal tissues was detected. Expression level of caspase-3 was higher in normal tissues than in carcinoma.Patients with higher expression of survivin had worse histological grades and pathological stages. Expression of caspase-3 was significantly associated with histological stages, but not with the pathological stages. Although survivin expression in carcinoma was not inversely related to caspase-3, patients with survivin (-) and caspase-3(+) had the maximum apoptosis index.CONCLUSION: Expression level of survivin was associated with histological grades and pathological stages of the tumor,indicating that survivin may be a poor prognosis factor for gastric carcinoma. Unlike caspase-3, survivin (an apoptosis inhibitor) can markedly inhibit the apoptosis of tumor cells.展开更多

Rabbits were ...Objective To investigate the roles of sympathetic and vagus nerves in hypotension and bradycardia induced by fentanyl. Methods Fourteen rabbits were divided into 2 groups: normal and vagotomized rabbits. Rabbits were anesthetized, paralyzed, and artificial ventilated. Right renal sympathetic nerve was exposed and prepared for recording electrical activity. Fentanyl was injected intravenously in incremental doses of 1, 4, 15, 30, and 50 μg/kg at 10 minutes intervals. Results Fentanyl significantly reduced the spontaneous activity of renal sympathetic nerve, mean arterial pressure, and heart rate above a total dose of 20 μg/kg in both normal and vagotomized rabbits. However, normal rabbits spontaneous sympathetic nerve activity and mean arterial pressure were more depressed than vagotomized rabbits at total doses of 50 and 100 μg/kg. There were no significant difference in the reduction of heart rate between normal and vagotomized rabbits. Conclusion Fentanyl induction of bradycardia and hypotension in rabbits is mainly due to depression of sympathetic nerve activity.展开更多
Renewable energy exploitation is among the development strategies set by the government of Rwanda on the roadmap to 2023/2024 universal electricity access and theUnitedNations plans by 2030.Numerous previous studies o...Renewable energy exploitation is among the development strategies set by the government of Rwanda on the roadmap to 2023/2024 universal electricity access and theUnitedNations plans by 2030.Numerous previous studies oncleanenergy technologies inRwandahavemostly focusedonhouseholds’usagebut there are currentlynostudies describing the feasibility of clean energy technologies for financial institutions.The skepticism on renewable energy in Africa was previously reported by some personnel.Having realized that most SACCOs(Savings and Credit Co-Operatives)in Rwanda use diesel technology for backup/emergency electricity supply,taking consideration of abundant solar resources in Rwanda,having seen the viability and feasibility studies from other countries of renewable energy for different institutions(financial included);thiswork uses theHOMEREnergy Software and the electricity load profile of a typical SACCO in Rwanda to analyse the affordability and viability of on-site renewable energy generation for SACCO in Rwanda.The results reveal that a solar PV systemwith storage can be the optimal solution(with levelized cost of electricity(LCOE)of 0.713$/kWh which is cheaper than 0.73$/kWh for diesel technology)for SACCOs located in both off-grid areas and grid-connected areas(with 0.041$/kWh LCOE which is lower than the current electricity tariff in Rwanda).The findings in this work can serve as basic tools/materials for policy drafters in Rwanda on how financial institutions can contribute to climate change mitigation through self-renewable energy exploitation.展开更多

Methods: The viability of Cloundman melanoma S91 cells treated with As 2O 3 was measured by MTT [3-(4, 5-dimethyl-2-thiazolyl)-2, 5-diphenyl-2H-tetrazolium bromide] assay. The apoptosis was determined by transferase-mediated dUTP nick end labeling (TUNEL) detection, and the morphology of apoptotic cells was observed through transmission electron microscopy (TEM). The cells growth phase were analyzed by flow cytometry (FCM). Results: A time-and dose-dependent decrease in cell viability was induced in S91 cells after treatment with As 2O 3 at the concentration of 1-5 μmol/L respectively. The TUNEL indices were 0.033±0.018, 0.062±0.012, 0.102±0.016, 0.132±0.031, and 0.162±0.027 respectively, which were much higher compared with the control group (0.017±0.004, P<0.01). The flow cytometry showed that hypodiploid peak after treatment with 3 μmol/L and 5 μmol/L of As 2O 3 for 48 h were 9.99% and 17.59% respectively, which increased significantly compared with the control cells (3.05% P<0.01,). The apoptotic morphology observed by transmission electron microscope showed the chromatin became condensed and attached to the inner surface of nuclear membrane. Conclusion: Arsenic trioxide can induce melanoma Cloundman S91 cell apoptosis at the concentration of 1-5 μmol/L, which will provide enhanced benefit in melanoma therapy.展开更多
Pyrrolizidine alkaloids(PAs)and their N-oxides(PANOs)are phytotoxins produced by various plant species and have been emerged as environmental pollutants.The sorption/desorption behaviors of PAs/PANOs in soil are cruci...Pyrrolizidine alkaloids(PAs)and their N-oxides(PANOs)are phytotoxins produced by various plant species and have been emerged as environmental pollutants.The sorption/desorption behaviors of PAs/PANOs in soil are crucial due to the horizontal transfer of these natural products from PA-producing plants to soil and subsequently absorbed by plant roots.This study firstly investigated the sorption/desorption behaviors of PAs/PANOs in tea plantation soils with distinct characteristics.Sorption amounts for seneciphylline(Sp)and seneciphylline-N-oxide(SpNO)in three acidic soils ranged from 2.9 to 5.9μg/g and 1.7 to 2.8μg/g,respectively.Desorption percentages for Sp and SpNO were from 22.2%to 30.5%and 36.1%to 43.9%.In the mixed PAs/PANOs systems,stronger sorption of PAs over PANOs was occurred in tested soils.Additionally,the Freundlich models more precisely described the sorption/desorption isotherms.Cation exchange capacity,sand content and total nitrogen were identified as major influencing factors by linear regression models.Overall,the soils exhibiting higher sorption capacities for compounds with greater hydrophobicity.PANOs were more likely to migrate within soils and be absorbed by tea plants.It contributes to the understanding of environmental fate of PAs/PANOs in tea plantations and provides basic data and clues for the development of PAs/PANOs reduction technology.展开更多
