摘要
目的:研究红景天甙(salidroside,sal)对免肺动脉平滑肌细胞(PASMC)增殖、DNA 合成和 c-fos、c-myc 原癌基因表达的影响,并初步探讨其作用机理。方法:应用离体培养的兔肺动脉平滑肌细胞,采用四唑盐比色试验(MTT)、~3H-胸腺嘧啶核苷(~3H-TdR)掺入和斑点杂交的方法,观察平滑肌细胞的增殖、DNA 合成和 c-fos、c-myc 原癌基因表达。结果:sal(2.0×10^(-5)~64×10^(-5)mol/L)可明显抑制离体培养的兔肺动脉平滑肌细胞增殖、~3H-TdR 掺入和 c-fos、c-myc 原癌基因表达.sal(64×10^(-5)mol/L)可明显抑制去甲肾上腺素(10μmol/L)对 PASMC的促增殖作用(P<0.05),但 sal 与心得安(propranolol)合用并不增加对 PASMC 的抑制效应(P>0.05)。结论:红景天甙可抑制离体培养的兔肺动脉平滑肌细胞的增殖、DNA 合成和 c-fos、c-myc 原癌基因表达。
Objective:To investigate the effects of salidroside on the proliferation,the DNA synthesis and c-los,c-myc oncogene expression of rabbit pulmonary arterial smooth muscle cell(PASMC).Methods:Techniques of cell culture,methyl thia- zolyl tetrazolium test,~3H-TdR incorporation and dot blot test were used.Results:2.0×10^(-5)~64×10^(-5)mol/L salidro- side inhibited significantly the proliferation and ~3H-TdR incorporation in cultured rabbit PASMC(P<0.05).Salidroside also inhibited the c-los,c-myc oncogene expression of PASMC.64×10^(-5)mol/L salidroside inhibited the proliferation and ~3H-TdR incorporation induced by norepinephrine(10umol/L),but when salidroside was combined with propranolol, these effects had not been enhanced(P>0.05).Conclusion:Salidroside inhibited the proliferation,DNA synthesis and c-fos,c-myc oncogene expression of cultured rabbit PASMC,but its mechanism need to be further investigated.
